The simple immunoprecipitation method was used to isolate tilapia immunoglobulin (Ig) for immunization in order to produce monoclonal antibodies (MAbs) specific to tilapia Ig. First, the tilapia antiserum against bovine serum albumin (BSA) was prepared by peritoneal injection of BSA into tilapia, and the tilapia anti‐BSA antiserum was used to precipitate BSA to form the Ig/BSA immune complex. The Ig/BSA immune complex was then injected into Swiss mice for hybridoma production. After fusion, three hybridoma clones producing MAbs specific to the tilapia antibody were selected by dot blot and Western blot. All MAbs (101A, 59G, and 11A) were bound specifically to the heavy chain of immunoglobulin M (IgM). The MAbs 101A and 59G demonstrated twofold higher affinity than MAb 11A and the commercialized antibody. However, MAbs 11A could also bind to the heavy chain of IgM in Asian seabass, Lates calcarifer, as well. These MAbs can be used to monitor the immune responses of individual fish by indirect ELISA upon exposure to various antigens.  相似文献

Sargassum horneri (S. horneri), an edible brown alga, has been proposed as a functional food with an improvement effect on abnormal skin immune responses. The present study investigates the anti-allergic effect of an ethanol extract from S. horneri (SHE) on immunoglobulin E (IgE)/bovine serum albumin (BSA)-mediated activation in bone marrow-derived cultured-mast cells (BMCMCs) and passive cutaneous anaphylaxis (PCA) reaction in mice. SHE markedly and dose-dependently suppressed the degranulation of BMCMCs by reducing the β-hexosaminidase and histamine release without cytotoxicity. In addition, SHE significantly decreased the FcεRI expression on the surface of BMCMCs and its IgE binding. Moreover, SHE reduced the mRNA expression and the production of allergic cytokines; interleukin (IL)-1β, IL-4, IL-5, IL-6, IL-10, IL-13; interferon (IFN)-γ and/or tumor necrosis factor (TNF)-α; and a chemokine, thymus and activation-regulated chemokine (TARC), by suppressing the activation of Src-family kinases and nuclear factor (NF)-κB signaling. In further study, the application of SHE reduced the PCA reaction in an IgE/BSA-induced type I allergic mice model. Taken together, we suggest that SHE has an anti-allergic effect in type I allergic responses.  相似文献

Adverse events can occur after rabies post‐exposure prophylaxis (PEP), and linkage to causality is often difficult to determine. We report a case of recurrent temporary paralysis that began immediately after the initiation of rabies PEP in a man exposed to a bat. The recurrent temporary paralysis first occurred in the patient after his initial dose and then again after day 3 of his rabies PEP. The PEP was terminated prior to a serologic response. The patient continued to experience numerous discrete episodes of temporary paralysis for over two years.  相似文献

牛奶中含有丰富的活性营养物质,包括乳铁蛋白、免疫球蛋白、乳过氧化物酶、α-乳白蛋白、β-乳球蛋白等。传统工艺生产的酸奶经过高温杀菌,活性营养物质几乎完全损失。为了避免这一情况,本文推出一种新的酸奶生产工艺,生产出的酸奶可保留大部分活性营养物质。首先将牛奶的乳脂进行分离,乳脂部分进行125 ℃、5 s杀菌,脱脂牛奶部分进行75 ℃、15 s杀菌,将两部分料液混合后加入菌种发酵至酸度75~85°T,破乳后冷却后熟8 h以上。经测定,新工艺酸奶中α-乳白蛋白、β-乳球蛋白、乳铁蛋白、免疫球蛋白、乳过氧化物酶含量分别为966 mg/L、1 963 mg/L、23.4 mg/L、102 mg/L、1 025 U/L,远高于传统工艺酸奶的68 mg/L、200 mg/L、0.7 mg/L、14 mg/L、0 U/L。通过对比新工艺与传统工艺酸奶保质期内的活菌数、黏度、酸度和口感,并无显著差异。  相似文献

