本文报道了按照森林土壤标准物质要求采集的二个土壤样品(碱性土-碱灰性褐土;酸性土-黄红壤)和一个植物样品(杉木叶)的制备过程,粒径2mm的土壤样品采用Y型混样,0.149mm的土壤和0.177mm的植物样品采用高铝瓷球磨制备,土壤样品用高湿灭活,植物样品用^60Co辐射灭活,在二年测定标准样品稳定性和同时进行标准样品中微生物类群和数量的分析。土壤、植物标准样品的均匀性和稳定性采用数理统计方法检验,其结果均达到标准物质要求。  相似文献

It is desirable to develop an objective Soil Quality Index (SQI) to guide sustainable agronomic intensification, thereby promoting socio‐economic well‐being. This study pioneers the use of Ward's cluster and principal component regression methods to evaluate soil homogeneity and construct a SQI (expressed as %). Field data were acquired from five different sites within Ohio, USA, that were under no‐till (NT), conventional till (CT) management and natural vegetation (NV) land use. Soil pH, carbon/nitrogen (C/N) ratio, nitrate and soil organic carbon (SOC) concentrations were identified as primary drivers of soil quality. Based on Ward's cluster method, the soil properties of croplands were not significantly different from those under NV land use. However, SQI ranked surface soils under CT management as higher in quality than NV and NT managed soils, respectively. The coefficient of determination (R2) between SQI and corn (Zea mays L.) and soya bean [Glycine max (L.) Merr.] yields was 0.7 and 0.9, respectively, implying this SQI effectively relates soil properties, a function of anthropogenic land management practices, with crop yields. In future, time series analyses will be used to assess SQI versus crop yield dynamics, with key socio‐economic and climate variables.  相似文献

SLO1 potassium channels are pivotal to many aspects of spermatogenic cell. Experiments were conducted to assess physiology and function of SLO1 potassium channels in different developmental stages of spermatogenic cell in Eriocheir sinensis. First, the expression of SLO1 protein was examined via Western blot, RT‐PCR, immunohistochemistry and immunofluorescence assays. The results showed that the expression of the SLO1 protein was not uniform in the spermatogenic cells of E. sinensis. Second, whole‐cell patch clamp technique was used to record the potassium current of spermatogenic cells and to analyse the electrophysiological characteristics of the potassium channels with the aid of inhibitors. It is proved that the potassium current in E. sinensis germ cells is associated with intracellular Ca2+, and the calcium‐activated potassium channel mediated by SLO1 protein is mainly large‐conductance Ca2+‐activated K+ channels (BKCa). Based on these researches, the cDNA of SLO1 from testis was cloned and sequenced. The SLO1 protein contained domains bound to calcium ions, and the spatial structure formed by its tetramers constituted potassium channels. Phylogenetic analysis revealed that SLO1 was much closer to Scylla paramamosain than other examined species. Finally, iberiotoxin (IbTX) and CdCl2 were used to inhibit the acrosome reaction (AR) induced by A23187 and to explore the role of SLO1 potassium channels in the AR of E. sinensis. The experimental results showed that SLO1 potassium channels were expressed in E. sinensis spermatogenic cells and played an important role in the AR of crab sperm (SP).  相似文献

According to the structural characteristics of bridge engineering, the mix optimization design method of bridge fair-faced concrete was presented, and C30~C50 concrete with high cohesion and package, outstanding fluidity and excellent mechanical properties were prepared. Through the test of air content of fresh concrete, micro hardness and micro structure in different parts of hardened concrete specimens, effects of viscosity-modifying admixtures on homogeneity of concrete were discussed. The results showed that the composition of bridge fair-faced concrete were relatively homogeneous, and the microstructure were relatively dense. When C30 concrete mixed with 5 × 10 -5 cellulose ether or 6% silica fume (mass fraction of cementitious material), air content of fresh concrete were less than 2%, and slurry viscosity values were between 1 900 and 2 000 MPa·s. Also, the micro hardness values in different parts of hardened concrete specimens were closed. The results were applied in practical bridge engineering, which generally meets the apparent quality requirements of bridge fair faced concrete.  相似文献

该文介绍了作者设计并研制的新型翻转式双轴桨叶混合机,阐述了其工作原理。为了提高双轴桨叶混合机的混合性能,采用二次回归正交旋转组合试验,对设计样机进行混合均匀度及机内残留率的试验研究,得出了其工作参数的合理组合:混合时间为180 s,充满系数为0.75,桨叶轴转速为56 r/min;机内残留量极小(约为0.015%),满足其作为配合饲料生产,尤其是预混合饲料生产的工艺要求,也为进一步改进设计提供了依据。  相似文献

Protein heterogeneity in European wheat landraces and obsolete cultivars   总被引:2,自引:0,他引:2  
Identity and present degree of genetic homogeneity and heterogeneity, respectively of 52 European wheat accessions, maintained in the collection of wheat genetic resources, have been characterized using analyses of glutenins by sodiumdodecylsulphate polyacrylamide gel electrophoresis (SDS-PAGE). Six of the analyzed wheat accessions were observed to be homogeneous, while 46 (88.5%) of them were heterogeneous in protein profiles. Heterogeneous accessions possessed 2 to 13 different protein lanes. Together, 17 high molecular weight glutenin subunit (HMW-GS) alleles have been found. The most frequent HMW-GS alleles at the Glu-A1, Glu-B1, and Glu-D1 complex loci were 1, 7+9, and 2+12, respectively. However, also low frequented HMW-GS alleles or allelic combinations, such as 7+15, 13+16, 20, 6, 7, and 9 were observed. Furthermore, another new allele encoding HMW glutenin subunit with relative molecular weight 98.6 kDa has been found in one of the lines of the cultivar Eritrospermum 917. The Glu-score in the examined accessions varied in broad range, some of the lines reached the maximum value 10.  相似文献

A study on the distribution of free iron and manganese oxides was conducted in soils developed on calcareous alluvial deposits under subhumid climatic conditions, in Western Greece. Soil samples from two well drained soils and from two poorly drained soils, classified as Alfisols, were collected and used in this study. After certification of soil homogeneity the acid ammonium oxalate and dithionite-citrate-bicaxbonate methods were used to extract free iron and manganese oxides from the samples. Iron oxides extracted by the dithionite-citrate-bicarbonate method (Fed) were significantly higher than the iron oxides extracted by the ammonium oxalate method (Feo), indicating that a considerable fraction is present in crystalline forms, independent of drainage status. A confirmation of free iron oxides and fine clay was detected. The ratios Feo/Fed and (Fed-Feo)/total Fe (Fet) could not be used to distinguish the well drained soils from the poorly drained soils. Manganese movement in a soluble form is independent of the fine clay.  相似文献

以种子蛋白图谱为指标,分析了由系谱法、回交法所选育的一套遗传梯度不同的玉米自交系和姊妹系的差异。结果表明,用PAGE法所测定的种子蛋白谱带能鉴别出遗传成分差异在13%以上的两个自交系及自交后代材料的纯合程度的遗传稳定性,可以作为玉米自交系选育的一个辅助指标。  相似文献
