Summary Meiotic observations in PMCs were made in eight kinds of interspecific hybrids in Lilium. Three hybrids, 6134-S × L. cernuum. L. regale × L. leichtlinii maximowiczii and L. auratum platyphyllum × L. henryi showed respectively the mean chromosome association frequencies of 1.4II, 0.8II and 1.5II per cell at MI which were the lowest values hitherto reported in Lilium hybrids. In L. longiflorum × L. henryi, 33.3° of the cells had 1 or 2 multivalents of three to five chromosomes in addition to uni- and bivalents. 12° bivalents and 12 univalents were invariably observed in the triploid hybrid L. longiflorum × L. cernuum. The hybrids between the species belonging to the different sections of the genus generally showed high pollen sterility, with some exceptions. A more or less remote genomic homology was found between the different sections in Lilium. 相似文献

Summary The length was estimated of the chromosome segment with the desired marker gene introgressed from a donor by backcrossing into a recurrent parent. It was found that, for instance, for a chromosome with a length of 100 cM the length of the segment is 32 cM in BC6.These estimates were compared with those made by Hanson (1959). Two corrections proved to be necessary.The implications for breeders and researchers are discussed. Differences between two near-isogenic lines often ascribed to the difference in genotype at the marker locus, may also be caused by genes linked to the marker gene and dragged into the backcross product. 相似文献

为发掘和利用荆州黑麦所携抗梭条花叶病基因,综合利用分子细胞遗传学与分子标记技术结合多年抗性鉴定,从高感梭条花叶病小麦地方品种辉县红与荆州黑麦杂交后代(F7~F9)中选育出二体异附加系5个(分别添加1R、2R、R3、5R和R7)、5RS端二体异附加系1个和多重异附加代换系2个(染色体组成分别为20’’+2R(2D)’’+4R’’和19’’+1R(1B)’’+2R(2B)’’+4R’’)。鉴定表明,双二倍体荆辉1号高抗梭条花叶病,表明黑麦抗性基因可在小麦背景中稳定表达,2R、R7二体异附加系及2个含2R的多重异附加代换系均表现高抗,推测2R和R7上可能携带抗病基因。这些材料是研究荆州黑麦抗性基因遗传及小麦抗病育种的新种质。 相似文献

The colour of plant organs is a useful trait in crop breeding. The pod colours of soybeans primarily include black, brown and tan types, which are controlled by two classical genetic loci, L1 and L2. Most wild soybeans have black pods, which reflect a possible role in adaptation to the natural environment. Here, an improved chromosome segment substitution line (CSSL) population SojaCSSLP3 was established to identify the L1 gene. The segment on the 19th chromosome represented by the SSR marker Satt313 was found to link with locus L1. The region was further delimited three times with increased SSR and InDel markers using a population derived from a heterozygous plant of CSSL124 from SojaCSSLP3. The L1 gene was finally located in a 184.43‐kb region between SSR_19p09 and Indel_19P7. Thirteen putative genes in this region were analysed with qRT‐PCR. The expression level of Glyma19 g27460, which is a member of the SANT superfamily with a MYB DNA‐binding domain, was significantly upregulated in black pods and was recognized to be the most likely candidate for the L1 gene. 相似文献

Summary Eight grain pearl millet (2n=14) accessions were crossed as male to hexaploid spring wheat cv. Fukuho (2n=6x=42). An average of 80% wheat pistils showed pearl millet pollen tube entry in the ovules, compared to 56% in wheat x maize cv. Seneca 60 cross. Of the 15 embryos, obtained through in vitro immature seed culture from wheat x pearl millet crosses, 3 plantlets were produced and grown to maturity. These three were of the somatic chromosome constitution 2n=42, 21 and 22, respectively. Haploid wheat plant (2n=21) apparently originated from pearl millet chromosome elimination during embryogenesis. The 22 chromosome plant had retained a single pearl millet chromosome at tillering stage, but this chromosome was eliminated from pollen mother cells prior to and also during gamete formation. Triticum sharonense was hybridized with autotetra-ploid T. speltoides, T. longissimum and T. bicorne. Meiotic analysis of these hybrids showed that T. sharonense is almost equally related to both T. speltoidcs and T. longissimum, while it is comparatively distant from T. bicorne. Summary The association of genes for purple pigment in the coleoptile with the chromosomes of the winter wheat variety Mironovskaya 808 was investigated using monosomic F2 analysis. The segregation ratio for F2 hybrids of Chinese Spring monosomics x Mironovskya 808 seems to indicate that the purple colour of the coleoptile is determined by two dominant genes, Rc3 and Rc4, which are located on the chromosomes 7D and 6B respectively, and which reinforce each other. Apart from these two genes, suppressors found on the chromosomes 2A, 2B, 2D, 4B and 6A also play a role in the intensity of the purple colour.With the aid of a Chinese Spring telocentric chromosome marker it was observed that the Rc3 gene is located on the chromosome arm 7DS, at a distance of 16±4.23 crossover units from the centromere. 相似文献

