中国18 省市番茄晚疫病菌生理小种的鉴定   总被引:26,自引:2,他引:26  
 利用5 个含有不同单显性抗番茄晚疫病基因的番茄材料Ts19、Ts33、W1Va700、LA1033 和L3708为鉴别寄主, 对我国18 省、市、自治区的番茄晚疫病菌201 个纯化分离物进行了生理小种鉴定。结果共鉴定出8 个小种, 即生理小种T0、T1、T1 ,2 、T1 ,2 ,3 、T1 ,2 ,3 ,4 、T1 ,4 、T1 ,2 ,4 和T3。其中, 小种T1 和T1 , 2是主流小种, 地理分布最广和发生频率最高, 分别在13 和11 个省市出现, 占样本总数的28.8 %和28.4 %;其次是小种T0 和T1 ,2 ,3 , 分别在8 和7 个省市出现, 分别占1819 %和819 %; 再其次是小1 ,2 ,3 ,4 , 在4个省市有发生, 占710 %; 小种T1 ,2 ,4 、T1 ,4 和T3 分布均少, 仅在1~2 个省市发生, 分别占3.0 %、2.5 %和2.5 %。这是我国的首次报道, 为番茄晚疫病的抗源材料筛选和抗病育种提供了病理学依据。  相似文献

营养元素对木霉菌防治甜瓜枯萎病效果的影响   总被引:13,自引:1,他引:13  
为明确各种营养元素在木霉菌防治甜瓜枯萎病生防体系中的作用,研究了9种营养元素铜、锌、铁、硼、钼、钙、锰、镁及钾对甜瓜枯萎病菌菌丝生长、木霉菌T23菌丝生长及产孢量、甜瓜种子萌发及根系生长的影响.结果表明:10~1000μg/mL的硫酸铜、硫酸锌、硫酸亚铁及硼酸对镰孢菌菌丝生长有较强的抑制作用,而对木霉菌的抑制作用远小于镰孢菌.适量加入钼酸铵、硫酸亚铁、硫酸钙及磷酸二氢钾有利于木霉菌菌丝生长及孢子形成,对甜瓜种子萌发及胚根生长也有较强的刺激作用.木霉菌结合钼酸铵、硫酸锰和硫酸钙处理土壤能显著降低甜瓜枯萎病的病情指数,相对防效分别为73.95%、73.77%和71.36%.  相似文献

 在云南西双版纳的红掌上发现一种由细菌侵染引起的病害,从叶片的病组织上分离到具有致病性的杆状细菌,将分离的病原菌接种于健康的红掌上,表现出与田间一致的症状。感病初期在叶缘或叶脉间出现水渍状小点,后期病斑多呈棕褐色至黑色坏死,病健交界处多呈黄色。从接种发病的病斑与田间标样上分离的病原菌菌落形态完全相同。通过菌株的培养性状、常规生理生化特性及透射电镜下菌体形态观察结果,将病原菌初步鉴定为黄单胞菌属(Xanthomonas)。BIOLOG鉴定和致病性测定结果进一步鉴定病原菌为地毯草黄单胞菌花叶万年青致病变种Xanthomonas axonopodis pv. dieffenbachiae(Xad)(McCulloch & Pirone) Vauterin et al.  相似文献

Simple diagnosis by ethanol immersion (SDEI) to detect Glomerella cingulata was used to detect three other fungi that also cause latent infection of strawberry plants. Signs on strawberry leaves with asymptomatic latent infection by Colletotrichum acutatum became visible using SDEI. Salmon-pink conidial masses were produced in the acervuli on the treated leaves 5 days after incubation at 28°C. In the case of Dendrophoma obscurans, pycnidia with amber conidial masses formed 5 days after incubation at 28°C. The pycnidia were observed mainly on the ribs, and conidial masses exuded from the ostiole. These macroscopic conidial masses were similar to those of G. cingulata and C. acutatum. When water was dripped onto a lesion caused by D. obscurans, the pycnidia exuded white filamentous conidial masses, making the distinction of D. obscurans from G. cingulata or C. acutatum. On petioles with latent infection by Fusarium oxysporum f. sp. fragariae, white aerial hyphae grew out from the vascular tissues on the cut surface 3 days after incubation at 28°C and were easily observed by eye or with a loupe. Thus, SDEI was also useful for diagnosing latent infection of strawberry plants by C. acutatum, D. obscurans, and F. oxysporum f. sp. fragariae.  相似文献

