Influence of death struggle on the structural changes in chub mackerel muscle during chilled storage

ABSTRACT: Chub mackerel (34–35 cm, approximately 500 g), which were caught by fishing with a rod and line at the Bungo Channel, Oita prefecture, were rested overnight in a fish preserve and either killed by decapitation (control group) or allowed to struggle in air for 30 min (struggled group). Muscle samples were excised every 4 h, and measurements on breaking strength and histological observations were done for both groups. The breaking strength of muscle in the control group was significantly higher than that in the struggled group, whereby a decrease in breaking strength was delayed for 12 h compared to the struggled group. Light microscopy showed space extension among muscle cells in association with a decrease in breaking strength. Especially in the struggled group, the extended area was larger and the difference in area was significant at the time when breaking strength showed a significant difference. Using electron microscopy, the extended area showed cut and/or disappeared collagen fibrils. From these results, it was demonstrated that struggling to death promoted the degradation of collagen fibrils and the weakening of connective tissue and, resultantly, led to the faster softening of muscle of chub mackerel. 相似文献

ABSTRACT: Until now, six crustacean hyperglycemic hormones (CHH) designated Pej-SGP-I, -II, -III, -V, -VI and -VII have been characterized in the kuruma prawn Penaeus japonicus . All CHH consist of 72 amino acid residues and have an amidated carboxyl (C)-terminus. In the present study, we expressed Pej-SGP-III in methylotrophic yeast Pichia pastoris in order to obtain a large quantity of recombinant CHH possessing biological activity. A cDNA encoding Pej-SGP-III that had been previously cloned was processed by polymerase chain reaction (PCR) and the resulting product was ligated into an expression vector. Pichia pastoris was transformed with this vector after which a recombinant Pej-SGP-III was expressed having an additional amino acid residue (glycine) at the C-terminus (rPej-SGP-III-Gly), a form considered to be a putative precursor of this hormone. rPej-SGP-III-Gly secreted into the culture medium was purified by reversed-phase HPLC, and amidated using a peptidylglycine alpha-amidating enzyme. The amidated rPej-SGP-III (rPej-SGP-III) showed hyperglycemic activity in in vivo bioassay almost comparable to that of the natural Pej-SGP-III. rPej-SGP-III thus obtained will be a useful tool not only for its physiological study but also for the determination of its 3-D structure. 相似文献

The effect of different periods of starvation (0, 2, 4 and 8 days) followed by re-feeding on growth, feed utilization, oxygen consumption and some immune indexes [reactive oxygen intermediates (ROIs), activities of superoxide dismutase (SOD) and catalase (CAT)] was evaluated over an 18-day experimental period in shrimp Macrobrachium nipponense (De Haan) that had an initial body weight of 0.52 g. As a result of compensatory growth, indicated by an increase in specific growth rate (SGR), feeding rate (FR) and feed conversion efficiency (FCE) after re-feeding, final body weight of shrimp starved for 2 days (0.63 g) and 4 days (0.65 g) did not differ ( P >0.05) from the control group (0.64 g), with feed withholding for 8 days presenting a significant lower value (0.63 g). Oxygen consumption rate (OCR) decreased during the starvation period in all groups, followed by a gradual increase to a similar ( P >0.05) value than found in the control group (0.47 mg kg−1 h−1 ) at the end of the experiment. Although ROIs and the activity of SOD and CAT fluctuated during starvation in the feed-deprived groups, values at the termination of the experiment were comparable ( P >0.05) to those found for the control group. 相似文献

The yellow European eel (Anguilla anguilla L.) may adopt a sedentary lifestyle in inland freshwaters
Abstract – We analysed the movements of the growing yellow phase using a long-term mark–recapture programme on European eels in a small catchment (the Frémur, France). The results showed that of the yellow eels (>200 mm) recaptured, more than 90% were recaptured at the original marking site over a long period before the silvering metamorphosis and downstream migration. We conclude that yellow European eels >200 mm may adopt a sedentary lifestyle in freshwater area, especially in small catchment. 相似文献

Long‐term cryopreservation of the giant freshwater prawn, Macrobrachium rosenbergii, spermatophores using glycerol (Gly) and ethylene glycol (EG) as cryoprotective agents (CPAs) was studied. The tolerance of sperm to cryopreservation was evaluated on the basis of sperm survival and fertilizing ability. The survival of the sperm was determined by trypan blue staining, while the fertilizing ability was assessed from artificial insemination of the cryopreserved spermatophores. The rates of embryo survival on day 5 after spawning and of spermatophores capable of producing embryos survived to hatching were determined. Storage of spermatophores at ?20°C without CPA for a short period of up of 1–5 days decreased the sperm survival significantly and did not preserve fertilizing ability. Preservation at ?20°C in the presence of 10% or 20% Gly or of 10% or 20% EG offered a simple and efficient short‐term storage up to 10 days. For a long‐term storage, cryopreservation in the presence of 20% EG at ?196°C was more efficient than at ?20°C. High sperm survival rates and high fertilizing ability were recorded from those cryopreserved at ?196°C for up to 150 days. High sperm survival rates with moderate levels of fertilizing ability were obtained from those cryopreserved at ?20°C for not more than 30 days. The results indicate that preservation at ?196°C with 20% EG is a suitable procedure for long‐term storage of the giant freshwater prawn spermatophores. 相似文献

ABSTRACT Recent developments in prawn production technologies (i.e., added substrate, increased stocking densities, size grading, and increased feed rates) have increased production rates from 900-1,000 kg/ha to over 2,500 kg/ha. While prawn can receive substantial nutritional benefit from natural foods at the lower biomass densities, at higher production rates prawn are likely to be more dependent on prepared diets. To ensure that maximum production is being achieved by these new production technologies, production rates must not be nutritionally constrained. This study was conducted to compare the current recommended technology of phase feeding of different quality feedstuffs to prawns of different sizes with the feeding of a high-quality penaeid diet throughout the production season. Two treatments were evaluated: Treatment 1 was phase feeding (current technology-control) where prawns were fed unpelleted distillers' grains with solubles (DDGS) for the first four weeks, then a 28%-protein prawn diet for weeks 5-12, and finally a 40%-protein penaeid diet for weeks 13-16. In Treatment 2, prawn were fed the 40%-protein penaeid diet throughout the entire production period. Feeding rates in both treatments were based upon a feeding table. Three 0.04 ha ponds were used for each treatment. All ponds were stocked at 59,280 juveniles/ha and were provided with artificial substrate in the form of a polyethylene “safety fence” oriented vertically to increase available surface area by 50%. After 106 culture days, no significant differences (P >0.05) were observed between treatments in terms of yield, average individual weight, food conversion ratio (FCR), or survival, which averaged 2,575 kg/ha, 46 g, 2.3, and 94%, respectively, overall. Due to the higher cost of the penaeid diet (US$0.84/kg), feeding costs for the penaeid diet treatment (Treatment 2) were 38% higher than those for Treatment 1. No benefit to using higher protein diets during the first 12 weeks of prawn pond production was observed. 相似文献
