The serum and fecal testosterone (T) concentrations and testicular sizes of two male bharals (Pseudois nayaur) were determined for approximately one year. The profiles of the fecal T concentrations showed a similar tendency as the profiles of serum T concentrations, and there was a significant correlation between serum and fecal T concentrations (r=0.72). T concentrations rose drastically in October and decreased gradually until January. The maximum testicular size was observed between November and January. Semen collected between December and January was excellent in quality and comparable to domestic sheep and goats. The active periods of the testes were synchronized with the early breeding season of females.  相似文献

In vitro fertilization (IVF) is widely used in reproduction research, but the sperm of some inbred strains of mice yield low fertilization rates in IVF. To determine the cause of this problem, we examined the effect of epididymal sperm morphology, in particular, tail bending and the presence and type of cytoplasmic droplet (CD), on fertilizability in vitro. Sperm suspensions were obtained from the following five strains: C57BL/6J, BALB/cA, C3H/HeN, DBA/2J, and 129 x 1/SvJ. The sperm were fixed in 10% formalin and three parts of the sperm, namely the head, tail, and CD, were examined. We recorded the proportion of abnormal sperm heads and hairpins at the neck; tails were categorized as straight, proximal bent, or distal bent; and the CDs were categorized as none, light-type, and heavy-type. Based on these parameters, we determined the correlations between sperm morphology and fertilizability in vitro, as judged by IVF using ICR oocytes. The proportion of sperm with a hairpin neck was higher in strain C57BL/6J, while abnormal head morphology occurred significantly more often in strain BALB/cA. The percentage of sperm with a proximal bent tail was highest in strain DBA/2J and lowest in strain 129 x 1/SvJ. A heavy-type CD was observed more frequently in the 129 x 1/SvJ and C57BL/6J strains than in the other three strains in which a light-type CD predominated. The rank order of the fertilization rates was 129 x 1/SvJ < C57BL/6J < C3H/HeN < BALB/cA < DBA/2J. In addition, fertilization rate was positively correlated with a proximal bent tail, but negatively correlated with a heavy-type CD and distal bent tail. This new classification system establishes that the morphological characteristics of epididymal sperm differ among inbred strains of mice and that tail and CD morphology are closely related to fertilization ability in IVF. Thus, our results provide a novel method for assessing the quality of mouse sperm used for IVF.  相似文献

A 1-year- and 11-month-old spayed female toy poodle had showed progressive ataxia and paresis in the hindlimbs since 11 months old. Magnetic resonance imaging revealed high signal intensity on T2-weighted and fluid-attenuated inversion recovery images at the thoracic and lumbar spinal cord. The dog’s neurological condition slowly deteriorated and flaccid tetraparesis was exhibited. At 4 years and 11 months old, the dog died of respiratory failure. On postmortem examination, eosinophilic corkscrew bundles (Rosenthal fibers) were observed mainly in the thoracic and lumbar spinal cord. Histological features were comparable to previously reported cases with Alexander disease. This is a first case report to describe the clinical course and long-term prognosis of a dog with Alexander disease.  相似文献

To evaluate the coexistence of agricultural managements and wetland ecosystem conservation, the Bibai mire, an ombrotrophic mire in Hokkaido, Japan, was selected as an experimental site that had been affected by neighboring agricultural managements. Since the lowering of the level of the groundwater table in the peripheral area of the mire had threatened indigenous vegetations, keeping the groundwater table shallow by trench irrigation seemed to be an effective measure to recover the original vegetation. The objective of the present study was to quantify the amount of water and the effective area of trench irrigation required in a bamboo-invading area in a pristine mire. We constructed a trench 28 m long and irrigated at the rate of 2.22 m3 d−1 in a bamboo-invading area in the mire. And to analyze the hydro-meteorological conditions under the trench irrigation, we measured the saturated hydraulic conductivity of the peat layer (3.8 × 10−3 cm s−1), the evapotranspiration rate (2.80 mm d−1), the depth of the non-irrigated groundwater table (0.15 m) and the surface gradient (0.00493). In addition, using the mass conservation equation and Darcy's law, we derived a steady state model of the level of the groundwater table formed by trench irrigation, which required the five parameters mentioned above. The irrigated water spread over a distance of only about 15 m to both sides of the trench. The model also estimated that the distance for the irrigated area was 14.6 m and we concluded that the irrigated area was limited within a distance of 20 m distances to both sides of the trench and that the irrigation rate per unit trench length did not exceed 0.12 m2 d−1 for realistic values of the evapotranspiration rate and the saturated hydraulic conductivity in peatland.  相似文献

Five compounds oxidizing canine erythrocytes were isolated from an aqueous ethanol garlic extract by silica gel column chromatography and preparative thin-layer chromatography. On the basis of nuclear magnetic resonance, infrared spectroscopy, and mass spectrometry, they were identified as three known compounds: bis-2-propenyl trisulfide (1), bis-2-propenyl tetrasulfide (2), and bis-2-propenyl pentasulfide (3) as well as two novel compounds, bis-2-propenyl thiosulfonate (4) and trans-sulfuric acid allyl ester 3-allylsulfanyl-allyl ester (5). A mixture of compounds 1-3 and compounds 4 and 5 induced methemoglobin formation in canine erythrocyte suspension in vitro resulting in the oxidation of canine erythrocytes. These groups of characteristic organosulfur compounds contained in garlic probably contribute to oxidations in blood. The constituents of garlic have the potential to oxidize erythrocytes and hemoglobin, suggesting that foods containing quantities of garlic should be avoided for feeding dogs.  相似文献