Eight plants of the putative double monosomic addition line (DMAL, 2n= 20) were developed by crossing a monosomic chromosome addition line of radish [f(A)‐type monosomic addition line (MAL) (2n= 19)] carrying the f chromosome of Brassica rapa (2n= 20, AA) with another [e(C)‐type MAL (2n= 19)] having the echromosome of Brassica oleracea (2n= 18, CC). The homoeological relationships between the two alien chromosomes were investigated by morphological, cytogenetic and random amplified polymorphic DNA (RAPD) analysis. Seventeen morphological traits that were not present in the radish cv. ‘Shogoin’ were observed in both MALs and these traits were substantially exhibited in DMAL plants. At the first metaphase of pollen mother cells (PMCs), the two parental MALs showed a chromosome configuration of 9II +1I, demonstrating impossibility of recombination between the R and the added chromosomes. The DMALs formed 10II in approximately 73% of PMCs, with one bivalent showing loose pairing between two chromosomes differing in size. In an attempt to identify the two MALs by RAPD‐specific markers using 26 selected random primers, 13 and 20 bands were specific for the f(A)‐type and the e(C)‐type MALs, respectively; 12 bands were common to both MALs (26.7%). In conclusion, the f chromosome of B. rapa is homoeologous to the e chromosome of B. oleracea. The genetic domain (genes) for 17 morphological traits are linked to each homoeologous chromosome bearing 27% of the corresponding RAPD markers. 相似文献

In cereals, chromosome doubling of microspore-derived haploid plants is a critical step in producing doubled haploid plants.
This investigation was undertaken to study the effect of incorporation of colchicine in the induction medium for anther culture,
and the effect of colchicine on anther culture-derived plants of triticale grown under controlled greenhouse conditions. In
the latter case, chromosome doubling of adult sterile plants derived from anther culture of fourteen triticale populations
was attempted, where androgenetic plants with non-dehiscent anthers were cloned and subjected to the colchicine treatment,
and then grown with the aid of hydroponics. The hydroponic system provided optimal conditions for recovery of the affected
haploids from the toxic effects of colchicine treatment and all colchicine-treated plants survived. A topcross-F1 (TC1F1) population with timopheevii cytoplasm produced the highest percentage of plants with seed-set either due to chromosome doubling
by colchicine (98%) or spontaneous doubling of chromosome number (15%). Colchicine-treated anthers performed inferior than
control in both induction and regeneration phases. One of the key observation of this study was the reversal from reproductive
stage back to the vegetative stage which in turn enabled further cloning of haploid plants under hydroponic conditions once
they were identified as sterile. The one hundred percent survival rate of in vitro-derived plants, 100% survival rate of colchicine treated haploid plants and the high chromosome doubling success rate (X
= 82.3) observed in this study imply that a temperature-controlled greenhouse with an hydroponic system provides an efficient
environment for inducing chromosome doubling of haploid plants in cereals.

该文探讨白桦愈伤组织染色体的制片方法,并对白桦愈伤组织及其再生植株的染色体数目变异进行了研究。结果表明:取继代培养1周的白桦愈伤组织,用0.2%的秋水仙素溶液预处理1.5~2 h,1 mol/L HCl室温(18~20℃)解离10~15 min或2%纤维素酶和果胶酶混合液室温解离30 min,压片后易获得分散、染色效果好的细胞分裂中期染色体图像。随着继代次数增加,白桦愈伤组织染色体数目变异的频率及范围增大。在愈伤组织分化过程中,二倍体细胞占有优势,能够分化为正常的植株,愈伤组织再生植株的变异频率低于愈伤组织。 相似文献
