不同温度下圆叶决明降解过程红壤可溶性氮及氮水解酶动态变化研究 总被引:1,自引:0,他引:1
为了探明圆叶决明(Chamaecrista rotundifolia)降解过程果园红壤供氮水平的变化规律,本研究采用模拟培养试验,研究15℃和25℃培养果园红壤硝态氮、铵态氮、可溶性总氮、可溶性有机氮含量的变化及脲酶、蛋白酶、天冬酰胺酶活性的变化。结果表明,圆叶决明降解过程果园红壤4种可溶性氮含量均显著提高,且在培养140 d达到最大值,25℃培养的效果更佳。其中,可溶性总氮、可溶性有机氮和硝态氮含量随时间的动态变化可用三次曲线方程来拟合。圆叶决明降解过程还能显著提高脲酶、蛋白酶和天冬酰胺酶的活性,其中蛋白酶活性和脲酶活性的变化可用指数方程和三次曲线方程来拟合。本研究认为圆叶决明降解过程可提高果园红壤可溶性氮含量,因此可通过翻压圆叶决明提高土壤的供氮水平。 相似文献

During the past two decades, food safety issues in China not only posed serious threats to Chinese consumers but also damaged the image of Chinese products internationally. In China, food safety is not only about scientific discoveries, advanced laboratories, and sanitation equipment; it is more about the role of different players in the food supply chain. The poultry meat supply chain is instrumental in the spread of the avian influenza A virus (H7N9), raising questions about how policymakers respond to such threats and whether industries need to be restructured to manage and control this epidemic so that it does not recur. As a short-term measure, to prevent the spread of this disease, government authorities enforced the closure of live bird markets (LBM) in disease-affected areas of China. However, in the long term, the poultry meat supply chain needs to be restructured. The aim of the current study was to analyze distribution channels for chicken meat in China and then describe arrangements in poultry meat sectors that incorporate small- and medium-scale producers into the supply chain while responding to shifts in LBMs. We also assessed the role of LBMs in spreading H7N9 and how these interventions affect the poultry meat supply chain in the Chinese market. 相似文献

Elimination of the CRISPR/Cas9 constructs in edited plants is a prerequisite for assessing genetic stability, conducting phenotypic characterization, and applying for commercialization of the plants. However, removal of the CRISPR/Cas9 transgenes by genetic segregation and by backcross is laborious and time consuming. We previously reported the development of the transgene killer CRISPR (TKC) technology that uses a pair of suicide genes to trigger self-elimination of the transgenes without compromising gene editing efficiency. The TKC technology enables isolation of transgene-free CRISPR-edited plants within a single generation, greatly accelerating crop improvements. Here, we presented two new TKC vectors that show great efficiency in both editing the target gene and in undergoing self-elimination of the transgenes. The new vectors replaced the CaMV35S promoter used in our previous TKC vector with two rice promoters to drive one of the suicide genes, providing advantages over our previous TKC vector under certain conditions. The vectors reported here offered more options and flexibility to conduct gene editing experiments in rice. 相似文献

为探明引起贵州省某鸭场雏鸭发病的病原及其致病性和耐药情况,本研究对该鸭场送的疑似细菌感染病鸭进行剖检,取鼻黏膜、心脏和肝脏等组织器官接种于培养基中进行细菌分离鉴定,通过对分离菌进行药敏试验、动物回归试验和毒力基因检测研究其耐药情况和致病性。结果显示,分离菌在血琼脂培养基上生长16 h后呈现为边缘整齐、有光泽的乳白色菌落,伴有β-溶血现象,经革兰氏染色后在生物显微镜下呈两端钝圆、弧状、排列无规则的革兰氏阴性短小杆菌,与霍乱弧菌相符;16S rDNA基因序列同源性及系统进化树显示,该分离菌与霍乱弧菌同源性高达99.6%~99.7%聚为一支;药敏试验结果显示,分离菌对大部分药物都表现为耐药,其中对氨苄西林、克林霉素、复方新诺明、苯唑西林和克林霉素等抗菌药耐药性较强,对头孢哌酮和头孢曲松敏感;动物回归试验显示,分离菌可导致试验组雏鸭5 d内全部发病死亡,表明该分离菌对雏鸭具有较强的致病性;毒力基因PCR检测结果显示,检测的霍乱弧菌相关毒力基因hlyA、ompW和chxA为阳性,而检测的O1群rfb、O139群rfb、tcpA及ctxA基因为阴性,表明本次分离的霍乱弧菌携带有致病基因,但不属于O1和O139血清群。结果表明,该鸭场雏鸭发病的疫情病原为非O1/O139血清群霍乱弧菌,该菌致病性强且对多种抗菌药物耐药。本试验结果为贵州省鸭霍乱弧菌病的防控提供了参考依据。 相似文献

