外源共生基因对紫云英根瘤菌共生效率的影响   总被引:2,自引:0,他引:2  
 将豌豆根瘤菌共生质粒pJB5JI、含紫云英根瘤菌共生基因的重组质粒pRaZ15及含苜蓿根瘤菌共生基因的重组质粒pRmSL26导入紫云英根瘤菌野生型菌株7653R,然后对3种转移接合子的共生固氮效率进行了比较全面的测定。结果表明:转移接合子7653R(pJB5JI)的共生固氮能力较出发菌株7653R大幅度提高,而且表现出较高的竞争结瘤能力,转移接合子7653R(pRmSL26)在固氮酶活性、植物干重及结瘤数方面亦显著高于对照菌株7653R;转移接合子7653R(pRaZ15)虽然在固氮酶活性及植物干重方面没有发现显著性差异,但结瘤数显著减少。  相似文献

Abstract

A field experiment was conducted during two consecutive growing seasons (2013 and 2014) to evaluate the effects of inoculations with Rhizobium and Azotobacter on the growth and yield of two chickpea (Cicer arietinum L.) varieties under saline (5.8 dS m?1) arid condition. The single treatment of either Rhizobium or Azotobacter exhibited to promote the growth of chickpea to some level, however, co-inoculation produced more effects and increased the shoot dry weight (30.3 and 26.4%), root dry weight (17.5 and 26.3%), nodule number (79.1 and 43.8 piece per plant), nitrogen content in roots (9.62 and 10.9%), in shoots (12.6 and 8.3%) and seed protein (7.1 and 4.3%) in both Flip06-102 and Uzbekistan-32 chickpea varieties compared to the control. Our studies showed that the highest yield response of 429 (27.9%) and 538 (23.9%) kg?ha?1 over the control was revealed by the co-inoculation with Rhizobium and Azotobacter inoculants in Flip 06-102 and Uzbekistan-32, respectively. A new introduced Flip 06-102 chickpea variety was more salt tolerant and had higher root nodulation than the local Uzbekistan-32 chickpea variety. Nitrogen (N), phosphorus (P), and potassium (K) contents in the shoots and roots were significantly (p?Rhizobium plus Azotobacter could be applied to improve the vegetative growth and yield of chickpea and to alleviate the effects of salt stress.  相似文献

Siratro (Macroptilium atropurpureum (DC) Urb.) is a vigorous perennial forage legume with good potential for improving pastures in the extensive neotropical regions of the world. It is well adapted to a wide range of soil and climatic conditions. The objective of these studies was to determine effects of Glomus fasciculatum colonization, rigorous defoliation, and soil fertility treatments to a Psammentic Paleustalf (Eufaula) soil on growth, regrowth, nodulation, and nitrogenase activity (C2H2 red.) of Siratro inoculated with Rhizobium leguminosarum Frank. Top growth increased significantly with soil K and P amendment and with mycorrhiza colonization. Nodulation and nitrogenase activity were correlated with highly significant increases from G. fasciculatum, P treatments and K additions to 300 mg K kg‐1 soil. Growth and peduncles of nonclipped plants increased about 4 fold from 90 to 225 day age with mature seed yield increasing about 10 fold; nodule mass and nitrogenase activity levels approximately doubled. Regrowth response of plants defoliated at 45 day intervals, following their initial 90 day age, was somewhat constant between clippings for magnitude of regrowth 12.3–13.8g, development in number of peduncles 4.0–6.8, seed yield 1.4–2.6g, nodulation 2.9–3.7g, and nitrogenase activity 73.9–95.8μ mol C2H4g‐1 nodule. Multiple regression for nitrogenase = 0.55 g top wt. + 0.63 g nodule wt. + 1.91 day age ‐ 0.07 peduncle no., R2 = 0.85 and C.V. = 14.3%. Favorable tripartite symbiosis with both effective Rhizobium and endophyte mycorrhiza were essential for high levels of symbiotic nitrogen fixation.  相似文献

A field experiment was conducted to evaluate the effect of iron (Fe) chlorosis on the mineral composition of lemon tree leaves cultivated in calcareous soil in the region of Murcia, Spain. A Fe‐polyflavonoid was employed as fertilizer to control Fe deficiency. The evolution of the leaf content in Fe, potassium (K), sodium (Na), magnesium (Mg), calcium (Ca), phosphorus (P), manganese (Mn), and zinc (Zn) was monitored during all the sampling cycle. A high correspondence between total Fe and chlorophyll content could be appreciated. A multivariance analysis was performed with the data obtained, showing that Fe was correlated with P, Mn, and K.  相似文献

Plant growth-promoting bacteria (PGPB) Pseudomonas lurida-NPRp15 and Pseudomonas putida-PGRs4 possessing multiple plant growth-promoting traits were isolated from rhizoplane of pea and rhizosphere of garlic, respectively. The effects of individuals and combinations of Pseudomonas spp. with effective root nodulating symbiotic nitrogen fixing Rhizobium leguminosarum-FB1 on plant growth, nutrient uptake and yield of the rajmash plant were studied under greenhouse conditions. Bacterial inoculation resulted in significantly higher values for plant dry biomass, N, P, K, Zn and Fe contents as compared to the uninoculated control. Furthermore, dual inoculation of P. lurida-NPRp15 with R. leguminosarum-FB1 significantly increased root and shoot dry weight, nodulation, nutrient uptake, pod yield, and nutrient content of pods of rajmash VL63 compared to controls, single and triple inoculation. The results of the study indicate the potential of harnessing the benefit of plant growth-promoting and nitrogen-fixing microorganisms to improve the growth and yield of rajmash.  相似文献

