Urea (U), sodium hypochlorite (H) and sodium chloride (S) alone or combined were applied upon high‐fibre forages (HFF) in three experiments. Experiment (Exp) I: tall wheatgrass (TW), weeping lovegrass (WL), deferred sorghum (DS) and barley straw (BS) as substrates were untreated (U0) or treated with 4 g 100 g?1 DM (U4), and stored for 45 days. Within each forage, CP and IVDMD were increased by U4 (p < 0.01). Exp II: TW was treated with U and H: U (as in Exp I) and H (at 0, 0.25, 0.50, 0.75 and 1 g active Cl 100 g?1 DM), alone or combined. The interaction U*H was significant for ADF, ADL and CP. For U4+H0.25 ADF and ADL decreased (p < 0.01). For U0, ADL was reduced from H.025 through H1.00 (p < 0.05). U4 increased IVDMD and CP (p < 0.01). Exp III: oat straw (O) was treated with U0 and U4, H0 and H1, S0 and S4 (4 g S 100 g?1 DM), including all combinations at 3 storage times (T = 10, 20 and 30 days). Interactions U*S*H*T for ADL, U*H*T for NDF and CP and S*H*T for IVDMD (p < 0.05) were found. ADF was reduced by U4, H1 and S1 (p < 0.01). The greatest decrease (4.5%) was with U4. Principal components analysis showed U4S4H1 highly related to IVDMD for all T. All treatments had little or no impact on NDF, ADF and ADL content. Lower quality forages had greatest improvements in digestibility and N‐retention.  相似文献

The potential role of Boophilus microplus as a natural tick vector of Babesia equi and Babesia caballi in Brazilian horses was assessed using nested polymerase chain reaction (PCR)-based marker assay. B. equi merozoite-specific 218bp gene fragment was detected in almost 96% of horse blood samples, and 45.3-62.5% of females, eggs, larvae, and nymphs of B. microplus collected from 47 horses at Campo Grande in the State of Matto Grosso, Brazil. Except for the partially-fed female ticks, the B. caballi-specific 430bp gene fragment was amplified from horse blood samples, and all developmental stages. Parasite DNA from both species was detected in horse blood samples and B. microplus, with the preponderance of B. equi DNA. No DNA samples were positive solely for B. caballi parasite. Only 32% of the Giemsa-stained thin blood smears were positive for Babesia parasites, as against detection of B. equi parasite DNA in 95.7% of the blood samples by nested PCR. We have obtained molecular evidence that strengthens earlier experimental and ultrastructural studies in Brazil incriminating B. microplus as a natural vector of B. equi, and possibly of B. caballi. The detection of B. equi and B. caballi DNA in eggs and larvae of B. microplus is likewise suggestive of the possibility of both transovarial and transstadial parasite transmission in this tick vector.  相似文献

The enzyme-linked immunosorbent assay (ELISA) was used to examine cross-reactivity of Neospora caninum with Toxoplasma gondii and Hammondia heydorni. Anti-T. gondii mouse and cat sera cross-reacted with N. caninum soluble antigen (NLA), but not with the recombinant surface antigen (NcSRS2). Anti-H. heydorni dog sera showed no cross-reactivity with either the NLA antigen or the NcSRS2. Lack of cross-reactivity between anti-H. heydorni sera and N. caninum antigens, and the cross-reactivity of anti-T. gondii sera with the NLA suggest that N. caninum has common antigens to T. gondii except for NcSRS2 based on serology. In light of several studies suggesting a closer relationship between N. caninum and H. heydorni than with T gondii, examination of serological cross-reactivity with N. caninum may be necessary to further classify the parasites in addition to molecular and morphological studies and clarification of the life cycle.  相似文献

Proteases have been proposed as part of the invasion strategies of some pathogenic fungi. In this work, a serine protease produced by the phytopathogenic fungus Fusarium solani f.sp. eumartii was purified and characterized. Purification of the enzyme was accomplished by gel filtration through a Superose 12 column, followed by hydrophobic interaction chromatography in Phenyl Superose and gel filtration chromatography through Superdex 75. Analysis of the purified enzyme by SDS/PAGE without heat treatment, revealed a single band, which corresponded to the proteolytic activity detected by zymogram. When this protein was subjected to denaturing conditions, two major polypeptides of approximately 30 and 33kDa were revealed. The N-terminal amino acid sequence of one of these polypeptides showed a high similarity with fungal mature serine proteases of the subtilisin family. This protease hydrolysed in vitro, specific polypeptides of potato intercellular washing fluids and cell walls. The protease was also able to degrade pathogenesis-related proteins from the intercellular washing fluids. The role of this serine protease as part of the fungal strategy to colonize potato tuber tissues is discussed.  相似文献

