Investigations of 1167 dairy cows out of 105 herds with fertility disorders on secretion of Coxiella burnetii (C. burnetii) by the genitals as well as serological studies of these animals using ELISA demonstrated that C. burnetii infections are significantly associated with abortions but not with repeated inseminations without success or vaginal excretions. The demonstration of an increased risk of infections for persons caring for those dairy herds could be shown by a total of 253 serological studies. A comparison of these studies of farmers caring for dairy herds suffering from abortions a seroprevalence of > or = 20% showed that these persons revealed significantly more frequent antibodies against C. burnetii than farmers of the group compared with. Further studies showed that in herds suffering from abortions a seroprevalence of > or = 20% means an additional risk of infections of farmers. Vice versa abortions of the cows in herds with a seroprevalence of > or = 20% imply an additional infectious risk. 相似文献

The development and use of modern techniques, such as intracytoplasmic sperm injection (ICSI), gene knockout and sperm fluorescence in situ hybridization with chromosome- specific probes, have significantly increased our knowledge about sperm defects. We describe a new oligoasthenoteratozoospermic defect in a bull. Because of its morphological characteristics the defect was named the multinuclear-multiflagellar sperm defect. All spermatozoa in the ejaculate were abnormal. Many of the spermatozoa had multiple nuclei and multiple sperm tails. All spermatozoa lacked an acrosome, and only seldom did spermatozoa have a mitochondrial helix in the midpiece area. Meiosis and spermiogenesis were severely affected in this otherwise phenotypically normal bull. The sperm defects resembled the phenotype of a targeted gene knockout Hrb(-/-) (HIV-1 Rev-binding/interacting protein) mutant mouse strain, which is expressed as sterility in males, while females remain fertile. Since the father of this bull has been extensively used in at least three countries the defective gene has possibly become widespread in the red and white breeds (Ayrshire, Swedish Red and White, Norwegian Red) in the Nordic countries. The present study aims to ascertain the influence of gamma-amino butyric acid (GABA)(A or B) receptors on arginine vasopressin (AVP) release in vitro and determine whether E(2) modulates GABA-AVP interaction. Within 10 min of ewe killing, saggital midline hypothalamic slices (from the anterior preoptic area to the mediobasal hypothalamus along with the median eminence, 2-mm thick, two per ewe) were dissected, placed in oxygenated minimum essential media (MEM)-alpha at 4 degrees C and within 2 h were singly perifused at 37 degrees C with oxygenated MEM-alpha (pH 7.4; flow rate 0.15 ml/min), either with or without E(2) (24 pg/ml). After 4-h equilibration, 10-min fractions were collected for 4 h interposed with a 10-min exposure at 60 min to a specific GABA(A or B) receptor agonist or antagonist at various doses (0.1-10 mm). GABA(A) (muscimol; no E(2), n = 7 perifusion chambers, with E(2), n = 11) or GABA(B) (baclofen; no E(2), n = 8, with E(2), n = 15) agonists (10 mm) did not influence AVP concentrations. However, AVP release increased (p < 0.05) 20-30 min after exposure to 10 mm GABA(A or B) antagonists (bicuculline, no E(2), n = 7: from 4.6 +/- 0.7 to 33.0 +/- 0.4, with E(2), n = 17: from 11.9 +/- 1.4 to 32.8 +/- 6.0; CGP52432, with E(2), n = 14: from 14.0 +/- 2.6 to 28.8 +/- 3.9 pg/ml). At the end of the collection period, hypothalamic slices responded to KCl (100 mm) with AVP efflux (p < 0.05). GABA(B) but not GABA(A) antagonist-stimulated AVP release was enhanced in the presence of E(2). Although low water use landscaping is becoming common in arid regions, little is known about drought tolerance and drought responses of many ornamental plants, especially herbaceous perennials. Drought responses were assessed for six herbaceous ornamental landscape perennials in a 38 l pot-in-pot system in northern Utah over a 2-year period. The first year was an establishment period. During the second year, drought responses were evaluated for established Echinacea purpurea (L.) Moench, Gaillardia aristata Pursh, Lavandula angustifolia P. Mill., Leucanthemum × superbum (J.W. Ingram) Berg. ex Kent, ‘Alaska’, Penstemon barbatus Roth var. praecox nanus rondo, and Penstemon × mexicali Mitch. ‘Red Rocks’. Plants were irrigated at frequencies of 1 (control), 2, or 4 weeks between June and September, simulating well-watered conditions, moderate drought, or severe drought. Osmotic potential (Ψs), gas exchange, visual quality, leaf area, and dry weight were assessed. In a confined root zone, P. barbatus showed the greatest tolerance to all levels of drought, avoiding desiccation by increasing root:shoot ratio and decreasing stomatal conductance as water became limiting. L. angustifolia and P. × mexicali showed tolerance to moderate drought conditions, but died after exposure to the first episode of severe drought. Neither G. aristata nor L. superbum were able to regulate shoot water loss effectively. Instead, both species displayed drought avoidance mechanisms, dying back when water was limiting and showing new growth after they were watered. Compared to control plants, G. aristata shoot dry weight was reduced by 50% and 84%, and L. superbum shoot dry weight was reduced by 47% and 99% for the 2- and 4-week irrigation intervals, respectively. Root dry weights were affected similarly for both species. E. purpurea exhibited poor visual quality at all irrigation intervals, in particular wilting severely in both drought treatments, but regaining turgor when watered again. P. barbatus is recommended for ornamental landscapes that receive little or no supplemental irrigation, while E. purpurea is not recommended for low water landscapes because of low visual quality under even mild drought. 相似文献

