以籼稻、粳稻两种类型的成熟种子为外植体研究了不同浓度的AgNO3在水稻农杆菌转化中的作用。结果表明,在农杆菌转化前添加AgNO3可以提高愈伤组织诱导率,最适浓度为8~10mg/L,但对愈伤组织的分化没有明显促进作用;转化后添加低浓度的AgNO3可以减轻愈伤组织的褐化,提高抗性愈伤率和绿苗分化率,最适浓度为2-8mg/L。同时发现,无论是转化前还是转化后,添加高浓度的AgNO3对水稻愈伤组织的分化均具有毒害作用。  相似文献

The sensitivity of a bioassay in detecting soil inoculum of Colletotrichum coccodes and Helminthosporium solani was examined using potato minitubers and microplants. Tests were conducted on soils which were collected from fields in which the interval after a previous potato crop differed, and which were also artificially infested with conidia or microsclerotia. For C. coccodes , determining plant infection based on the occurrence of infected roots after 9–12 weeks was a sensitive method for detecting and quantifying the amount of inoculum in soil. Infestations of less than 0·4 microsclerotia per g soil were detected in artificially infested soils. A semiselective medium, developed for isolating C. gloeosporioides from pepper, detected soil infestations by C. coccodes as low as nine conidia or one microsclerotium per g soil in artificially infested soil. For H. solani , infection on minitubers was a sensitive measure, with soil inoculum of fewer than 10 conidia per g soil being detected. Soil infestation could be quantified by assessing the percentage surface area of minitubers covered by sporulating lesions, which was strongly related to the amount of soil infestation. The results of these bioassay tests were compared with published results for real-time quantitative PCR assays on the same soils. The two methods were in good agreement in artificially infested soils, but the bioassay appeared to be more sensitive with naturally infested soils.  相似文献

目的 鞋印是刑事侦查的重要物证之一,如何对积累的大量鞋印花纹图像进行自动归类管理是刑事技术迫切需要解决的问题之一。与其他类图像不同,鞋印花纹图像具有种类多但数目未知、同类花纹分布不均匀且同类花纹数目少的特点。基于鞋印花纹图像的这些特点,用目前典型的聚类算法对鞋印花纹图像集进行聚类,并不能取得很好的效果。在对鞋印花纹图像进行分析的基础上,提出一种K步稳定的鞋印花纹图像自动聚类算法。方法 对已标记的鞋印花纹图像进行统计发现,各类鞋印花纹之间在特征空间上存在互不相交的区域(本文称为隔离带)。算法的核心思想是寻找各类鞋印花纹之间的隔离带,来将各类分开。过程为:以单调递增或递减的方式调整特征空间中判定两点为一类的阈值,得到数据集的多次划分;若在连续K次划分的过程中,某一类的成员不发生变化,则说明这K次调整是在隔离带中进行的,即聚出一类,并从数据集中删除已标记的数据;选择下一个阈值对剩余的数据集进行划分,输出K步不变的类;依此类推,直到剩余数据集为空,聚类完成。结果 在两类公开测试数据集和实际鞋印花纹数据集上进行实验,本文算法的主要性能指标都超过典型算法,其中在包含5792枚实际鞋印花纹数据集上的聚类准确率和F-Measure值分别达到了99.68%和95.99%。结论 针对鞋印花纹图像特点,提出了一种通过寻找各类之间的隔离带进行自动聚类的算法,并在实际应用中取得了很好的效果。且算法性能受参数的变化以及类的形状影响较小。本文算法同样适用于具有类似特点的其他数据集的自动聚类。  相似文献

The purpose of this study was to determine the influence of temperature applied during short day-induced budset on induction of dormancy in six ecotypes of Betula pubescens Ehrh. and two ecotypes of Betula pendula Roth. Seedlings were grown in a phytotron at constant temperatures of 9–21°C under a 12 h photoperiod (SD) during dormancy induction. Induction of dormancy was monitored by following bud flushing and shoot growth after transfer to long photoperiod conditions (24 h) at 18°C. Chilling requirement was studied in seedlings exposed to 10 weeks of SD. In both species induction of bud dormancy developed most rapidly at 15–18°C, and both 9–12°C and 21°C delayed the induction of dormancy. Raising the temperature (from 9 to 21°C) applied during induction of dormancy significantly increased the chilling requirement. These responses were noted for all ecotypes tested, but in general the northern ecotypes entered dormancy more quickly than the southern ones. No such trend was recorded for chilling requirement, although a B. pubescens ecotype from Iceland and another from the coast of northern Norway appeared to require a longer chilling treatment than the other ecotypes. In conclusion, induction and depth of bud dormancy in birch are significantly affected by temperature conditions and these effects may explain some of the annual variation in dormancy and chilling requirement observed in nature.  相似文献

