从患有腐皮病的锦鲤(Cryprinus carpio)肝、肾、脾、血液里分离到1株优势菌,对该菌进行纯化培养并经人工感染试验,确定该菌为病原菌。经16S r RNA序列分析,鉴定该病原菌为蜡样芽孢杆菌(Bacillus cereus)。用不同浓度福尔马林和不同温度灭活蜡样芽孢杆菌,筛选其灭活效果并制得灭活疫苗。将灭活疫苗免疫健康锦鲤后测定其抗体效价变化及免疫保护率。结果表明,0.2%的福尔马林溶液在28℃下灭活24 h及4℃灭活36 h为蜡样芽胞杆菌的适宜灭活条件。首次免疫后第20天,免疫Ⅰ组、Ⅱ组、Ⅲ组、Ⅳ组血清抗蜡样芽孢杆菌的抗体效价达到高峰,分别为1∶128、1∶64、1∶64、1∶32;第30天同样剂量加强免疫后,各免疫组血清抗体效价达到更高水平,分别为1∶256、1∶256、1∶128、1∶64;免疫后第50天时攻毒,Ⅰ组、Ⅱ组、Ⅲ组、Ⅳ组免疫保护率分别为66%、66%、55%、44%。  相似文献

Arabinogalactan protein JC from Jatropha curcas seed endosperm (mean molecular weight 140 kDa) was isolated by cold water extraction and characterized concerning sugar and amino acid composition. At 10 and 100 µg/mL JC stimulated mitochondrial activity (MTT test) of human skin cells (HaCaT keratinocytes, fibroblasts) and the ATP status of primary keratinocytes. JC did not influence the cellular proliferation, while primary keratinocytes were triggered into differentiation status. Investigations on a potential mode of action of JC were performed on complex organotypic skin equivalents. JC induced the production of HGF, KGF and TGFβ, with TGFβ being the main inductor for the differentiation-inducing effect of JC. Also the expression of GM-CSF was stimulated strongly by JC.  相似文献

ABSTRACT:   This study assessed the stock-recruitment relationship (SRR) for the Japanese sardine Sardinops melanostictus in the North-western Pacific. Of the 20 SRR models investigated, the Akaike information criterion (AIC) was the minimum (AIC = 551.2) when the data were separated into two groups (A and B) and the log-normal distribution was applied as the error term. Group A was constructed with SRR data from 1976–1987 and 1992–2004. Group B consisted of data from 1988–1991. The AIC minimum model was R  = 22.8 S  ×  e ε for Group A, where R , S , and ε denote the recruitment of sardine (individual number of 0-year old fish), spawning stock biomass (SSB), and error term, respectively. This model indicated that recruitment was proportional to the SSB and that no density-dependent effect operated over the range of SSB investigated (51 000–11.3 million t). Recruitment was markedly higher (lower) when the sea surface temperature (SST) of the Kuroshio Extension area in February was low (high). The essential SRR can simply be expressed as R  = 22.8 S  ×  e ε with the level of recruitment deviating from the model to a greater or lesser degree depending on the environmental conditions.  相似文献

RPS11是核糖体小亚基40S的组成部分,属于核糖体蛋白S17p家族,由RPS11基因所编码,主要存在于真核生物中。本研究通过PCR扩增RPS11基因,构建pET43a-RPS11和pGEX-4T-1-RPS11原核表达载体。将重组质粒转入E.coli DH5α,序列测定正确后,将其转入E.coli BL21(DE3),加入IPTG诱导表达。表达蛋白经SDS-PAGE分析后,利用亲和层析法纯化蛋白。结果成功克隆RPS11基因,构建了原核表达载体pET43a-RPS11和pGEX-4T-1-RPS11,转化宿主菌E.coli BL21(DE3)中进行了表达。SDS-PAGE分析证实表达目的蛋白正确。通过Ni-TNA和GSH-Sepharose层析柱获得纯化蛋白,为进一步研究该蛋白的生物学功能奠定了基础。  相似文献

