人白细胞介素-4基因在家蚕杆状病毒表达系统中的表达

采用RT-PCR法从经LPS诱导刺激的人外周血淋巴细胞中克隆了人白细胞介素-4(human interleu kjn-4,hIL-4)基因。为高效表达hIL-4,促进其临床应用,将hIL-4插入家蚕杆状病毒转移载体pBacPAK8中,构建重组载体pBacPAK-hIL-4,并与线性化病毒Bm-BacPAK6 DNA共转染家蚕BmN细胞,获得重组病毒BacPAK-hIL-4。将重组病毒感染家蚕5龄幼虫和蚕蛹(1×10~5PFU/头),表达产物以ELISA、SDS-PAGE和Western blotting方法检测,用活化的人外周血T淋巴细胞测定表达产物体外生物活性。ELISA法结果表明hIL-4在感染病毒后96h的蚕血淋巴和120h的蚕蛹中表达量最高,分别达到2．3μg/mL蚕血淋巴和10．2μg/mL体液;SDS-PAGE、Western blotting分析表明表达产物分子量约20kD;表达产物对活化的T淋巴细胞增殖具有明显促进作用。     

类蚕丝丝素结晶区蛋白表达载体的构建与表达

为探讨蚕丝结构与性能的关系,设计了几种类似蚕丝丝素结晶区高度重复序列结构(GAGAGX)n的基因序列,并构建了大肠杆菌表达载体,在BL-21菌株中成功诱导表达了4种约由110个氨基酸组成的小分子蛋白。对表达产物进行W estern印迹分析和ELISA检测的结果显示,(GAGAGA)n、(GAGAGY)n、(GAGAGV)n3种蛋白都有小于14.4 kD的大小相同的条带,而(GAGAGS)n有一条稍大于20.1 kD的条带。以超声波破菌的缓冲液作空白对照,4种蛋白的ELISA的结果均比对照的BL-21(无质粒)高,显示为阳性。     

当前猪呼吸道病的治疗与原因分析

近段时间,笔者所在区域内猪呼吸道疾病频繁发生,断奶仔猪、育肥猪发病率高,偶有哺乳仔猪发病。虽然发病率高,但是死亡率低。由于呼吸系统疾病的顽固性、反复性,且由于广大养殖户药物防治的盲目性,使其付出了很大的防治代价。笔者就近来诊治的典型案例进行总结,不当之处请大家指正。     

浅谈南方旱养肉鸭的技术关键

Isolation,Identification and Analysis of Serotype and Antibiotic Susceptibility of Pathogenic Salmonella from Chickens

To investigate the situations of predominant strain and antibiotic resistance of pathogenic Salmonella from chickens in Guangxi Zhuang Autonomous region, Salmonella was pre-enriched and isolated from tissues of clinical suspected chickens, and the Salmonella isolates were identified by biochemical test using ID 32E System for the identification of Enterobacteriaceae of VITEK System ATB Expression, serotypes were determined by slid agglutination test, antibiotic susceptibility was analyzed by ATB VET Susceptibility Test Strip of Enterobacteriaceae antibiotics.The results showed that 34 Salmonella strains were obtained from 310 clinical samples, of which 1 strain belonged to A serogroup, 1 to C2 serogroup, 15 to B serogroup, 14 to D serogroup and 3 to untyped serogroup, and S.typhimurium of B serogroup and S.pullorum of D serogroup were the predominant serotypes.All Salmonella isolates were sensitive to meropenem, imipenem, spectinomycine and apramycine, and the resistance rates to chloramphenicol, kanamycine and gentamicine were less than 10%, and the resistance rates to amoxicillin, streptomycine, flumequine, oxolinique, sulfamethizol, tetracycline and nitrofurantoine ranged between 50% and 90%, while the resistance rates to penicilline, oxacilline, fusidique, rifampicine and metronidazole reached to 100%.The results indicated that the serotype distribution of pathogenic Salmonella from chickens exhibited regional characteristics, and S.typhimurium and S.pullorum were the predominant serotypes in Guangxi Zhuang Autonomous region, and antibiotic resistance of Salmonella isolates was very serious which should be highly concerned.     

Study on Biological Characteristics and Drug Sensitivity of Haemophilus parasuis Isolated in Sichuan Province

The study was aimed to investigate the biological characsteristics and drug sensitive tests of Haemophilus parasuis isolated in Sichuan province.This paper reported the biological characteristics,16S rRNA gene PCR and drug sensitive test of the isolates by technique of microbiological and molecular biology.The results showed that the isolates had micro anaerobic and could produce satellite phenomenon,the isolates were just same in biochemical characteristics with field isolates in other area.All the isolates amplified 821 bp 16S rRNA gene.And drug sensitive tests showed that the isolates were highly sensitive to cefradine,ampicillin,cefradine,amoxicillin,cefaclor,ofloxacin,cefotaxime and furadantin,the sensitive rate was 75.61% to 87.80%,but the isolates were high resistance to lincomycin,carbenicillin,enrofloxacin,amikacin and sulfamethoxazole,and multiple-drug resistance was found among the isolates,the number of bacterial strains resistant to antibiotics were more between five and eight,the resisitance strains were 21.95% to 30.49%,7 isolates were even resistant to 13 antibiotics.The results showed the phenomenon of drug resistance of the isolates was very serious.     

猪致病性大肠埃希氏菌的药物敏感性试验

本实验采用世界卫生组织推荐的(K—B法)纸片琼脂扩散法,对从本地区猪场分离出的10株猪致病性大肠埃希氏菌,用10种临床上常用的抗菌药物对其进行了敏感性测定。结果:耐药现象严重,多重耐药性普遍存在。对猪致病性大肠埃希氏菌作用较强的依次是阿莫西林/克拉维酸(AMX/CA)、头孢唑啉(CZ)、氯霉素(C)、庆大霉素(GM),抑菌作用较差的是四环素(TC)、环丙沙星(CIP)、氨苄西林(AM)、氧氟沙星(DFL),链霉素(S)、磺胺(SMX)为中度敏感。
