Continuous Plant Regeneration from Established Embryogenic Cell Suspension Cultures of Italian Ryegrass and Tall Fescue 总被引:8,自引:0,他引:8
The suitability of different protocols was compared for entire plant regeneration by somatic embryogenesis, of the forage plants Lolium multiflorum Lam. (Italian ryegrass) and Festuca arundinacea Schreb. (tall fescue). In the first protocol, miniature embryos were used as starting material, while mature seeds were retained in the other two. Whichever the considered protocol, undifferentiated calli were produced on Murashige and Skoog MS medium supplemented with 2,4-D. The calli were subcultured in the dark on solid MS agar medium, containing 5 mg/1 2,4-D (protocol 2) or on solid MS medium followed by transfer to a rotated liquid MS medium with 2 mg/1 2,4-D (protocol 1). In these conditions, induction of somatic embryogenesis occurred, and whole plants were regenerated during a limited lapse of time, upon transfer in the light, to MS medium supplemented with BAP but devoid of 2,4-D. The simultaneous elimination of 2,4-D and transfer to light appeared essential for full regeneration of the plants. Using this characteristic, an additional step was added to a new protocol (protocol 3) in which microcalli, cultured on liquid MS medium containing 5 mg/1 2,4-D, were transferred to the same medium with 2 mg/1 2,4-D, in the dark. In these conditions, the suspensions kept their embryogenic potential for months. In all cases, plantlets were successfully transferred into the soil. An evaluation of the somaclonal variation potential of the plants issued from each protocol is now underway. 相似文献

Characterization of the durable resistance to yellow rust in old winter wheat cultivars in the Netherlands 总被引:2,自引:0,他引:2
Summary Winter wheat cultivars released in the Netherlands before 1930 carried durable resistance to yellow rust. Cultivars released in the period between 1930 and 1950 often were durably resistant while recent cultivars infrequently showed durable resistance. This durable resistance was not difficult to transfer to new cultivars. Twenty nine older cultivars with durable resistance and eight recent non-durably resistant cultivars were tested in the seedling stage and in the adult plant stage against 12 West-European yellow rust races and against some non-European races in the seedling stage only. The adult plant tests were carried out in race nursery tests in the Flevopolder. Per race nursery all 37 cultivars, planted in hills of about 20 plants on both sides of the highly susceptible cv. Michigan Amber, were exposed to one race.The infection type of each cultivar-race combination was scored on 0 to 9 scale once in the seedling stage and twice in the adult plant stage. In the race nurseries the percentage leaf area affected was evaluated three times to be used to calculate the area under the disease progress curve (AUDPC). This AUDPC multiplied with the mean infection type in the field gave the susceptibility index (SI).The infection types were classified into resistant (R), intermediate (I) or susceptible (S) when the infection types were 0 to 3, 4 to 6 or 7 to 9, respectively. Four categories of resistance were discerned on the basis of the three infection type scores: 1) RRR, overall resistance; complete or near-complete resistant at all stages. 2) SRR, adult plant resistance, complete- or near-complete resistant at the adult plant stage only. 3) SRS and SSR, temperature sensitive resistance, the resistance changed from the one evaluation data to the other. 4) SSS and an SI lower than that of Michigan Amber, partial resistance.The frequencies of overall, adult plant and temperature sensitive resistance were 1.4, 52 and 54% in the older cultivars and 40, 62 and 22% in the recent ones, respectively. Among the older cultivars all had a fair to high level of partial resistance, the SI being on average only 20% of that of Michigan Amber, while most cultivars also seemed to carry temperature sensitive resistance. The partial resistance of the recent cultivars was of a much lower level with a mean SI compared to that of Michigan Amber of 61%. Partial resistance was highly correlated (r = –0.94) with the mean resistance scores from the Dutch Recommended Cultivars Lists. Summary Fifty-five accessions of Triticum species were analyzed for content of hydroxamic acids (Hx), a natural resistance factor against various organisms. Hx were found in all accessions analyzed. Extreme values were found in wild diploid species: highest in T. speltoides (16.0 mmol/kg fr. wt) and lowest in T. tauschii (0.21). Modern polyploid wheats sharing the same genome did not show substantial variations in Hx levels. The data suggest possible sources of high Hx levels for wheat breeding programs. 相似文献

