基因组印记的研究进展

综述基因组印记的可能发生机制及人和鼠中常见的印记基因，论述了且印记作用的生物学意义。     

葡萄属植物染色体DNA的提取、纯化及RFLP鉴定

利用ＪｅｎｎｉｆｅｒＡ．ＣｏｕｃｈａｎｄＰａｕｌＪ．Ｆｒｉｔｚ的改良方法提取葡萄嫩叶染色体ＤＮＡ，并进行ＲＦＬＰ分析。结果表明，该方法提取的ＤＮＡ均未降解；以Ｓｅｐｈａｒｏｓｅ２Ｂ层析柱纯化后可进行ＲＦＬＰ分析；核糖体ｒＤＮＡ的ＰＶｖｃ一Ａ片段制备的探针与ＨｉｎｄⅢ酶组合，使所有供试样品都显示多态性。     

Detection of Allelic Variation in Chinese Wheat (Triticum aestivum L.) Germplasm with Drought Tolerance Using SSR Markers

Allelic variation in two domestic wheat landraces, Pingyaobaimai and Mazhamai, two cornerstone breeding materials and their derived cultivars with drought tolerance was detected by SSR (simple sequence repeat) markers. The clustering of 25 accessions showed that the similarity between Pingyaobaimai and Yanda1817, the latter was developed from the former, was 0.71, the highest one of all accessions, but the similarities were very low between these two accessions and other accessions including their derived cultivars. A similar situation was revealed between Mazhamai and its derived cultivars. Pingyaobaimai and its three derived cultivars shared three alleles at loci Xgwm526, Xgwm538 and Xgwm126 on chromosome arms 2BL, 4BL and 5AL, respectively. There were six shared alleles in Mazhamai and its derived cultivars, in order of Xgwm157,Xgwm126, Xgwm212, Xgwm626, Xgwm471 and Xgwm44 on chromosome arms 2DL, 5AL, 5DL, 6BL, 7AS and 7DC, respectively. Only one shared allele was detected between the pedigrees of Pingyaobaimai and Mazhamai. The difference of shared alleles in two cornerstone breeding materials and their derived cultivars revealed the diversity in Chinese wheat germplasm with drought tolerance and the complication in genetic basis of drought tolerance in wheat.

复合地板是极具竞争力的新型内装饰材料，本文介绍了复合地板的特性及生产工艺，着重阐述了表板侧向预先胶合三层板坯结构常规加热生产工艺与表板在压机内挤紧五层板坯结构高频加热生产工艺二者的区别。     

~(14)C示踪研究不同小麦品种开花、灌浆期的光合效率及同化物的运转与分配

应用~(14)C示踪技术测定三个籽粒饱满度不同的小麦品种,结果表明:籽粒饱满度与品种的净光合率、光合持续期和同化物在籽粒中的分配率有关;光合作用后30分钟测定,~(14)C同化物的分配,叶片最高,叶鞘其次,麦穗再次,茎中最低;开花期~(14)C同化物在成熟麦穗中的分配,约占引入量的25—32％,灌浆期~(14)C同化物在成熟麦穗中的分配,对籽粒饱满的两个品种,占引入量的50％以上,而籽粒不饱满的品种,分配率还不到30％。     

Role of NF-κB in inducing HEL cell proliferation and apoptosis during human cytomegalovirus infection

AIM:To investigate whether human cytomegalovirus(HCMV) regulate human embryonic lung fibroblast(HEL) cell proliferation and apoptosis by activating NF-κB.METHODS:Immunohistochemistry and Western blot analysis were used to detect the NF-κB translocation and/or Bcl-2 and the levels of I-κBα during HCMV infection. Apoptotic cell were examined by flow cytometry, and the HEL cell proliferation was determined by MTT.RESULTS:The levels of NF-κB in the nucleus reached highly 48 h postinfection, and the levels of I-κBα were low 24 h postinfection. The activity of NF-κB was inhibited 120 h postinfection. The levels ofbcl-2was accorded with the activity of NF-κB. HCMV promoted HEL cells to proliferate before 72 h postinfection and induced apoptosis 120 h postinfection.CONCLUSION:NF-κB plays a role in HEL cell proliferation and apoptosis during HCMV infection, and it involves in the pathological mechanisms of diseases associated with HCMV infection.     

烟草高分子量核DNA提取方法比较

高分子量(HMW)核DNA的制备是构建BAC文库关键环节之一。为构建高质量的烟草BAC文库,作者研究了一次选择法和二次选择法提取烟草高分子量核DNA的效果。研究结果表明:虽然二次选择法得到的高分子量核DNA浓度较低,但此法省略了对细胞核连续3￣5次的漂洗,而且得到含DNA的凝胶块(plugs)数量是用一次选择法的3倍,因此得到高分子量DNA的总量更高;同时二次选择法得到的plugs纯度更高,排除了酶切时细胞杂质对HMWDNA的干扰,此外在节约材料等方面也优于一次选择法。

SR92鸡的OPAY02型标记初步研究

测定了SR92A系鸡初生、28、49、75日龄体重,屠体、半净膛、全净膛重等生产性能,同时用OPAY02随机引物对其DNA进行RAPD扩增,分析OPAY02型标记与鸡群早期增重及75日龄屠宰性能之间的关系。结果表明,片段长度为1 660.8bp和2 326.0bp的两条带所代表的位点,在SR92A系中出现A、B、C三种类型,其中C型母鸡75日龄体重超过A型607.4g,初步表明OPAY02C型标记在SR92A系中可作重要遗传标记。
