Vibrio infections are common among marine fish and lead to serious problems in the aquaculture sector. This study reports a recent occurrence of Vibrio species (spp.) isolated from cultured groupers in Peninsular Malaysia using the gyrB and pyrH genes. A total of 147 Vibrio strains were successfully isolated from 77 (64%) groupers using culture method and subjected to gyrB and pyrH sequencing for species identification and confirmation. Results showed that 89% of Vibrio strains were identified and clustered to six groups of Vibrio spp., while 11% were not clustered to any Vibrio spp. using the gyrB sequences. Meanwhile, by analysis of the pyrH sequences all the 147 Vibrio strains (100%) were successfully identified and clustered into 11 groups of Vibrio spp., including the gyrB non‐identified strains. The pyrH gene provides a better resolution for identification of Vibrio spp. compared with the gyrB gene. Thus, the pyrH gene was more suitable for a rapid determination of Vibrio spp. distribution in Peninsular Malaysia. Using the pyrH gene, our study found higher prevalence of Vibrio vulnificus (33%), V. alginolyticus (24%) and V. parahaemolyticus (22%), followed by V. rotiferianus (5%), V. harveyi (3%), V. tubiashii (2%), V. campbellii (2%), V. ponticus (1%), V. diabolicus (1%), V. owensii (1%) and others Vibrio sp. (7%). Thus, the results of this study revealed that the occurrence of pathogenic vibrios among grouper fish is still high in Malaysian aquaculture. In addition, the pyrH gene was proved as a suitable marker for rapid identification of Vibrio species compared with the gyrB gene.  相似文献

测定和分析了4种鱚属(Sillago)鱼类:斑鱚(Sillago aeolus)、亚洲鱚(S. asiatica)、多鳞鱚(S. sihama)和少鳞鱚(S. japonica)线粒体细胞色素b基因序列片段,比较了不同鱚属鱼类种间的序列差异,探讨了彼此间系统发育关系和分类地位。结果显示,4种鱼类平均核苷酸组成为T 29.9%、C 29.2%、A 21.5%、G 19.3%,种间平均净遗传距离在0.157到0.280之间。最大似然法构建的系统树显示,少鳞鱚和亚洲鱚首先聚类、再与多鳞鱚和斑鱚相聚,斑鱚是最晚分化出的鱼类。基于Cyt b基因序列核苷酸分歧速率计算得出4种鱼类发生遗传分化时间在距今785—1400年前的第三纪中新世(Miocene)。  相似文献

为了研究珠母贝属的分类地位和系统演化情况,通过聚合酶链式反应技术(PCR)扩增并直接测序出雷州半岛珠母贝属中的马氏珠母贝、大珠母贝、珠母贝、斑珠母贝和黑珠母贝的3种线粒体基因(12S r RNA、16S r RNA和COⅠ)部分序列,分析其碱基组成和种间遗传距离。同时结合Gen Bank上发表的白珠母贝序列,构建珠母贝属的分子系统树。结果显示,5种珍珠贝的3种基因部分序列碱基AT含量大于GC含量,与其他无脊椎动物线粒体DNA序列基本一致,序列都处于高度饱和的状态。系统分析显示,构建的分子系统树与传统的形态分类基本一致。在黑珠母贝和白珠母贝亲缘关系上,3种线粒体DNA片段在碱基组成、种间遗传距离和系统进化树上都显示黑珠母贝和白珠母贝非常相近,可以认为是同一种中的2个亚种。在斑珠母贝的亲缘关系上,系统分析显示斑珠母贝与黑珠母贝和白珠母贝更为接近。研究表明,大珠母贝和珠母贝在系统进化中较早的分离出来,是一个比较原始的种类。3种分子标记对马氏珠母贝亲缘关系分析上存在一定差异,根据现有的数据尚不足以得出结论,需进一步做分子系统研究并结合形态特征和解剖结构进行分析。  相似文献

Amplified fragment length polymorphism (AFLP) markers and multilocus sequence analysis (MLSA) were used to analyse 63 bacterial strains, including 30 soft‐rot‐causing bacterial strains collected from Syrian potato fields and 33 reference strains. For the MLSA, additional sequences of 41 strains of Pectobacterium and Dickeya, available from the NCBI GenBank, were included to produce a single alignment of the 104 taxa for the seven concatenated genes (acnA, gapA, proA, icd, mtlD, mdh and pgi). The results indicate the need for a revision of the previously classified strains, as some potato‐derived Pectobacterium carotovorum strains were re‐identified as P. wasabiae. The strains that were classified as P. carotovorum during the analyses demonstrated high heterogeneity and grouped into five P. carotovorum highly supported clusters (PcI to PcV). The strains represented a wide range of host plants including potatoes, cabbage, avocados, arum lilies, sugar cane and more. Host specificity was detected in PcV, in which four of the six strains were isolated from monocotyledonous plants. The PcV strains formed a clearly distinct group in all the constructed phylogenetic trees. The number of strains phylogenetically classified as subspecies ‘P. c. subsp. brasiliensis’ in PcIV dramatically increased in size as a result of the characterization of new isolates or re‐identification of previous P. carotovorum and P. atrosepticum strains. The P. carotovorum strains from Syria were grouped into PcI, PcII and PcIV. This grouping indicates a lack of correlation between the geographical origin and classification of these pathogens.  相似文献

