旨在探究宿主蛋白程序性细胞死亡因子10(programmed cell death factor 10,PDCD10)通过抑制Ⅰ型干扰素表达进而促进口蹄疫病毒(foot-and-mouth disease virus,FMDV)的复制。首先,本研究验证了过表达和沉默PDCD10对FMDV复制的影响,接着利用双荧光素酶报告系统探究PDCD10对Ⅰ型干扰素信号通路活化的影响,最后,利用实时荧光定量PCR探究PDCD10对Ⅰ型干扰素通路下游刺激基因(IFN-stimulated genes,ISGs)转录的影响。结果表明,过表达PDCD10显著促进FMDV的复制,沉默PDCD10显著抑制FMDV的复制。与对照相比,过表达PDCD10后感染仙台病毒(Sendai virus,SeV)的细胞培养液上清液显著促进FMDV复制,进一步,PDCD10显著抑制SeV诱导的IFN-β启动子以及NF-κB的激活且呈剂量依赖性,并且PDCD10负调控Ⅰ型干扰素通路信号分子转录,最后还发现PDCD10负调控Ⅰ型干扰素下游ISGs转录。本研究结果为深入探究PDCD10在抗病毒天然免疫中的作用积累了资料。  相似文献

为研究大麦籽粒支链淀粉、直链淀粉和β-葡聚糖积累特性,及淀粉各组分与β-葡聚糖的关系,以2个皮大麦、1个裸大麦和1个糯裸大麦为试验材料,测定花后7、14、21、28d淀粉各组分及β-葡聚糖含量。结果表明,甘啤6号淀粉各组分含量随灌浆推进均逐渐升高,在成熟期达到最大值;甘垦啤7号和甘垦6号均呈先升高后降低趋势,在花后21d有1个峰值。糯大麦C 2-1直链淀粉含量显著低于非糯大麦;支链淀粉含量显著高于非糯大麦,整个灌浆期呈先升高后降低趋势。β-葡聚糖含量均随灌浆时间延长逐渐升高,成熟期含量最高;整个灌浆期糯大麦C 2-1含量显著高于非糯大麦。相关分析表明,直链淀粉/支链淀粉比值与β-葡聚糖含量呈极显著负相关,可以将直链淀粉/支链淀粉比值作为高β-葡聚糖品种选育的一个指标。Logistic方程拟合发现,直链淀粉、β-葡聚糖最终积累量与积累起势与有效积累时间有关,支链淀粉最终积累量取决于最高积累速率和平均积累速率。  相似文献

旨在探究陈皮、青皮、木瓜、厚朴、红景天等5种中药材提取液以及饲喂了陈皮和青皮的猪尿液胶体金免疫层析法检测结果呈阳性的原因。建立测定猪尿及中药提取物中辛弗林的LC-MS/MS检测方法,测定陈皮、青皮的提取物和饲喂陈皮、青皮后猪尿中的辛弗林浓度。用小鼠肝微粒体孵育CGIA检测呈阳性的陈皮、青皮、木瓜和厚朴4味中药提取物。饲喂陈皮和青皮后的猪尿液检出辛弗林,浓度分别为1.36和1.65 μg·mL-1;在陈皮和青皮的提取复溶液中检出辛弗林,浓度分别为132.6和312.7 μg·mL-1。厚朴和木瓜两种中药提取物经过肝微粒体孵育后,在2~5 h逐渐由CGIA检测阳性转为阴性,陈皮和青皮孵育后,0~6 h的结果均为CGIA检测阳性;阴性中药组CGIA检测均呈阴性,而添加辛弗林的阳性对照组0~6 h CGIA检测均呈阳性。猪较大剂量使用陈皮和青皮会引起猪尿β-受体激动剂CGIA检测出现假阳性,该假阳性现象跟中药成分辛弗林有关,体外试验中辛弗林不易被小鼠肝微粒体代谢。  相似文献

Whole grain oats, as a rich source of β-glucan, have been shown beneficial to glycemic control. In the current study, the impact of native form β-glucan in oat grains (NFO-glucan) on starch digestion and postprandial glycemia was investigated. The dry-milling prepared NFO-glucan sample was enriched with native form β-glucan (15.6%), and an in vitro starch digestion assay of NFO-glucan (0.5% starch equivalent) showed a significant decrease of starch digestion rate compared to oat starch (0.5%, w/v). However, pretreatment by either β-glucanase or pepsin significantly increased the starch digestion. Consistently, an in vivo examination on the postprandial glycemia of the cooked NFO-glucan sample using a mouse model displayed a significant decrease of postprandial glycemia compared to gelatinized oat starch. Further experiment on the pasting property of NFO-glucan sample by a rapid visco-analyser demonstrated both β-glucan and protein affected its viscosity profiles. Scanning Electronic Microscope (SEM) observation revealed a network-like native structure of β-glucan that might encapsulate protein and starch to reduce the enzyme accessibility and so the digestion of starch. Novel food processing technologies to maintain the native form of β-glucan in oat grains might be a better way to modulate the postprandial glycemia of oat-based whole grain foods.  相似文献

