ABSTRACT Infection of soybean (Glycine max) plants inoculated with particles of Alfalfa mosaic virus (AlMV) isolate 425 at 12 days after germination was monitored throughout the life cycle of the plant (vegetative growth, flowering, seed formation, and seed maturation) by western blot analysis of tissue samples. At 8 to 10 days after inoculation, the upper uninoculated leaves showed symptoms of virus infection and accumulation of viral coat protein (CP). Virus CP was detectable in leaves, stem, roots, seedpods, and seed coat up to 45 days postinoculation (dpi), but only in the seedpod and seed coat at 65 dpi. No virus accumulation was detected in embryos and cotyledons at any time during infection, and no seed transmission of virus was observed. Soybean plants inoculated with recombinant AlMV passaged from upper uninoculated leaves of infected plants showed accumulation of full-length chimeric AlMV CP containing rabies antigen in systemically infected leaves and seed coat. These results suggest the potential usefulness of plants and plant viruses as vehicles for producing proteins of biomedical importance in a safe and inexpensive manner. Moreover, even the soybean seed coat, treated as waste tissue during conventional processing for oil and other products, may be utilized for the expression of value-added proteins.  相似文献

Strawberry (Fragaria × ananassa Duch.) is rich source of dietary antioxidants, minerals and nutrients. Dietary antioxidants have been known as beneficial for enhancing the fitness, preventing certain diseases and even mitigating the effects of ageing. The objectives of the present study were to determine variability and inheritance of antioxidants, to identify antioxidant rich and productive genotypes, and to suggest suitable breeding approaches. The genotypes, namely Ofra, Chandler, Festival and Camarosa showed higher concentrations of dietary antioxidants and therefore could be useful in future breeding. Results indicate that the effect of the genotypes on antioxidant contents is stronger than that of the environment. The high heritability (>80%) and low genetic advance as percentage of mean (<40%) for ascorbic acid and β-carotene contents could be improved by heterosis breeding. However, selection and hybridization would be effective tools to enhance the phenols and anthocyanin content, and yield potential as these traits showed high heritability (>80%) and high genetic advance as percentage of mean (>40%). Positive direct effect on fruit yield was highest for phenol content (0.609) which is also fairly close to its correlation coefficient (0.765) indicating that a direct selection based on phenol content would be most effective and that the phenol content could be used as a reliable biochemical marker to identify the productive genotypes having higher amounts of dietary antioxidants. The information could also be used for developing antioxidant rich cultivars, i.e. ‘Breeding Strawberry for High Antioxidants’.  相似文献

The effect of concentration and addition method of glycerol on the quality of cryopreserved mithun (Bos frontalis) spermatozoa was investigated. Semen samples were collected from five healthy mithun bulls through rectal massage method and cryopreserved in liquid nitrogen. The samples were diluted in Tris–egg yolk–glycerol extender, equilibrated for 4 h at 4 °C and loaded into 0.50‐ml straws. The straws were then frozen in liquid nitrogen vapour for 10 min and finally plunged into liquid nitrogen for storage. The required amount of glycerol was added into the diluted samples either in a single dose (3%, 4%, 5%, 6% or 7%; added at 37 °C immediately before equilibration) or in split doses (5%, 6% or 7%; the total amount was divided into four equal parts, and a part was added at 37 °C immediately before equilibration, and the remaining parts were added subsequently at 1, 2 and 3 h of equilibration at 4 °C). In the single‐dose addition method, following freeze‐thawing, greater (p < 0.05) motility (%) and proportion of live spermatozoa with intact acrosome (LSIA, %) in 5% glycerol (40.6 ± 1.7 and 43.4 ± 1.8 respectively) and lesser (p < 0.05) total morphological abnormalities (%) in 5% (14.1 ± 0.8) and 6% (13.7 ± 1.0) glycerol were observed compared to the other glycerol concentrations. In the split‐dose addition method, following freeze‐thawing, greater (p < 0.05) motility (%) and LSIA proportion (%) were found in 5% (50.2 ± 1.9 and 53.3 ± 1.8 respectively) compared to 6% or 7% glycerol, but the total morphological abnormalities were not different among the glycerol concentrations. In addition, in all the glycerol concentrations, better (p < 0.05) post‐freeze‐thaw motility and LSIA proportions were observed when glycerol was added in split doses compared to a single dose. In conclusion, Tris–egg yolk extender with 5% glycerol added in split doses was found most suitable for cryopreserving mithun sperm.  相似文献