The effects of cytokinins, carbohydrate sources and cold pretreatment on the conversion of protocorm-like bodies (PLBs) to shoots were investigated for the enhancement of micropropagation of Dendrobium huoshanense C.Z. Tang et S.J. Cheng, an endangered medicinal plant in China. The effects of cytokinins and carbohydrate sources on the conversion of PLBs to shoots depended on their types and concentrations. The best results in terms of shoot development from PLBs occurred on 1/2 MS medium supplemented with 20 μM kinetin and 10 g l−1 maltose. Cold pretreatment at 10 °C for 1–2 weeks significantly enhanced the conversion of PLBs to shoots, and over 1300 shoots were obtained from one gram of PLBs after 3 months of culture. The developed shoots were rooted on growth regulator-free MS medium supplemented with 8 g/l banana paste to give complete plantlets, which were successfully acclimatized with a survival rate of approximately 65%. The results indicate that a suitable cold pretreatment (10 °C for 1 week) followed by the use of 20 μM kinetin and 10 g/l maltose in 1/2 MS medium would produce a large number of shoots from PLBs for plantlet regeneration of D. huoshanense. 相似文献

Aquaporins (AQPs) are essential membrane protein channels for the transport of water across membranes. Fluid movement in the epididymis is important for modulation of the luminal environment, in which sperm mature and reside. This study was designed to understand the morphology and localization of AQPs in ram efferent ducts (ED) and epididymis. For this purpose, the epididymis of seven animals were removed for histologic and immunohistochemical analyses. AQP1 immunoreactivity was observed in the apex of the ED, and AQP9 was found adjacent to the nuclei of the epithelial cells of the ED. The epithelial lining of ram epididymis is pseudostratified columnar and presents principal, basal, apical and narrow cells. In the initial segment (IS), a moderate reaction for AQP1 was observed in the apical cytoplasm of epithelial cells. An intense reactivity for AQP1 was noted over the microvilli of principal cells and in spermatozoa in the caput. In the corpus and cauda, AQP1 was noted only over the endothelial cells of vascular channels located in intertubular spaces. A weak‐to‐moderate reaction for AQP9 was observed in the nuclei of epithelial cells in the IS, caput and corpus of the epididymis. In the cauda, an intense reaction to AQP9 was observed in the epithelial border. In the IS, caput and corpus, the reactivity for AQP9 differed from those observed in domestic animals. The cauda showed a pattern similar to that previously described. These results indicate that AQPs 1 and 9 have reversed locations and roles in rams, suggesting activity variations related with fluid and solute absorption throughout the epididymis. 相似文献