采用微卫星DNA标记对甘肃高山细毛羊、滩羊等5个绵羊群体(125只个体)的5个微卫星座位等位基因进行检测,分析5个绵羊群体的遗传多样性和亲缘关系。结果表明:5个绵羊群体中共发现78个等位基因,其中在甘肃高山细毛羊肃南县群体中发现的等位基因数最多(57个),滩羊最少(43个)。多态信息含量、期望杂合度和观察杂合度的数据分析表明:在研究的5个绵羊群体中甘肃高山细毛羊天祝县群体和肃南县群体的遗传多样性较丰富,而滩羊的遗传多样性相对较低。DA遗传距离和DS遗传距离构建的邻接(NJ)系统发育树均表明:甘肃高山细毛羊天祝县群体和肃南县群体与藏羊的亲缘关系较近,为一类;而滩羊与小尾寒羊的遗传距离较近,为另一类。  相似文献

从80个随机扩增多态性DNA(RAPD)随机引物中筛选出22个谱带清晰且重复性好的引物用于PCR扩增,探讨夹竹桃科(Apocynaceae)10属12种植物的遗传关系。获得162个位点用于计算Nei’s遗传相似性系数,并应用NTSYS程序UPGMA法构建了系统发育树。黄蝉(Allamanda schottiiPohl.)与软枝黄蝉(A.catharticaL.)、红鸡蛋花(Plumeria rubraL.)与鸡蛋花(P.rubraL.‘Acutifolia’)之间的遗传相似性最高;在遗传相似性系数0.73处将12种植物划分为10个属,并在0.58处分为3支,但并不完全支持3个独立的亚科。RAPD分析结果与形态学的分析结果基本一致,可为夹竹桃科系统分类学提供分子生物学的证据。  相似文献

Summary Sixteen types of mandarins were screened for their leaf flavonoid patterns with the help of cellulose thin-layer chromatography. In general, Tanaka's classification of mandarins into 36 species is untenable, as was already concluded by Swingle (1948), Hodgson (1965) and Singh (1967). It is likely that a fine loose skinned mandarin was first evolved. Viral haemorrhagic septicaemia virus (VHSV) is a rhabdovirus that infects over 48 species of teleosts and is lethal in many. VHSV threatens marine and aquatic fisheries. VHSV was first discovered outside Europe in 1988 in fish from the Pacific coast of North America. In 1994, VHSV was discovered in Newfoundland. In 2003, VHSV was isolated from fish in Lake St. Clair (Michigan and Ontario). In this study, we used 46 nucleotide sequences for the glycoprotein gene from 12 studies and 150 nucleotide sequences for the nucleoprotein gene from nine studies. We combined phylogenetics and a geographic information system to visualize the transmission paths of VHSV lineages. We also reconstructed the spread of VHSV lineages through optimization of geographic data for viral isolates on phylogenetic trees. We demonstrate that VHSV was transmitted from the North Atlantic Ocean and/or Baltic Sea to the Atlantic coast of North America and Japan in independent events. From the Atlantic coast, the virus was transmitted independently to the Laurentian Great Lakes and the Pacific coast of Canada and the contiguous United States. From the Pacific Northwest, the virus was transmitted to Asia and Alaska in independent events. These results clarify the debate ongoing in the literature on the geographic spread of VHSV.  相似文献

 Wheat blue dwarf(WBD) is a disease caused by phytoplasma and only reported from China. A fragment about 1.3 kb in protein translocation gene, secY was amplified by PCR from the total DNA of di-seased wheat sample with primer pair secYF/secYR, which was designed based on secY gene sequence of known 16SrI group members. Nucleotide acid sequence analysis of amplified fragment indicated that the length was 1 240 bp. A phylogenetic tree based on secY gene sequences was constructed and showed that wheat blue dwarf phytoplasma was clustered into the Candidatus Phytoplasma asteris, subgroup 16SrI-C. Wheat blue dwarf phytoplasma showed high homology with clover phyllody phytoplasma strains based on sequence comparison and phylogenetic analysis.  相似文献