A study on the rice–wheat cropping system was conducted at Bidhan Chandra Krishi Viswavidyalaya, West Bengal, India, to assess the effects of long-term manuring and fertilization on transformation of the inorganic phosphorus (P) fraction in soil after 22 years of the crop cycle. Soil samples were collected after Kharif from seven treated plots having different types of organic amendments like farm yard manure, paddy straw and green manuring with 50% substitution of nitrogen levels in rice crop only. The result showed that the yield trend of rice was maintained due to the buildup of P from various organic inputs. Although cultivation for 22 years without adding any fertilizer caused a significant decrease in almost all the forms of P viz. avail-P, saloid P, iron phosphorus fraction (Fe–P), aluminum phosphorus fraction (Al–P), calcium phosphorus fraction (Ca–P) and total P in control. Partial substitution of inorganic fertilizer N (50%) with organics, however, caused a significant increase in almost all the P fractions in soil over the control. The relative abundance of all the fractions of inorganic P irrespective of treatments was as follows: Fe–P > reductant soluble P fraction > occluded P > Al–P > Ca–P > saloid P. Saloid and Fe–P were the dominating fractions responsible for 92% variation of available P and total P levels, respectively.  相似文献

To test the hypotheses of modern human origin in East Asia, we sampled 12,127 male individuals from 163 populations and typed for three Y chromosome biallelic markers (YAP, M89, and M130). All the individuals carried a mutation at one of the three sites. These three mutations (YAP+, M89T, and M130T) coalesce to another mutation (M168T), which originated in Africa about 35,000 to 89,000 years ago. Therefore, the data do not support even a minimal in situ hominid contribution in the origin of anatomically modern humans in East Asia.  相似文献

Southeast Asia hosts a great diversity of different rattan genotypes. There are 5 genera and 60 different species of rattan in India and the Northeastern region is a natural sanctuary for 4 different genera and 16 different species. The natural reserves of this species have come under the threat of genetic erosion due to overexploitation. This investigation was directed at characterizing 15 rattan genotypes of the genera Calamus, Plectocomia and Daemonorops which yield rattans of commercial importance, based on RAPD fingerprints. From 20 different random decamer primers, 12 primers gave reproducible amplification profiles and 104 polymorphic bands. A considerable degree of polymorphism (98.1%) was detected among the genotypes. Jaccard’s coefficient of similarity ranged from 0.146 to 0.632 with a mean of 0.320±0.082, signifying extensive genetic divergence among the genotypes studied. UPGMA cluster analysis clearly distinguished P. assamica Griff. and C. erectus Roxb. The other 13 genotypes were grouped into two distinct clusters, one cluster involving two C. tenuis genotypes along with C. inermis T.Anders. and C. acanthospathus Griff. and the other cluster involving the rest of the Calamus genotypes along with D. jenkinsianus (Griff.) Mart. Unique fingerprints for 7 Calamus and 1 Daemonorops genotypes were detected. The results presented in this paper demonstrated the utility of RAPD markers in elucidating patterns of genetic variation among genotypes of the three main rattan genera of Northeast India and in identifying individual genotypes, which may serve as potential sources of unique genetic material for genetic improvement and conservation.  相似文献

A field experiment was conducted during two consecutive years of 2010–2011 and 2011–2012 to study the effect of biofertilizers in conjunction with organic and inorganic sources of nutrient management on productivity, quality and soil health on field pea at ICAR RC for NEH Region, Nagaland Centre Jharnapani, Nagaland, India. The experiment was laid out in split plot design with five nutrient sources in main plots and four treatment of biofertilizers with zinc in sub plots. Results indicated that the application of 100% recommended dose of fertilizer (RDF) through inorganic + 50% recommended dose of nitrogen (RDN) through vermicompost significantly improved root nitrogen (N) content, cation exchange capacity (CEC) of roots, NA activates, seed yield (1153 and 1262 kg ha?1), straw yield (2182 and 2332 kg ha?1) in the year of 2010–2011 and 2011–2012, respectively. Nutrients (N, P, K, S and Zn) uptake by seed and straw, protein content, protein harvest, soil organic carbon (SOC), available N, P, K, S, Zn and economics significantly higher with 100% RDF through inorganic + 50% RDN through vermicompost during both the years. Seed inoculation with biofertilizers along with 5 kg Zn ha?1 markedly enhanced the root N content, CEC of roots, nitrogenase activities (NA), seed yield (1080 and 1193 kg ha?1), straw yield (1978 and 2128 kg ha?1), nutrients [N, phosphorus (P), potassium (K), sulfur (S) and zinc (Zn)] uptake, soil organic carbon (SOC) (%), and available N, P, K, S, and Zn of pea in both the years, respectively.
