以未成熟合子胚为外植体,建立杂种落叶松胚性愈伤组织诱导和植株再生体系.从日本落叶松5×长白落叶松77.3和日本落叶松5×兴安落叶松9两个家系诱导出胚性愈伤组织,授粉后65天左右采集的合子胚在添加0.5 mg·L-1 2,4-D+0.5 mg·L-1 6-BA+0.5 mg·L-1 KT的S培养基上胚性愈伤组织诱导率最高,为10%.从胚性愈伤组织中筛选出3个具有稳定分化能力的胚性细胞系,其中授粉后65天采集的日本落叶松5×长白落叶松77-3合子胚诱导的胚性细胞系的每克胚性愈伤组织形成的体胚数、体胚萌发率、植株再生率最高,分别为18.1个,77.O%,28.1%.再生植株的移栽成活率为41.8%. 相似文献

Abstract A population of Limonium stocksii was studied for one year at the upper end of Manora Creek along the Arabian Sea coast, near Karachi, Pakistan. The community was divided into three zones. Soil pH varied from 8.1 to 8.5. Soil conductivity was highest (191 dS m?1) in September and lowest (55 dS m?1) in June. Soil water content was relatively low and varied from 1.6% in February to 9.5% in June. Limonium stocksii maintained a persistent seed bank, which peaked at 5,887 seeds m?2 in May. Very slow growth was observed; however, plants in the intermediate zone showed better growth than the other two zones. Stem and leaf tissue water content were affected by salinity fluctuations, but root tissue water remained unaffected. Sodium (Na+) and chloride (Cl?) accumulated in stems and leaves more than other ions. Leaves also accumulated a very high amount of magnesium (Mg++) compared to roots and stems. 相似文献

Abstract Wound models attempt to simulate the natural healing processes in wounds. However, all models have significant limitations due to the complexity of the tissue repair process. Much can be learned from wound models in vitro by the use of cell culture techniques. The horse can provide a suitable naturally occurring model of chronic wound healing because it has many similarities to wound healing encountered in human medicine. The tissue architecture was investigated with regard to extracellular matrix and growth factor distribution during wound healing and growth factors were consistently present in the wound area. Biochemical investigations revealed increased levels of hydroxyproline, collagen, and TGFβ1 in exuberant granulation tissue. Equine wound models were established in vitro using cell culture techniques and growth factors had significant effects on the growth of the cells and their ability to synthesize collagen. Two gelatinases (MMP-2 and MMP-9) were detected in the tissues and wound fluid samples investigated. Zusammenfassung Wundmodelle haben zum Ziel, die natürlichen Heilungsprozesse in Wunden zu simulieren. Allen Modellen sind durch den komplexen Gewebsheilungsprozess deutliche Grenzen gesetzt. Durch die Verwendung von Zellkulturtechniken kann von Wundmodellen in vitro viel abgeleitet werden. Das Pferd stellt wegen der vielen Ähnlichkeiten zur humanmedizinischen Wundheilung ein geeignetes Modell für chronische Wundheilung dar. Die Gewebsarchitektur wurde bezüglich der Verteilung von extrazellulärer Matrix und von Wachtumsfaktoren während der Wundheilung untersucht; Wachtsumsfaktoren waren ständig in der Wunde vorhanden. Biochemische Untersuchungen ergaben erhöhte Hydroxyprolin-, Kollagenund TGFβ1-Spiegel. Wundmodelle beim Pferd in vitro und Zellkulturtechniken wurden entwickelt; Wachstumsfaktoren hatten deutliche Wirkung auf die Zellen und ihre Fahigkeit zur Kollagensynthese. Sin embargo, todos 10s modelos presentan limitaciones considerables debido a la complejidad del proceso de curación de heridas. Se puede aprender mucho de modelos de heridas in vitro mediante el uso de técnicas de cultivo celular. El caballo puede suponer un modelo natural adecuado de curación crónica de heridas ya que se asemeja a la curación de heridas en medicina humana. Se investigó la arquitectura tisular en referencia a la matriz extracelular y la distribución de factores de crecimento viendo que los factores de crecimiento se encontraban presentes de forma constante en el área de la herida. Las investigaciones bioquímicas revelaron incremento en los niveles de hidroxiprolina, colágeno y TGFbl en tejido de granulación exhuberante. Cependant tous les modèles ont des limitations significatives dues à la complexité des processus de réparation des tissus. Beaucoup peut être étudié sur des modèles de plaies in vitro suite à l'utilisation des techniques de culture cellulaire. Le cheval peut constituer un modèle nature1 fiable de cicatrisation chronique de plaie de par les nombreuses similarités qu'il partage avec la cicatrisation des plaies en médecine humaine. L'architecture tissulaire a étéétudiée concernant la matrice extracellulaire et la distribution des facteurs de croissance pendant la cicatrisation, et les facteurs