Summary Seedlings of Trifolium repens showed considerable variation with regard to the morphology and growth of their calli, and their ability for in vitro differentiation of shoots. One of the lines selected for regeneration in primary callus cultures also showed shoot formation from protoplasts. Somatic embryogenesis in callus cultures of T. pratense and T. arvense occurred only in selected seedling lines. Summary Improvement of durum wheat at the International Maize and Wheat Improvement Center (CIMMYT) was achieved by exploiting the variability of the tetraploid species in large numbers of combinations, and multi-location testing of germplasm on a world wide base. High yielding, widely adapted durums with short straw, good fertility, and adequate earliness were selected for distribution to major durum growing countries. Present attempts to improve durum wheat are directed at adjusting agronomic traits such as leaf angle and straw strength, and increasing spike fertility and spike size, thereby reaching higher grain numbers per unit area and higher yield potential. Summary Seedling emergence was closely correlated with coleoptile length and plant height among parents, F2 and F3, populations of crosses involving dwarf wheats Olesen Dwarf (CI 14497), Norin 10 derivative D6301, Tom Thumb derivative D6899, and the standard-height varieties Ramona 50 and Nainari 60. Genetic mechanisms that governed plant height also influenced coleoptile length, but the relative effects of genes showing dominant or epistatic effects appeared to be different. With respect to the two parents involved in each of 15 crosses, mean F2 coleoptile lengths were consistently closer to the low parent value than were corresponding mean F2 plant heights. A slight curvilinear relationship was also found between coleoptile length and plant height of F3 lines. The results suggest that selection of semidwarf wheats with long coleoptiles and improved emergence properties from crosses involving the dwarf wheats of this study would be unlikely. 相似文献

Summary In vitro pollen germination at 10°C, 14°C and 22°C of four groups of two pure line tomato varieties was compared with their plant growth at 19°C D/10°C N under controlled environmental conditions. Generally, pollen germination was slow at 10°C but after 6 h the percentage of germination was similar to that at 22°C. Maximum germination was obtained at 14°C already after 4 h. The longest pollen tubes occurred at 22°C. The two varieties within each group differed significantly from each other for percentage of pollen germination. For one group, this difference was greater at 10°C than at 22°C. The varieties in two groups also differed significantly for pollen tube growth. These differences in pollen tube growth rate were greater at 22°C than at 10°C. There was no clear relationship between pollen germination, pollen tube growth and plant growth in any of the four pairs of varieties. The results are discussed with regard to the possibility of pollen selection at low temperature in order to improve the efficiency of breeding for growth at low temperature. 相似文献

Selection for lodging resistant cultivars in cereal breeding programs is difficult due to the challenge of screening for this
trait under natural field conditions. The identification of easily measurable culm traits related to lodging resistance would
simplify the selection process. The present study was conducted to determine if differences in culm anatomy exist among modern
wheat genotypes differing in lodging susceptibility, and to determine the association between culm characters and lodging.
From a 2-year field study conducted in Edmonton, Alberta, 13 spring wheat cultivars were chosen based on predetermined susceptibility
to artificially induced lodging. Morphological and anatomical culm measurements were made visually and with an environmental
scanning electron microscope. Genotypes differed (P < 0.05) for plant height, number of internodes per culm, basal internode length and diameter, culm wall thickness and the
number of vascular bundles, but not for adventitious root frequency, lumen diameter or sclerenchyma ring thickness. Mean genotype
field scores for artificially induced lodging were correlated (P < 0.05) with plant height (r= 0.51) and the length of the fourth basal internode (r= 0.51). Short, wide basal internodes and thick culm walls were characteristic of three lodging tolerant genotypes: Kohika,
Sapphire and Olso. Nevertheless, despite such apparent genotype specific association between culm anatomy and field lodging,
general applicable associations were not observed for most traits. The most practical and easily selectable trait for lodging
resistance within a wheat breeding program remains plant height.

A wheat microsatellite locus, Xgwm 261, whose 192-bp allele closelylinked to the dwarfing gene Rht8, on chromosome 2D, was used toscreen 71 wheat cultivars from 13 countries to assess the variation at thislocus. Screening of this wheat collection showed that a 165-bp allele anda 174-bp allele were the most frequent. None of the New Zealand cultivarspossessed a 192-bp allele specific to Rht8, while only one cultivarfrom the US produced this important allele. The frequency of a 192-bpallele among these wheat cultivars was 5.63%. The highest allelefrequency was observed for a 174-bp fragment (52.11%) followed by a165-bp fragment (26.76%). The only durum wheat `Cham 1', did notshow any amplification due to the absence of D genome. The objective was to study the genetic basis of adult plant resistance to powdery mildew of the winter wheat line RE714 by quantitative trait loci (QTL) analysis and to investigate the stability of the QTL detected in two different genetic backgrounds. Two DH populations from the crosses between RE714 and the susceptible parents ‘Festin’ and ‘Hardi’ were used. Reaction of the DH lines to powdery mildew was assessed in different environments in Belgium under natural disease infection. Considering both populations and according to the environment tested, one to seven QTL were detected. Among them, residual effects of the race‐specific resistance genes Pm4b and MIRE were found. Two major QTL were very stable (on chromosome 5D and at the MIRE locus), since they were detected in both populations and over all environments tested. The QTL detected varied according to the susceptible parent used, and a residual effect at the Pm4b gene was not observed with the genetic background of ‘Hardi’. 相似文献
