Summary Somaclonal variation is a tool that can be used by plant breeders. The review examines where this tool can be applied most effectively and the factors that limit or improve its chances of success. The main factors that influence the variation generated from tissue culture are (1) the degree of departure from organised growth, (2) the genotype, (3) growth regulators and (4) tissue source. Despite an increasing understanding of how these factors work it is still not possible to predict the outcome of a somaclonal breeding programme. New varieties have been produced by somaclonal variation, but in a large number of cases improved variants have not been selected because (1) the variation was all negative, (2) positive changes were also altered in negative ways, (3) the changes were not novel, or (4) the changes were not stable after selfing or crossing. Somaclonal variation is cheaper than other methods of genetic manipulation. At the present time, it is also more universally applicable and does not require containment procedures. It has been most successful in crops with limited genetic systems and/or narrow genetic bases, where it can provide a rapid source of variability for crop improvement. 相似文献

Protoplasts were isolated from young inflorescence-derived suspension cultures of a japonica rice cultivar ‘Taipei 177’. The isolated protoplasts which were cultured either in liquid, agar on Sea plaque agarose underwent sustained division. Maximum plating efficiency of 1.06% occurred in a medium containing macroelements of KM, microelements and vitamins of B5, 0.5 % Sea plaque agarose, 1.0 mg/l of 2,4-D, and glucose as an osmotic stabilizer. The cytological status of plantlets regenerated from shoot apical meristems of Pisum sativum was investigated. Chromosome counts in root apices of in vitro regenerated plants showed a preponderance of diploid cells. Moreover, the karyotypes of root-tips from plants derived from culture and from normal plants were basically the same. Parasitic angiosperms cause great losses in many important crops under different climatic conditions and soil types. The most widespread and important parasitic angiosperms belong to the genera Orobanche, Striga, and Cuscuta. The most important economical hosts belong to the Poaceae, Asteraceae, Solanaceae, Cucurbitaceae, and Fabaceae. Although some resistant cultivars have been identified in several crops, great gaps exist in our knowledge of the parasites and the genetic basis of the resistance, as well as the availability of in vitro screening techniques. Screening techniques are based on reactions of the host root or foliage. In vitro or greenhouse screening methods based on the reaction of root and/or foliar tissues are usually superior to field screenings and can be used with many species. To utilize them in plant breeding, it is necessary to demonstrate a strong correlation between in vitro and field data. The correlation should be calculated for every environment in which selection is practiced. Using biochemical analysis as a screening technique has had limited success. The reason seems to be the complex host-parasite interactions which lead to germination, rhizotropism, infection, and growth of the parasite. Germination results from chemicals produced by the host. Resistance is only available in a small group of crops. Resistance has been found in cultivated, primitive and wild forms, depending on the specific host-parasite system. An additional problem is the existence of pathotypes in the parasites. Inheritance of host resistance is usually polygenic and its transfer is slow and tedious. Molecular techniques have yet to be used to locate resistance to parasitic angiosperms. While intensifying the search for genes that control resistance to specific parasitic angiosperms, the best strategy to screen for resistance is to improve the already existing in vitro or greenhouse screening techniques. 相似文献