de croissance se sont montrés présents de façon consistante dans la zone de la plaie. Des investigations biochimiques ont révélé une élévation des taux d'hydroxyproline, collagène, et TGFβ1 dans la granulation tissulaire exubérante. Des modèles équins de plaies ont étéétablis in vivo en utilisant des techniques de culture cellulaire et les facteurs de croissance ont montré des effets significatifs sur la croissance des cellules et leur capacité de synthétiser le collagène. Abstract In the past 5 years advances in the management of wounds in dogs and cats have included the development of materials and techniques used to enhance the healing of both open and closed wounds. New topical and locally injected medications as well as new bandaging materials have been evaluated. The use of omental flaps to enhance wound healing has been found to be promising. Ultrasonography has been found useful in assessing wound tissues to guide treatment. Because tension is a major factor in dealing with wound closure, skin expanders, skin stretchers, presutures and adjustable horizontal mattress sutures have been described as means of overcoming tension. Recently completed research projects at the Scott-Ritchey Research Center studied the woundhealing effects of fibrin sealants, topical bovine collagen, omega-3 fatty acid-enriched diets and magnetic field therapy. Projects underway are evaluating the healing effects of a new biological tissue adhesive, a macrophage stimulant and a thromboxane synthestase inhibitor. Parameters for evaluating wound healing have included histopathology, tensiometry, planimetry, laser Doppler image scanning, biochemistry and immunohistochemistry. Resumen En los últimos 5 años, algunos de los avances en la curación de heridas en perros y gatos ha incluido el desarrollo de materiales y técnicas utilizados para mejorar la curación tanto de heridas cerradas como abiertas. Se han evaluado nuevas medicaciones tópicas y sistémicas asi como nuevos marteriales de vendaje. El us0 de injertos omentales para estimular la curación de heridas ha mostrado resultados prometedores. La ecografia es un método éutil en la valoración de los tejidos lesionados para guiar el tratamiento. Debido a que la tensión es un factor importante en el caso de cerramiento de heridas, se describen como métodos de evitar la tensión productos como expansores cutáneos, estiradores cutáneos, presuturas y suturas cutáneas horizontales acolchonadas ajustables. Algunos proyectos de investigación recientemente finalizados en el Centro de Investigación Scott-Ritchey estudiaron los efectos sobre la curación de heridas de selladores de fibrina, colágeno tópico bovino, dietas enriquecidas con ácido graso omega-3 y terapia de campo magnético. Otros proyectos en realización están evaluando los efectos curativos de un nuevo adhesivo tisular biológico, un estimulante de macrófagos y un inhibidor de la tromboxano-sintetasa. Neue lokale und systemische Medikamente sowie neue Verbandsmaterialien wurden untersucht. Die Verwendung von Omentumlappen zur Verbesserung der Wundheilung scheint vielversprechend zu sein. Ultraschalluntersuchungen waren von Nutzen, um Wundgewebe zu beurteilen und die Behandlung zu lenken. Spannung ist ein wesentlicher Faktor im Wundverschluss, Hautexpander, Hautdehner, vorbereitende Nähte und regulierbare, horizontale Matratzennähte wurden zur Überwindung der Wundspannung beschrieben. Kürzlich fertiggestellte Forschungsprojekte am Scott-Ritchie Research Center erforschten die Effekte von Fibrindichtungsmitteln, lokalem bovinen Kollagen, Futtermitteln mit Omega-3 Fettsäurenzusatz und Magnetfeldtherapie. De nouvelles médications topiques et systémiques ainsi que de nouveaux matériaux de bandages ont étéévalués. L'utilisation de lambeaux d'éplipon s'est révélée prometteuse dans l'amélioration de la cicatrisation des plaies. L'ultrasonographie s'est montrée utile dans le traitement de guidance de l'évaluation des plaies tissulaires. La tension étant un facteur major lors de la fermeture de plaies, expansseurs cutanés, étireurs cutanés et surjets de matelassier ajustables ont tous été décrits comme moyens permettant de vaincre la tension. 以银脉单药花(Aphelandra squarrosa)当年生幼嫩茎段为外植体,根据其茎段幼嫩程度不同分为三级(未木质化、半木质化、木质化)进行进瓶诱导并建立了无菌系;在此基础之上对其进行了丛芽增殖及生根诱导研究,实验结果表明:半木质化茎段为最佳的外植体;而且不同的外植体其最佳的灭菌条件有所不同:未木质化:φ=75%酒精30 s 1 g/L升汞5 min;半木质化:φ=75%酒精30 s 1 g/L升汞7.5 min;木质化:φ=75%酒精30 s 1 g/L升汞8.5 min;较好的增殖培养基为:MS KT0.5 mg/L IBA0.1 mg/L;较好的生根诱导培养基为:MS NAA0.5 mg/L。 相似文献
