利用高脂饲料诱导建立巴马小型猪代谢疾病模型并分析肝脏转录组变化。选取1月龄巴马小型猪24头,随机分为对照组与高脂诱导组,对照组饲喂普通饲料,高脂诱导组饲喂含2.0%胆固醇饲料,定时监测体重、血常规及生化指标,6个月后试验结束,作心脏冠状动脉造影、器官组织学检测及肝脏转录组分析。结果表明,高脂诱导组个体饲喂4个月体重显著高于对照组,诱导期后血液生化指标中甘油三酯显著高于对照组,饲喂6个月时冠状动脉造影及器官组织切片未见显著病变,肝脏转录组分析显示高脂诱导组与对照组相比有2 468个差异表达基因,主要与代谢途径相关。为期6个月的高脂饲喂未致巴马小型猪明显代谢性疾病及心血管病变,但血液甘油三酯水平显著升高,显著影响肝脏代谢途径相关基因表达。文章系统探讨高脂诱导模型,结合血液生化、组织学、冠脉造影及肝脏转录组分析,进一步获得代谢通路相关潜在标记基因,为建立理想小型猪代谢疾病模型提供参考。  相似文献

The atherosclerosis (AS) model was prepared in Guangxi Bama mini-pig,and the atherosclerosis index (AI) was preliminarily identified in occurrence of AS to provide the basis for the preparation of related models.20 Guangxi Bama mini-pigs were randomly divided into control group and experimental group which were fed with high fat and cholesterol diet to prepare the AS model.Blood biochemical indexes were detected in the process of model preparation and the correlation between AI and the results of vascular slices was analyzed to initially draw up AI of AS.The results of vascular slices showed that the incidence rates of AS of Guangxi Bama mini-pigs in experimental group and control group were 20% and 0,respectively.The association analysis between the results of vascular slices and AI in the pathogenic Guangxi Bama mini-pigs preliminarily suggested that AI was above 3.8 and lasted for more than 3 months during the attack.  相似文献

本试验旨在研究饲粮添加脯氨酸对妊娠环江香猪体成分的影响。选取32头妊娠15 d的环江香猪,随机分为2组,每组8个重复,每个重复2头猪。2组试验猪分别饲喂在基础饲粮中添加0.77%L-丙氨酸(等氮对照组)和1.00%L-脯氨酸(脯氨酸组)的饲粮。分别于妊娠第45和70天时,每个重复选择1头母猪屠宰,测定其体成分。结果表明:与等氮对照组相比,妊娠第45天时脯氨酸组环江香猪的脂肪率显著降低(P0.05),皮肤率呈增加趋势(0.05≤P0.10);妊娠第70天时脯氨酸组的脂肪率显著增加(P0.05),皮肤率呈降低趋势(0.05≤P0.10),背最长肌中脯氨酸的含量显著增加(P0.05),精氨酸和丙氨酸的含量呈增加趋势(0.05≤P0.10)。与妊娠第45天时相比,妊娠第70天时脯氨酸组的脂肪率显著增加(P0.05),瘦肉率与皮肤率显著降低(P0.05)。由此可见,饲粮添加脯氨酸可增加妊娠环江香猪机体脂肪和精氨酸的沉积量,妊娠中期的机体脂肪沉积量比妊娠前期有所增加,从而有利于胎儿的生长发育。  相似文献

从广西巴马小型猪的卵巢中提取总RNA,并以其为模板,应用逆转录一聚合酶链式反应(RT—PCR),在合成的特异性引物引导下,扩增得到巴马小型猪卵泡抑素(Follistatin) cDNA的全序列,长度为1035bp。该扩增片段经纯化后,连接到pMD18-T载体上扩增。经序列分析结果表明,本研究中克隆的猪卵泡抑素cDNA序列与GeneBank中已报道的家猪卵泡抑素同源性高达99.4%,与奶牛、家鼠、挪威鼠和马等其他物种的卵泡抑素cDNA序列间同源性也大于89%。  相似文献

This study was designed to compare the blood physiological and biochemical parameters between plateau Juema minipigs and plain Bama minipigs,and provide a theoretical basis for Juema minipigs served as experimental animals in the future.The blood samples of Juema minipigs were taken in Gannan,and the blood samples of the Bama minipigs were taken in Xi'an,24 blood physiological and biochemical parameters were tested using an automatic analyzer of blood cells and biochemistry.Between the plateau Juema minipigs and Bama minipigs,there were significant differences in PLT,RDW-CV and RDW-SD (P< 0.05) and extremely significant differences in RBC,HGB,HCT,MCH,MCMH,GGT,GLU and CRE (P< 0.01).The plateau Juema minipig had an altitude acclimatization,so it was better than Bama minipig in plateau medical science.  相似文献

利用微量液体组织块贴壁法原代培养分离新生广西巴马小型猪肾成纤维细胞,进行常规传代培养、冷冻和复苏;通过接触抑制和解除抑制研究该类细胞的增殖潜力,免疫荧光进行鉴定;并通过脂质体介导对分离到的新生广西巴马小型猪肾成纤维细胞分别进行EGFP-N1和DsRed-N1基因转染。结果成功分离到新生广西巴马小型猪肾成纤维细胞,体外可以大量传代和冷冻,目前已经传至50代以上,生长良好;冷冻复苏和接触抑制解除后仍可以正常培养和传代;细胞免疫荧光检测,波形蛋白表达阳性,角形蛋白表达阴性;转染48h后荧光显微镜下可以观察到绿色荧光和红色荧光,表明瞬时转染新生广西巴马小型猪肾成纤维细胞可以获得成功,其中转染DsRed-N1基因的新生广西巴马小型猪肾成纤维细胞已经传至60代以上。结果表明,本试验成功建立了新生广西巴马小型猪肾成纤维细胞的体外培养体系,在体外能稳定培养和传代,并可以成功获得转基因的新生广西巴马小型猪肾成纤维细胞。  相似文献

试验主要对广西巴马香猪耳皮成纤维细胞的分离和培养进行了研究.利用组织块法能够成功分离培养广西巴马香猪耳皮成纤维细胞,其大小范围在1~3 mm3,去除表皮有利于组织块贴壁和细胞的游出;而采用0.25%胰蛋白酶消化液4℃冷处理过夜,不仅有利于去表皮操作,减少取材处理时间,而且操作时间自由度大.在酶消化法中采用胰蛋白酶消化30 min,胶原酶继续消化30~40 min,能够成功分离培养广西巴马香猪耳皮成纤维细胞.广西巴马香猪皮肤成纤维细胞要经历滞留期、对数期、平台期,分别为0~2 d、2~8 d、8~12 d.  相似文献

The introduction of Bama minipigs from Guangxi Bama Yao autonomous county for breeding, we observed the change of adaptability and the blood physiological and biochemistry indicators in the Northeast environment.After two years observation and inspection, we confirmed that the introduced Bama minipigs had good adaptability, normal reproductive performance, sexual maturity of 2 to 3 months of age, pregnancy period of 114 to 120 days, the number of piglets of 3 to 7 primiparous head, and the number of multiparity of 5 to 10 piglets head.The numbers of piglets were significantly related to the weights of sow (R2=0.96).The values of WBC, HGB, AST, Urea, Cr, CK, LDH, GLU, T-Ch, TG, CHS, Na, Cl, AMS and CO2CP of Liaoning minipigs were lower than the data of original pig farm (P<0.05), other values were not significantly different (P>0.05).The results provided a reference for the test of Bama minipigs in Liaoning area.  相似文献