Colletotrichum gloeosporioides (Penz.) Penz. & Sacc. is an important fungal pathogen known to cause glomerella leaf spot (GLS). The objective of this study was to analyze the infection of C. gloeosporioides on leaves of apple cv. Fuji (resistant) and cv. Gala (susceptible) and apply proteomics techniques to study the apple defense responses 48 h after inoculation (h.a.i.). On both of cultivars, C. gloeosporioides started to germinate at 3 h.a.i. on adaxial surface and produced appressoria adhering to epidermal cell juxtapositions. Histological analysis showed more stratified parenchyma in leaves of cv. Fuji than cv. Gala associated with differences in the chemical composition of cell walls. Total and unique proteins expressed by cvs. Fuji and Gala at 3, 12, 24 and 48 h.a.i. were detected by comparative proteomes analysis. A total of 42 unique proteins expressed at 24 and 48 h.a.i. were identified by MALDI/TOF mass spectrometry, and most of these proteins were identified as directly involved in defense responses. 相似文献

AIM: To investigate the effect of paricalcitol (P) on renal tubulointerstitial fibrosis and the underlying mechanisms in diabetic nephropathy (DN).METHODS: DN rat model was induced by a single intraperitoneal injection of streptozotocin after fasting. The animals were randomly divided into 2 groups:the DN rats in paricalcitol-intervened group (group P) were injected intraperitoneally with paricalcitol dissolved in propylene glycol after the day when the model was induced successfully at a dose of 0.4 μg/kg (3 times a week); the DN rats in DN group (group D) were given isopyknic propylene glycol. Normal control group (group C) was also set up. The samples of blood, urine and renal tissue were collected after intervention of paricalcitol for 12 weeks. The biochemical indexes were measured. The renal tissues were used for pathologic observation and determining the expression of transforming growth factor-β1 (TGF-β1), Wnt-4, β-catenin and Klotho by immunohistochemistry and Western blotting. In addition, the correlation among the above indexes was analyzed.RESULTS: (1) Scr, BUN and 24 h urine protein increased significantly in group D compared with group C, while decreased in group P compared with group D (P<0.05). (2) The area of renal tubulointerstitial fibrosis increased in group D compared with group C, while decreased in group P compared with group D (P<0.05). (3) The expression of Klotho decreased, while the expression of TGF-β1, Wnt-4 and β-catenin increased in group D compared with group C (P<0.05). Compared with group D, the expression of Klotho increased, while the expression of TGF-β1, Wnt-4 and β-catenin decreased in group P (P<0.05). (4) The expression of Klotho was negatively correlated with the fibrosis area, TGF-β1, Wnt-4 and β-catenin (P<0.05).CONCLUSION: Paricalcitol inhibits renal tubulointerstitial fibrosis in DN by promoting the expression of renal Klotho, and inhibiting Wnt/β-catenin signaling pathway activation and TGF-β1 synthesis. 相似文献

利用树轮学方法,以长白山雌雄异株植物红豆杉、山杨和水曲柳为研究对象,通过判定雌雄异株树种生长释放/抑制事件及其与气候因子的关系,以期揭示物种属性和性别因素对树木生长释放/抑制事件的影响及其对气候响应机制的差异性。结果表明:3种雌雄植株的生长释放事件大多发生在幼龄阶段,生理特征差异也可能导致不同雌雄植株发生生长释放事件的时间和频率存在一定差异性。性别因素对山杨和红豆杉雌雄植株的生长释放存在显著影响,山杨雌株和红豆杉雄株对外界环境干扰更为敏感,水曲柳雌雄株对环境干扰的响应差异不明显;山杨更易受到外界环境干扰发生生长释放事件;水曲柳植株发生生长释放/抑制事件可能与林窗干扰有关。红豆杉和山杨的生长释放事件与气候因子具有显著相关关系,红豆杉雄株与温度、降水则存在显著正相关关系(P0.05),山杨雄株与温度、降水则存在显著负相关关系(P0.05),山杨雌株与降水呈显著负相关关系(P0.05),植株的生长释放/抑制事件对气候的响应与树木径向生长--逐月气候因子的关系有所不同。因此应当考虑生长--气候因子之间的非线性关系以及物种和性别因素对生长--气候关系的影响,以期完善植株生长对气候的响应机制。 相似文献
