Studies have shown that the three subunits of β-conglycinin are the main potential allergens of soybean sensitive patients.And β-conglycinin has adverse effects on nutrition and food processing.So solation and production of lines with lower β-conglycinin content has been the focus of recent soybean breeding projects.Soybean lines with deficiency in one or all subunits of β-conglycinin have been obtained.An effective and rapid system to identify such mutations will facilitate genetic manipulation of the β-conglycinin subunit composition.Here,two segregating F_2 populations were developed from crosses between Cgy-1/cgy-1 (CC),an α′-lacking line (Δα′),and DongNong 47 (DN47),a wild-type (Wt) Chinese soybean cultivar with normal globulin components,and Cgy-2/cgy-2 (CB),an α-lacking line (Δα),and DN47.These populations were used to estimate linkage among the cgy-1 (conferring α′-null) and cgy-2 (α-null) loci and simple sequence repeat (SSR) markers.Seven SSR markers (Sat_038,Satt243,Sat_307,Sat_109,Sat_231,Sat_108 and Sat_190) were determined to co-segregate with cgy-1,and six SSR markers (Satt650,Satt671,Sat_418,Sat_170,Satt292 and Sat_324) co-segregated with cgy-2.Linkage maps being composed of seven SSR markers and cgy-1 locus,and six SSR markers and the cgy-2 locus were then constructed.It assigned that the cgy-1 gene to chromosome 10 at a position between Sat_307 and Sat_231,and the cgy-2 gene to chromosome 20 at a position between Satt650 and Satt671.These markers should enable map-based cloning of the cgy-1 and cgy-2 genes.For different subunit-deficiency types[α′-null,α-null and (α′+α)-null types],the two sets of SSR markers could also detect of polymorphism between three normal cultivars and seven related mutant lines.The identification of these markers is great significance to the molecular marker-assisted breeding of soybean β-conglycinin subunits.  相似文献

自2015年开始,北京市大力选派机关优秀干部到京郊各村镇任“第一书记”,工作成效显著。北京市第三批“第一书记”的工作给进驻的村庄带来了许多新的变化,在驻村工作中,驻村书记重视调查研究和基层党建、加强基础设施建设、注重打造产业品牌和建设特色乡村文化。但是,工作也存在一些问题,比如过于注重经济建设,对村子队伍建设关注度不够;过于依赖派出单位,以致后期村庄的可持续发展动力不足等。因此,建议持续加大乡村治理人才的培养力度、充分发挥市场作用以及让有条件的村庄联动发展等,使“第一书记”的工作更加富有成效。  相似文献

[目的]分析牛栏江流域上游保护区水土流失治理模式,以期为流域水土流失治理提供新的思路。[方法]基于DEM数据在ArcGIS内进行子流域自动提取,结合遥感影像进行人工交互解译、修正以确定子流域;以划分的子流域为基本单元进行定性与定量赋分并计算综合得分,依据综合得分确定流域水土保持分区及其主导功能;根据确定的水土保持分区主导功能进行“四型”小流域治理模式配置。[结果]牛栏江流域上游保护区共划分9个子流域,确定4个水土保持分区;在各水土保持分区分别构建以生态安全型、生态景观型、生态经济型和生态清洁型为主的“四型”小流域治理模式。[结论]子流域可作为牛栏江流域上游保护区“四型”小流域水土流失治理模式配置的基本单元,水土保持主导功能可作为“四型”小流域模式配置的依据,提供因型施策、精准至区的水土流失科学治理模式。  相似文献

为适应精准农业的发展趋势,提高测产计量系统的精度,开发一套基于IMM UKF算法的冲量式测产系统。该系统由冲量传感器模块、北斗定位模块、DTU传输模块和上位机等组成。系统通过传感器模块进行信号的采集和处理,采用CAN通信方式将处理后的数据发送至上位机。上位机使用IMM UKF滤波算法对传感器上传的数据进行滤波,最终根据标定曲线得到谷物产量。通过车辆空载试验,采用IMM UKF算法与KF和UKF算法进行对比验证,结果表明IMM UKF算法的滤波效果更好,均方差为0.000 42 N。通过传感器标定试验,确定传感器的检测值与实际谷物重量的关系。通过大田测产试验,得出每个地块的产量分布图,试验结果表明该测产系统的平均误差达到3.911%。  相似文献

Whole-genome sequencing of “Candidatus Liberibacter asiaticus” (Las) indicated some polymorphic gene regions enabling the molecular characterization of this bacterium. Although the population diversity of Las in China has been previously studied, no reports have used a combination of a prophage region and short tandem repeat (STR) loci for phylogenetic relationship characterization. In this study, we investigated the genetic diversity and structure of 667 Las strains from nine provinces in southern China using multiple genetic loci including three type-specific prophage loci, two STR loci, and a miniature inverted-repeat transposable element (MITE) region. The results indicated that the genetic diversity varied according to the gene loci used. The prophage regions, including the MITEs, revealed significant genetic differences of Las populations in Yunnan and Guizhou from other provinces, while the STR loci only indicated a difference of Las population in Guizhou from those of other provinces. In particular, the Las population shown to be diverse in Jiangxi at the CLIBASIA_01215_STR locus was not diverse when measured using the other loci. Considering all loci, the Las populations from Yunnan and Guizhou were different, those from Guangdong and Guangxi were complex, while the Las population in Jiangxi was comparatively simple. Las populations from five neighbouring provinces, including Guangdong, Guangxi, Hunan, Fujian, and Jiangxi were grouped in a big cluster. The Las population structure changed with age according to the prophage type. This study evaluated the method used for studying the molecular diversity of Las populations and provided more detailed information on the geographical origin and epidemic trend of Chinese Las populations.  相似文献

