3种水质调控方式下参池沉积物酶活性的比较研究   总被引:1,自引:0,他引:1  
2015年10月至2016年9月对自然纳潮、微孔曝气、养水机3种水质调控方式下海参池塘沉积物中淀粉酶、蛋白酶、碱性磷酸酶、脱氢酶的活性进行了跟踪比较研究。研究结果显示,3种水质调控方式下池塘沉积物中的淀粉酶活性年变化为0.126~0.880 mg/g,年均值(0.410±0.180) mg/g,蛋白酶活性年变化为0.024~0.472 mg/g,年均值(0.190±0.103) mg/g,碱性磷酸酶活性年变化为0.068~1.042 mg/g,年均值(0.340±0.196) mg/g,脱氢酶活性年变化为12.092~52.794 mL/g,年均值(26.980±8.295) mL/g。3种水质调控方式下池塘沉积物中蛋白酶、碱性磷酸酶及脱氢酶活性均在自然纳潮池塘中均值最高,变化幅度最大;淀粉酶活性均值则在自然纳潮池塘中最低,养水机池塘最高,这与养水机池塘有机质最低,自然纳潮池塘最高,养水机池塘沉积物的细菌多样性最高,真菌数量最多有关。表明养水机能够快速去除沉积物中氮、磷有机化合物,有利于池塘的正常物质循环。本研究从沉积物酶活性的角度,探讨了养水机的作用效果与其他两种水质调控方式产生差异的机理。  相似文献

本研究采用响应面法(RSM)对南极海洋芽孢杆菌(Bacillus sp.)N11-8液体发酵产蛋白酶PBN11-8的发酵条件进行快速优化。通过单因素实验初步确定南极海洋芽孢杆菌N11-8产蛋白酶的最佳碳源和氮源分别为可溶性淀粉和蛋白胨;并通过Plackett-Burman(PB)设计对影响其产酶相关因素进行评估,筛选出具有显著效应的3个因素:温度、氮源和碳源;以最陡爬坡实验逼近至上述因子最大响应区域,进而采用RSM法对其最佳水平范围进行研究,确定最优发酵条件为:可溶性淀粉3 g/L,蛋白胨13.1 g/L,酵母浸粉2.9 g/L,NaCl 5 g/L,KH2PO4 1 g/L,FeCl3·6H2O 2 mmol/L,初始pH值为7.0,温度为34.0℃,转速为200 r/min,装液量为50 ml/250 ml,接种量为4%,培养时间为60 h。最终优化后的酶活达到90.5 U/ml,比初始酶活提高了23.6%。  相似文献

The transformation of natural landscapes into impervious built-up surfaces through urbanization is known to significantly interfere with the ecological integrity of urban landscapes and accelerate climate change and associated impacts. Although urban reforestation is widely recognised as an ideal mitigation practice against these impacts, it often has to compete with other lucrative land uses within an urban area. The often limited urban space provided for reforestation therefore necessitates the optimization of the ecological benefits, which demands spatially explicit information. The recent proliferation of tree stands structural complexity (SSC) and topographic data offer great potential for determining the ecological performance of reforested areas across an urban landscape. This study explores the potential of using topographic datasets to predict SSC in a reforested urban landscape and ranks the value of these topographic variables in determining SSC. Tree structural data from a reforested urban area was collected and fed into a tree stand structural complexity index, which was used to indicate ecological performance. Topographic variables (Topographic Wetness Index, slope, Area Solar Radiation and elevation)- were derived from a Digital Elevation Model (DEM) and used to predict SSC using the Partial Least Squares (PLS) regression technique. Results show that SSC varied significantly between the topographic variables. Results also show that the topographic variables could be used to reliably predict SSC. As expected, the Topographic Wetness Index and slope were the most important topographic determinants of SSC while elevation was the least valuable. These results provide valuable spatially explicit information about the ecological performance of the reforested areas within an urban landscape. Specifically, the study demonstrates the value of topographic data as aids to urban reforestation planning.  相似文献

Aquaponics is generally regarded as a sustainable practice, but its environmental burdens were not yet deeply investigated. In this study, Life Cycle Assessment (LCA) was used to assess the environmental impacts of two hypothetical coupled aquaponics systems (CAPS): Raft System (RAFT) and Media-Filled Beds System (MFBS). Rainbow trout (Oncorhynchus mykiss) and lettuce (Lactuca sativa) were considered as cultivated species in both systems. The Simapro© software V.8.0 was used for calculation. The comparison between the two virtual systems indicated the floating technique as the less impacting one. Even though energy consumption appears to be higher in the floating system, LCA results were markedly influenced by the extensive use of inert materials in MFSB. In both systems, contribution analyses underlined that the main environmental impacts are related to infrastructures, electricity and fish feed. The LCA analyses carried out in this study highlights that the choice of less impacting materials, and the optimization of management practices, should be taken as priorities in order to reduce environmental impacts of this activity.  相似文献

The purpose of this study was to investigate the effects of inbreeding on the immune responses and antioxidant status of Portunus trituberculatus juveniles. Results showed that inbreeding affected the total haemocyte count, and phagocytic, pro‐phenoloxidase (propo), phenoloxidase and antibacterial activities decreased after the seventh generation. Antioxidant status showed a similar pattern: total antioxidant capacity, superoxide dismutase (SOD) activity and GSH/GSSG in the cell‐free haemolymph and hepatopancreas decreased, while catalase activity in the cell‐free haemolymph increased (P < 0.05). There were no significant differences in α2‐macroglobulin and bacteriolytic activities in the cell‐free haemolymph and glutathione peroxidase activity in the cell‐free haemolymph and hepatopancreas among nine inbreeding generations. Gene expression levels of proPO and crustin in haemocytes and SOD in haemocytes and hepatopancreas also decreased significantly as the inbreeding generations increased. The results suggest that a high level of inbreeding could severely reduce the physiological health of P. trituberculatus. Our obtained data would be particularly useful for P. trituberculatus breeding programmes.  相似文献

