Initial results demonstrating the feasibility of a multiplexed liquid array immunoassay for foot-and-mouth disease viral antigen detection and simultaneous serotype differentiation are presented. Serotype-specific antibodies from rabbit and guinea pig hyperimmunesera were isolated and prepared for use in a multiplexed, bead-based assay. The performance of all of the available antibodies as both capture and detector reagents was evaluated in the multiplexed system to establish a combination exhibiting the highest homotypic responses and lowest heterotypic reactions. The multiplexed assay was evaluated against inactivated cell culture supernatant samples of the same subtype as the virus used to raise the capture and detector antibodies. Distinct serotype differentiation was observed, except in the case of serotype SAT1. Subsequently, cell culture supernatant samples from a larger pool of viral subtypes were analyzed. Distinct serotype differentiation was obtained when analyzing cell culture supernatant samples from viral serotypes C, Asia, and SAT3, irrespective of the subtype. However, limitations of the current antibody pairs were realized in some inconclusive results obtained when analyzing samples from a broader range of O, A, and SAT2 subtypes. The results obtained in this initial study will be used to further optimize the assay using polyvalent or monoclonal antibodies and move toward the analysis of clinical samples. 相似文献

Numerical simulations of Neptune's arcs show that self-gravity between macroscopic arc particles can prevent interparticle impacts and thereby stabilize their resonant confinement by Galatea, a satellite of Neptune. Stable subkilometer arc particles provide a source for replenishing the observed dust and explain the clumpy substructure seen in arcs. A few confining kilometer-sized particles between the major arc components can account for the observed arc widths spanning several resonance sites. The modeled distribution of dust is consistent with observations and helps to explain how embedded satellites may affect the structure and evolution of planetary ring systems. 相似文献

Utilization Potential of Peats – a Study on Peat Biodegradability Determined by Respirometric Method

The biodegradation of different peat types was studied with a manometric respirometric test. Compaction peat and sphagnum peat samples were analysed, and the effect of peat pH on biodegradation behaviour was evaluated. Only minor (BOD/ThOD?<?0.4%) biodegradation was observed with compaction peat samples, and the stable state, in which biodegradation stopped, was achieved during a two month period. As expected, sphagnum peat samples with a lower decomposition rate degraded more than compaction peat samples. Alkalinity (pH between ca. 4–9) of the peat was noticed to reduce the degree of biodegradation and accelerate the achievement of the stable state. 相似文献

Four different treatments of bovine ketosis, using 3 different pharmaceutical preparations, were monitored. The main antiketogenic and glucogenic ingredients of the preparations were as follows: invert sugar (Metabol), prednisolone and dexamethasone (Predasen), the former preparations combined (Metabol & Predasen) and propylene glycol and dexamethasone (Dexaprol). Blood samples were drained from the ketotic cows prior to the treatment. The animals were sampled again 2 and 7 days after the first sampling. The whole blood concentrations of acetoacetate (AA concn), β-hydroxybutyrate (BHB concn) and glucose (glue concn) were determined. The measures of the antiketogenic and glucogenic efficacy of the various treatments applied were their ability to reduce the AA concn and BHB concn and to elevate the gluc concn.Invert sugar (Metabol) alone had no antiketogenic efficacy, although in some cases it led to a subjective clinical improvement as reported by the owners of the animals. Invert sugar and glucocorticoids (Metabol & Predasen) dexamethasone and prednisolone (Predasen) and dexamethason plus propylene glycol (Dexaprol) were equally effective in bringing the AA, BHB and glue concns to normal range. The mean glue concn of the cows treated with Dexaprol was higher than that of the cows in the other treatment groups at 2 days (P < 0.01). No other differences between the 3 antiketogenic treatments could be shown. 相似文献

The study develops a bio-economic crop management model that internalizes the environmental cost of nitrate pollution, accounts for stochastic weather, and includes an option for split fertilization. The integrated model is designed to indicate whether a producer can benefit from applying fertilizer several times during the growing season, in response to crop needs, rather than a single time, at sowing. The model is parameterized for the cultivation of spring malting barley (Hordeum vulgare L.) in Southern Finland. The costs of negative externalities from nitrogen leaching are internalized in the landowner’s decision problem through a pollution tax. The results indicate that without a pollution tax a single application of fertilizer gauged to meet the needs of the entire season is optimal. With a tax, the benefits of split application - applying varying amounts of fertilizer at selected stages of the growing season - increase significantly. In comparison to a single application of fertilizer at sowing, split fertilization improves yields, increases the total amount of fertilizer used, and reduces nitrogen leaching. 相似文献

Multi-location data collected over 30 years in Finland on major cereal crops (spring barley, oat and wheat, and winter wheat and rye) provide an opportunity to characterise variation, interrelation and relative importance of the grain yield determining components, grain number per square meter (GNO) and single grain weight (SGW). In addition to evaluating differences among cereal crops in yield determination, changes among different age groups (AG) were compared. Field experiments were carried out in 1970–2001 at 25 locations in Finland. Grain yield was recorded (at 15% moisture) and SGW (mg) and GNO (number m−2) determined. Frequencies of different GNO and SGW combinations were analysed for all crops and component means in different yield groups as well as among four AGs. In most cases GNO dominated SGW, representing the major yield component determining grain yield. GNO was highly responsive to favourable growth conditions. The change in GNO was especially high when recurrent, relatively low yield groups were compared, while at higher yields the role of SGW became increasingly important, but did not exceed that of GNO. The degree of GNO domination varied according to crop. Increased grain yield in spring oat and winter rye was associated with higher numbers of set grain, while spring barley and winter wheat responded to yield favouring conditions also through higher SGW. Despite GNO dominated SGW due to its responsiveness to growing conditions, its role in yield improvement was not obvious when the entries (n ≥ 30) were classified into four AGs. The main goals of the wheat androgenesis experiment were to study the main phenomena of in vitro androgenesis in anther culture of F2 populations (10) and their parents (6), to compare the usage of P-4 and C-17 media for the formation of embryo/callus and to demonstrate a new plant regeneration system. The P-4 induction medium was found to be significantly better than the C-17 in the number of responsive anthers (RA) and calli induced (CI) at the 1 % and 0.1 % level, respectively. Genotypic effect was evident in both RA and CI. The yields of F2 populations in RA and CI were significantly higher than those of their parents regarding both media. The data confirmed the existence of heterosis for RA and CI in F2 populations. The ratio of green/albino plant regeneration was more favourable in the C-17 derived embryo/calli than in the P-4 derived ones. The frequency of plantlet regeneration was enhanced in the group of unresponsive calli by the application of the multiple-step regeneration system. In this system the calli lacking well developed morphogenic structure were transferred to a new regeneration medium, containing a higher concentration of the same cytokinin, other cytokinin or basic medium, before the occurrence of irreversible changes in their physiology. 相似文献