Research for developing silvopastoral systems includes tree species selection and management, and interactions of trees with pastures. In the Caribbean humid lowlands of Costa Rica, total height, diameters at breast height (DBH), survival, basal area and bole volume were compared among three plantations with four species each, in pure stands as well as in the mixture of the four species. In Plantation 1, at 86 months old the pure plantations of Jacaranda copaia and Vochysia guatemalensis, and the mixed plantations (Jacaranda copaia + Vochysia guatemalensis + Calophyllum brasiliense + Stryphnodendron microstachyum) had the largest DBH. Vochysia guatemalensis is one of the most preferred species in the region for reforestation of degraded pastures, while Jacaranda copaia is not preferred because it has low local economic value. In Plantation 2, at 81 months, the greatest DBH was found in Virola koschnyi, Terminalia amazonia and the mixed plantations (Virola koschnyi + Terminalia amazonia + Dipteryx panamensis + Pseudosamanea guachapele). Dipteryx panamensis has good economic value and has a relatively open canopy that produces moderate shade. Virola koschnyi and Terminalia amazonia have denser canopies, are preferred by farmers, and they could be planted in low density in silvopastoral systems. In Plantation 3, at 70 months, the greatest DBH was found in Vochysia ferruginea, Hieronyma alchorneoides and the mixed plantation (Vochysia ferruginea + Hieronyma alchorneoides + Genipa americana + Balizia elegans). Most of these species are currently used in silvopastoral systems in the region. Mixed plantations may combine beneficial effects of the different species' characteristics, with the additional advantage of product diversification. We described the transmission dynamics of Fasciola hepatica at its southern distribution range. Studies of prevalence and egg output in cattle and population dynamics and infection in snails were performed in a farm in the Andean Patagonian valleys, Argentina, between December 1998 and February 2002. Snail surveys were conducted from spring to autumn. Infection was diagnosed coprologically in the whole herd at the beginning and end of the study, and in a cohort of heifers at the beginning and end of 2001. A twice-a-year anthelmintic treatment was implemented in 1999. The relationship of the variables mentioned above with temperature and rainfall was determined. Lymnaea viatrix showed a life-span of about 15 months and an annual pattern of population dynamics. Specimens were frequently found in temporary environments and lagoons, and rarely in streams. Snail abundance and soil-water availability were directly related in temporary environments and inversely related in lagoons. Overall prevalence in L. viatrix was 0.67% (range: 0.9-14%) and infection was detected in summer and autumn. At the beginning of the study, calves were the least infected age group (15%). Prevalences and median egg counts in grazing animals were similar at the beginning (heifers: 81%, 3.3 epg; cows: 60%, 1.3 epg) and end of the study (heifers and cows: around 51%, 1 epg). Likewise, the prevalence in the cohort of heifers remained similar (around 40%) between surveys. Transmission to cattle was highly effective despite of the short activity period and the low infection rate of snails, and the regular anthelminthic treatment. There would be two seasonal transmission peaks, one in summer-autumn, when infected snails were present, and the other in early spring due to overwintering metacercariae. Some recommendations based on the climatic conditions of the region are provided to minimize snail infection and ultimately to reduce the incidence of fasciolosis in cattle.  相似文献

The aims were (a) to quantify the number of Duddingtonia flagrans chlamydospores per gram of faeces (CPG) recovered from sheep administered with different oral doses and, (b) to describe the relationship between CPG and eggs per gram of faeces (EPG) on the efficacy to reduce Haemonchus contortus infective larvae. Three doses of chlamydospores per kg BW were orally administered during seven days: (T1) non treated control group, (T2) 1 × 106, (T3) 2.5 × 106 and (T4) 5 × 106. Three lambs, infected with H. contortus, were used per group. Faeces were obtained from the rectum of each lamb during the fungal administration period (days 0–6) and for six days after that period. Four coproculture replicates were made from each animal in days 2, 4, 6, 8 and 10. A higher chlamydospore dose produced higher CPG in faeces (p < 0.05), but a clear dose dependent effect was not found either in the larvae reduction or in the CPG:EPG ratio. When ratios were re-analyzed, independently of the treatment groups of origin, a better efficacy was obtained with a ratio from 5 to 10 CPG:EPG and a higher ratio (>10 per egg) showed a lower reduction efficacy (p < 0.05). The binomial analysis showed that for each unit of increment in CPG:EPG ratio there was a reduction of larvae number until a point (between 5 and 10 CPG:EPG) where no further reduction was detected. The surface response test indicated that the number of larvae was reduced by CPG until possible saturation. The highest CPG:EPG ratios did not necessarily improve efficacy of D. flagrans.  相似文献

In most goat breeds, testosterone serum concentration and semen quality decrease during the nonbreeding season. However, bucks reproductive activity may be stimulated with the administration of equine chorionic gonadotropin (eCG). Therefore, the aim of this study was to determine whether the repeated administration of eCG stimulates the reproductive status of bucks during the nonbreeding season. The study was performed with 19 bucks that were assigned to a group that was treated with eCG (GeCG) and an untreated control group (GCon). The GeCG bucks received an initial dose of 800 IU of eCG (Day 0), followed by four doses of 500 IU administered every 5 days beginning on Day 5. Serum testosterone and anti‐eCG antibody concentrations, testicular and seminal traits were determined until Day 60. Testosterone concentration (from Day 3 to 21: p < 0.0001), anti‐eCG titre (from Day 12 to 44: p ≤ 0.01), percentage of motile spermatozoa (Day 6: p = 0.006 and 14: p = 0.001) and of spermatozoa with progressive motility (Day 6: p = 0.01 and 14: p = 0.002) and the percentage of spermatozoa with functional membrane (Day 6: p = 0.02 and 22: p = 0.008) were higher in GeCG than in GCon bucks. Also in frozen‐thawed samples, the percentage of motile spermatozoa tended to be higher in GeCG than that of GCon bucks (p = 0.07). In conclusion, the administration of eCG during the nonbreeding season stimulated the secretion of testosterone and improved fresh and possibly frozen‐thawed semen quality. However, it also resulted in an increase in anti‐eCG antibody titre.  相似文献
