通过非灭菌土盆栽接种试验,观察了Glomus mosseae(GM)、G.intraradices(GI)、G.aggregatum(GA)和G.atunicatum(GE)4种AM真菌对油桐幼苗生长和氮磷吸收的影响,结果表明接种GM、GI和GE后菌根侵染率显著提高,侵染率分别为77.4%、92.3%和96.7%,比对照分别提高20.4%、43.5%、50.4%。接种GA与对照无显著差异;接种GM和GI后植株的干质量增加,而接种GA和GE对全株及各器官干质量影响不明显;接种4种真菌对油桐幼苗的氮素营养影响不大;接种GM、GI和GE显著提高了根部磷含量。综合考虑,接种GM效果最好。  相似文献

Arbuscular mycorrhizal (AM) development in different soil types, and the influence of AM fungal hyphae on their original soil were investigated. Plantago lanceolata, which can grow in soils of a very wide pH range, was grown in two closely related limestone soils and an acid soil from rock habitats. Plants were colonised by the indigenous AM fungal community. The use of compartmented systems allowed us to compare soil with and without mycorrhizal hyphae. Root colonisation of P. lanceolata was markedly higher in the limestone soils (30-60%) than in the acid soil (5-20%), both in the original habitat and in the experimental study. Growth of extraradical AM fungal hyphae was detected in the limestone soils, but not in the acid soil, using the signature fatty acid 16:1ω5 as biomass indicator. Analysis of signature fatty acids demonstrated an increased microbial biomass in the presence of AM fungal hyphae as judged for example from an increased amount of NLFA 16:0 with 30 nmol g−1 in one of the limestone soils. Bacterial activity, but not soil phosphatase activity, was increased by around 25% in the presence of mycorrhizal hyphae in the first harvest of limestone soils. AM fungal hyphae can thus stimulate microorganisms. However, no effect of AM hyphae was observed on the soil pH or organic matter content in the limestone soils and the available P was not depleted.  相似文献

Mobilization of non‐exchangeable ammonium (NH ) by hyphae of the vesicular‐arbuscular mycorrhizal (VAM) fungus Glumus mosseae was studied under controlled experimental conditions. Maize (Zea mays) and parsley (Petroselinum sativum) were grown either alone or in symbiosis with Glomus mosseae in containers with separated compartments for roots and hyphal growth. In one experiment, 15NH was added to the soil to differentiate between the native non‐exchangeable NH and the non‐exchangeable NH derived from N fertilization. Non‐exchangeable NH was mobilized by plant growth. Plant dry weight and N uptake, however, were not significantly influenced by mycorrhizal colonization of the roots. The influence of root infection with mycorrhizal fungus on the mobilization of non‐exchangeable NH was negligible. In the hyphal compartment, hyphal uptake of N resulted in a decrease of NH in the soil solution and of exchangeable NH . However, the NH concentration was still too high to permit the release of non‐exchangeable NH . The results demonstrate that, in contrast to roots, hyphae of VAM fungi are not able to form a non‐exchangeable‐NH depletion zone in the adjacent soil. However, under conditions of a more substantial depletion of the exchangeable NH in the mycorrhizal sphere (e.g., with longer growth), an effect of mycorrhiza on the non‐exchangeable NH might be found.  相似文献

In an earlier study we reported the apparent stabilization of a low fungal biomass in ex-arable lands during the first decades after abandonment. It was hypothesized that the lack of increase in fungal biomass was due to constraints on development of fungi with persistent hyphae such as lignocellulolytic basidiomycetes and ericoid mycorrhizal fungi. With respect to the former group, the slow increase of the pool of lignocellulose-rich organic matter was expected to be the major constraint for their development. To study this, we enriched soil samples of one arable land, of two recently abandoned arable lands, of one older abandoned arable land and of heathland with carbon substrates that differed in composition (glucose, cellulose and sawdust). In addition, we combined the effect of carbon addition on fungal biomass development in arable and recently abandoned lands with inoculation of 1% of soil from the older abandoned site and the heathland. All treatments induced a fast increase and a subsequent rapid decline in fungal biomass in the arable and ex-arable fields. Denaturing Gradient Gel Electrophoresis (DGGE) band patterns and enzyme activities did show differences between the carbon treatments but not between the recent and older abandoned field sites, indicating a similarly responding fungal community even after three decades of land abandonment and irrespective of soil inoculation. Identification of fungi by sequencing and culturing confirmed that decomposition processes were mostly dominated by opportunistic fungi in arable and ex-arable fields. In the heathland, only a very slow increase of microbial activity was observed after addition of carbon and sequencing of DGGE bands showed that ericoid mycorrhiza (ERM) fungi were responsible for carbon decomposition. We conclude that an increase of enduringly present fungal hyphae in ex-arable land may only be possible when a separate litter layer develops and/or when suitable host plants for ERM fungi become established.  相似文献

