The present work deals with production of ethanol from sweet sorghum bagasse by a zygomycetes fungus Mucor hiemalis. The bagasse was treated with phosphoric acid and sodium hydroxide, with or without ultrasonication, prior to enzymatic hydrolysis by commercial cellulase and β-glucosidase enzymes. The phosphoric acid pretreatment was performed at 50 °C for 30 min, while the alkali treatment performed with 12% NaOH at 0 °C for 3 h. The pretreatments resulted in improving the subsequent enzymatic hydrolysis to 79-92% of the theoretical yield. The best hydrolysis performance was obtained after pretreatment by NaOH assisted with ultrasonication. The fungus showed promising results in fermentation of the hydrolyzates. In the best case, the hydrolyzate of NaOH-ultrasound pretreated bagasse followed by 24 h fermentation resulted in about 81% of the corresponding theoretical ethanol yield. Furthermore, the highest volumetric ethanol productivity was observed in the hydrolyzates of NaOH pretreated bagasse, especially after ultrasonication in pretreatment stage.  相似文献

The chopped sweet sorghum stalk was thin-layer-dried for long-term storage and ethanol production. The drying kinetics and the effects of drying temperature on the qualities of sweet sorghum stalk were investigated in this work. The results showed that the drying process could be simulated well by Wang and Singh's model. The diffusivity constant (D0) and active energy (Ea) were estimated as 4.4 × 10−5 m2/s and 21.4 kJ/(mol K) for drying the chopped fresh stalk. According to the sugar composition, browning degree, and fermentability of the dried stalk obtained at various temperatures, the approximate drying temperature could be suggested as 50-60 °C for application. In this range, the moisture of the chopped fresh stalk could drop below the safe moisture for storage in 7-5.5 h with 12.1-9.7% total sugar loss during the drying process.  相似文献

Marker-assisted selection (MAS) for resistance genes (R-genes), identified using molecular markers and quantitative trait loci (QTL) analysis, is now possible in many crops. MAS can be used to pyramid several R-genes into a single host genotype. However, this may not provide durable genetic resistance because the pathogen is exposed to a full homozygous pyramid during hybrid seed production and to a full heterozygous pyramid in the resultant hybrid. Alternative gene deployment strategies that generate genetic variability were analysed, for hybrid cereal cultivars of pearl millet, maize, sorghum and rice, using maintainer lines (B-lines) with two smaller complementary pyramids. An F1 seed parent, produced on two such B-lines, can be used to produce a three-way hybrid. All target loci are heterozygous for resistance alleles in the F1 seed parent, and the pathogen is exposed in the hybrid to a host population that is heterogeneous and heterozygous for alleles at the resistance loci targeted by MAS. Alternatively, single-cross hybrids can be made on seed parents that are maintained by two B-lines that differ for the complementary resistance gene pyramids. In a cross-pollinated crop, the B-lines are allowed to intermate to produce a synthetic B-line. In an inbreeding crop, the B-lines are equivalent to a two-component multiline variety. In inbreeding crops, because there is no intermating between the B-line components, the resultant synthetic seed parents have a higher frequency of genotypes with resistance alleles (R-alleles) at several resistance loci. However, in both cross-pollinated and inbreeding crops the genotypic structure in the hybrids is almost the same. All alternatives to a single-cross hybrid having a full pyramid produce hybrid cultivars having lower frequencies of resistance alleles. The frequency of genotypes having R-alleles at several loci increases greatly in both seed parent and hybrid when the overall frequency of R-alleles in the maintainer lines increases. This is simply done by adding a maintainer line that has a full pyramid or by the component lines having overlapping pyramids. Parasitic flowering weeds of the genus Striga aremajor biotic constraints to sorghum (Sorghum bicolor(L.) Moench) production in sub-Saharan Africa. The agar-gel assay was used to evaluate stimulation ofStriga hermonthica (Del.) Benth. seed germination by a complete F1 diallel involving nine sorghum cultivars and inbred lines. Striga populations from Mali and Niger were employed. The same genetic materials were planted in pot trials in both countries to observe striga plant emergence. Variation in hybrid performance was determined by general (GCA) and specific combining ability (SCA) effects, with preponderance of GCA, for both germination distance in the agar-gel assay and number of emerged striga. Reciprocal effects were significant only in the agar-gel assay and were unstable across striga populations. For lines and hybrids, estimates of broad-sense heritabilities were 0.97 and 0.91 for germination distance, and 0.38 and 0.58 for emerged striga, respectively. Only a weak positive relationship existed between in vitro germination distance and emerged striga number in the pot trial. Although selection for low germination distance has merit, valuable material with resistance mechanisms other than low stimulant production may be lost if these traits are not additionally assessed. Laboratory assays which allow a non-destructive, quick and economical screening for resistance mechanisms other than the low stimulant character are likely to increase the efficiency of breeding programs for striga resistance. The significant contribution of SCA effects indicates that thorough screening of testcrosses is indispensable for selection in hybrid sorghum breeding programs. The growth and production of sweet sorghum [Sorghum bicolor (L.) Moench] crops under semi-arid conditions in the Mediterranean environment of southern Italy are constrained by water stress. The effects of temporary water stress on growth and productivity of sweet sorghum were studied during three seasons at Rutigliano (Bari, Italy). The aim of this research was to evaluate the sensitivity of phenological stages subjected to the same water deficit. In a preliminary study it was observed that stomata closed when pre-dawn leaf water potential (Ψb) became lower than −0.4 MPa. This criterion was used in monitoring plant water status in three different plots: one never stressed and two stressed at different phenological stages (‘leaf’ and ‘stem’) when mainly leaves or stems were growing, respectively. An evaluation of the sensitivity of phenological stages subjected to identical water stress was obtained by comparing the above-ground biomass and WUE of drought crops with those of the well-irrigated crop (up to 32.5 t ha−1 of dry matter and 5.7 g kg−1). The sensitivity was greatest at the early stage (‘leaf’), when a temporary soil water stress reduced the biomass production by up to 30% with respect to the control and WUE was 4.8 g kg−1 (average of three seasons). These results help quantify the effects of water constraints on sweet sorghum productivity. An irrigation strategy based on phenological stage sensitivity is suggested.  相似文献