Four foliar and two stem-base pathogens were inoculated onto wheat plants grown in different substrates in pot experiments. Soils from four different UK locations were each treated in three ways: (i) straw incorporated in the field at 10 t ha−1 several months previously; (ii) silicon fertilization at 100 mg L−1 during the experiment; and (iii) no amendments. A sand and vermiculite mix was used with and without silicon amendment. The silicon treatment increased plant silica concentrations in all experiments, but incorporating straw was not associated with raised plant silica concentrations. Blumeria graminis and Puccinia recondita were inoculated by shaking infected plants over the test plants, followed by suitable humid periods. The silicon treatment reduced powdery mildew ( B. graminis ) substantially in sand and vermiculite and in two of the soils, but there were no effects on the slight infection by brown rust ( P. recondita ). Phaeosphaeria nodorum and Mycosphaerella graminicola were inoculated as conidial suspensions. Leaf spot caused by P. nodorum was reduced in silicon-amended sand and vermiculite; soil was not tested. Symptoms of septoria leaf blotch caused by M. graminicola were reduced by silicon amendment in a severely infected sand and vermiculite experiment but not in soil or a slightly infected sand and vermiculite experiment. Oculimacula yallundae (eyespot) and Fusarium culmorum (brown foot rot) were inoculated as agar plugs on the stem base. Severity of O. yallundae was reduced by silicon amendment of two of the soils but not sand and vermiculite; brown foot rot symptoms caused by F. culmorum were unaffected by silicon amendment. The straw treatment reduced severity of powdery mildew but did not detectably affect the other pathogens. Both straw and silicon treatments appeared to increase plant resistance to all diseases only under high disease pressure.  相似文献

Fusarium head blight (FHB) is an important disease of wheat, which can result in the contamination of grains with mycotoxins such as deoxynivalenol (DON). Artificial inoculation of flowering ears with conidial suspensions is widely used to study FHB diseases. Our goal was to compare four inoculation treatments in which a conidial suspension was sprayed on flowering ears and to study the effect of the application of moisture during kernel setting and filling with a mist-irrigation system. Ten wheat genotypes were inoculated with a DON-producing Fusarium culmorum strain. Inoculation treatments varied in time of application of the inoculum (morning or evening) and in the method of controlling humidity during inoculation (bagging or mist irrigation). A wet season was simulated with a mist-irrigation system, keeping the crop canopy wet for at least 26 days after flowering. The severity of FHB symptoms (area under disease progress curve (AUDPC)), yield loss and DON contamination in the grains were determined. AUDPC data obtained with the different inoculation treatments were highly correlated (r=0.85–0.95). Mist irrigation after inoculation resulted in a higher mean disease severity, but in a overall lower toxin contamination as compared to the non-irrigated treatments. Genotypic differences in DON accumulation were present: for one wheat line toxin contamination significantly increased when irrigated, while two genotypes accumulated significantly less toxin. The closest relationships (r=0.73–0.89) between the visual symptoms and the DON content were obtained under moderate mean infection pressure. This relation between visual symptoms and the DON content deteriorated at higher infection levels.  相似文献

