The quality and safety of agricultural products is the technique focus of the strategy of rural vitaliza⁃tion. Carbamate pesticide residues are seriously threatening the safety of agricultural product and human life and health. Nowadays, the common detection methods are instrument-based for carbamate pesticide residue, which dis⁃ satisfy the request of an on-site rapid, sensitive and simultaneous detection for multiple carbamate pesticide resi⁃ dues. Based on the home-made monoclonal antibodies against carbofuran and carbaryl respectivity, a microarray for rapid and simultaneous detection of multiple carbamate pesticide residues was developed within 30 min. Under the optimized conditions, the limits of detection(LODs)were found to be 0.11 and 0.09 ng/g, while the linear ranges were 0.33-3000 and 0.18-1500 ng/g for carbofuran and carbaryl respectively. By using spiked peanut samples, it recorded recoveries of 88.4%-98.3%, 90.1%-118.8% for carbofuran and carbaryl respectively. The RSD of intraarray were found at 9.0%, 3.5%, while the RSD of inter-array were recorded at 12.3%, 16.3% respectively. The de⁃
tection results of the microarray were highly consistent with that of HPLC-MS/MS. This proposed microarray provid⁃ ed an on-site high-sensitivity, simultaneous and rapid detection of carbofuran and carbaryl, and could be widely used in the fields of agricultural product quality safety, food safety and environmental monitoring. 相似文献

油菜黑胫病是由Leptosphaeria biglobosa引起的一种真菌病害。这种病害在我国油菜产区广泛发生,并造成一定的经济损失。为通过获取突变体来研究L. biglobosa的生态适应性机制及致病机制,本文优化了影响农杆菌介导转化(ATMT)油菜黑胫病菌L. biglobosa菌株Lb731的因素,评估转化子质量,并筛选相关突变体。结果明确了农杆菌介导转化菌株Lb731的最佳因素:潮霉素B浓度为50 μg/mL,转化受体(分生孢子)培养时间为15 d (20℃),浓度为2 × 107~8孢子/mL,农杆菌-受体共培养温度为25℃,共培养时间为72 h。在最适条件下的转化效率达到80个转化子/百万分生孢子。T-DNA插入基因组的频率为100%,单拷贝插入频率为72.7%,转化子抗潮霉素性状能稳定遗传。从2136个转化子中获得了11个菌丝生长减缓突变体,7个色素产生缺陷突变体和14个分生孢子产生缺陷突变体,并从这些突变体中鉴定出7个致病力丧失突变体。采用hiTAIL-PCR技术,从3个突变体中获得了T-DNA插入位点侧翼序列。上述结果为深入研究L. biglobosa的生态适应性机制及致病机制提供了材料和线索。 相似文献

以重庆市江津区为研究区,采集2019年生产的63份大米籽粒和64份玉米籽粒,用电感耦合等离子质谱仪(ICP–MS)和原子荧光光谱仪(AFS)测定大米和玉米籽粒中砷(As)、钡(Ba)、镉(Cd)、铜(Cu)、铁(Fe)、锰(Mn)、钼(Mo)、镍(Ni)和锌(Zn)的质量分数,并利用环境健康风险模型评价江津区大米和玉米中重金属的健康风险。结果表明:江津区大米和玉米籽粒中As最大质量分数分别为176.0、162.8 μg/kg,低于国家食品卫生As标准(<200 μg/kg);江津区3份大米和6份玉米样品中Cd质量分数超过国家食品卫生Cd标准(大米Cd质量分数<200 μg/kg、玉米Cd质量分数<100 μg/kg);大米籽粒中As和Mo质量分数均值显著高于玉米籽粒的(P<0.05),Cd质量分数均值略高于玉米籽粒的;大米籽粒中As、Cu、Mn和Zn的单因子污染指数均小于1,属于清洁水平,3份大米籽粒样品中Cd属于轻度污染;大米和玉米籽粒重金属综合污染指数分别为1.09和1.57,属于轻度污染,江津区玉米籽粒中重金属污染程度大于大米籽粒;大米籽粒中重金属As和Cd的致癌风险分别占健康总风险的82.7%和16.8%,表明江津区大米籽粒中As含量虽低于国家食品卫生标准安全值,但其健康风险较高,其次为Cd。可见,重庆地区需重点监控农作物中As和Cd的健康风险。 相似文献

