临床上大部分抗血小板药物存在继发性出血等副作用,亟需寻找一种安全有效的抗血小板药物。二磷酸腺苷(ADP)和花生四烯酸(AA)是介导血小板聚集的主要物质,文章探讨成纤维细胞生长因子-21(FGF-21)是否抑制ADP或AA诱导大鼠血小板聚集和活化。将大鼠分为正常对照组、正常鼠血小板活化组、阿司匹林干预后血小板活化组、FGF-21高、中、低剂量干预后血小板活化组。给药干预后提取各组血小板,分别用ADP或AA处理,观察处理后血小板聚集情况以及P选择素和血栓素(TXB2)表达水平。与正常对照组相比,正常鼠血小板经ADP或AA处理活化后,血小板聚集率显著升高,血浆中P选择素和TXB2含量明显上升;与正常鼠血小板经ADP或AA处理活化后相比,经阿司匹林和FGF-21干预后分别经ADP或AA处理活化后的血小板聚集率显著下降,血浆中P选择素和TXB2含量显著下降;FGF-21干预组经ADP或AA活化后,血小板聚集率、P选择素和TXB2含量下降水平呈明显剂量依赖性。目前国内外尚未发现FGF-21对血小板聚集与活化作用的相关报道,研究首次证明FGF-21具有抑制ADP和AA诱导血小板聚集和活化作用及明显的抗血凝作用,填补FGF-21在抗血凝研究领域空白,为开发安全有效的抗血凝药物提供理论依据。 相似文献

以不同血缘及不同世代的遗传育种材料为母本、高频孤雌生殖诱导系辽诱1和CAU5等为父本杂交诱导产生的单倍体子粒为实验材料,研究秋水仙素浓度为0.625 mg/mL的条件下,用喷淋加倍设备对试验材料进行8 h喷淋的加倍效果。结果表明,喷淋秋水仙素后,郑单958的S2代的材料加倍率最高,为33.75%;第2位的是L3258×H71,是NSS种质S0代材料,其加倍率为13.02%;第3位的是D299/PH4CV,为NSS种质S1代材料,加倍率为10.34%。研究表明,喷淋秋水仙素的加倍效果与遗传背景关系紧密,喷淋秋水仙素加倍率总体由高到低的顺序为郑单958NSS种质(SS+NSS)种质SS种质欧洲杂交种,喷淋秋水仙素对全部试验材料的加倍率都有提高作用,比自然加倍提高0.83~5.08倍。利用该喷淋设备加倍单倍体子粒,具有节约用药量、操作简便、便于大规模生产应用、减少人员对秋水仙素的接触、废液回收方便、有利于保护环境等优点。 相似文献

The Ryukyu long-furred rat, Diplothrix legata, is a large rodent
distributed only on Amami-ohshima Island, Tokuno-shima Island and Okinawa-jima Island,
Japan. This animal is endangered as a result of deforestation, predation by introduced
carnivores and mortality caused by vehicles. We performed theriogenological examinations
of 32 male and 25 female Ryukyu long-furred rats carcasses collected from wild populations
on northern Okinawa-jima Island from December 2005 to September 2013. Adult males had
remarkably large preputial glands. Seminiferous diameter of adult was significantly small
(136 ± 28 µm, n=8) from April to August. Numerous spermatozoa were
observed from September through February, and seminiferous diameter was significantly
large (216 ± 27 µm, n=12) during this time in adults; testes length
changed in a similar pattern. These findings indicate that the mating season may occur
from September through February. Size (body length) at sexual maturity was estimated to be
>560 mm in both sexes. From observation of corpora lutea and placental scars, litter
size was estimated to range from 2 to 12 (average=6, n=4). These results provide
fundamental knowledge that will be beneficial for in situ and ex

The involvement of the lung during the septic systemic inflammatory response elicited by administration of lipopolysaccharide (LPS) was investigated. Eight-week-old male Sprague–Dawley rats were injected i.p. with 15 mg/kg LPS. After 24 h, the lungs were excised to evaluate the cellular responses to LPS. Matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF) analysis revealed that type VI collagen (ColVI) was extremely upregulated during sepsis in the rat lung within the first 24 h of LPS administration. Upregulation of ColVI protein and its mRNA was demonstrated by Western blot analysis, real time PCR, and immunohistochemistry. To the best of our knowledge, this is the first report demonstrating the activation of ColVI in the rat lung at the early stage of systemic inflammation. Activation of ColVI might be involved in sepsis-mediated lung fibrosis at an early stage. 相似文献

To evaluate the effectiveness of sodium bicarbonate (SB) in removing uranium and protecting animals from uranium toxicity, we intramuscularly administered 1 mg/kg of uranyl nitrate to 8-wk-old male SD rats, and 20 min after administration of uranyl nitrate, the animals were given a single oral administration of SB at 0.1, 0.3 or 1 g/kg. The SB treatment at a dose of 0.3 g/kg or more raised the pH of the rats’ urine until 4 h after treatment, and it significantly reduced the uranium amounts in the kidneys at 1 day after treatment. In another experiment, rats were intramuscularly administered 1 mg/kg of uranyl nitrate, and 20 min later, the animals were treated with sodium bicarbonate (0.1 or 1 g/kg). The rats were autopsied at 1, 3 and 7 days after uranium treatment. High-dose SB resulted in a significant increase in urinary uranium excretion in the first 24 h and a reduction of uranium deposition in the kidneys and femurs, and it also significantly suppressed uranium-induced renal toxicity, as shown by both histopathology and clinical chemistry at 3 days after uranium treatment. Low-dose SB did not show such marked effects. Our findings demonstrated that the uranium decorporation effect of sodium bicarbonate was observed at the dosage showing urine alkalinization in rats and that decorporation effect of sodium bicarbonate might be beneficial if it is administered immediately after incorporation of soluble uranium. 相似文献

The Spontaneously Diabetic Torii (SDT) fatty rat, a model for obese type 2 diabetes,
shows bone quantitative abnormalities, namely low bone mineral density (BMD). The
objective of this study was to evaluate bone morphological changes, in particular
identifying the bone qualitative abnormalities, in the SDT fatty rat. Male SDT fatty rats
showed increases in total trabecular area and trabecular number and decreases in
trabecular thickness in cancellous bones of the proximal tibia, indicating trabecular
miniaturization. The SDT fatty rat is useful for investigation of pathophysiological
changes in bone quality in diabetic osteoporosis. 相似文献

应用胰蛋白酶分次消化法分离乳鼠心肌细胞,以差速贴壁法纯化心肌细胞,α-sarconme-actin抗体免疫细胞化学染色鉴定心肌细胞。心肌细胞在无血清无酚红培养基中培养48h后,用双氢睾酮(DHT)诱导心肌细胞肥大,建立心肌细胞肥大模型。24h后检测心肌细胞肥大的指标心肌细胞表面积;BCA法测定心肌细胞蛋白含量;半定量RTPCR两步法检测心肌细胞肥大的特征性基因—心房利钠因子(atrial natriuretic factor,ANF,β-肌球蛋白重链(β-myosin heavy chain,β-MHC)mRNA的表达。结果显示免疫细胞化学染色显示培养的心肌细胞纯度达到90%以上,心肌细胞分离良好。与对照组相比,10-8 mol/L的DHT能显著的增加心肌细胞表面积、蛋白质含量、ANP和β-MHC基因表达的增加(P0.01),心肌细胞肥大模型建立成功。 相似文献
