Analysis of Mycosporine-Like Amino Acids in Selected Algae and Cyanobacteria by Hydrophilic Interaction Liquid Chromatography and a Novel MAA from the Red Alga Catenella repens

Mycosporine-like amino acids (MAAs), a group of small secondary metabolites found in algae, cyanobacteria, lichens and fungi, have become ecologically and pharmacologically relevant because of their pronounced UV-absorbing and photo-protective potential. Their analytical characterization is generally achieved by reversed phase HPLC and the compounds are often quantified based on molar extinction coefficients. As an alternative approach, in our study a fully validated hydrophilic interaction liquid chromatography (HILIC) method is presented. It enables the precise quantification of several analytes with adequate retention times in a single run, and can be coupled directly to MS. Excellent linear correlation coefficients (R² > 0.9991) were obtained, with limit of detection (LOD) values ranging from 0.16 to 0.43 µg/mL. Furthermore, the assay was found to be accurate (recovery rates from 89.8% to 104.1%) and precise (intra-day precision: 5.6%, inter-day precision ≤6.6%). Several algae were assayed for their content of known MAAs like porphyra-334, shinorine, and palythine. Liquid chromatography-mass spectrometry (LC-MS) data indicated a novel compound in some of them, which could be isolated from the marine species Catenella repens and structurally elucidated by nuclear magnetic resonance spectroscopy (NMR) as (E)-3-hydroxy-2-((5-hydroxy-5-(hydroxymethyl)-2-methoxy-3-((2-sulfoethyl)amino)cyclohex-2-en-1-ylidene)amino) propanoic acid, a novel MAA called catenelline.     

3种含氯消毒剂对柑橘黄龙病菌室内防治效果研究

柑橘黄龙病是柑橘生产上最具毁灭性的病害，目前没有防治特效药也没有较好的抗性品种。本文以含氯消毒剂作为柑橘黄龙病菌防治的候选药剂开展药剂筛选试验。将染病接穗分别浸泡3种含氯消毒剂再嫁接到健康枳壳砧木上，利用定量PCR技术定量分析药剂处理前后黄龙病菌含量的变化，建立柑橘黄龙病菌药剂防治效果评价体系。试验结果表明，漂白粉（CH）不适合利用嫁接技术进行候选药剂筛选；二氯异氰尿酸钠（NaDCC）和三氯泡腾消毒片（TCCA）的抑菌效果随处理浓度的降低而减弱，在有效氯含量为5000mg/L时，病菌减退率最高，分别为99.1%和99.5%，相对防效最佳，分别达到了97.9%和98.8%，与对照药剂盐酸四环素(TET)的抑制效果相当，说明NaDCC和TCCA可以作为柑橘黄龙病化学防治的候选药剂。     

Expression Analysis of THBS3 Gene in Different Tissues and Skeletal Muscles at Different Periods from Landrace and Tongcheng Pigs

To study the expression pattern of THBS3 gene in different tissues and during skeletal muscle development, the THBS3 gene expression in different tissues and skeletal muscles during prenatal periods (33, 45, 65, 70 and 90 d) and postnatal periods (0, 9, 30, 60, 120 and 160 d) from Landrace and Tongcheng pigs were detected by Real-time quantification PCR.The results showed that THBS3 gene widely expressed in all tissues examined, exhibiting similar spatial expression patterns with expression peaks in lung in both pig breeds except in stomach and intestine.Moreover, although THBS3 gene showed a significant higher expression level in gestation than after birth in Landrace and Tongcheng pigs (P<0.05), it exhibited different expression patterns between Landrace and Tongcheng pigs, the expression peak was detected at gestation day 45 in Landrace pig, while was detected at gestation day 65 in Tongcheng pig.The results suggested that THBS3 gene involved in skeletal muscle growth and development in pigs, as well as the regulation of asynchronization of skeletal muscle development in different pig breeds.     

覆盖方式对旱地小麦花后旗叶抗氧化生理及粒重形成的影响

于2017—2018年和2018—2019年,在西北中部旱作雨养农业区以冬小麦‘康庄974’为试验材料,设秸秆带状覆盖(SM)、地膜覆盖(PM)和无覆盖对照(CK)共3个栽培处理,分析不同覆盖方式对小麦灌浆期土壤水分和温度及抗氧化能力的影响,探讨花后干物质积累量和粒重形成的关系。结果表明,随生育期推进,花后旗叶相对含水量(RWC)逐渐降低,丙二醛(MDA)含量逐渐升高,且PM较SM降、升幅度明显。与CK相比,覆盖显著提高花后旗叶抗氧化酶活性,增加渗透调节物质含量;开花时间越长,SM抗氧化酶活性升高的幅度越大,而PM主要提高花后7 d旗叶抗氧化能力。SM和PM的粒重分别较CK增加14.3%和19.1%(P<0.05)。相关分析表明,土壤水分(SW)是影响旗叶生理活性的关键因子,提高SW,有利于增加RWC(r=0.84^**),从而提高旗叶抗氧化能力,其中RWC和抗坏血酸酶(ASA)活性呈极显著正相关关系(r=0.82^**);ASA活性和脯氨酸(Pro)含量呈极显著正相关关系(r=0.94^**),和MDA含量呈极显著负相...     

基于斜率比法评定断奶仔猪对不同铜源的相对生物学利用率

本试验旨在研究不同铜源（硫酸铜、碱式氯化铜和柠檬酸铜）和铜添加水平（20和30 mg/kg）对断奶仔猪生长性能、血清铜含量与含铜酶活性以及组织铜含量的影响,探讨断奶仔猪对不同铜源的相对生物学利用率。试验采用3×2双因子随机区组设计,选取平均体重为（8.98±0.48） kg的杜×长×大三元杂交断奶仔猪112头,随机分为7组,每组4个重复,每个重复4头猪。对照组饲喂玉米-豆粕型基础饲粮（铜含量为7.80 mg/kg）,各试验组分别在基础饲粮中添加20或30 mg/kg硫酸铜、碱式氯化铜或柠檬酸铜（均以铜含量计）。试验预试期3 d,正试期28 d。结果表明：1）试验组与对照组断奶仔猪平均日增重、平均日采食量和料重比均无显著差异（P>0.05）。2）饲粮铜源和铜添加水平对断奶仔猪血清铜含量具有显著影响（P<0.05）,但对血清铜蓝蛋白和铜锌超氧化物歧化酶活性均无显著影响（P>0.05）。3）饲粮铜源和铜添加水平对断奶仔猪肝脏铜含量具有显著影响（P<0.05）,但对心脏、肾脏、胰脏和跖骨铜含量均无显著影响（P>0.05）。4）根据多元线性回归斜率比法计算,以血清铜...     

Solution the Polarization Field Strength inside Ellipsoid in a Homogeneous Dielectric

The solution of polarization field strength in ellipsoid and a homogeneous dielectric in condition of a constant external field is obtained with strict derivation and ellipsoidal coordinate. The conclusion that some textbooks given: the direction of polarization field strength with the direction of external field holds strict anti-parallel must be supplemented with additive certain conditions, otherwise it will not be correct. By means of the latticework and the step change calculation, the angle value of polarization vector with the external field is calculated and the changing situation about the angle value with the change of three half axes of ellipsoid is analysied.     

水库汛期分期方法研究及其应用

应用矢量统计法和相对频率法，基于年最大值取样以及超定量取样进行汛期分期计算，并与现有的方法进行比较研究。实例应用结果表明，这两种方法均可用于汛期的分期计算中，具有一定的实用价值。
