为改善有刺黄龙果在引种地区的生长势,提高其抗逆性,本研究探讨了不同砧木和嫁接方法对有刺黄龙果嫁接育苗的影响,以期筛选到适宜有刺黄龙果的砧木和嫁接方法。结果表明,不同砧木和嫁接方法对有刺黄龙果的成活率、出芽率和新芽长势有显著影响。嫁接30天时,芽接、楔接、套接和平接法的愈合率显著高于插接和靠接法。嫁接45天时,平接、靠接和楔接法的出芽率较高,分别是76.7%、76.7%和66.7%。嫁接苗田间定植2个月时,平接、靠接和楔接法嫁接苗长势较快。其中楔接法嫁接效率高,适合工厂化批量嫁接。白肉品种‘无刺黄龙’和红肉品种‘金都1号’、‘富贵红’是有刺黄龙果嫁接的优良砧木,嫁接2个月时出芽率分别为86.7%、90.0%和86.7%,且新芽高度和宽度均大于其他砧木。综上所述,适宜有刺黄龙果规模化嫁接育苗的嫁接方法是楔接法,砧木是‘金都1号’、‘富贵红’和‘无刺黄龙’。  相似文献

选用6个羟基红花黄色素A(HSYA)含量差异较大的亲本,采用双列杂交方法配制杂交组合,测定2015年和2016年2年亲本及其后代F1和F2红花中的HSYA含量。运用双子叶植物种子数量性状遗传模型和统计分析方法,分析胚、细胞质和母体植株3套遗传体系的基因效应和环境互作效应。结果发现:在HSYA含量的遗传体系中,母体遗传效应影响最大,胚效应次之,细胞质效应影响最小。3套遗传体系均表现出基因主效应大于环境互作效应。机误方差较大,说明HSYA含量还受环境机误或抽样误差的影响。亲本遗传效应分析表明,豫红花1号(P1)做亲本表现稳定,有利于增加杂交后代HSYA含量,达到提高品质、改良品种的效果。胚显性方差和母体显性方差均达到极显著水平,表明同时存在种子杂种优势和母体杂种优势,而且其主效应基因不受环境影响。综合考虑遗传主效应、胚显性效应和母体显性效应,亲本组合(P1×P5)有利于提高后代杂交品种的HSYA含量。该研究结果可为后代材料在杂种优势利用中的亲本选择提供理论支持。  相似文献

使用RT-PCR方法从吉林省疑似感染胡葱黄条病毒(Shallot yellow stripe virus,SYSV)的分蘖洋葱(毛葱)叶片中获得了SYSV吉林毛葱分离物SYSV-JL(MN607702)的全基因组序列。SYSV-JL的全长序列为10?427 nt,与GenBank已登录的3条SYSV全长或近全长序列在多个基因上保持着较高的序列一致性,但在P1基因上核苷酸和氨基酸序列一致性较低,分别为69.5%~84.2%和60.9%~76.9%,进一步通过变异分析发现,P1基因包含369个变异位点,表现出较为明显的高变异特征。系统发育分析显示,不同SYSV分离物具有较为明显的地区差异,SYSV-JL分离物与多个中国分离物系统发育关系较近。  相似文献

India is one of the world’s largest producers of papaya. Viruses, mainly begomoviruses and potyviruses, cause a significant loss in papaya production. The study described here has identified a new species of begomovirus and a new species of betasatellite infecting Carica papaya in India. The sequences of the begomovirus and betasatellite show 90.03% nucleotide sequence identity to an isolate of Radish leaf curl virus and 92.25% identity to an isolate of Tomato leaf curl betasatellite, respectively. Maximum likelihood phylogenetic tree of begomovirus sequence of isolate DP2 (KX353622) showed distant relationships with previously characterised begomoviruses. Recombination analysis proposed six recombination breakpoints in begomovirus genome with other geographical begomovirus isolates.  相似文献

为探讨生物炭长期施用对酸化茶园土壤改良和真菌群落结构的影响,分析了按生物炭用量0、2.5、5、10、20、40 t·hm-2施用5年后的茶园土壤性状和真菌群落结构变化。结果表明,施用生物炭5年后的茶园土壤pH提高了0.16~1.11,可溶性有机碳含量提高了52.6%~92.3%,而铵态氮和硝态氮含量以10 t·hm-2处理最高。施用生物炭5年后的土壤性质变化,进一步影响了真菌群落结构,表现为Chao指数、ACE指数和Shannon指数随生物炭用量增加呈先增加后降低的趋势;提高生物炭施用量对茶园土壤次要作用的真菌(LDA值<3.50)丰度的增加效果高于优势真菌(LDA值>3.50)的效果,其中被孢霉属、木霉属、毛壳菌属的相对丰度增加,黑盘孢属的相对丰度降低。  相似文献

The polymerase chain reaction–denaturing gradient gel electrophoresis (PCR‐DGGE) of 16S ribosomal RNA gene was used to investigate bacterial communities in the intestines of large yellow croaker at six different ages (12 d, 18 d, 26 d, 40 d, 3 mo, and 1 yr old) as well as within the corresponding feed and culture water. In addition, Illumina Miseq sequencing was utilized to compare intestinal microbiota between 12‐d‐old and 1‐yr‐old individuals. PCR‐DGGE results revealed that the culture water had the highest bacterial diversity, followed by the feed, while the intestines had the lowest diversity. The intestinal microbiota at six ages changed severely; however, the change did not follow any trend. The large yellow croaker intestines harbored specific bacterial communities that differed from those in both feed and water. Illumina Miseq sequencing results revealed that the diversity of intestinal bacteria in 12‐d‐old fish was higher than that in 1‐yr‐old fish, and the bacterial composition differed significantly between them. γ‐Proteobacteria and Pseudoalteromonas supplied the most abundant phylum and genus in the 12‐d‐old fish intestine. However, in the 1‐yr‐old fish intestine, Firmicutes and Clostridium were the most dominant, respectively. The study may contribute to a better understanding of gut microbiota and dynamics of the large yellow croaker and the relationship with their surrounding environment.  相似文献