Abstract

Methyl jasmonate (MeJA) has recently been shown to act as a plant-to-bacteria signal. We tested the hypothesis that pre-induction of Rhizobium leguminosarum bv. phaseoli cells with genistein and/or MeJA would at least partially overcome the negative effects of low root zone temperature (RZT) on bean nodulation, nitrogen fixation and plant growth. Otebo bean plants were grown at constant air temperature (25oC) and two RZT regimes (25 and 17oC) and inoculated with R. leguminosarum bv. phaseoli pre-induced with MeJA and/or genistein. Our results indicate that low RZT inhibited nodulation, nitrogen fixation and plant growth. The plants growing at low RZT began fixing nitrogen seven days later compared to those at higher RZT. The low RZT plants had fewer nodules, lower nodule weight, less N fixation, slower plant growth, fewer leaves, smaller leaf area, and less dry matter accumulation comared to plants at a higher RZT. Rhzobium leguminosarum bv. phaseoli cells induced with genistein and/or MeJA enhanced bean nodulation, nitrogen fixation and growth at both optimum and suboptimum RZTs. The results of this study indicate that MeJA improves bean nitrogen fixation and growth at both optimum and suboptimum RZTs, and can be used alone or in combination with genistein to partially overcome the low RZT induced inhibitory effects on nodulation and nitrogen fixation.  相似文献

Abstract

Characteristics of Brown Forest soils developed under different bio-climatic conditions from low to high eleyations in the Kinki District were studied with special reference to their pedogenetic processes. The Brown Forest soils at high elevations were characterized by a lower bulk density, higher capacities to adsorb organic matter, phosphate, and moisture, which were correlated with the ratio of the amorphous content to the clay content (the value of the ratio of (Feo + Alo)/clay), as compared to those at low elevations. Considering the fact that the value of the (Feo + Alo)/clay ratio was not correlated with the volcanic glass index, the formation of an amorphous fraction at high elevations was considered to proceed according to the following mechanism.

Low temperature at high elevations (above 700 m) may retard the crystallization of oxide minerals. The amorphous oxides with variable positive charge thus formed may adsorb organic matter, confering a darker color and high moisture and high phosphate retention capacities to the subsoil. Adsorbed organic matter stabilizes these amorphous oxides, thus enhancing the amorphous properties and inhibiting crystallization. A1 translocation due to the weak podzolization may contribute to the increase of the content of amorphous materials.

Characterization of the B horizons in forest soils in Japan, in terms of the values of the ratios of (Feo + Alo)/clay, (Fed-Feo)/Fet, and Alo/Ald, (CEC -ECEQ/CEC and STPT-ZPC, suggested that forest soils might be classified into four groups.  相似文献

ABSTRACT

Common bean (Phaseolus vulgaris L.) is relatively poor in dinitrogen (N2) fixation, so selecting compatible host cultivar and Rhizobium strain combinations may offer an improvement. The effectiveness of six rhizobial strains was evaluated using five bean cultivars of bean (three pinto and two black bean) in a growth-room experiment. We subsequently selected the three best strains to assess whether multi-strain inoculation had advantages over single-strain inoculation in growth-room and field experiments. In the first-growth-room experiment, Rhizobium strains UMR 1899, RCR 3618, and USDA 2676 were selected for high nodulation, plant dry weight, shoot nitrogen (N), and N2 fixation. In a second growth-room experiment, the individual strains and a mixture of the three strains generally did not differ in the parameters evaluated. Total shoot N accumulated ranged from 172.9 to 162.8 mg plant?1, of which 32.1% to 33.6% (equivalent to 54.0 to 59.2 mg plant? 1) was fixed. In field experiments, plant biomass and seed N2 fixed did not differ among the inoculants at any site. These results suggest that the three strains were equally effective and that the multi-strain inoculant offered no consistent advantage over the single-strain inoculants.  相似文献

4种生物质对大豆双共生系统、土壤微生物及产量的调控   总被引:1,自引:1,他引:0  
通过田间试验研究施入生物质对根瘤菌、丛枝菌根真菌与大豆双共生系统、土壤微生物及大豆产量的影响。生物质设置秸秆、土豆皮、食用菌废弃料、大豆浸出液4个处理。分别在植株生长第4片复叶展开(V5)、结荚始期(R3)和鼓粒盛期(R6)取样测定。研究结果表明: 外施4种生物质对大豆双共生系统均有不同程度促进作用, 且生物质对大豆菌根形成的促进作用较对根瘤形成的促进更显著, 持续时间更长。在4种生物质中, 秸秆处理表现效果最佳, 不仅对大豆双共生系统、土壤细菌、真菌数量有增效作用, 同时对大豆产量有协同促进作用。在R6期大豆有效根瘤数量、丛枝菌根真菌侵染率、土壤细菌数量分别较对照增加238.46%、26.28%、131.99%, 产量较对照增加69.78%, 且秸秆容易获得, 施用方便, 是有效提高大豆产量的生物质。大豆浸出液处理对大豆双共生系统促进显著, 在R6期有效根瘤数量较对照增加89.74%, 同时丛枝菌根真菌侵染率较对照增加31.23%, 但由于此处理播种前采用浸种方式, 对大豆子叶造成较大损伤, 使保苗率严重降低从而导致产量相对较低。食用菌废弃料和土豆皮处理在3个生育时期对大豆双共生系统的作用不尽一致, 但R6期2个处理的丛枝菌根真菌侵染率和土壤中真菌数量都极显著高于对照。  相似文献

快生型大豆根瘤菌G113是从吉林省、公主岭黑土上生长的野生大豆G3的根瘤中分离获得。野生大豆G3在有快慢生型大豆根瘤菌同时存在时,选择G113和其共生。G113菌株和吉林20大豆共生优于轻碱土中的土著大豆根瘤菌。因此在轻碱土上、吉林20大豆接种G113菌株有增产趋势。  相似文献
