旨在对制备的甘露糖修饰的壳聚糖聚乳酸-羟基乙酸共聚物[poly (D,L-lactide-co-glycolide),PLGA]纳米微球作为口蹄疫病毒(foot-and-mouth disease virus,FMDV)核酸疫苗递送载体进行评价。采用西佛碱反应和元素分析制备具有一定取代度的甘露糖修饰的壳聚糖衍生物(mannose modified chitosan,MCS),然后,经双重乳化挥发法制备得到甘露糖修饰的壳聚糖PLGA纳米微球(MCS-PLGA-NPs)。采用纳米粒径仪检测MCS-PLGA-NPs粒径分布和表面电势(zeta)、扫描电镜考察其形态、琼脂糖凝胶电泳观察其对质粒的吸附和吸附质粒后抵抗核酸酶降解能力、CCK-8法检测MCS-PLGA-NPs的细胞毒性、激光共聚焦观察巨噬细胞对MCS-PLGA-NPs-质粒DNA复合物的摄取、荧光显微镜和Western blot验证MCS-PLGA-NPs加载质粒DNA在细胞中的表达。元素分析结果表明,成功制备了取代度为5%~10%的MCS。纳米粒径测定和扫描电镜结果表明,MCS-PLGA-NPs的zeta为正值、粒径分布均匀且形态规则呈球形。琼脂糖凝胶电泳结果显示,MCS-PLGA-NPs吸附质粒的能力随着其质量的增加而增强并且可以在一定程度上抵抗核酸酶降解质粒DNA。在细胞毒性试验中,不同浓度的MCS-PLGA-NPs与RAW264.7细胞共孵育24 h后,细胞存活率仍在85%以上。在细胞摄取试验中,用激光共聚焦显微镜可以明显观察到质粒DNA结合到纳米微球表面被RAW264.7细胞摄取。荧光显微镜和Western blot试验证明MCS-PLGA-NPs加载质粒DNA可以在细胞中进行表达。综上表明,本研究成功制备了MCS以及具有递送核酸疫苗能力的MCS-PLGA-NPs,为FMDV核酸疫苗的递送研究提供了新的方向和见解,也为该递送载体携带特定抗原靶向抗原递呈细胞表面甘露糖受体以及应用于动物免疫的研究奠定基础。  相似文献

Alginates are naturally occurring polysaccharides extracted from brown marine algae and bacteria. Being biocompatible, biodegradable, non-toxic and easy to gel, alginates can be processed into various forms, such as hydrogels, microspheres, fibers and sponges, and have been widely applied in biomedical field. The present review provides an overview of the properties and processing methods of alginates, as well as their applications in wound healing, tissue repair and drug delivery in recent years.  相似文献

提高农药的有效利用率是植保工作者非常关心的问题。本文阐述了农药有效利用率的广义和狭义涵义,并分析了农药使用中存在的药剂浪费、有效利用率低的问题。根据喷雾技术中的“剂量传递”过程,分析了农药有效利用率的狭义涵义,在春季果园和作物苗期,常规喷雾法的农药有效利用率只有20%~30%;在夏秋季果园和作物中后期,随着作物叶面积系数(LAI)的增加,农药的有效利用率可达到50%~60%。论文分析了造成农药有效利用率低的原因,提出喷雾技术的优化措施:(1)大田喷雾时采用机动喷杆喷雾替代背负式手动常规喷雾,可以改善雾滴沉积分布的均匀性;(2)添加喷雾助剂可以提高药液在靶标表面的润湿性;(3)优化雾滴粒径,采用细雾滴替代粗雾滴可以提高雾滴的中靶率;(4)降低施药液量可以减少药液流失;(5)加装挡板可以减少雾滴飘失等。通过以上技术的优化,可以大幅度提高农药的有效利用率,达到减量增效的目的。  相似文献