Fusarium culmorum is a phytopathogenic, toxigenic fungus causing seedling diseases, foot rot and head blight of cereals. For estimating competition effects in mixtures of two single-spore isolates, two winter rye single crosses were tested with either four isolates individually or four 1 : 1 mixtures of the same isolates in six field environments. Two isolates (FC46, FC64) were highly aggressive deoxynivalenol (DON) and 3-acetyl DON-producers, the other two (FC30, FC71) were medium aggressive nivalenol-producers. Rye heads were inoculated during flowering with conidia of pairs of isolates expressing similar (FC46 + FC64, FC30 + FC71) or contrary (FC46 + FC71, FC30 + FC64) levels of aggressiveness and similar or different concentrations and chemotypes of mycotoxins, respectively. Head blight rating and yield components relative to the non-inoculated plots were recorded as aggressiveness traits. Additionally, mycotoxin concentrations were measured in the rye grain. Random pathogen samples were re-isolated from heads at the onset of symptom development and analysed by molecular markers (RAPD–PCR) in one environment. Aggressiveness of the isolate mixtures was significantly lower than that of the isolates applied individually on both rye genotypes. Similarly, mycotoxin concentrations were significantly lower in the mixtures in seven out of eleven comparisons. Among the re-isolates, the component genotypes of a mixture significantly deviated from the inoculated 1 : 1 ratio when a particular isolate (FC46) was present in the mixture. This isolate displayed a superior competitive ability irrespective of the aggressiveness or mycotoxin profile of the mixing partner illustrating that pathogenic fitness is caused by additional factors that have not, as yet, been identified.  相似文献

A defined medium was developed in which to monitor deoxynivalenol (DON) accumulation, fungal growth and expression of genes involved in trichothecene biosynthesis (designated Tri genes). In liquid culture, DON accumulated shortly after maximum expression of Tri6 and coincident with expression of Tri5. This was generally 96 h after inoculation. The effects of sublethal concentrations of the fungicides azoxystrobin, trifloxystrobin, kresoxim-methyl and tebuconazole on biosynthesis of the trichothecene DON by Fusarium culmorum were studied using this medium. The strobilurin fungicides trifloxystrobin and azoxystrobin significantly reduced the accumulation of DON in culture medium at a range of concentrations. Kresoxim-methyl, also a strobilurin, and tebuconazole, a triazole, did not significantly reduce the accumulation of DON, although levels were lower than those in nonamended cultures. Trifloxystrobin significantly reduced the accumulation of DON when added to cultures before initiation of trichothecene biosynthesis. RT-PCR assays of the expression of Tri6 and Tri5 genes indicated that trifloxystrobin acted by inhibiting the initiation of trichothecene biosynthesis.  相似文献

The population of Phytophthora infestans in Brazil consists of two clonal lineages, US-1 associated with tomatoes and BR-1 associated with potatoes. To assess whether host specificity in these lineages resulted from differences in aggressiveness to potato and tomato, six aggressiveness-related epidemiological components – infection frequency (IF), incubation period (IP), latent period (LP), lesion area (LA), lesion expansion rate (LER) and sporulation at several lesion ages (SSLA) – were measured on detached leaflets of late blight-susceptible potato and tomato plants. Infection frequency of US-1 was similar on potato and tomato leaflets, but IF of BR-1 was somewhat reduced on tomato. Incubation period was longer on both hosts with US-1, although this apparent lineage affect was not significant. Overall there was no host effect on IP. On potato, BR-1 had a shorter LP (110·3 h) and a larger LA (6·5 cm2) than US-1 (LP = 162·0 h; LA = 2·8 cm2). The highest LER resulted when isolates of BR-1 (0·121 cm2 h−1) and US-1 (0·053 cm2 h−1) were inoculated on potato and tomato leaflets, respectively. The highest values of the area under the sporulation capacity curve (AUSC) were obtained for isolates of US-1 inoculated on tomato leaflets (6146) and for isolates of BR-1 on potato leaflets (3775). In general, higher values of LA, LER, SSLA and AUSC, and shorter values of LP were measured when isolates of a clonal lineage were inoculated on their original host than with the opposite combinations. There is evidence that there are quantitative differences in aggressiveness components between isolates of US-1 and BR-1 clonal lineages that probably contribute to host specificity of P. infestans populations in Brazil.  相似文献