在丛生竹林下对不同基质配方和硒浓度菌棒进行埋棒栽培以及覆土方式栽培试验,结果表明,菌棒不同基质配方对竹荪产量及其营养成分有显著影响,而且三列浅沟形和双列龟背形2种覆土方式有利于提高竹荪产量。改良基质配方菌棒竹荪产量比常规配方提高约50%;在添加外源硒质量分数为0~2.0 mg/kg的浓度范围内,竹荪子实体产量先随着硒浓度的增加而逐渐增加后再有所降低,基质中添加1.5 mg/kg硒肥比不添加产量提高了195.30%。基质中添加硒质量分数为1.0~2.0 mg/kg的硒肥可以较显著地提高竹荪花的硒含量,其干物质中硒含量平均值从约2.50 mg/kg提高到8.05~13.30 mg/kg,外源硒肥利用率达到9.70%~15.36%。菌棒不同基质配方对竹荪子实体的粗蛋白及粗多糖含量有明显影响,改良配方竹荪蛋与竹荪花的粗蛋白含量是常规配方的1.21和1.29倍,其粗多糖含量是常规配方的4.81和1.35倍;基质中添加硒肥与不添加硒肥相比,竹荪蛋与竹荪花中粗蛋白含量分别增加了40.90%和14.30%。在硒质量分数为1.0 mg/kg的竹荪菌棒林下覆土栽培试验中,三列浅沟形覆土方式单位面积鲜竹荪蛋产量最高,为10.27 kg/m2;双列龟背形覆土方式单个菌棒的鲜竹荪蛋产量最高,达1.40 kg。三列浅沟形和双列龟背形2种覆土方式基质生物转化率分别为93.00%和94.14%,达到了较高水平。 相似文献

以福建特色水果‘油㮈’为试材,在等肥料成本条件下,进行水肥一体化施肥和常规施肥下‘油㮈’树枝梢生长长度和粗度、果实品质、产量表现及施肥成本的比较研究,以期为山地水肥一体化施肥提供参考。结果表明,水肥一体化施肥可在一定程度上促进当年枝梢长度和粗度的增长,提高果实可溶性糖、可溶性固形物及Vc含量,降低可滴定酸含量,提高果实平均单果重、可食率、产量。等价施肥条件下,水肥一体化施肥节省人工成本1104.0元/hm2。建议结合降水量等气候条件及山地土壤特点、生态环境等对水肥一体化施肥技术进行优化。 相似文献

This study used two different collectors made of polypropylene (PP) rope and polyethylene (PE) fishing net to determine Mediterranean mussel's settlement, growth, condition index (CI), meat yield (MY) and shell thickness index (STI) during the period spent between June 2017 and November 2018. With this regard, chlorophyll a, water temperature, salinity, pH and particulate matter were defined through water samples that were collected on monthly basis triplicate. The mean chlorophyll a level was recorded as 0.32 ± 0.31 μg/L, while water temperature was 19.73 ± 5.14°C, salinity was 35 ± 2.070‰, pH was 8.12 ± 0.04, and TPM was 14.91 ± 10.48 mg/L. As a result of the cultivation period of 8 months spent following the first intense grip, the length of the mussels is measured as 31.79 ± 6.20 mm. Based on the Pearson correlation analysis results, the most important environmental parameter affecting Mediterranean mussels growth in length on PP rope collectors is the temperature (p < .05). The STI and CI were determined to be related with environmental temperature parameters, while no determinations could be obtained concerning the correlation of MY with environmental parameters. Assessment of Mediterranean mussels’ growth rate in PP rope collectors proved significant differences (p < .05) between March and November 2018. As no Mediterranean mussels are observed on PE fishing nets during the samplings performed throughout the study period, it is determined that PE fishing nets are not suitable collector types for Mediterranean mussel cultivation. 相似文献

The western tarnished plant bug, Lygus hesperus Knight, overwinters as a diapausing adult in response to short day lengths. Once environmental conditions are favorable, the bugs revert to an active reproductive state. To determine the impact on life-history traits of diverting resources toward diapause rather than oogenesis during early adulthood, diapausing and nondiapausing L. hesperus females were reared from the same cohorts. Body mass, ovarian maturation, ovipositional activity, and survivorship were monitored starting either at the time of release from diapause-inducing conditions or at adult eclosion for diapausers and nondiapausers, respectively. Females that had gone through 2 wk of diapause were larger and able to mobilize the resources necessary for oogenesis faster than nondiapausers, initiating oogenesis and ovipositing sooner and at a faster initial rate. However, lifetime egg production and average daily rates were similar for both groups. Postdiapausers lived longer than nondiapausers by an average of 19 d, which is five more than the 2-wk period when they were reproductively senescent. Overall, the results indicate that short-term diapause does not have a negative impact on life history. Furthermore, the extra endogenous resources stored during diapause may be able to enhance the alacrity with which the female can take advantage of improved environmental conditions and may prolong life by shielding the females against environmental stressors such as temperature extremes, oxidative agents, or food deficits. 相似文献