Effects of Popping on the Endosperm Cell Walls of Sorghum and Maize   总被引:1,自引:0,他引:1  
The structure of the vitreous endosperm of raw and popped grains of popcorn maize and sorghum has been examined by light and scanning electron microscopy. In both cereals, popping produces everted grains consisting of expanded endosperm foam attached to the pericarp and embryo tissue. As previously reported, each bubble of the foam is formed from an individual starch granule inflated by internal steam pressure. Large fissures may contribute significantly to the expansion of the endosperm foam. The cell walls of the vitreous endosperm of both cereals are shattered into small fragments, which separate slightly as the cell contents expand during popping. Despite this, the endosperm cells retain their polygonal outline. Intact cell walls of raw endosperm, wall fragments from popped endosperm foam, and fragments isolated after treatment of the foam withalpha -amylase, were visualised through the autofluorescence of their ferulic acid content. The in vitro digestibility of popped sorghum was unchanged compared to raw sorghum, whereas that of wet-cooked sorghum was greatly reduced. It is suggested that popping-induced wall fragmentation improves the accessibility of the protein and starch reserves of the endosperm to digestive enzymes.  相似文献

影响高粱高产的因素很多,包括品种、水肥、土质及气候等因素,为探究高粱的高产性能,寻找影响高粱高产的突破口,本研究运用单因素随机区组设计的方法对粒用高粱拔节期和灌浆期的最大需水量及其一生的耗水特性进行了研究。结果表明:随着灌水量的增加,耗水量增加,穗长和枝梗数显著性地增加,但灌水量120~140mm的穗长和枝梗数差异不显著;籽粒千粒重和产量显著性地增加,灌水量100~140mm的千粒重和产量差异不显著。高粱拔节期最大需水量为120mm;开花灌浆期最大需水量为100mm;高粱拔节前耗水48.74mm,占总耗水量的10%~15%;拔节~抽穗耗水140.51~177.12mm,占总耗水的35%~43%,抽穗-成熟耗水137.65~272.27mm,占总耗水的42%~55%;水分生产效率随耗水量的增大,呈现先升后降的趋势,灌浆期灌水100mm处理的水分生产效率为最高,最高可达29.49kg/(hm2·mm)。  相似文献

Arabinose to xylose ratio (A/X), phenolics, antioxidant capacity and gelling properties were evaluated in glucoarabinoxylans (GAX) extracted from white (W-GAX), red (R-GAX) and high tannin (T-GAX) sorghum brans (SB). The characterization of arabinoxylans from corn fiber (CFAX) was used as benchmark. Sorghum GAX had higher branched structure (A/X 1.08–1.41) than CFAX (0.59). Nine 3-deoxyanthocyanins (3-DAs) were identified in SB and two glycosylated forms remained associated to the R-GAX and T-GAX extracts. T-GAX was the only that contained tannins (0.41 mg catechin equivalents (CE)/g dry basis (db)) and exerted the highest antioxidant capacity (81.75 mM Trolox equivalents (TE)/g db) followed by R-GAX (48.49 mM TE/g db), which contained the highest amount of 3-DAs (0.11 mg Luteolinidin equivalents (Lut eq)/g db), and W-GAX (35.45 mM TE/g db) that was not significantly different from the CFAX (25.83 mM TE/g db). Among sorghums, only the W-GAX gelled but it formed a weaker gel compared to CFAX likely due to its lower hydroxycinnamic acids (HCA) concentration. The presence of 3-DAs in the structure of R-GAX and T-GAX affected negatively their solubility and gelling properties. The different SB showed potential as sources of GAX with antioxidant capacity.  相似文献

甜高粱茎秆切割装置的研究,对于提高其割茬整齐度和糖份利用率均有重要意义。通过分析滚刀式切割器切割原理,基于ANSYS/LS-DYNA软件建立甜高粱茎秆切割装置有限元模型,模态分析结果显示,刀片的固有频率为1 908.7 Hz,能够避免滚刀高速旋转情况下动刀片共振情况的发生。研究甜高粱切割过程中刀片的受力情况与动刀片参数的关系,以整机前进速度、刀片安装角度和滚刀转速为3个因素,以切割力为试验指标,利用正交试验的方法,得出当刀片切割力最小时,刀片安装角度为140°,滚刀转速为1 400 r/min,整机前进速度为4 km/h,为甜高粱茎秆切割装置的研究提供依据。  相似文献
