为探究体细胞克隆荷斯坦奶牛出生后死亡的原因,对新生后死亡的克隆荷斯坦公牛和自然繁殖的荷斯坦公犊的主要组织器官进行比较。通过解剖和石蜡切片-HE(hematoxylin-eosin staining)染色技术,对主要的组织器官结构进行观察和分析。结果表明,新生后死亡的克隆荷斯坦公牛肺部结构清晰;肝脏的肝细胞明显肿大,出现脂肪轻度变性;肾小管上皮细胞出现变性;心肌的肌纤维间空隙增大;骨骼肌纤维间隙明显,空泡变性,这可能导致该克隆牛犊肌肉无力并功能不全;淋巴结皮髓质界限不分明,淋巴小结细胞较稀疏,生发中心不明显,淋巴窦细胞较少;脾脏内红细胞较少,这说明该克隆牛的造血功能可能不完善;胸腺的皮质部分和髓质部分界限不明显,嗜酸性胸腺小体不易辨认,可能发育不全。该克隆公牛免疫器官出现不同程度的发育不全现象较严重,这有可能是其出生后死亡率高的主要原因。 相似文献

为了阐明肠道气呼吸对泥鳅的生理作用,本研究通过抑制大鳞副泥鳅肠道气呼吸,探究其主要呼吸器官的组织病理变化。结果显示,被抑制肠道气呼吸的大鳞副泥鳅通常在1周左右死亡。当被抑制肠道气呼吸的大鳞副泥鳅出现垂死时,采集对照组和实验组的鳃、皮肤、前肠、中肠、后肠以及直肠进行苏木素—伊红(H.E)染色、阿利新蓝—高碘酸雪夫氏(AB-PAS)染色组织切片观察和扫描电镜观察,主要的病理变化:①H.E染色结果显示实验组大鳞副泥鳅鳃丝末端充血,背部皮肤表皮层的毛细血管收缩并减少,且真皮层细胞呈畸形,前肠黏膜褶膨大,后肠浆膜层有血红细胞渗出,后肠、直肠结缔组织显著增厚;②AB-PAS染色结果显示,实验组大鳞副泥鳅鳃、背部皮肤、后肠、直肠组织中嗜酸性空泡细胞均增多,前肠和中肠固有层酸性黏蛋白含量增多,黏膜下层中性黏蛋白含量减少;③扫描电镜结果显示,实验组大鳞副泥鳅鳃丝鳃小片表面皱缩,表皮受损脱落,背部皮肤表面分泌孔增加,中肠内腔表面突起增多,后肠和直肠絮状颗粒增多。研究表明,抑制大鳞副泥鳅肠道气呼吸会引发其主要呼吸器官上皮组织出现黏液细胞增多、血红细胞溢出等病理变化,甚至导致机体死亡,由此可知,肠道气呼吸行为是大鳞副泥鳅的必要生理活动。本研究将为大鳞副泥鳅的健康养殖及幼苗培育提供新的参考。 相似文献

Minimally invasive techniques used to evaluate canine peripheral lymphadenopathy (PLN), including fine needle aspiration biopsy with cytological evaluation (FNAB‐C) and flow cytometry (FC), have benefits and limitations. The cell block (CB) method is an alternate processing technique in which fine needle aspirate biopsy samples are concentrated, fixed, and embedded in paraffin for routine histological processing/staining. Utilizing three observers, we determined the diagnostic value of the CB in evaluating canine PLN across six categories (non‐diagnostic, reactive, inflammatory/infectious, probable lymphoma and lymphoma, metastatic neoplasia) and correlated findings to immunophenotypic and clonal antigen receptor rearrangement results in canine nodal lymphoma. Eighty‐five paired FNAB‐C and CB samples were evaluated from canine patients presenting to the University of Minnesota Veterinary Oncology or Internal Medicine services. Diagnostic quality samples were obtained in 55/85 (65%) CB and 81/85 (95%) FNAB‐C samples, respectively, and nodal pathology impacted CB diagnostic yield. Overall percent agreement between diagnostic‐quality FNAB‐C and CB samples was 86%, but increased to 95% if the categories of lymphoma and probable lymphoma were combined. There was 100% agreement for both the diagnoses of metastatic neoplasia and reactive lymph nodes and 92% agreement for the diagnosis of lymphoma/probable lymphoma. Using immunohistochemistry (IHC), CB samples correctly immunophenotyped 22/23 (96%) cases of B‐cell lymphoma, but only 1/6 (17%) cases of T‐cell lymphoma. IHC was not completed on nine cases of lymphoproliferative disease because of insufficient cellularity. When the CB method (CBM) yielded diagnostic quality samples there was good to excellent agreement with FNAB‐C samples and CB samples were suitable for some IHC tests. 相似文献