以抗病家系与易感家系半滑舌鳎为材料,进行哈维弧菌感染实验,并对易感家系感染前(CsSU)、易感家系感染后(CsSC)、抗病家系感染前(CsRU)、抗病家系感染后(CsRC)4组进行转录组测序,根据RNA-seq数据挖掘半滑舌鳎长链非编码RNA信息;通过生物信息学分析,筛选出与抗哈维弧菌病相关的差异长链非编码RNA(long non-coding RNA, lncRNA)。结果显示,共识别出4 584个lncRNA座位,包含5 714个转录本;其基本特征与编码基因的比较分析,lncRNA的GC含量低于编码基因,单外显子基因数多于编码基因,转录本的平均长度长于编码基因,基因表达量低于编码基因。对4组样品进行两两比较(CsRU vs CsSU、CsRC vs CsSC、CsRC vs CsRU、CsSC vs CsSU)分别筛选出818、813、261、140个差异表达lncRNA,其中CsRU与CsSU之间、CsRC与CsSC之间lncRNA数目差异最多,通过聚类分析确定了各实验组的表达模式之间的联系,CsRU与CsSU之间的表达模式最为相近。通过共表达分析,预测出lncRNA和274个编码基因可能存在14 539种相互关系,并进行了功能注释,进而筛选出7个关键lncRNA。qRT-PCR结果显示,差异表达lncRNAs的表达模式和转录组数据得到的基本一致。研究结果为揭示lncRNA在半滑舌鳎抗哈维弧菌免疫调控反应中的作用提供重要的参考数据。  相似文献

Oriental river prawn (Macrobrachium nipponense) has been widely cultured in Asian countries. However, its nutritional studies are very limited. In the present 8‐week study, we investigated the effects of dietary protein to energy ratio (P/E ratio) on the growth, feed utilization and body composition in juvenile M. nipponense (initial weight 0.302 ±0.03 g). Two‐factor experiment was designed and nine semi‐purified diets were formulated to contain three lipid levels (20, 80 and 140 g kg?1) and three protein levels (330, 380 and 430 g kg?1), producing P/E ratios from 16.5 to 23.4 mg KJ?1 protein. The results indicated that the growth, survival rate and protein efficiency were dose dependently improved by the increased dietary lipid, but not dietary protein content. Increased dietary lipid content and/or protein content increased lipid accumulation in whole body, hepatopancreas and muscle, but did not change the feed intake and hepatopancreas weight. In conclusion, our present study indicated that M. nipponense is a species with relatively high‐energy requirement. It could utilize dietary lipid content up to 140 g kg?1, while the dietary protein with more than 330 g kg?1 would not promote growth and protein efficiency. Taken together, 330 g kg?1 dietary protein and 140 g kg?1 dietary lipid level with P/E ratio 16.49 could be optimum for M. nipponense.  相似文献

本研究以十足目虹彩病毒(Decapod iridescent virus 1, DIV1)主要衣壳蛋白基因为靶序列设计引物,建立了DIV1的环介导等温扩增(Loop-mediated isothermal amplification, LAMP)检测方法,并以pMD18-DIV1质粒标准品为模板对该方法的检测灵敏度、检测特异性等进行了评估。结果显示,此方法最适反应温度为64.4℃,优化后的25 μl反应体系中包含2.5 μl 10×Isothermal amplification buffer、4.0 mmol/L Mg2+、1.2 mmol/L dNTPs、6.4 U Bst 2.0 WarmStart® DNA聚合酶、0.8 μmol/L EvaGreen®和4.4 μl ddH2O。该方法检测灵敏度下限为3.54×102拷贝/反应;与虾肝肠胞虫(EHP)、致急性肝胰腺坏死病副溶血弧菌(VpAHPND)、对虾偷死野田村病毒(CMNV)、传染性皮下及造血组织坏死病病毒(IHHNV)、白斑综合征病毒(WSSV)、桃拉综合征病毒(TSV)和黄头病毒(YHV)等主要虾类病原没有交叉反应;具有较好的重复性和稳定性。以GeneFinder®替换EvaGreen®并将其预置于反应管内,结合上述扩增方法可实现对DIV1的现场快速高灵敏检测。本研究建立的DIV1-LAMP实时荧光定量和现场检测方法具有灵敏、特异和快速等特点,为近几年新发虾类病原DIV1的定性、定量以及现场快速检测提供了新的技术选择,有利于对虾养殖业中开展DIV1的监测、预警和防控。  相似文献

2018年4月10日、6月11日、8月14日在桓仁水库的上游(江南九队)、中游(砬砬岗子)和下游(泗河大地)3个站位用500 mL具塞磨口瓶盛装采水器采集的水库水样,进行限制性营养盐原位试验,同时测定水中溶解氧和初级生产力,并检测分析水体理化指标,以查明辽宁桓仁水库氮磷分布、营养盐限制和初级生产力状况,保护桓仁水库生态系统的结构和功能完整性。试验结果显示,营养盐加富试验中,0.5 mg/mL氮+0.3 mg/mL磷的添加组可显著提高水体中的溶解氧水平(P<0.05);上游和中游氮磷比为2∶1、下游氮磷比为4∶1可显著提高水体中的溶解氧水平(P<0.05);初级生产力随季节变化特征明显,原始P/R系数均小于试验组。根据试验结果分析得知,桓仁水库营养盐限制因素受季节变化的影响显著,具有氮、磷双限制等特征;根据初级生产力判断桓仁水库为自养代谢型水体。  相似文献
