This study characterized the genetic diversity of 18 Tunisian fig cultivars using random amplified polymorphic DNA (RAPD) and inter simple sequence repeats (ISSR). Both random and ISSR primers tested generated a total of 116 RAPD and 47 ISSR markers. Considerable genetic variation was observed among fig cultivars sampled from two regional Tunisian collections with an average diversity of 4.57. RAPD and ISSR banding patterns and genetic distances values reflected the high level of diversity among the collections and lower variability between the two collections. The correlation between the RAPD and ISSR similarity matrices computed for the 153 pairwise comparisons among the 18 varieties was lower and significant. An analysis of molecular variance showed that 92% of the total genetic diversity resided within collections, whereas only 8% between collections. The results indicated that in the local fig germ plasm the information provided by RAPD and ISSR is not analogous, most likely as a consequence of the fact that the two classes of markers explore, at least in part, different portions of the genome.  相似文献

This study evaluated the effects of barley inclusion and glucanase supplementation on the productive performance and digestive function in laying ducks. The experiment used a randomized design with a 5 × 2 factorial arrangement of 5 graded levels of barley (0%, 15%, 30%, 45% and 60%) with or without 1.5 g/kg β-1,3-1,4-glucanase (15,000 U/kg). During the experimental period of 120 d, the weight and total number of eggs within each pen were recorded daily, and egg quality was determined every 4 wk. At the end of the experiment, 3 randomly selected ducks within each replicate were sacrificed, then duodenal digesta and jejunal mucosa was collected. Dietary inclusion of barley had no effects on egg production, daily egg mass or FCR, but supplementation with glucanase improved egg production and FCR (P < 0.01). Barley did not affect feed intake of laying ducks, but glucanase tended to increase feed intake (P = 0.09). Neither barley nor β-glucanase had effects on the egg quality variables, except for yolk color score, which was decreased with increasing barley supplementation. Glucanase, but not barley, increased the activity of chymotrypsin and amylase in duodenal digesta. Barley inclusion affected the activity of alkaline phosphatase and maltase in jejunal mucosa (P < 0.05), but β-glucanase had no effects on the activity of these brush border enzymes. Barley inclusion increased the glucan content in duodenal digesta, but supplementation of glucanase to barley-based diet reduced digesta glucan content and reduced total volatile fatty acids and increased the proportion of acetic acid in cecal contents. The results indicate that, without glucanase, the optimal dietary barley level in the diets of laying ducks is about 13% for maximal production performance; glucanase supplementation of the barley diets improved production performance, probably through enhancing digestive function.  相似文献

The pharmacokinetics of marbofloxacin (MAR) was compared in geese (Anser Anser domesticus) after single intravenous (IV) and intramuscular (IM) (thigh and pectoral muscles) administrations of 5 mg/kg. Serum concentrations of MAR were determined with high-performance liquid chromatography (HPLC) method. Serum MAR concentrations versus time were analyzed by a noncompartmental method. After IV administration, MAR showed high volume of distribution at steady state (Vdss) of 5.24 ± 1.08 L/kg. The serum body clearance (Cl) and elimination half-life (T1/2λz) of MAR were 0.79 ± 0.07 L hr−1 kg−1 and 6.94 ± 1.12 hr, respectively. The peak of MAR serum concentrations Cmax achieved at one and 0.50 hr after thigh and pectoral IM sites of injections, respectively, were 1.20 and 0.91 μg/ml. Significant differences were found in the mean absorption time (MAT), the systemic bioavailability (F%), and elimination parameters of MAR between two sites of injections, indicating that the absorption was fairly slow and complete after thigh IM injection. The pharmacokinetics of MAR in geese diverged according to the site of IM injection following a parallel study design. We recommend the thigh muscle as IM site of injection to obtain maximum concentrations of the administered drug in geese.  相似文献

Background: Negative energy balance during late pregnancy in ewes is an important cause of hyperketonemia. Ketone bodies can generate superoxide radicals and cause oxidative stress and cellular dysfunction, as noted in cows with subclinical ketosis or in diabetic people. Objective: The aim of this study was to evaluate the role of hyperketonemia in initiating the process of lipid peroxidation. Methods: The study included 10 pregnant ewes (aged 3.5–6 years) with pregnancy toxemia, 10 clinically healthy pregnant ewes, and 10 clinically healthy nonpregnant ewes. Serum concentrations of β‐hydroxybutyrate (BHB), cortisol, and glucose, plasma activities of glutathione peroxidase, superoxide dismutase, and catalase, and plasma concentrations of thiobarbituric acid reactive substances (TBARS), markers of lipid peroxidation, and reduced glutathione were measured. Data from the 3 groups were statistically analyzed and compared. Results: Serum concentrations of BHB, cortisol, and TBARS were significantly higher in ewes with pregnancy toxemia when compared with concentrations in healthy pregnant and nonpregnant groups (P≤.05). In ewes with pregnancy toxemia, a strong positive correlation was found between concentrations of TBARS and BHB (r=.80; P=.002) and between concentrations of BHB and cortisol (r=.76; P=.005). Conclusions: Oxidative stress and lipid peroxidation are involved in the development and complications of pregnancy toxemia. An association between hyperketonemia and the products of lipid peroxidation has also been demonstrated, suggesting that ketosis is a risk factor in the development of lipid peroxidation and oxidative stress in ewes affected by pregnancy toxemia.  相似文献