为探究来源于牛呼吸道疾病综合征发病牛的皮特不动杆菌对小鼠的致病性,对其进行相关毒力基因的检测和小鼠致病性试验,试验内容主要包括临床症状观察、病理剖检、组织病理学观察,以及半数致死量(LD50)和各器官载菌量的测定。结果显示,牛源皮特不动杆菌ZZCNC1807-6和ZZCSF1807-9菌株对小鼠均具有较强的致死性,对小鼠的LD50分别为4.487×107 CFU·mL-1和3.177×108 CFU·mL-1。剖检死亡小鼠可见,牛源皮特不动杆菌主要造成小鼠皮下充血、肺淤血肿胀、肠道积气积液等症状,可引起心、肝、脾、肺、肾、脑等多种器官感染。组织病理学观察显示,小鼠各器官病变均较轻微,主要表现在肝、肺中有少量炎性细胞浸润,脾中白髓细胞坏死和纤维增生,其中ZZCNC1807-6攻菌组的病变较ZZCSF1807-9攻菌组相对严重。载菌量分析结果显示,2株菌对小鼠肝、心、脾、肺和肾的侵袭力差异不大,但ZZCNC1807-6对脑的侵袭力强于ZZCSF1807-9。综上,ZZCNC1807-6菌株的毒力强于ZZCSF1807-9。 相似文献

Amoebic gill disease (AGD) is a pathogenic disease in salmonids caused by Neoparamoeba perurans. Treatment of AGD infection has been through freshwater bathing of the fish. However, as the availability of fresh water is often limited, hydrogen peroxide has been introduced as an alternative treatment. This study investigated the effect of hydrogen peroxide as treatment for AGD‐infected salmon (Salmo salar L.,) at different seawater temperatures and hydrogen peroxide dosages. In total, 600 fish were challenged with N. perurans and the severity of the AGD infection was measured using a gill score scale. After challenge and disease development, the fish were distributed into 12 tanks. The treatment was performed at different seawater temperatures (8°C, 12°C, 17°C) using different hydrogen peroxide doses. Each temperature included an untreated control group. Linear models were used to analyse gill score. A significant effect of treatment was found (?0.68 ± 0.05) regardless of dose and temperature, suggesting that hydrogen peroxide was effective in treating AGD. When the model included dose, a negative linear relationship between dose and gill score was found. The study proved that treatment of AGD with hydrogen peroxide was successful, as gills partially recovered following treatment and further disease development was delayed. 相似文献

采用透射电镜技术和生化分析等方法,对患“水瘪子”病病蟹的超微病理和病理生理进行研究。超微结构观察表明,病蟹肝胰腺、鳃、肌肉等组织细胞发生了不同程度的病变,主要表现为肝细胞变性、坏死,脂滴锐减,微绒毛肿胀、断裂,细胞内线粒体肿胀,内嵴紊乱、减少甚至消失形成空泡,内质网扩张或断裂为片层状结构;鳃上皮细胞角质膜变薄,细胞核异染色质化,角质层面伸出的指状突起破裂消失,上皮细胞顶膜形成的微绒毛杂乱、断裂甚至消失,角质膜下空腔增大,线粒体畸变,数量减少,内嵴减少,形成空泡,溶酶体数量增多,并与空泡和空泡化的线粒体形成自噬体,严重病变的鳃上皮细胞中出现细菌颗粒,未见包涵体等病毒样颗粒;肌肉的病变主要是肌纤维松弛、断裂,肌质网溶解消失或者形成小泡,线粒体数量减少、体积变小,细胞核固缩且边缘化。病理生理分析结果显示,病蟹血淋巴中血糖含量和谷草转氨酶活力显著高于健康蟹,碱性磷酸酶和谷丙转氨酶活力极显著高于健康蟹,甘油三酯的含量极显著低于健康蟹;病蟹肝胰腺中肝糖原含量极显著低于健康蟹,碱性磷酸酶和酸性磷酸酶活力显著低于健康蟹。 相似文献

