为了提高链霉菌菌株JH108-2的发酵滤液对植物寄生线虫的毒力,采用正交试验设计,优化其发酵培养基。使用优化培养基[大豆粉20g、(NH4)2SO4 6g、蛋白胨3g、葡萄糖40g、酵母粉6g,NaCl 2g、K2HPO4 0.6g.CaCO3 2g、蒸馏水1000mL、pH 6.0]所得的发酵滤液,稀释10倍,处理南方根结线虫(Meloidogyne incognita)二龄幼虫,24h后校正死亡率达95.4%,比对照培养基的发酵滤液所产生的校正死亡率高6.9%,差异达到极显著水平(P=0.01)。 相似文献

从一系列灵芝(Ganoderma sp.)菌种中筛选出多糖产量高、生长快的紫灵芝菌株(G.japoncium),研究了不同氮源、碳源及金属离子对紫灵芝产多糖的影响。结果表明,以2%的蔗糖为碳源,0.2%的豆饼粉为氮源,0.2%的FeSO4为培养基可获得较高多糖。发酵罐放大实验表明,采用同样的培养基,每100mL发酵液胞外粗多糖含量可高达181.7mg,每100mL发酵液菌丝体含量可高达151.0mg,发酵过程中pH值的变化比较缓和,相对摇瓶生长,发酵生产可获得更多的灵芝多糖。 相似文献

Black foot disease of grapevines is a decline and dieback disease caused by a soilborne pathogen complex including Cylindrocarpon liriodendri , C. macrodidymum , Campylocarpon fasciculare and Campyl. pseudofasciculare . These pathogens cause primary infections of roots and basal ends of grafted cuttings in nursery soils. Thirteen fungicides were screened in vitro for mycelial inhibition of these pathogens. Prochloraz manganese chloride, benomyl, flusilazole and imazalil were the most effective fungicides tested, and were subsequently included in semi-commercial field trials. Basal ends of grafted cuttings were dipped in various chemical and biological treatments prior to planting in open-rooted nurseries. Black foot pathogens were not isolated from grafted cuttings prior to planting. Additional treatments involved soil amendments with Trichoderma formulations and hot water treatment of dormant nursery grapevines. Field trials were evaluated after eight months. Isolations from uprooted plants revealed low levels of black foot pathogens in the roots of untreated control plants, and significantly higher levels in basal ends of rootstocks. The incidence of black foot pathogens, as well as that of Petri disease pathogens, was not significantly and/or consistently reduced by the majority of chemical or biological treatments. However, these pathogens were not isolated from uprooted plants that were subjected to hot water treatment. It is therefore recommended that hot water treatment of dormant nursery plants be included in an integrated strategy for the proactive management of these diseases in grapevine nurseries. 相似文献

The fungal pathogen Monilinia fructicola causes blossom blight and fruit brown rot of stone fruits in California. In this study, spore densities in the air were monitored
in six orchard/year combinations with Burkard spore traps. A real-time PCR assay was developed to efficiently quantify the
dynamics of spore density in these orchards during the growing season. Different patterns of dynamics of spore density were
observed in these orchards. A linear relationship between numbers of spores counted with a compound microscope and those determined
with the real-time PCR assay was obtained, using the same samples of spore traps. Spore density in five of six orchard/year
combinations ranged from 0.0 to 0.05 spores l−1, except for that in orchard 4, which showed much higher values of spore density in the air, as well as higher values and

Root and stem rot with wilt of above ground parts of cultivated chrysanthemums was first found in Ibaraki, Toyama and Kagawa
prefectures, Japan in 2002 and 2003. Pythium species were isolated from the diseased tissues and identified as P. dissotocum, P. oedochilum, P. sylvaticum, P. ultimum var. ultimum and asexual strains of P. helicoides based on their morphologies and sequences of rDNA-ITS region. The genetic diversity of 71 Pseudomonas savastanoi pv. savastanoi strains isolated from different host species and from diverse geographical regions was determined by fluorescent amplified fragment length polymorphism (f-AFLP) analysis. The study was carried out using three different selective primer combinations. Strains of P. syringae pv. syringae , P. syringae pv. phaseolicola , P. syringae pv. glycinea , P. syringae pv. tagetis and P. amygdali were also included as outgroups. Based on cluster analysis of f-AFLP data, all P. savastanoi pv. savastanoi strains showed a high degree of similarity, grouping in a cluster and forming a taxon clearly separate from outgroup strains. AFLP analyses failed to support placing strains of P. savastanoi pv. savastanoi , P. syringae pv. phaseolicola and P. syringae pv. glycinea in the same species. Strains of P. savastanoi pv. savastanoi formed subclusters that correlated with the host species. Strains identified within these subclusters were related to the geographical region where the strains were isolated. Strains of P. savastanoi pv. savastanoi from olive were divided into two subclusters. Strains from oleander were differentiated from those from ash and were divided into two additional subclusters, distinct from olive strains. Three strains isolated from jasmine showed a high level of similarity among them but, at a lower Dice similarity coefficient, were linked to a subcluster including olive strains. One hundred and sixty strains of Pseudomonas syringae subsp. savastanoi from Olea europaea, Olea europaea var. sylvestris, Nerium oleander, Fraxinus angustifolia and Retama sphaerocarpa, and four type strains of other pathovars were studied, investigating 102 phenotypic traits, among which we include biochemical characteristics, assimilation of different carbon sources, sensitivity or resistance to antibiotics and indoleacetic acid (IAA) production. Results were analysed with an affinity dendrogram via the Jaccard coefficient. They indicate an influence of environmental factors on the formation of the 15 phenons obtained, since isolated (knot) strains from the same species but different geographical areas are segregated. Segregation, also detected in strains from different hosts within the same area, added to the pathogenicity test helps to characterise these strains as different pathovars. 相似文献
