为安全有效地提高虹鳟苗种成活率,应用1种生物制剂——抗菌肽对虹鳟苗种进行浸泡处理。通过比较苗种成活率,检测SOD、ALP、CAT、IL、IFN、TNF等6种免疫相关酶表达量,采用扫描电镜检测苗种体表细菌情况,确定抗菌肽浸泡对虹鳟成活率的影响。试验结果显示,自破膜期开始,用40%抗菌肽浸泡10次,试验组6种免疫相关酶表达量明显高于对照组,试验组苗种的成活率高达95%以上,对照组苗种的成活率则低于30%。电镜观察结果显示,试验组仔鱼体表细菌数量显著减少。试验结果表明,用抗菌肽浸泡法能显著提高虹鳟苗种成活率。  相似文献

A 60‐day feeding trial was conducted to examine the effects of different levels (0, 100, 200, 400 and 600 mg/kg) of antimicrobial peptides on growth, protease activity of foregut, the morphology of foregut villi and related genes mRNA expression level in the common carp (Cyprinus carpio). The results showed that the feed of antimicrobial peptides promote common carp growth, and the optimal dosage of antimicrobial peptides is 200–333 mg/kg in the common carp feed. The protease activity of 200 and 400 mg/kg groups were significantly higher than the control and other groups (p < 0.05). The foregut villus height with 100, 200 and 400 mg/kg antimicrobial peptide groups were significantly higher than control group (p < 0.05). The crypt depth of 200 and 400 mg/kg antimicrobial peptide groups were significantly lower than control group (p < 0.05). The ratio of villus height and crypt depth of 100, 200 and 400 mg/kg antimicrobial peptide groups were significantly higher than control group (p < 0.05). The ratio with 600 mg/kg group was significantly lower than the control group (p < 0.05). The IGF‐I gene expression level of 200 mg/kg and 400 mg/kg groups were significantly higher than the control group and 600 mg/kg group (p < 0.05). The IL‐1β gene expression level of 100 mg/kg and 200 mg/kg groups were significantly higher than the control group (p < 0.05). These results indicated up‐regulation of growth and immune related genes in antimicrobial peptides fed common carp. Correlation analysis showed that IGF‐I mRNA and IL‐1β mRNA were positively correlated with SGR. IL‐1β mRNA and FCR were significantly negative correlated. It indicated that growth and immune gene common regulated the growth of the carp under antimicrobial peptides intervention. In conclusion, antimicrobial peptides can improve growth and related genes mRNA expression in the common carp. Further studies using molecular biological technique or immunologic methods are required to conclude that antimicrobial peptides are beneficial in common carp.  相似文献

油菜籽中含有多种具有抗氧化活性的物质,为抵御机体的氧化损伤起到了积极作用。本文综述了油菜籽来源的诸多抗氧化活性成分及抗氧化活性的评价方法。油菜籽主要的抗氧化成分有肽类、多酚类、多糖类、生育酚和其它抗氧化成分,其中的肽类主要有三种产生方式:非消化酶酶解、消化酶酶解和发酵。针对油菜籽来源的活性物质抗氧化活性的评价方法,包括体外测定方法(化学检测)和细胞内抗氧化实验和动物实验。综述还探讨了油菜籽抗氧化活性成分研究中存在的问题和解决方法,旨在为油菜籽活性成分的开发和利用提供参考。  相似文献

The antimicrobial peptides (AMPs) are a class of defense peptides that widely exist in nature, which are important components of the innate immune system. Compared to antibiotics, AMPs have some good quality, such as small molecular weight, good water-solubility, thermal stability and unique antibiotic mechanism. They can also restrain virus, fungus and even tumour. With the appearance of multidrug-resistant (MDR) bacteria and the increasing of new AMPs, the study focuses on it’s mechanism of biological activity. This paper summarizes the biological activity mechanism and molecular modification of AMPs, so as to provide theory basis for the molecular modification and the application of AMPs.  相似文献

为了探究灵芝活性组分灵芝多肽、多糖的降血糖机理,采用四氧嘧啶建立糖尿病小鼠模型,经不同组分灵芝提取物灌胃连续4周后测定空腹血糖值、胰岛素浓度、肝糖原、肌糖原含量、肝脏SOD活性MDA含量。结果表明,灵芝多糖能有效增加小鼠血清胰岛素浓度(P0.05);灵芝多肽和多糖可显著降低糖尿病小鼠空腹血糖含量,降糖率分别为19.1%和29.8%;显著增强小鼠肝脏SOD活性(P0.05),升高率分别为13.4%和19.7%;降低肝脏MDA含量(P0.05),且分别降低了27.8%和35.1%。  相似文献

为研究发酵床饲养模式下饲粮中添加抗菌肽对仔猪生长性能、抗氧化指标、免疫指标及盲肠主要菌群数量的影响,选用108头35日龄,体重13kg左右的健康苏钟猪,随机分为3组,每组3重复,每重复12头猪,分别饲喂基础饲粮(对照组)、基础饲粮+40mg/kg杆菌肽锌+20mg/kg硫酸抗敌素(抗生素组)和基础饲粮+300 mg/kg抗菌肽(抗菌肽组),试验期49d。结果表明:(1)与对照组相比,饲粮中添加抗菌肽能够显著提高仔猪血清和肝脏中谷胱甘肽过氧化物酶活性(P0.05),极显著降低肝脏丙二醛含量(P0.01);抗菌肽组肝脏谷胱甘肽过氧化物酶活性比抗生素组极显著提高(P0.01)。(2)与对照组相比,饲粮中添加抗菌肽能够极显著提高仔猪十二指肠上皮内淋巴细胞数量(P0.01);抗菌肽组仔猪血清IgA含量分别比对照组和抗生素组提高4.03%、2.27%,抗菌肽组空肠上皮内淋巴细胞数量比对照组提高10.70%,但差异均不显著(P0.05)。(3)与对照组相比,饲粮中添加抗菌肽能够显著降低仔猪盲肠中大肠杆菌数量(P0.05);细菌总数降低5.98%,但差异不显著(P0.05)。综上所述,本试验条件下,饲粮中添加300mg/kg的抗菌肽能够提高仔猪抗氧化和免疫功能;减少仔猪盲肠中大肠杆菌数量,而对盲肠有益菌群无不良影响。  相似文献