This study was performed for the purpose of investigating the prevalence of Sarcocystis spp. in buffaloes in Beni-Suef Governorate, Egypt. Both macroscopic (Sarcocystis fusiformis) and microscopic (Sarcocystis levinei) cysts were recognized, and were differentiated by their morphological features and location in the tissues. Of 379 buffaloes examined in abattoirs in Beni-Suef, 299 were found to be infected, with an overall prevalence of 78.9%. Depending on age, three categorized groups of naturally infected buffaloes were examined: male buffalo calves aged 1.5–2 years, adult females aged 2–5 years, and females older than 5 years. Among these groups, infection rates were 74.5%, 82.3%, and 81.2%, respectively. Organs examined included esophagus, tongue, and heart. Macroscopic cysts were examined by the naked eye through meat inspection in abattoirs, while the pepsin-digestion method and the histological technique were applied to detect microscopic cysts. It has been found that esophagus showed the highest rate of infection among the infected organs, with both macroscopic and microscopic cysts seen in the infected buffaloes. Moreover, results of the pepsin-digestion method proved more accurate than those produced by the histological technique in terms of infection rates for the microscopic cysts. Our findings indicated that infected buffaloes aged 2–5 years showed the highest mixed infection rate (82.3%) for both types of cysts. The high prevalence of microscopic Sarcocystis spp. in Beni-Suef Governorate reflects a significant role played by stray dogs, rather than cats, in the transmission of these parasites.  相似文献

The technique of microwave-assisted acid hydrolysis was applied to wholegrain wheat (Triticum durum Desf. cv. Balcali 2000) flour in order to speed the preparation of samples for analysis. The resultant hydrolysates were chromatographed and quantified in an automated amino acid analyzer. The effect of different hydrolysis temperatures, times and sample weights was examined using flour dispersed in 6 N HCl. Within the range of values tested, the highest amino acid recoveries were generally obtained by setting the hydrolysis parameters to 150 °C, 3 h and 200 mg sample weight. These conditions struck an optimal balance between liberating amino acid residues from the wheat matrix and limiting their subsequent degradation or transformation. Compared to the traditional 24 h reflux method, the hydrolysates were prepared in dramatically less time, yet afforded comparable ninhydrin color yields. Under optimal hydrolysis conditions, the total amino acid recovery corresponded to at least 85.1% of the total protein content, indicating the efficient extraction of amino acids from the flour matrix. The findings suggest that this microwave-assisted method can be used to rapidly profile the amino acids of numerous wheat grain samples, and can be extended to the grain analysis of other cereal crops.  相似文献

Purpose

Gasification process is regarded as a viable option for disposal of various types of organic waste. Its application to meat and bone meal (MBM) and dried distillers grains (DDG) is a new concept that is recently receiving more attention as possible strategy for safe recycling of these materials to benefit from their energy and nutrient value. Besides, this process produces ash that is rich in phosphorus (P) and may be suitable for utilization as a P fertilizer. Therefore, the objective of this study was to evaluate the effectiveness of these specific types of ashes as P fertilizer via evaluating their direct effect on canola growth, P uptake, apparent P recovery and changes in some selected soil chemical properties after their addition to a P-deficient soil.

Materials and methods

A growth chamber experiment was set up to meet the study objective. The experimental treatments included meat and bone meal ash (MBMA) and dried distillers grains ash (DDGA) applied at three rates (25, 50, or 100 kg P ha?1) in comparison to a mineral (mono-calcium phosphate) fertilizer (MP) applied at the same rates in addition to non-P-treated soil (control).

Results and discussion

After a growth period of 5 weeks, the DDGA was the most effective ash type and provided biomass yield, P uptake, and apparent P recovery better or similar to that of MP, indicating high availability of its P. The MBMA had a limited effect on measured crop variables, suggesting that a significant portion of this ash P is present in insoluble form and is not as readily available for plant uptake. This was also indicated by its lesser effect on enhancing extractable available P remaining in soil after harvest in comparison to MP or DDGA. Application of all ash material caused a slight but significant change in soil content of inorganic N as well as soil pH and EC; however, this change was more evident with DDGA treatments.

Conclusions

Ash derived from gasified DDG was the most effective P fertilizer and was comparable to mineral fertilizer. The results of this study demonstrated that the effectiveness of organic material ash as a P fertilizer is controlled by the type of gasified feedstock. The positive results obtained from this study should stimulate further research on utilization of these ashes as a source of P for different crops in different soil types, especially repeated application under field conditions.  相似文献
