将牛分枝杆菌哺乳动物细胞侵入蛋白4A(Mammalian cell-entry protein 4A,Mce4A)基因克隆至原核表达载体pET30a( )中,得到重组表达载体pET30-mce4A,转入宿主菌BL21(DE3)中,获得大量表达,表达量占菌体蛋白总量的52%,大小约为43 Ku。经Ni-NTA琼脂糖亲和层析获得Mce4A重组蛋白;圆二色谱(CD)测定结果表明,Mce4A重组蛋白α螺旋含量为31.1%,β-折叠含量为0%,无规卷曲含量为61.0%,转角含量为7.9%。为进一步开展牛分枝杆菌致病机理的研究和寻找新的药物作用靶位点奠定了基础。 相似文献

This paper summarises the clinical findings of 9 cases of disseminated alimentary mycobacteriosis in horses presented at a Finnish referral equine hospital 2009–2014. Four of 9 horses were Standardbreds and 8/9 horses were male. The median age was 2 years, ranging from 6 months to 15 years. The duration of clinical signs before admission ranged from 2 weeks to 6 months. All horses demonstrated deterioration of the clinical signs after a protracted period of the disease and were finally subjected to euthanasia after poor response to multiple medical therapies. The most common complaints on admission were weight loss and diarrhoea (9/9), pyrexia (7/9), ventral oedema (7/9), lethargy (7/9) and inappetance (6/9). The most common clinicopathological abnormalities were hypoalbuminaemia and hyperfibrinogenaemia, which were present in all horses. Rectal biopsy specimens were examined from 5/9 horses and specimens were stained with Ziehl‐Nielsen (ZN). At rectal biopsy, mild multifocal neutrophilic or mild granulomatous proctitis was recognised in all 5 horses, but the ZN stain for mycobacteria was positive in only one biopsy. A liver biopsy was taken from one horse in which hepatomegaly was observed clinically and revealed marked granulomatous hepatitis with the presence of mycobacteria. The rectal biopsy from this horse was ZN negative. At post mortem examination, chronic, multifocal to coalescing granulomatous typhlocolitis and lymphadenitis were found in all horses with the small intestine less frequently involved. At histopathological examination of post mortem samples, a ZN stain was performed and intracellular acid‐fast bacilli were identified in macrophages and multinucleated giant cells in the large intestine, liver and lymph nodes in 9/9 horses and in the small intestine in 5/9 horses. Mycobacterium avium ssp. hominissuis was isolated in 5/9 horses from post mortem samples. 相似文献

Paratuberculosis (Johne's disease) is a widespread and costly disease. This consensus statement will summarize recommendations regarding diagnosis, control, and treatment of Johne's disease in cattle and other species. Each section of recommendations is followed by a statement that subjectively characterizes the strength of the supporting evidence. The role played by Mycobacterium avium subsp. paratuberculosis (MAP) in the pathogenesis has been a matter of controversy for many years. A male Thoroughbred fetus was
aborted on day 251 of pregnancy. Gross and histological examinations detected systemic
granulomatous lesions in many superficial and visceral lymph nodes and organs including
the liver, tonsils, lungs, thymus, spleen, right thyroid gland and gastrointestinal tract,
and suppurative placentitis, pyogranulomatous amnionitis and intralesional acid-fast
bacilli were also detected. An examination of the DNA base sequence of the β subunit of
RNA polymerase demonstrated that Mycobacterium avium strain 104 had
infected several organs. To the best of our knowledge, this is the first report of equine

盐穗木根际土壤产ACC脱氨酶细菌的筛选与鉴定 总被引:1,自引:0,他引:1
【目的】研究从新疆盐碱地的盐穗木根际土壤中筛选到产ACC脱氨酶活性较高的植物根际促生菌。为ACC脱氨酶活性菌株的植物促生作用的研究奠定基础。【方法】以新疆盐碱地的盐穗木根际土壤为样品源,ACC为唯一氮源,利用筛选培养基定向富集的方法,筛选产ACC脱氨酶活性菌株。通过扫描电镜进行形态学的观察;革兰氏染色、硝酸盐还原试验和柠檬酸盐利用等生理生化试验,用Biolog Gen III板微孔鉴定及16S rDNA序列同源性分析对分离的菌株鉴定。【结果】筛选了三株具有ACC脱氨酶活性的菌株,用扫描电镜可以清晰的观察到三株菌株均为短杆菌,革兰氏染色都为阴性,均有芽孢,硝酸盐还原试验均为阳性,都能利用柠檬酸盐。三株菌株分别为1# 、3# 和5#,三株菌株的ACC脱氨酶活性的比活力分别为0.012,0.012,0.014 U/mg;1# 和5#,3# 与5# 之间酶活力差异显著(P<0.05),1#和3#之间酶活力差异不显著(P<0.05),用Biolog Gen III板微孔和16S rDNA序列同源性分析对分离的菌株1#、3# 和5# 菌株分别鉴定为Klebsiella pneumoniae、Klebsiella pneumoniae、Klebsiella oxytoca。【结论】 相似文献
