ABSTRACT Squid processed products such as dried-seasoned squid turn brown during storage, causing a change in quality. The main cause of this browning is melanoidin produced by the Maillard reaction. In this study, four types of squid were processed at different heating temperatures between 50 and 90°C and examined for suppression of the production of D-ribose, the main component of melanoidin. Furthermore, the color change of the squid meat with each heat treatment during storage of 32 days at 30°C was investigated. Bigfin reef squid and cuttlefish with a relatively high freshness decreased D-ribose generation as the heating temperature increased, and the browning during storage also decreased accordingly. In contrast, Humboldt squid with a low freshness did not generate new D-ribose at any heating temperature during storage. Nevertheless, the browning was reduced as the heating temperature increased. It is thought that the Maillard reaction was also reduced by the heat denaturation of muscular proteins. These results suggest that browning due to the Maillard reaction of squid meat is greatly affected by initial freshness and heating temperature. 相似文献

Mammalian sexual fate is determined by the presence or absence of sex determining
region of the Y chromosome (Sry) in the “bipotential” gonads.
Recent studies have demonstrated that both male and female sexual development are induced
by distinct and active genetic pathways. Breeding the Y chromosome from Mus m.
domesticus poschiavinus (POS) strains into C57BL/6J (B6J) mice
(B6J-XYPOS) has been shown to induce sex reversal (75%: bilateral ovary, 25%:
true hermaphrodites). However, our B6N-XYPOS mice, which were generated by
backcrossing of B6J-XYPOS on an inbred B6N-XX, develop as males (36%: bilateral
testis with fertility as well as bilateral ovary (34%), and the remainder develop as true
hermaphrodites. Here, we investigated in detail the expressions of essential sex-related
genes and histological features in B6N-XYPOS mice from the fetal period to
adulthood. The onsets of both Sry and SRY-box 9 (Sox9) expressions as determined
spatiotemporally by whole-mount immunohistochemistry in the B6N-XYPOS gonads
occurred 2–3 tail somites later than those in B6N-XYB6 gonads, but earlier than
those in B6J-XYPOS, respectively. It is possible that such a small difference
in timing of the Sry expression underlies testicular development in our
B6N-XYPOS. Our study is the first to histologically show the expression and
ectopic localization of a female-related gene in the XYPOS testes and a
male-related gene in the XYPOS ovaries. The results from these and previous
experiments indicate that the interplay between genome variants, epigenetics and
developmental gene regulation is crucial for testis development. 相似文献

A portion of the minute chylomicrons less than 75 nm in diameter are transcytosed from
the extravascular tissue into the subepithelial blood capillaries (sBC) in the villous
apices of the rat jejunum. However, the details of the transportation mechanism have not
been clarified. In this study, the endothelial receptor involved in the transportation of
minute chylomicrons into the sBC’s lumina was immunohistochemically and
histoplanimetrically examined in intestinal villi of the rat jejunum. Immunopositivity for
very low density lipoprotein (VLDL) receptor was detected on the luminal and basal
surfaces of the endothelial cells of sBC in approximately 68% of those apices of jejunal
villi that possessed numerous chylomicrons in the lamina propria, while VLDL receptor was
detected on the endothelial cells of sBC in only approximately 8% of intestinal villi that
possessed few or no chylomicrons in the lamina propria. No immunopositivity for LDL
receptor was detected in the sBC of all intestinal villi. These findings suggest that VLDL
receptor is expressed by the endothelial cells of the sBC in conjunction with the filling
of the lamina propria of jejunal villi with many chylomicrons produced by the villous
columnar epithelial cells and that the VLDL receptor mediates the transportation of minute
chylomicrons, maybe VLDL, into the subepithelial portal blood from the extravascular
tissue of the rat jejunal villi. 相似文献

Effect of element type on the internal bond quality of wood-based panels determined by three methods
Three mechanical tests with different loading modes were conducted to evaluate the effect of element type on the internal bond quality of wood-based panels. In addition to the internal bond test, which is commonly used for mat-formed panels, interlaminar and edgewise shear tests were used to test oriented strandboard (OSB), particleboard, medium-density fiberboard (MDF) of two thicknesses, and plywood. The following results were obtained. Epoxy resin proved to be suitable for determining the interlaminar shear modulus instead of hot-melt glue. There was a linear relation between panel density and interlaminar shear modulus and a linear correlation between the interlaminar shear strength and internal bond (IB) strength for the mat-formed panels tested. OSB had the highest edgewise shear modulus, and MDFs had the highest edgewise shear strength in this study. The modulus/strength ratio also depended on both panel type and loading mode. The relation between the shear moduli determined from the edgewise and interlaminar tests indicated the characteristics of the shear properties of panels made of different elements.Part of this paper was presented at the Fourth International Wood Science Symposium, Serpong, Indonesia, September 2002 相似文献