目的:建立夹竹桃花挥发油GC-MS的色谱分离鉴定方法,分析夹竹桃花挥发油的化学成分和对豚鼠离体子宫平滑肌的作用。方法:采用挥发油提取器提取夹竹桃花挥发油,以GC-MS法进行分析鉴定,建立了豚鼠离体子宫模型。结果:分别从无酶和有酶的夹竹桃花挥发油提取物中确证了58和57个化合物,挥发油对豚鼠离体子宫平滑肌有收缩作用。结论:对夹竹桃的挥发油化学成分进行了比较分析结果表明,β-葡萄糖苷酶对夹竹桃花有较弱增香作用,也能增加豚鼠离体子宫平滑肌收缩。  相似文献

The possibility of using naked barley for food products is gaining popularity due to its dietary fibre content, especially β-glucans. The technological process (dough preparation, fermentation and baking) influences bread quality but also may contribute to degradation or preservation of valuable grain components. The aim of the study was to investigate the effects of different wholemeal barley flour share and bread production method on the quality of bread and non-starch polysaccharides content and solubility.Barley enriched bread contained more both soluble and insoluble dietary fibre and β-glucans, products of 40% barley share contained 67% more total dietary fibre and 160% more β-glucans than control. However, barley incorporation decreased the amount of soluble arabinoxylans. High barley contents contributed to the breads’ volume reduction by 14% and change in their crumb and crust colour. However, barley enriched breads gained higher ratings of taste than wheat bread. Barley sourdough fermentation improved breads’ volume, colour and sensory properties. Sourdough fermentation also resulted in higher concentration of dietary fibre, arabinoxylans and β-glucans. The beneficial effect of barley addition to wheat bread may be successfully enhanced by using barley wholemeal sourdough fermentation.  相似文献

建立了香蕉中吡虫啉和高效氯氰菊酯多残留分析方法。样品经1%乙酸乙腈提取,N-丙基乙二胺(PSA)和石墨化炭黑(GCB)净化,以甲醇和2 mmol/L乙酸铵水溶液为流动相,进行梯度洗脱分离,采用电喷雾正离子源模式 (ESI +),多反应监测(MRM)采集数据,基质匹配外标法定量。吡虫啉和高效氯氰菊酯在0.001~0.1 mg/L范围内线性关系良好,相关系数r > 0.9991,方法检出限(LOD)为1 pg,定量限(LOQ)为0.01 mg/kg。在0.01、0.05、0.10 mg/kg 3个添加水平下,吡虫啉和高效氯氰菊酯在香蕉中的回收率为84.91%~104.78%,相对标准偏差(RSD)为1.23%~9.69%。该法操作简单、灵敏度高、重现性好,适用于香蕉中吡虫啉和高效氯氰菊酯的残留量测定。  相似文献

Weaning may cause oxidative injury, immune response impairment, apoptosis and other injuries in piglets. Oxidative and endoplasmic reticulum stress (ERS) can elicit inflammatory responses, and persistent oxidative and ERS also may lead to apoptotic cascades, which is associated with the pathogenesis of multiple diseases. β-carotene, a natural carotenoid, has potential anti-inflammatory and antioxidant functions. However, the effect of β-carotene on apoptosis in weaned piglets and the detailed molecular mechanism remain unclear. In this study, we found that β-carotene decreased malondialdehyde (MDA) levels and increased the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in piglet serum. β-carotene could inhibit the mRNA levels of caspase-3 significantly, but had no significant inhibitory effect of the mRNA levels of caspase-9 and caspase-12 in the piglet jejunum. In addition, β-carotene decreased the activation of GRP78, CHOP, and JNK/p38 MAPK and the ratio of Bax/Bcl-2. Furthermore, β-carotene had a significant influence on the activation of ERS and apoptosis-related signals in TG-induced IPEC-J2. In the present study, β-carotene pre-treatment attenuated the ratio of Bax/Bcl-2 and prevented TG-induced increases in the level of PERK-CHOP and IRE1-JNK/p38 MAPK pathway activation in a dose-dependent manner. Overall, these findings indicate that β-carotene may protect weaning-induced apoptosis through inhibiting ERS.  相似文献