Detailed knowledge of the evolutionary genetics of virulence is needed to understand and predict host–pathogen dynamics. This study used a virulence assay based on digital image analysis and treated virulence as a quantitative rather than a binary trait. Such quantitative data may better reflect the genetic underpinning of virulence in many pathogen systems and provide better resolution in statistical investigations. A greenhouse experiment based on a common garden design was conducted to measure virulence (% of leaf area covered by lesions) of 126 genetically distinct isolates of the barley scald pathogen, Rhynchosporium commune, originating from nine field populations around the world. Virulence in this pathosystem was found to be a quantitative trait with a continuous distribution in all populations. By comparing population genetic differentiation for virulence and neutral microsatellite markers (i.e. a QST/GST comparison), evidence that virulence is under stabilizing selection across populations was found. Heritability values were high and ranged from 0·52 to 0·96 with a mean heritability of 0·84. Virulence was positively correlated with spore production as predicted by the trade‐off theory of virulence evolution. Furthermore, an association analysis between virulence and sequence haplotypes of three known necrosis‐inducing effector genes (NIP1, NIP2 and NIP3) revealed a significant effect of NIP2 haplotypes and NIP1 deletions. Overall, the results support a quantitative model for virulence in the R. commune–barley pathosystem and very high evolutionary potential for this trait.  相似文献

This experiment was conducted to assess the effects of antimicrobial peptides at different levels (Diet 1 (0%), Diet 2 (0.1%), Diet 3 (0.2%), Diet 4 (0.4%), Diet 5 (0.6%) and Diet 6 (1%)), on growth, serum biochemical indices and antioxidant effect, feed utilization and disease resistance in Litopeneaus vannamei. There were four replicates in each group in the experiment (mean weight = 0.21 ± 0.00 g) and also fed with their respective diets for 8 weeks. Growth performance compared with the control group (0%) significantly increased at first and then decreased among treatment groups (p < .05) whereas the survival rate ranged from 78% to 96%. No significant differences were observed in terms of moisture, crude protein and ash content, but there was a significant increase in crude lipid (p < .05). In serum, acid phosphatase, alkaline phosphatase, polyphenol oxidase, triglyceride, glucose and total cholesterol changed as compared with 0%. Total antioxidant capacity, glutathione peroxidase, catalase, superoxide dismutase and malondialdehyde were also different from 0%. Disease resistance was increased in shrimp among treatment groups with 0.4% recording the lowest mortality percentage of 37% after the challenge test. The results from the present study suggested that supplementation of AMP at 0.4% in shrimp diet can improve growth performance, antioxidants activities and innate immune response of Pacific whiteleg shrimp.  相似文献

Based on their composition, marine algae, and namely red seaweeds, are good potential functional foods. Intestinal mucosal barrier function refers to the capacity of the intestine to provide adequate containment of luminal microorganisms and molecules. Here, we will first outline the component of seaweeds and will summarize the effects of these on the regulation of mucosal barrier function. Special attention will be paid to unique components of red seaweeds: proteins and derived peptides (e.g., phycobiliproteins, glycoproteins that contain “cellulose binding domains”, phycolectins and the related mycosporine-like amino acids) together with polysaccharides (e.g., floridean starch and sulfated galactans, such as carrageenans, agarans and “dl-hybrid”) and minerals. These compounds have been shown to exert prebiotic effects, to regulate intestinal epithelial cell, macrophage and lymphocyte proliferation and differentiation and to modulate the immune response. Molecular mechanisms of action of peptides and polysaccharides are starting to be elucidated, and evidence indicating the involvement of epidermal growth factor receptor (EGFR), insulin-like growth factor receptor (IGFR), Toll-like receptors (TLR) and signal transduction pathways mediated by protein kinase B (PKB or AKT), nuclear factor-κB (NF-κB) and mitogen activated protein kinases (MAPK) will also be summarized. The need for further research is clear, but in vivo experiments point to an overall antiinflammatory effect of these algae, indicating that they can reinforce membrane barrier function.  相似文献

利用基因工程技术在毕赤酵母中高效表达乳源抗高血压七肽,为大规模生产抗高血压肽奠定基础。根据毕赤酵母密码子偏爱性,人工合成了具有较高活性的抗高血压七肽串联基因序列。将串联基因克隆至表达载体pPIC9上,并转化毕赤酵母GS115,经PCR检测获得2株重组菌株。重组菌经甲醇诱导,表达的分泌蛋白经胰蛋白酶酶解,测定酶解产物抑制ACE酶活性。结果表明,表达蛋白的胰蛋白酶酶解产物抑制ACE酶活性达到65.12%。IC50值为0.0203 mg.mL-1。研究成功合成了串联的13拷贝抗血压七肽基因,并实现了其在毕赤酵母中的高效表达,表达蛋白的胰蛋白酶酶解产物具有抗高血压肽活性。  相似文献