Anthers of Brassica nigra, excised from fresh as well as cold-pretreated (3 days at 3 ± 2°C) buds cultivated on modified B5 medium (Gamborg et al. 1968) containing sucrose level varying from 2 % to 10 %, along with 1O?6M BAP (benzylaminopurine) and 9 × 10?6M 2,4-D (2,4-dichlorophenoxyacetic acid), developed calli and/or embryos. The latter response was observed only in anthers reared on media containing 6 % or higher levels of sucrose. On media containing two or four per cent sucrose, the anthers produced calli, exclusively. The growth of embryos was inhibited or else they started callusing if left on the media containing higher levels of sucrose. However, on transfer to MS medium (Murashige and Skoog 1962), containing 2 % sucrose, embryos started callusing and subsequently a few secondary embryos differentiated. Such embryos were sub-cultured on MS + 5 × 10?6M BAP + 2 % sucrose, wherein numerous shoots developed from embryos. Long-term continuous selection is essential for germplasm improvement. However, choice of germplasm for long-term genetic improvement might limit the success of germplasm enhancement programs. The objective of this research was to report the response to long-term selection in early maturing North Dakota (ND) synthetic varieties. We wanted to determine whether the performance of three ND maize synthetic varieties was improved by long-term mass selection (M) and if the performance of one of them was improved by long-term modified ear-to-row (MER) selection. The evaluation of long-term selection response was performed at two plant densities. An experiment in a randomized complete block design with split-plot arrangement was used to evaluate NDSM(M), NDSAB(M), NDSCD(M), and NDSAB(MER) under 75,000 and 42,500 plants per hectare across seven environments. Long-term mass selection for grain yield and stalk lodging resistance in NDSM(M), NDSAB(M), and NDSCD(M) was not successful, since there were no significant changes in grain yield or stalk lodging in these populations at either low or high densities. On the other hand, long-term modified ear-to-row selection was effective for grain yield improvement in NDSAB(MER). Grain yield increased non-linearly from 3.9 Mg ha−1 in cycle 0 to 5.0 Mg ha−1 in cycle 12 at a rate of 2.5% per cycle. Interaction between plant density and genotype was not detected even though selection was performed at relatively low densities (20,000 plants ha−1 for mass selection and 50,000 plants ha−1 for ear-to-row selection). The confirmation of a lack of interaction between plant density and genotype suggests that selection at low plant densities might still be able to provide high-density stress resistance through density-independent genotypes, allowing progeny testing across multiple locations with better accuracy and fewer resources. Selection methods that emphasize both additive and dominance effects such as full-sib recurrent selection are recommended to maximize genetic improvement of advanced population cycles of early maturing synthetics. 相似文献

Summary A total of 749 genotypes from a number of white clover (trifolium repens L.) cultivars and populations, many collected from dryland areas, were cloned and grown in field titles. Several morphological characters, including leaf size, number and diameter of large nodal roots (taproots), and proportion of root weight that was classified as taproot were measured. There was large variation between lines and genotypes for all characters measured, and differences between genotypes within lines are also reported. Summary A commercial horticultural industry that establishes plant-breeding nurseries for an exotic species throughout a regional
economy will expand until the marginal profit of the last nursery established is zero. However, a regional government concerned
with social welfare will take into account not only the profits of the horticultural industry but also any expected costs
of an accidental invasion. The latter costs will consist of the discounted expected social damages due to an increase in the
rate of invasion over time and the increase in expected damage cost per hectare caused by an additional nursery. A government
can employ an “introducers' pay” tax equal to the latter social costs to ensure that the plant breeding industry establishes
the optimal number of nurseries. We illustrate this outcome with the example of purple loosestrife (Lythrum salicaria) in North America. In the absence of any tax, the horticultural industry will establish n = 3528 nursery operations, and the expected damages from invasion are US$ 28.2 million per year. With the tax, only n = 300 nurseries are established but the expected damages from invasion are reduced to US$ 1.3 million per year. Although
profits for the horticultural industry are lower from the tax, the net gains in overall social welfare more than offset the
losses. Although these results are illustrative only, they show that the problem of plant invasives initiated by commercial

In chemistry and many related fields, electronegativity (EN) is an important fundamental concept and its scale is a useful physcochemical parameter. Here, calculations of both ionization potentials and electron affinities are extended toward 107 elements and done by density functional theory at the local density approximation(LDA) level and the LDB level, i.e., the local density approximation level with further non-local corrections for exchange and correlation included self-consistently as well as the modified Slater transition-state method. The definite-differentiation method is employed into calculations of the electronegativity scale and the related parameters of 107 elements with very good results due to the consideration of relativistic effects. The calculation presented is to examine both the LDA and LDB approximations in calculations for the ionization potential and electron affinity of the elements with an improved or modified Slater transition-state method, and relativistic effects have also been taken into account for 107 elements compared with 103, 86 or less in the previous report under a spin polarized density function theory with some approximations to the exchange-correlation function. The calculation results for the various quantities represent an obviously improvement over some previous calculations. It is shown that the results calculated by the extended technique and the improved Slater transition-state method in general agree well with experimental values presented by Pearson, and are better than the reported values in many previous literatures. The developed new electronegativity scale will widely be applicable in many fields such as molecular structural parameterization expression, chemobiological activity optimization prediction, structure-activity quantitation modeling, functional chemical adaptization designing, and so on. 相似文献