After a cell wall protein fraction (CWP) of Pythium oligandrum was sprayed on sugar beet leaves, we screened leaves for induced expression of defence-related genes and for resistance against
Cercospora leaf spot. In a western blot analysis, the CWP was primarily retained on the surface of leaves without degradation
for at least 48 h after spraying. In northern blot analyses, four defence-related genes (β-1, 3-glucanase, acidic class III
chitinase, 5-enol-pyruvylshikimate-phosphate synthase and oxalate oxidase-like germin) were expressed more rapidly in CWP-treated
leaves compared to control leaves treated with distilled water (DW). When CWP was applied to a suspension of cultured cells
of sugar beet, an oxidative burst was observed that did not occur after the DW treatment. In growth chamber trials after inoculation
with Cercospora beticola, the severity of Cercospora leaf spot was significantly reduced in CWP-treated plants compared to the DW-treated controls.
In a field experiment, CWP treatment was also effective against the disease. CWP did not reduce growth rate of the pathogen
in plate tests. The results together suggest that the CWP from P. oligandrum can be retained on the leaf surface and induce expression of disease resistance genes, thereby reducing Cercospora leaf spot

Nitrogen and carbon dynamics in paddy and upland soils for rice cultivation and in upland soil for corn cultivation was investigated by using 13C and 15N dual-labeled cattle manure compost (CMC). In a soil with low fertility, paddy and upland rice took up carbon and nitrogen from the CMC at rates ranging from 0.685 to 1.051% of C and 17.6–34.6% of N applied. The 13C concentration was much higher in the roots than in the plant top, whereas the 15N concentration differed slightly between them, indicating that organic carbon taken up preferentially accumulated in roots. The 13C recovery in the plant top tended to be higher in upland soil than in paddy soil, whereas 15N applied was recovered at the same level in both paddy and upland soils. In the experiment with organic farming soil, paddy rice took up C and N from the CMC along with plant growth and the final recovery rates of 13C and 15N were 2.16 and 17.2% of C and N applied. In the corn experiment, a very large amount of carbon from the CMC was absorbed, accounting for at least 7 times value for rice. The final uptake rates of 13C and 15N reached about 13 and 10% of C and N applied, respectively. Carbon emission from the CMC sharply increased by 2 weeks after transplanting and the nitrogen emission was very low. It is concluded that rice and corn can take up an appreciable level of carbon and nitrogen from the CMC through roots. 相似文献

Ixodes persulcatus Schulze (I. persulcatus) is distributed in Russia and Far East Asia including Japan, and has been implicated as the vector of several human pathogens. In particular, I. persulcatus acts as the only tick vector for human lyme borreliosis in Japan. In order to elucidate the mechanism of transmission of I. persulcatus-borne pathogens, we developed a laboratory colony of I. persulcatus. Ticks were fed on Syrian hamster and engorged ticks that had dropped off the animals were collected and maintained to allow them to molt. Tick rearing was performed in incubator at 20 degrees C with 95% relative humidity and 12-hour light/dark photo-period regimen. We found out that adult females fed for 8 +/- 2 days and had a pre-oviposition period lasting for 7 +/- 2 days. The minimum egg incubation period was 1 month with the hatched larvae feeding for 3 +/- 1 days and molting to nymphs 3-4 months thereafter. Meanwhile, the nymphs fed for 4 +/- 1 days and molted to adult 2-3 months thereafter. For future analysis of gene expression profiles in I. persulcatus, we cloned and sequenced the actin gene (a housekeeping gene), and found that it is 92.7% to 98.6% homologous to the published sequences of related ixodid ticks. This laboratory colony of I. persulcatus will facilitate investigations on the role of tick-derived molecules on the transmission of I. persulcatus-borne pathogens and will be important for identification of potential anti-tick vaccine and acaricide target molecules. 相似文献

Gastric-derived peptide hormone ghrelin is known for its potent growth hormone (GH) stimulatory effects. The acyl-modification on N-terminal Ser(3) residue is reported to be important to stimulate the ghrelin receptor, GH secretagogue-receptor type1a (GHS-R1a). However, major portion of circulating ghrelin lacks in acylation, and some biological properties of des-acyl ghrelin have been reported in monogastric animals. In the present study, the responsiveness of plasma hormones and metabolites to ghrelin in steers was characterized, and role for des-acyl ghrelin in these changes was investigated. The repeated intravenous administrations of bovine ghrelin (1.0 microg/kg BW) every 2h for 8h to Holstein steers significantly increased the plasma acylated ghrelin, total ghrelin, GH, insulin and NEFA levels. The GH responses in peak values and area under the curves (AUCs) were attenuated by repeated injections of ghrelin, however, the responses of plasma total ghrelin were similar. Plasma insulin AUC decreased after fourth injection of ghrelin while plasma NEFA AUCs gradually increased by repeated injections of ghrelin. Pretreatment of des-acyl ghrelin (10.0 microg/kg BW) 5 min prior to the single injection of ghrelin (1.0 microg/kg BW) did not affect the ghrelin-induced hormonal changes. Moreover, the responses of plasma GH to bovine and porcine ghrelin, which differ in C-terminal amino acid residues, were similar in calves. These data show that (1) GH release was attenuated by repeated administration of ghrelin, (2) ghrelin regulates glucose and fatty acid metabolism probably via different pathway, and (3) des-acyl ghrelin is unlikely the antagonist for ghrelin to induce endocrine effects in Holstein steers. 相似文献