鱼类是最早产生免疫球蛋白(Immunoglobulin,Ig)的脊椎动物之一。鱼类IgM作为鱼类特异性体液免疫应答中最主要的免疫介质,目前已有较详细的研究报道。除此之外,近年来在鱼类还新发现了几个Ig,如IgD、IgW、IgNAR、IgZ和IgT等,笔者对其结构及功能进行简要综述,以期为国内同行提供科学参考。  相似文献

断奶日龄对五指山仔猪生长性能和血清生化指标的影响   总被引:1,自引:0,他引:1  
本试验旨在研究断奶日龄对五指山仔猪生长性能和血清生化指标的影响。选取日龄相近(±1 d)、体重[(0.56±0.07)kg]相近的五指山仔猪96头,随机分成4组,每组设4个重复,每个重复6头猪。4组仔猪分别在21、28、35和42日龄断奶。试验从8日龄开始,至56日龄结束,试验期间定期测定体重、采食量和血清生化指标。结果表明:各组仔猪断奶后平均日增重(ADG)出现不同程度的下降,且断奶日龄越早,ADG下降幅度越大;35日龄断奶组和42日龄断奶组21~28日龄、28~35日龄、35~42日龄、49~56日龄阶段的ADG显著高于21日龄断奶组(P0.05)。各组仔猪断奶后平均日采食量(ADFI)显著增加(P0.05),42日龄以后,各组ADFI无显著差异(P0.05)。21日龄断奶组、28日龄断奶组、35日龄断奶组血清总蛋白(TP)、白蛋白(ALB)、免疫球蛋白G(IgG)、免疫球蛋白M(IgM)含量以及谷丙转氨酶(ALT)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)活性在断奶后7 d均发生显著变化(P0.05),并以21日龄断奶组波动最大;与35日龄断奶组和42日龄断奶组相比,21日龄断奶组28、35日龄时血清TP、ALB、IgG、IgM含量显著降低(P0.05),ALT、AST、LDH活性显著提高(P0.05),到56日龄时,血清TP、IgG、IgM含量仍显著低于35日龄断奶组和42日龄断奶组(P0.05)。21日龄断奶组血清甘油三酯(TG)、胆固醇(CHOL)含量在28~56日龄间存在显著变化(P0.05),35日龄断奶组和42日龄断奶组血清TG、CHOL含量在整个试验期内均无显著变化(P0.05)。35日龄断奶组血清TP、ALB、IgG、IgM含量以及ALT、AST、LDH活性除在42日龄时与42日龄断奶组存在显著差异(P0.05)外,其余时间点与42日龄断奶组均无显著差异(P0.05)。由此得出,35或42日龄断奶的五指山仔猪在生长性能、血清生化指标及免疫机能方面优于21日龄断奶的五指山仔猪。  相似文献

Two kinds of specific chicken egg yolk immunoglobulins (IgYs), IgY‐WSSV and IgY‐VP28, were, respectively, raised against the 2 mM binary ethylenimine (BEI)‐inactivated white spot syndrome virus (WSSV) and a principal envelope protein VP28. The activity of purified specific IgYs was stable under the conditions of 20–70 °C, pH 3.0–10.0 and 0–700 g L?1 sucrose solution. In the neutralization assay, these high‐affinity IgY antibodies can specifically bind with the virus particles to protect shrimp (Fenneropenaeus chinensis) against WSSV infection. After oral delivery for 20 days, the IgY‐WSSV exerted a higher protection effect (RPS: 71.5%) than IgY‐VP28 (RPS: 63.7%). Moreover, an increase in RPS (79.2%) was found on addition of IgY‐WSSV:VP28 (0.1% IgY‐VP28 plus 0.2% IgY‐WSSV). This may indicate that neutralization of WSSV refers to the multiple‐hit model. By time‐course study of the levels of the specific IgYs in vivo, the data showed that the titre was enhanced to a relatively high level (P/N=8.35±0.45) at 3 days post administration, declined slightly (P/N=7.13±1.01) at 7 days post administration and then remained stable for further investigation. The stable antibody level potentially contributes towards blocking a large number of WSSV particles from entering and infecting on the major tissues at the early and late stages after challenge in shrimp.  相似文献