长期施肥对黄壤性水稻土有机碳矿化的影响   总被引:13,自引:0,他引:13  
以贵州省农业科学院内黄壤性水稻土长期(22年)定位施肥试验为对象,采用室内模拟培养试验研究了不施肥(对照,CK)、施化肥(NPK)、低量有机无机肥配施(0.5MNPK)、施牛粪(M)和常量有机无机肥配施(MNPK)对土壤有机碳矿化的影响。结果表明:NPK处理土壤有机碳含量(21.6 g kg~(-1))与CK处理(22.8 g kg~(-1))基本相同,而0.5MNPK、M和MNPK处理的土壤有机碳含量较CK处理分别提高了30.6%、72.9%和62.2%,其中,M和MNPK处理差异达显著水平(p0.05)。模拟培养条件下,CO2产生速率在培养的第2天达到最大值,然后迅速下降,而后缓慢下降(第4~24天),后期(第24~30天)趋于稳定;各处理土壤有机碳矿化速率大小依次为:MMNPK0.5MNPKCKNPK,各处理土壤有机碳矿化速率随时间的动态变化均符合对数函数关系(p0.01)。培养结束(30 d)时,各处理土壤有机碳累积矿化量为1.23~2.37 g kg~(-1),以M处理和MNPK处理较高,较CK处理(1.46 g kg~(-1))分别增加了62.6%和44.2%(p0.05);各施肥处理土壤有机碳的累积矿化率(土壤有机碳累积矿化量/土壤有机碳含量)较CK处理(6.4%)均有所下降,以M处理和MNPK处理下降较多,降幅分别为1.2%和0.9%。土壤有机碳累积矿化量随培养天数的动态变化可以用一级动力学方程拟合(p0.01),模拟结果表明,CK处理土壤潜在可矿化有机碳量为1.55 g kg~(-1),与CK处理相比,NPK处理下降了11.6%,但差异不显著(p0.05),而有机肥处理(0.5MNPK、M和MNPK)有不同程度的提高(21.3%~73.6%),其中,M和MNPK处理提高幅度较大(p0.05);同时,MNPK处理能够提高土壤有机碳的周转速率,减少周转时间。上述结果指示黄壤性水稻土长期施用有机肥(0.5MNPK、M和MNPK)能够提高土壤有机碳的矿化速率,在促进土壤有机碳积累的同时降低其累积矿化率(单位有机碳矿化水平),增强土壤固碳能力。  相似文献

The breadmaking quality of wheat is affected by the composition of gluten proteins and the polymerisation of subunits that are synthesised and accumulated in developing wheat grain. The biological mechanisms and time course of these events during grain development are documented, but not widely confirmed. Therefore, the aim of this study was to monitor the accumulation of gluten protein subunits and the size distribution of protein aggregates during grain development. The effect of desiccation on the polymerisation of gluten proteins and the functional properties of gluten were also studied. The results showed that the size of glutenin polymers remained consistently low until yellow ripeness (YR), while it increased during grain desiccation after YR. Hence, this polymerisation process was presumed to be initiated by desiccation. A similar polymerisation event was also observed when premature grains were dried artificially. The composition of gluten proteins, the ratios of glutenin to gliadin and high molecular weight-glutenin subunits to low molecular weight-glutenin subunits, in premature grain after artificial desiccation showed close association with the size of glutenin polymers in artificially dried grain. Functional properties of gluten in these samples were also associated with polymer size after artificial desiccation.  相似文献

The aim of this study is to investigate the composition of the fatty acids and phospholipids derived from large yellow croaker (Pseudosciaena crocea) roe. The composition of the total lipids and the molecular species of phospholipids were determined by gas chromatography–mass spectroscopy (GC–MS) and high performance liquid chromatography–evaporative light scattering detector (HPLC–ELSD), respectively. The results showed that large yellow croaker roe had high levels of the total lipid (19.6% ± 1.32%, w/w) and phospholipid (61.2% ± 1.22% of the total lipid). The phospholipid was rich in docosahexaenoic acid (31.0% ± 0.19% of the total phospholipids), and the major phospholipid molecular species was phosphatidylcholine (PC, 61.06% ± 0.02% of the total phospholipids, w/w). It was concluded that large yellow croaker roe is expected to be a good resource of phospholipids with a high content of PC.  相似文献

A comparative study on the volatile compounds in cooked muscle of wild and farmed large yellow croaker (LYC) was conducted. The two farmed LYC groups were fed with diets containing 44% (CF) and 25% (LF) of fish meal (FM) respectively. Results showed that 48 volatiles, including aldehydes, alcohols, ketones, hydrocarbons, aromatics, acids, esters, furans and miscellaneous compound, were detected in cooked fillets. The LF group had significantly lower amounts of total aldehydes and ketones, higher content of miscellaneous compound in cooked fillets than that in the CF and wild groups (< .05). Compared with the wild group, the LF group had significantly lower amounts of total alcohols, acids and esters, while the CF group had significantly lower amounts of total aldehydes, higher content of total ketones in cooked muscle (< .05). According to the principal component analysis (PCA), some volatiles (propanal, nonanal, etc.) could be considered as sensitive indicators to classify cooked muscle samples. In conclusion, differences in the volatiles in the cooked muscle between the wild and farmed LYC have been found. Low level of dietary FM (25%) changed the volatile profiles in cooked fillets of farmed LYC. A PCA may be useful to screen potential volatiles to classify cooked muscle samples in this study.  相似文献