Wheat stem rust, caused by the fungus Puccinia graminis f. sp. tritici (Pgt), is a notoriously damaging disease of wheat and barley. Pgt requires two hosts to complete its lifecycle; undergoing asexual reproduction on cereal crops and completing sexual reproduction on Berberis spp. The latter stage of its lifecycle is of particular importance in temperate regions such as western Europe, where asexual urediniospores are unable to survive cold winter weather. In the past, the crucial role of Berberis in the lifecycle of stem rust led to intensive eradication campaigns, initially carried out by farmers in the face of hostile scientific opinion. In the United Kingdom, common barberry (Berberis vulgaris) is today a relatively rare plant. Stem rust is, however, currently experiencing a resurgence; at the same time, there has been a general increase in the prevalence of barberry and an upsurge in its planting which, in the United Kingdom, is associated with attempts to encourage the endangered barberry carpet moth (Pareulype berberata). This article situates current developments within a broader chronological framework, examining changing attitudes towards barberry and rust in England in the past and the history of the plant's use and cultivation. It assesses how widespread B. vulgaris really was in the environment historically, and thus the scale of its eradication. We suggest that Berberis was never widely established as an archaeophyte in the United Kingdom. Current attempts to re-establish it are based on a misunderstanding of the plant's historical status and could potentially pose a serious threat to food security.  相似文献

借助MODIS NDVI数据和气象插值数据,采用一元线性回归、Hurst指数及相关分析等方法,从年际尺度探讨河南省21世纪以来植被覆盖时空变化规律,并分析其对气候因素的响应特征。结果表明:河南省植被覆盖区域差异较大,NDVI频度呈单峰结构;近16年来,河南省大部分地区植被覆盖呈现增加趋势,10年平均增速约为4%(P0.01),且阶段性明显;河南省植被变化的同向特征强于反向特征,植被覆盖整体上呈持续改善态势,但郑州、洛阳、焦作等市域局部地区存在持续减小现象;年平均温度的上升是平原区植被覆盖增加的主要驱动因子,但升温尤其是年平均最高温度增加抑制了山区植被生长,植被覆盖对气候因子的敏感程度地域异质性明显。  相似文献

为了探索福建省茎用芥菜1年2代繁育技术,选择6个茎用芥菜品种,分别进行冬春季正常繁育和夏秋季高山加代繁育的2代繁育试验,并对比各品种2代繁育的生育期、株高、株幅和种子产量。结果表明:所有参试茎用芥菜品种只要通过打破种子休眠、衔接好2代繁育的播种期都能完成1周年2代繁育,夏秋季高山加代繁育的全生育期比冬春季正常繁育平均缩短约50 d;冬春季繁育比夏秋季高山加代繁育株高增加31.2%~132.9%,株幅增加1.5%~21.8%,所有参试品种冬春季繁育的种子产量都极显著高于相应夏秋季高山加代繁育,种子产量增加17.8%~153.6%。  相似文献

This observational study aimed to determine MRSA prevalence using strain‐specific real‐time PCR at the pig level, stratified by age groupings, within a pig enterprise. A total of 658 samples were collected from individual pigs (n = 618) and the piggery environment (n = 40), distributed amongst five different pig age groups. Presumptive MRSA isolates were confirmed by the presence of mecA, and MALDI‐TOF was performed for species verification. All isolates were tested against 18 different antimicrobials. MRSA was isolated from 75.2% (95% CI 71.8–78.6) of samples collected from pigs, and 71% of the MRSA isolates from this source were identified as community‐associated (CA)‐MRSA ST93, while the remainder were livestock‐associated (LA)‐MRSA ST398. Amongst environmental isolates, 80% (CI 64.3–95.7) were ST93 and the remainder ST398. All MRSA isolates from pigs and the environment were susceptible to ciprofloxacin, gentamicin, linezolid, mupirocin, rifampicin, sulfamethoxazole–trimethoprim, teicoplanin and vancomycin. Phenotypic rates of resistance were penicillin (100%), clindamycin (97.6%), erythromycin (96.3%), ceftiofur (93.7%), chloramphenicol (81.2%), tetracycline (63.1%) and amoxicillin–clavulanate (63.9%). A low prevalence of resistance (9.2%) was observed against neomycin and quinupristin–dalfopristin. The probability of MRSA carriage in dry sows (42.2%) was found to be significantly lower (p < .001) when compared to other age groups: farrowing sows (76.8%, RR1.82), weaners (97.8%, RR 2.32), growers (94.2%, RR 2.23) and finishers (98.3%, RR 2.33). Amongst different production age groups, a significant difference was also found in antimicrobial resistance for amoxicillin–clavulanate, neomycin, chloramphenicol and tetracycline. Using the RT‐PCR assay adopted in this study, filtering of highly prevalent ST93 and non‐ST93 isolates was performed at high throughput and low cost. In conclusion, this study found that weaner pigs presented a higher risk for CA‐MRSA and antimicrobial resistance compared to other age groups. These findings have major implications for how investigations of MRSA outbreaks should be approached under the One‐Health context.  相似文献