Effects of food materials were investigated on removal of several kinds of thiols, sulfides, and disulfides, which arise from vegetables of Allium species during food preparation and eating. Methanethiol, propanethiol, and 2-propenethiol were captured by raw foods such as fruits, vegetables, and mushrooms or a mixture of their acetone powders and phenolic compounds. The odor of diallyl disulfide was remarkably reduced by kiwi fruit, spinach, cutting lettuce, parsley, basil, mushrooms, and, particularly, cow's milk, raw egg, boiled rice, and bovine serum albumin (BSA). This suggests that the removal of diallyl disulfide could be caused by a physical and chemical interaction between the disulfide and foods. Furthermore, milk and BSA captured propanethiol, 2-propenethiol, dipropyl sulfide, diallyl sulfide, dimethyl disulfide, and dipropyl disulfide very well. An enzymatic degradation of diallyl disulfide by spinach and asparagus was also observed. These results demonstrate that the deodorization with foods is achieved by multiple actions including physical and chemical interaction between volatile sulfur compounds and foods, enzymatic degradation of disulfides, and addition of thiols to polyphenolic compounds, catalyzed by polyphenol oxidases or peroxidases.  相似文献

Target therapy using the tyrosine kinase inhibitor imatinib is one of the new therapeutic approaches for canine mast cell tumors (MCTs). In the present report, we demonstrate a clinical response to imatinib in a dog with MCT carrying a c-kit c.1523A>T mutation. Moreover, the effect of this mutation on the phosphorylation status of KIT and the inhibitory potency of imatinib on the phosphorylation of the mutant KIT were examined in vitro. A dog with a MCT tumor mass on the right forelimb sole with lymph node metastasis and mastocytemia was treated with imatinib. The MCT mass markedly shrank and mastocytemia became undetectable with 2 weeks of treatment. The lymph node enlarged by metastasis became normal in size with 5 weeks of treatment. From the sequencing analysis of c-kit in tumor cells, a substitution mutation c.1523A>T that alters the amino acid composition (p.Asn508Ile) within the extracellular domain of KIT was identified. The mutant KIT expressed on 293 cells showed ligand-independent phosphorylation and imatinib suppressed this phosphorylation in a dose-dependent manner. From these findings, imatinib was considered to elicit a clinical response in a canine case of MCT via inhibition of the constitutively activated KIT caused by a c-kit c.1523A>T mutation.  相似文献

A genomic library of Xanthomonas oryzae pv. oryzae (X. o. pv. oryzae) T7174 was screened for 4-methylumbelliferyl β-D-glucoside-hydrolyzing (MUGase) activity. In subcloning of one of the MUGase-positive clones, an approximately 4.2-kb SacI-SphI fragment conferred not only MUGase activity but also 4-methylumbelliferyl β-D-cellobioside-hydrolyzing (MUCase) activity. Sequence analysis showed that the fragment contained an ORF of 2951 bp. The conceptual ORF product was significantly homologous with 1,4-β-D-glucan glucohydrolase D (CELD) from Pseudomonas fluorescens subsp. cellulosa, and was named CELDXo. Cell fractionation experiments suggested that CELDXo is localized in the cell-envelope fraction. We constructed a CELDXo-deficient mutant (74ΔCELD) from X. o. pv. oryzae. Little MUCase activity was detected in the cell-envelope fraction prepared from the mutant. The mutant 74ΔCELD did not grow in synthetic medium containing cellobiose as the sole sugar source. On the other hand, growth in rice leaves and pathogenicity of the mutant and the parental strain did not differ. These results suggested that CELDXo is involved in cellobiose utilization of X. o. pv. oryzae but that the gene is not required for bacterial growth in rice leaves. For 69 methyl N-phenylcarbamates having various benzene ring substituents, the fungicidal activity was determined against Botrytis cinerea resistant to benzimidazole fungicides by the agar medium dilution method. The structure-activity relationships were analyzed quantitatively using such physicochemical substituent parameters as hydrophobic π, steric B5, and HB (hydrogen bonding) with the Hansch-Fujita method (C. Hansch and T. Fujita, J. Amer. Chem. Soc.86, 1616 1964). The hydrophobicity of substituents was favorable to the acitivity. The effect was position-specific, the importance being in the order of ortho meta > para. The activity was related parabolically to the maximum width of one of the m-substituents having the larger dimension. The hydrogen acceptability of p-substituents enhanced the activity. The preventive activity of compounds against gray mold of cucumber caused by the resistant B. cinerea, which was determined by the foliar application in pot tests, was dependent on the magnitude of the fungicidal activity and hydrophobicity of the compounds as analyzed by the adaptive least-squares method.  相似文献

Functional traits are an important aspect of long‐term breeding strategies for dairy cattle. In this regard, it is necessary to develop simple methods for estimating the economic value of herd life. In this study, the economic daily value of herd life was estimated when survival rate varied between ?0.05 and 0.05 from the basal survival rate. The extension days per survival rate were 26.5 days in Hokkaido and 20.3 days in other regions. The increases in values of annual income per day of herd life were 95.18 yen in Hokkaido and 101.80 yen in other regions. The relative economic weights of milk yield to herd life per genetic standard deviation were 0.668 in Hokkaido and 1.03 in other regions. Estimated increments in yearly profits based on young sire selection for herd life were 963 yen in Hokkaido and 1,030 yen in other regions. The estimated increments in annual profits based on young sire selection for milk yield were 1,268 yen in Hokkaido and 2,097 yen in other regions. Given that economic value was linearly correlated with herd‐life length, the linear regression coefficients between these factors could be used to estimate the economic value of herd‐life length.  相似文献