Phthalates such as di‐n‐butyl phthalate (DBP) and their esters are widely used plasticizers, their ubiquitous presence in daily life, inevitably leads to their restricted use due to important environmental pollution and health impacts and endocrine disruption potential. The aim of this study was to examine the effects of a sublethal concentration of 10 mg L?1 DBP on haematocrit (HCT) values, gills and liver histology, malondialdehyde (MDA, 2‐thiobarbituric acid‐TBA reactivity) and reduced glutathione (GSH) levels in gills and liver tissue as oxidative stress biomarkers in the aquaculture fish species Nile tilapia (Oreochromis niloticus) after 24 (DBP‐24) and 96 (DBP‐96) h exposure. No differences were found between per cent HCT values in the 24 h exposure groups (P > 0.05). Response of antioxidant defence systems in liver and gill tissues of the fish were dependent on exposure duration and changed to a higher extent during 96 h. MDA levels in liver tissue increased in DBP treated fish in comparison to the control fish. However, the differences between the exposure and control groups were not significant (P > 0.05). A statistically significant decrease (P > 0.05) was recorded in gill MDA levels in the DBP‐96 group when compared to the control and DBP‐24 groups. The liver GSH levels were unchanged in the DBP treated fish. However, GSH levels were significantly lower in the gill tissue of the DBP‐96 group. Exposure to DBP caused several degenerative changes in the histology of gill and liver tissue. Gills displayed hyperaemia, epithelial lifting, oedema, talengiectasia, epithelial hyperplasia and fusion of secondary lamellae, whereas in liver several circulatory anomalies (hyperaemia, blood congestion and sinosoid dilatation) and vacuolization of hepatocytes were observed. Twelve quarters of six lactating cows were inoculated with Mycoplasma leachii strain GN407 through intramammary ductal infusion; another 12 quarters were inoculated with heat-inactivated M. leachii culture medium as negative controls. Multidisciplinary procedures, including clinical assessment, etiology assessment, pathology and immunohistochemistry (IHC), were used to elucidate the pathogenicity of M. leachii in bovine mastitis. From post-inoculation days (PIDs) 3 to 9, 12 inoculated quarters developed mild to severe clinical mastitis and mammary tissue histopathological changes, including inflammatory cell infiltration and architectural destruction of mammary gland ducts. The M. leachii antigen was also detected by IHC in the mammary gland epithelial cells of the inoculated quarters as a weak signal on PID 6 and as a strong signal on PID 9. The control quarters also developed mild mastitis and histopathological changes on PID 9, and M. leachii was also detected by IHC. Throughout the experimental period, the quarters of the negative control cow were clinically and pathologically normal, and the M. leachii antigen was not detected. In conclusion, direct histological and immunohistochemical evidence confirmed that M. leachii causes clinical bovine mastitis through histopathological lesions induced by invasion of the pathogen into mammary gland cells and through inflammatory cell infiltration. 相似文献

Since the 1940s, the anadromous allis shad, Alosa alosa (L.), has suffered population declines throughout its distribution range in Europe. In context of EU‐LIFE projects for the reintroduction of the allis shad in the Rhine system, a comprehensive study was started in 2012 to investigate infectious diseases occurring in allis shad. In course of the study, 217 mature and young‐of‐the‐year allis shad originating from the wild population from the Gironde–Garonne–Dordogne system (GGD‐system) and the Rhine system as well as 38 allis shad from the breeding population were examined by use of bacteriological and histological methods. In 2012 and 2014, an endocarditis valvularis thromboticans caused by a coccoid bacterium was detected in 16% and 25% of mature allis shad originating from the GGD‐system. Results of microbiologic examinations, including biochemical characteristics, matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS) and 16S rRNA gene sequence analysis, revealed Lactococcus lactis as causative agent of this infection. This is the first report of an endocarditis valvularis and parietalis thromboticans caused by Lactococcus lactis in fish. Possible sources of infection as well as the impact for the reintroduction programme are discussed. 相似文献

Pot experiments were carried out to characterize the response of two Cucumis metuliferus accessions (BGV11135 and BGV10762) against Mi1.2 gene (a)virulent Meloidogyne arenaria, M. incognita and M. javanica isolates and to determine the compatibility and the effect on physicochemical properties of fruit melons. In addition, histopathological studies were conducted. One week after transplanting, plants were inoculated with one J2 cm?3 of sterilized sand (200 cm3 pots) and maintained in a growth chamber at 25 °C for 40 days. The susceptible cucumber cv. Dasher II or melon cv. Paloma were included for comparison. The number of egg masses and number of eggs per plant were assessed, and the reproduction index (RI) was calculated as the percentage of eggs produced on the C. metuliferus accessions compared to those produced on the susceptible cultivars. The compatibility and fruit quality were assessed by grafting three scions, two of Charentais type and one of type piel de sapo, under commercial greenhouse conditions. The resistance level of both C. metuliferus accessions ranged from highly resistant (RI < 1%) to resistant (1% ≤ RI ≤ 10%) irrespective of Meloidogyne isolates. Melon plants grafted onto C. metuliferus accession BGV11135 grew as self‐grafted plants without negatively impacting fruit quality traits. Giant cells induced by Meloidogyne spp. on C. metuliferus were in general poorly developed compared to those on cucumber. Furthermore, necrotic areas surrounding the nematode were observed. Cucumis metuliferus accession BGV11135 could be a promising melon rootstock to manage Meloidogyne spp., irrespective of their Mi1.2 (a)virulence, without melon fruit quality reduction. 相似文献