The composition of arbuscular mycorrhizal fungi (AMF) communities found in agricultural systems has been found to be very different to that of forest. The implications of this, if any, for the restoration of indigenous forest on ex-agricultural land is poorly understood. This study investigated the effect that AMF communities isolated from ex-agricultural and forest soils have on the growth of an indigenous New Zealand tree species (Podocarpus cunninghamii). The forest AMF community was isolated from a remnant stand of P. cunninghamii forest and the ex-agricultural AMF from a retired grazing grassland. In addition, the study examined how the two AMF communities affected the competitiveness of P. cunninghamii when grown in competition with an invasive grass species (Agrostis capillaris), which is frequently dominant on ex-agricultural land in New Zealand. P. cunninghamii growth was significantly decreased by inoculation with ex-agricultural AMF compared to forest AMF. Furthermore, the forest AMF community was able to significantly increase P. cunninghamii root production when in competition with A. capillaris. The findings suggest that when attempting to restore indigenous forest on ex-agricultural land, inoculation of tree seedlings with appropriate forest AMF may improve their growth and survival.  相似文献

Summary Wheat cultivars assumed to be non-susceptible to vesicular-arbuscular (VA) mycorrhizae became colonized, and this effect persisted under different growth conditions. Colonization of all cultivars was similar regardless of the amount of inoculum and the time interval of inoculation. Different plant growth temperatures and the support given by the culture media, inoculation with different endophytes, and inoculation with sterilized and unsterilized spores affected VA colonization levels, although the level of colonization reached in cv. Champlein was similar to that reached in cv. 7-Cerros under each condition. VA mycorrhizal colonization was also affected by different plant growth conditions. After VA reinoculation, the plant dry weight of Castan and 7-Cerros increased, but not Negrillo and Champlein cultivars. VA mycorrhizae increased the shoot dry weight of 7-Cerros only, but not of Champlein, when grown at 35/24°C, and had no effect on the dry weight of either cultivar grown at 18/12°C and 42/24°C. Inoculation with Glomus mosseae increased the dry weight of the cultivars more than inoculation with G. fasciculatum or G. agregatum. The effect on the plant dry weight was greater in plants grown in soil than in sand/vermiculite pots. Inoculation with sterilized and unsterilized spores of G. mosseae, either in soil pots or in sand/vermiculite tubes, did not increase the plant dry weight. Our results indicate that there was no close relationship between the level of root colonization and the effect on plant growth. The effects of accompanying microorganisms in the VA inoculum on VA mycorrhizal symbiosis are discussed.  相似文献

A pot experiment was conducted to investigate the mobilization of sparingly soluble inorganic and organic sources of phosphorus (P) by red clover (Trifolium pratense L.) whose roots were colonized by the arbuscular mycorrhizal (AM) fungus Glomus mosseae and in association with the phosphate-solubilizing (PS) bacterium Bacillus megaterium ACCC10010. Phosphate-solubilizing bacteria and rock phosphate had a synergistic effect on the colonization of plant roots by the AM fungus. There was a positive interaction between the PS bacterium and the AM fungus in mobilization of rock phosphate, leading to improved plant P nutrition. In dual inoculation with the AM fungus and the PS bacterium, the main contribution to plant P nutrition was made by the AM fungus. Application of P to the low P soil increased phosphatase activity in the rhizosphere. Alkaline phosphatase activity was significantly promoted by inoculation with either the PS bacterium or the AM fungus.  相似文献

Arbuscular mycorrhizal fungi (AMF) produce a protein, glomalin, quantified operationally in soils as glomalin-related soil protein (GRSP). GRSP concentrations in soil can range as high as several mg g−1 soil, and GRSP is highly positively correlated with aggregate water stability. Given that AMF are obligate biotrophs (i.e. depending on host cells for their C supply), it is difficult to explain why apparently large amounts of glomalin would be produced and secreted actively into the soil, since the carbon could not be directly recaptured by the mycelium (and benefits to the AMF via increased soil structure would be diffuse and indirect). This apparent contradiction could be resolved by learning more about the pathway of delivery of glomalin into soil; namely, does this occur via secretion, or is glomalin tightly bound in the fungal walls and only released after hyphae are being degraded by the soil microbial community? In order to address this question, we grew the AMF Glomus intraradices in in vitro cultures and studied the release of glomalin from the mycelium and the accumulation of glomalin in the culture medium. Numerous protein-solubilizing treatments to release glomalin from the fungal mycelium were unsuccessful (including detergents, acid, base, solvents, and chaotropic agents), and the degree of harshness required to release the compound (autoclaving, enzymatic digestion) is consistent with the hypothesis that glomalin is tightly bound in hyphal and spore walls. Further, about 80% of glomalin (by weight) produced by the fungus was contained in hyphae and spores compared to that released into the culture medium, strongly suggesting that glomalin arrives mainly in soil via release from hyphae, and not primarily through secretion. These results point research on functions of glomalin and GRSP in a new direction, focusing on the contributions this protein makes to the living mycelium, rather than its role once it is released into the soil.  相似文献