Eight strains of nervous necrosis virus (NNV) isolated in Vietnam were used to detect the pathogenicity and immune response in sea bass (SB). All strains induced cytopathic effect in SB cell line, complete destruction of monolayer of cells appeared after seven days post infection (dpi). Virus titer was different for each strain, TCIDso ranged from 102.7 to 1069, and LDs0 from 1015 to 1075. Five NNV strains named QN 02, QN 05, QN 07, ND 11 and KH 05 had higher virulence than the other three, the first causing 100% mortality in experimental fish 3-5 dpi. NNV KH 05 had the highest antigenic similarity, and it was inactivated completely by 0.2% formalin, 0.002 mol/L binary ethylenimine (BEI) and 0.1% beta-propiolactone. The neutralization antibody titer obtained in fish of groups immunized by BEI 0.002 M and beta-propiolactone 0.1% inactivated virus was four to eight times higher than that of the group treated with the formalin inactivated virus. The antibody titer in fish immunized with beta-propiolactone inactivated virus was more persistent. The efficacy of vaccines developed from beta-propiolactone inactivated virus and aluminium hydroxide (AH) or aluminum phosphate (AP) was observed by intramuscularly immunizing Epinephelusfuscoguttatus size 1.5 cm. Neutralizing antibodies appeared in vaccinated fish on 10th day post-immunization (dpi) at a dilution of 1:16; 1:32 and highest levels were reached on 30-45 dpi, at dilutions of 1:256 and 1:512, after treatment with AH and AP vaccine, respectively. The relative percent of survival (RPS) of vaccine at 30 dpi was highest with challenge doses 0.2-1 × 10^6.8 TCIDs0, the RPS varied from 80%-83.3% in both groups of AH and AP immunization. This result provides the basis for developing a vaccine against NNA disease.  相似文献

为研究40S核糖体蛋白S15a(ribosomal protein S15a,RPS15a)在小麦多子房性状形成过程中的功能作用,以小麦(Triticum aestivum)多子房近等基因系构建的多子房性状SSH-cDNA文库中与RPS15a基因同源的EST序列为信息探针,采用RT-PCR技术从小麦多子房株系幼穗中克隆获得了RPS15a基因的cDNA与DNA序列,分析了该基因在近等基因系间的表达差异及其在多子房株系中不同时期幼穗和同一发育时期不同组织中的表达模式.结果表明,小麦RPS15a基因的cDNA序列(GenBank登录号:HM055513)长为413 bp,编码131个氨基酸;DNA序列(GenBank登录号:HM063421)长为642 bp,含有3个外显子和2个内含子.半定量RT-PCR分析显示,该基因在多子房株系2~4mm幼穗中的表达量高于单子房株系幼穗;在多子房株系不同时期幼穗中的表达量随着幼穗的发育呈现上升趋势,8~9 mm时表达量达到最高;该基因广泛存在于幼根、茎顶端、幼叶和幼穗组织中,在茎顶端表达量高于其他组织.研究推测RPS15a基因的表达上调,可能与小麦多子房性状的发生有关.  相似文献

Finkel-Biskis-Reilly murine sarcoma virus (FBR-MuSV) ubiquitously expressed (FAU) gene is down-regulated in human prostate, breast and ovarian cancers. Moreover, its dysregulation is associated with poor prognosis in breast cancer. Sponges (Porifera) are animals without tissues which branched off first from the common ancestor of all metazoans. A large majority of genes implicated in human cancers have their homologues in the sponge genome. Our study suggests that FAU gene from the sponge Suberites domuncula reflects characteristics of the FAU gene from the metazoan ancestor, which have changed only slightly during the course of animal evolution. We found pro-apoptotic activity of sponge FAU protein. The same as its human homologue, sponge FAU increases apoptosis in human HEK293T cells. This indicates that the biological functions of FAU, usually associated with “higher” metazoans, particularly in cancer etiology, possess a biochemical background established early in metazoan evolution. The ancestor of all animals possibly possessed FAU protein with the structure and function similar to evolutionarily more recent versions of the protein, even before the appearance of true tissues and the origin of tumors and metastasis. It provides an opportunity to use pre-bilaterian animals as a simpler model for studying complex interactions in human cancerogenesis.  相似文献

