An intravenous bile acid loading test was performed on 16 goats using various dosages and sampling protocols. A recently developed enzymatic bile acid assay provided an economical and technologically simple method for bile acid analysis. Intravenous injection of bile salts into goats was well tolerated. Clearance times were rapid. A protocol for performance of the test was suggested and a practical reference range for T 1/2 clearance of 4+/-1 minute was determined. Blood samples were collected every 2 h during a 24 h period from 6 cows of one herd and 10 cows of another herd. In a third herd 9 cows were sampled every 2 h from 6 a.m. to 8 p.m. Concentrations of total bile acids, acetoacetate, glucose and free fatty acids were determined in blood plasma. A marked difference in individual bile acid concentrations and patterns of diurnal variation was found. For most cows the highest bile acid values were observed between 2 and 6 a.m. (overall mean (+/- SD) at 6 a.m.: 104 +/- 84 mumol/l, range: 20-307 mumol/l). Fourteen cows with a bile acid value greater than 90 mumol/l at 6 a.m. ("high BA") were characterized as a group by showing a pronounced decrease in the mean bile acid concentration after morning feeding. In the group of 11 cows with a 6 a.m. bile acid value less than 90 mumol/l ("low BA") the time of day did not contribute significantly to the bile acid variation. For the "high BA" group a nearly synchronous variation between the mean values of the 3 feeding dependent parameters (acetoacetate, glucose and free fatty acids) and the mean values of bile acids was found. The within animal coefficients of correlation between bile acids and the feeding dependent parameters were significantly higher in the "high BA" group than in the "low BA" group. No direct connection was found between bile acid levels and the quantity of concentrates fed or the individual milk yield. 相似文献
There is a lack of laboratory-based embryonic chicken toxicity studies with the ecologically relevant low dose/s of endosulfan that utilizes a more practical approach such as the chorioallontoic membrane (CAM) injection. In this investigation, 2μg AR grade α-endosulfan/egg (40% of LD50 for embryos) was injected through the CAM in 12-day-old chicken embryos and the activities of glucose-6-phosphatase (G6Pase, EC 3.1.3.9), fructose 1,6-diphosphatase (FDPase, EC 3.1.3.11), adenosine triphosphatase (ATPase, EC 3.6.1.3) and succinic dehydrogenase (SDH, EC 1.3.99.1) and DNA and RNA content in liver and brain tissues and acetyl cholinesterase (AChE, EC 3.1.1.7) in the latter were determined at 24, 48, and 72 h post-exposure. The wet weight of the embryos did not differ between groups. Following endosulfan exposure, except increase in the hepatic ATPase activity (P < 0.01), there was a significant decrease in the following parameters: G6Pase activity in both the liver and brain (P < 0.01), SDH activity in the brain (P < 0.01), brain overall DNA and RNA concentration (P < 0.05), brain AChE activity (P < 0.01). Exposure of 18-day-old embryos to 2-μg endosulfan for 24 h caused decrease (P < 0.01) in the lymphocyte count and IgG content. Histopathology of thymus and bursa of Fabricius revealed a reduction in the population of thymic follicles, smaller thymocytes with the clear vacuoles in cytoplasm and fewer bursocytes accompanied by infiltration of erythrocytes in lymphoid follicles of the endosulfan-treated embryos. It was inferred that in ovo injection of 0.041 μg/g egg weight of α-endosulfan suppress gluconeogenesis (main energy source in embryonic life), nerve transmission, and immunity. 相似文献
The effect of true milk use in the diet of Spanish Brown Swiss male calves on animal performance, carcass and meat quality, and fatty acids composition was studied. In experiment 1, the effect of milk intake [ad libitum continuous (ADLIB) feed vs. restricted 0.7 during 75 days followed by ad libitum feed (RESTR)] and slaughter endpoint (225 kg vs. 5 month) were studied. In experiment 2, ad libitum concentrate feeding [grain-fed (GF)] was compared with milk supplementation until slaughter [milk-fed (MF)] in calves slaughtered at 345 kg. As regards to milk intake, carcass weight and degree of fatness were higher in the ADLIB group (P<0.05). The RESTR group revealed a higher percentage of saturated fatty acids (P<0.05). In experiment 2, the MF group exhibited a higher fat percentage (P<0.05), lower press and cooking losses (P<0.05), and higher scores for tenderness and juiciness (P<0.05) than the GF group. The percentage of saturated fatty acids was higher in the MF group (P<0.05). The results suggest that true milk use in veal production could be an advantageous alternative in terms on production costs, animal performance, and carcass and meat quality. 相似文献