Strawberry vein banding virus(SVBV)-infected strawberry cells contain cytoplasmic inclusions with isometric particles.To identify the components of the inclusions,green fluorescent protein(GFP)was fused to the carboxy-terminus(C-terminus)of SVBV open reading frames,these constructs were separately transformed into Agrobacterium tumefaciens and infiltrated into Nicotiana benthamiana leaves.Results showed that the SVBV P6 protein assembled into prominent and amorphous inclusion bodies(IBs).To investigate P6 subcellular localization,P6-GFP was ectopically expressed in N.benthamiana leaves by agroinfiltration and then stained with 4′,6-diamidino-2-phenylindole(DAPI).We found the P6 protein accumulated in the nuclei and also formed cytoplasmic IBs with different sizes.To further determine the location of P6 IBs in the cytoplasm,and explore whether the P6 IBs move freely or depend on cytoskeleton and endoplasmic reticulum(ER),the microfilament marker protein(GFP-ABD2-GFP),microtubules marker protein(m Cherry-MAP65-1)and ER marker protein(m Cherry-HDEL)were separately coexpressed with P6-GFP and into N.benthamiana leaves by agroinfiltration,exhibiting that P6 IBs aligned with cytoskeleton and endoplasmic reticulum.Meanwhile,coinfiltration of P1 and P6 indicated the P6colocalized with the P1 protein at periphery of cells.The P6 protein contains one C-terminal nuclear localization signal(NLS)region,a P6 protein mutant with a deleted NLS did not localize in the nucleus,did not form IBs,and was unable to facilitate exogenous GFP expression.These results demonstrate that the deleted NLS region is an important P6 domain required for biological functions.In summary,the mobile P6 IBs are associated with ER,microfilaments and microtubules and move along microfilaments to the SVBV P1 protein in the PD.  相似文献

Understanding farmers’ decision process on the adoption of biological control (BC) may help in promoting adoption. This study explored for the first time the impact of marketing mix elements (product, price, promotion, and place) on bolstering BC for the management of rice stem borer (Chilo suppressalis Walker) among rice farmers in Fuman County in northern Iran. Farmers using BC were familiar with this method primarily via village councils and rural municipality organizations (Dehyaris) and then via distributors of trichocards. Almost half of the farmers (48.3%) had weak to moderate level of contact with distributors of trichocards. All four elements (4Ps) of marketing mix (product, place, price, and promotion) were effective on the marketing of BC agents. Place (distribution) mix, followed by price mix, were the most important factors in marketing trichocards in farmers’ views. According to the sensitivity to 4Ps of marketing mix, most farmers were grouped as semi-sensitive (45.6%) and sensitive (39.4%). Overall, the availability of biological agent products in the market, the affordable price of the products, and the strong relationship of rural people with village councils and rural municipality organizations (Dehyaris) are crucial factors for BC promotion in paddy fields of Fuman.  相似文献

By the infection of Brucella virulent strain and attenuated strain in mice macrophage RAW264.7,the assay was aimed to explore the relationship between NF-κB signaling pathways and Brucella virulent strain and attenuated strain in intracellular survival.Use different MOI Brucella (2308,RB51,16M and M5) to infect mice macrophage RAW264.7,after 0,4,8 and 24 h infected,cracking cell and collecting supernatant,we detected the effect of Brucella on activation of NF-κB signaling pathway by Western blotting.Different concentrations of NF-κB signaling pathway inhibitor were incubated with mice macrophage RAW264.7,with different multiplicities of infection (MOI) of Brucella infecting cells,ELISA kits to detect the expressions of TNF-α,IL-1β and IL-6 cytokine;At the same time,count the number of intracellular bacteria of CFU.The results showed that rough cattle Brucella strains RB51 could strongly activate NF-κB signaling pathway,smooth cattle Brucella strains 2308 was weak in the activation;At the same time,the activation of NF-κB signaling pathway was concentration dependent.When the MOI was 80,infection time was 8 h,NF-κB activation degrees of rough cattle Brucella strains RB51 and smooth cattle Brucella strains 2308 were the strongest,and this pathway was involved in producing TNF-α and IL-6;NF-κB signaling pathway inhibitor BAY11-7082 affected Brucella intracellular survival.So rough cattle Brucella strains RB51 intracellular survival and NF-κB signaling pathway activity were closely related.The results laid the foundation for the further study of Brucella intracellular pathogenesis,also provided scientific basis for the research of new drugs to Brucella,and prevention and treatment of brucellosis.  相似文献