RPS3在体内具有多种功能,为了进一步揭示其结构与功能的关系,探索其编码区作为系统发育标记的可行性,本研究应用生物信息学、RT-PCR和3-′RACE方法首次克隆了猪RPS3基因(GenBank登录号:DQ660373)并进行了序列分析,同时利用半定量RT-PCR方法分析了RPS3基因的时空表达规律。序列分析表明RPS3开放读码框全长为732 bp,编码243个氨基酸残基的多肽链,含有核糖体蛋白S3标签,构建的系统发育树显示聚类结果与动物学上的分类一致。半定量RT-PCR分析表明RPS3在所检测的10种组织中均有较高水平的表达,心脏中的表达量最高,肺脏中的表达量最低,其它组织间差异不大;在仔猪中表达量高,随着日龄的增加表达量不断下降,在3月龄以后的猪中表达量趋于平稳。以上结果说明RPS3基因CDS适合构建种间的系统发育树,mRNA的表达水平与细胞的生长代谢速度有关。  相似文献

用福尔马林灭活哈维氏弧菌、鳗弧菌、迟钝爱德华氏菌、大菱鲆弧菌和溶藻胶弧菌制备成五联灭活疫苗,添加黄芪多糖作为佐剂。分别用添加黄芪多糖佐剂组和无佐剂组对大菱鲆进行腹腔注射,在第7、14、21、28、35、42、56天采取血样并检测抗体效价、溶菌酶活力和SOD活力,第56天进行人工攻毒实验,测定免疫保护力。结果表明,黄芪多糖佐剂组(免疫组2)和无佐剂组(免疫组1)两组的溶菌酶活力和SOD活力都在第28天达到最高值,且黄芪多糖佐剂组显著高于无佐剂组(P<0.05)。两个免疫组抗体效价也都在第28天达到最大值,但黄芪多糖佐剂组要显著高于无佐剂组(P<0.05),其中免疫组2的哈维氏弧菌、鳗弧菌、迟钝爱德华氏菌、大菱鲆弧菌和溶藻胶弧菌的抗体效价分别为27.25、26.75、28.5、28.75、26.25,免疫组1分别为26、25.75、26.75、27.5、25。用上述5种细菌分别进行人工攻毒实验,结果表明,免疫组2的免疫保护力依次为50.0%、50.0%、71.4%、62.5%、44.4%;免疫组1的免疫保护力依次为37.5%、30.0%、42.9%、50.0%、33.3%。实验结果证明,五联疫苗对大菱鲆具有免疫作用,而且添加黄芪多糖佐剂组的免疫效果显著好于无佐剂组。  相似文献

为确定嗜水气单胞菌(Aeromonashydrophila,Ah)J-1株温敏胞外蛋白酶EprJ1的表达产物是否具有酶活性和抗原性及测定对小鼠的相对保护率,根据eprJ1基因ORF阅读框的序列设计1对引物,扩增出不含信号肽序列的EprJ1成熟蛋白结构基因(850bp),经EcoRⅠ/SalⅠ双酶切后连接pET-32a( ),转化大肠杆菌(Escherichiacoli)BL21。在30℃经IPTG诱导后,重组菌冻融液在脱脂奶平板上经28℃和24h孵育检测有透明溶蛋白圈出现,经56℃和30min处理后则无。纯化的重组蛋白免疫小鼠后,用1.0×107cfu的AhJ-1(50LD50)腹腔注射攻击小鼠,重组蛋白对小鼠的相对保护率可达60%。免疫印迹显示,AhJ-1胞外产物的抗血清可以识别该融合蛋白,但AhJ-1的热稳定胞外蛋白酶ECPase54抗血清不能识别该融合蛋白。重组蛋白分子量约53.2kD,有温敏蛋白酶活性和抗原性。  相似文献