AIM:To investigate the role of expression of peroxisome proliferator-activated receptor α(PPAR α) in pathogenesis of rat fatty liver.METHODS:The rats were treated with a low dose of carbon terachloride (CCl4) and fed a high fat diet to produce fatty liver. We determined the concentrations of triglyceride (TG), total cholesterol (TC), free fatty acid (FFA) in liver and the alanine aminotransferase (ALT) activity, tumor necrosis factor-α (TNF-α), FFA in serum and the degree of hepatocytic steatosis. Total RNA of liver was extracted, and the expression of PPAR α were analyzed by semi-quantitative RT-PCR method.RESULTS:In model group, the hepatocytic PPAR α mRNA expression decreased to 0.41±0.28, compared to 1.41±0.29 in the control group (P<0.01). The contents of TG, TC, FFA in model rat liver were (1.88±0.20) mmol·L-1, (11.03±1.12) mmol·L-1 and (1 260.38±151.27) μmol·L-1, respectively, compared to (0.53±0.10) mmol·L-1, (1.25±0.25) mmol·L-1 and (334.30±27.09) μmol·L-1 in the control group (P<0.01). The activity of ALT, concentrations of TNF-α and FFA in serum were also increased remarkably in model group.CONCLUSION:Oxidation of fatty acid and utilization of lipids in liver are affected by reducing the expression of PPAR α, which result lipid accumulation in liver. 相似文献
AIM: To study the effects of tetrandrine(Tet) and fructose-1, 6-diphosphate(FDP) on the elevated intrasynaptosomal [Ca2+]i induced by excitatory amino acids(EAA). METHODS: A rapid method for preparing synaptosomes was used, and intrasynaptosomal free calcium([Ca2+]i) was measured by using the fluorescent indicator quin-2. RESULTS: L-glutamate(Glu, 100 μmol/L), aspartate(Asp, 100 μmol·L-1), N-methy1-D-aspartate(100 μmol/L) and Glu(50 μmol/L) plus Asp(50 μmol/L) all elevated intrasynaptosomal [Ca2+]i in a dose-dependent manner. Pretreatment with Tet(10, 30, 60 μmol/L), FDP(15, 30, 75, 150 μmol/L), MK-801(10, 20 μmol/L) and Tet(15, 30 μmol/L) plus FDP(15, 30 μmol/L) all attenuated the increase in intrasynaptosomal [Ca2+]i induced by EAAs mentioned as above in a dose-dependent manner, and the effect of Tet plus FDP was most significant. CONCLUSION: Both Tet and FDP inhibited a rise in intrasynaptosomal [Ca2+]i induced by EAAs, which may be one of mechanisms that Tet and FDP pretect cerebral tissues against ischemia injury. 相似文献
1. Studies were conducted with tall oil fatty acids (TOFA) to determine their effect on broiler chicken performance and ileal microbiota. TOFA, a product originating from coniferous trees and recovered by fractional distillation of side-streams from pulp production, mainly comprises free long-chain fatty acids (~90%) and resin acids (~8%). Conjugated linolenic acids and pinolenic acid are characteristic fatty acid components of TOFA.
2. TOFA products at 750 mg/kg feed were tested in two 35-day broiler chicken trials, each using a wheat soya-based diet and with 12 replicate pens per treatment. In both trials, TOFA improved body weight gain at all time points (P < 0.001) and feed conversion efficiency during the first 21 days (P < 0.01). Two different dry TOFA formulations (silica carrier and palm oil coating) were tested and showed performance effects similar to liquid TOFA.
3. Ileal digesta of the broiler chickens was analysed for total eubacteria, Lactobacillus spp., Enterococcus spp., Escherichia coli and Clostridium perfringens on days 14 and 35. TOFA significantly increased total eubacteria and lactobacilli density on day 14 (P < 0.05). There was a significant positive correlation between these bacterial groups and broiler body weight on day 14 (P < 0.01).
4. A numerical reduction in C. perfringens was observed. In vitro growth inhibition studies showed that C. perfringens was strongly inhibited by 10 mg/l TOFA (P < 0.001), while common lactobacilli were resistant to >250 mg/l. The in vitro results were thus in line with in vivo observations.
5. The mechanisms behind the bacterial shifts and their role in performance improvement are unknown. Further purification of TOFA components is needed to identify the effective agents. 相似文献