The occurrence of trichostrongylid nematodes in young cattle was studied in relation to climate and geographical location, by examination of herbage and faecal samples from 8 farms in 7 different regions in Norway, during 3 consecutive summers. Species of Ostertagia and Cooperia were dominant. Infective larvae, especially those of Ostertagia, were able to survive the winter in the pasture to a great extent in all the areas studied. The spring larval counts varied considerably from place to place, and from year to year, but were often at fairly high levels. This confirms previous findings that the larvae which overwinter on the pasture may be a major cause of losses. Overt clinical disease, however, was only seen at 1 farm during this investigation.The large number of larvae often seen in the autumn seems to be of less significance as a cause of disease, due to the late appearance, but is of major importance as a source of the larvae which survive the winter in the pasture and infect young cattle the following spring.There was no obvious correlation between pasture larval counts and the climate, except that there seemed to be higher counts in the warmer parts of the country, where the grazing season is also longer. The only place were both faecal egg counts and larval counts in the herbage were consistently low, was in the very far north.  相似文献

In evaluating radiographs of the limb joints and head, students encounter difficulty where superimposition occurs. By replacing calcium with silver salts in the bone, enhanced radiopacity can be produced. In this study, silver impregnation was used to increase the radiopacity of individual carpal and tarsal bones, selected bones of the skull and the sinuses, and guttural pouch of the horse. This provides an interpretation aid for teaching radiographic anatomy of these regions.  相似文献

The complete genomic sequences of several Pseudomonas spp. that inhabit the rhizosphere are now available, providing a new opportunity to advance knowledge of plant growth-promoting rhizobacteria (PGPR) through genomics. Among these is the biological control bacterium Pseudomonas fluorescens Pf-5. Nearly 6% of the 7.07 Mb genome of Pf-5 is devoted to the biosynthesis of secondary metabolites, including antibiotics toxic to soilborne fungi and Oomycetes that infect plant roots, and two siderophores involved in iron acquisition. Three orphan gene clusters, for which the encoded natural product was unknown, also were identified in the genome of Pf-5. The product synthesized from one of the orphan gene clusters was identified recently using a new ‘genomisotopic approach’, which employs a combination of genomic sequence analysis and isotope guided fractionation. Application of the genomisotopic approach to one orphan gene cluster in Pf-5 resulted in the discovery of orfamide A, founder of a new group of bioactive cyclic lipopeptides with a putative role in biological control of plant disease.  相似文献

ABSTRACT:   The effects of silvering state of wild female Japanese eels Anguilla japonica on the success of induced maturation and the following spawning were examined. Thirty-eight females, collected in Mikawa Bay, were divided into four stages based on their silvering state: yellow (Y1), late-yellow (Y2), silver (S1) and late silver eels (S2). Despite injections of salmon pituitary extract (SPE) through the standard technique, Y1 and Y2 eels did not respond to the treatment with undeveloped gonad (gonad-somatic index [GSI]: 0.3–0.9), and all these females died by 5 weeks, probably due to an abnormal physiological condition. Most S1 (81%) and S2 eels (100%) matured completely (GSI: 17.8–51.4), and finally spawned successfully (69% for S1, 89% for S2). S2 eels fully matured with oocytes of over 750 μm in diameter by significantly smaller number of injections of SPE (5–6 times) than the case of S1 eels (6–8 times). The amount of eggs released by S2 eels (0.65 ± 0.11 g/fish per body weight [BW]) was significantly larger than those by S1 eels (0.54 ± 0.09 g/fish per BW). There was no difference in fertilization and hatching rates between eggs released by S1 eels and those of S2 eels. These results indicate that the success of induced maturation and spawning in wild female Japanese eels depends on their silvering state, and matured eggs can be obtained efficiently through the use of S2 eels rather than other stages.  相似文献
