Protein expression of cyclin D2 and p16 in proliferation and differentiation of cultured cardiac myocytes

AIM:To investigate the protein expression of cyclin D₂ and p16 in proliferation and differentiation of cultured cardiac myocytes.METHODS:One-day-old Sparague-Dawley rats were used. Cardiac myocytes(CM) were collected by a trypsin-dispersal method and cultured. Cell growth line and fluorescence activated cell sorting (FACS) were used to investigate the proliferation of CM. Ultra-thin sections were made to observe the ultrastructure of CM under transmission electron microscope. The expression of cyclin D₂ and p16 in CM were measured using immunocytochemistry and image analysis.RESULTS:①Results of cell growth line and FACS analysis showed that cultured CM could proliferate in the first 3 cultured days, but the ability decreased quickly, concomitant with differentiation. CM was obseved quiescent in cell cycle three days later. The ultrastructure of CM showed the large amount of myofilaments and mitochondrion. ②The protein expression of cyclin D₂ in 3,4,5 day CM group was 0.89 times(P＜0.05),0.80 times (P＜0.05) and 0.56 times (P＜0.01) of that in 1 day group, respectively. The expression of p16 in CM was increased during the culture process, 2,3,4,5 day group were 1.63 times, 1.72 times, 1.99 times and 2.84 times (P＜0.01) of that in 1 day group, respectively.CONCLUSION:Cultured neonatal rat cardiac myocytes could proliferate during the first 3 days after incubation, but the ability of proliferation decreased, from the fourth day, concomitant with differentiation. Cyclin D₂ and p16 play the key roles in CM postnatal development. Downregulation of cyclin D₂ and upregulation of p16 may induce CM differentiation.     

钙、萘乙酸对亚精胺在猕猴桃贮藏期间作用效果的影响

以5年生中华猕猴桃品系(湘源81-2)为试材,进行了果实采收后Spd(亚精胺)、Spd+Ca2+、Spd+NAA浸果处理,研究钙、萘乙酸对多胺作用效果的影响。结果表明:不同处理均不同程度地抑制了果实的呼吸强度、果肉PG活性和细胞膜透性的增加,其中Spd+NAA浸果处理的作用效果最为显著,贮藏68d时,果肉硬度最高(0.259MPa),软果率较低(79%),好果率达100%;其次为Spd处理。     

一个马铃薯Y病毒山东分离物的分离与鉴定

 从具有典型花叶症状的马铃薯叶片中分离到马铃薯Y病毒(Potato virus Y,PVY)(本文称PVY-SD-TA分离物),扩繁后,提纯病毒,电镜下可观察到700~900 nm×11 nm的病毒粒体,病组织超薄切片观察可见风轮状的内含体结构,寄主反应特性研究表明其能侵染2科13种植物。SDS-PAGE电泳检测病毒编码的外壳蛋白亚基的分子量为33 kDa。以PVY-SD-TA基因组RNA为模板,应用RT-PCR方法和特异引物合成了外壳蛋白基因。对cDNA全序列分析表明,PVY-SD-TA CP基因核苷酸序列与N株系的同源性为96%,与GenBank中登录序列号为AJ390306的O株系分离物的同源性最高,为99%;与国内不同学者报道的PVY中国流行株的同源性分别为96%,97%和98%。通过以上生物学特性和分子水平的研究将PVY-SD-TA鉴定为普通株系(PVY^O株系)。     

热胁迫对辣椒花柱细胞中Ca2+ 分布的影响

 用焦锑酸钾沉淀法研究了热胁迫对辣椒开花前后柱头和花柱细胞中Ca²⁺分布的影响。结果表明: 常温下Ca²⁺ 分布在柱头细胞表面、花柱引导组织细胞间隙等质外体系统中, 且在花粉管上呈先增加后减少的规律性变化; 热胁迫后, 除细胞间隙等质外体系统外, 细胞质和细胞核中也出现Ca²⁺, 花柱引导组织中通过的花粉管发生扭曲、变形, 这可能与Ca²⁺ 的异常分布有关。     

‘禅寺丸’甜柿2n花粉形成机制的研究

 以2n花粉发生频率较高的甜柿品种‘禅寺丸’为试材，研究影响2n花粉形成的减数分裂行为。结果表明：2n花粉形成受融合纺锤体(fused spindles)，八字形纺锤体(triangle spindles)及后期Ⅱ染色单体不正常分离(abnormal disorientation of sister chromatid)3种减数分裂行为控制；融合纺锤体及八字形纺锤体形成的2n花粉占2n花粉总数的94.8%，在遗传上等同于FDR(first division restitution，第1次分裂重组)型配子；由姊妹染色单体不正常分离形成的2n花粉在遗传上等同于SDR (second division restitution，第2次分裂重组)型配子。甜柿2n花粉绝大多数为FDR型，因而在倍性育种中有重要的应用价值。     

硅对黄瓜幼苗生长及活性氧清除系统的影响

在水培条件下,研究了营养液中硅对黄瓜幼苗生长和活性氧清除系统的影响。结果表明,施硅处理可以促进黄瓜植株的生长,叶片中叶绿素含量增加,内源抗氧化物质抗坏血酸(AsA)含量增加,超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性升高,丙二醛(MDA)含量降低。     

A coat protein transgene from a Scottish isolate of potato mop-top virus mediates strong resistance against Scandinavian isolates which have similar coat protein genes

Resistance tests were made on seedlings of transformed lines of Nicotiana benthamiana which contain a transgene encoding the coat protein (CP) gene of a Scottish isolate of potato mop-top virus (PMTV). This transgene has been reported to confer strong resistance to the PMTV isolate from which the transgene sequence was derived and also to a second Scottish isolate. Plants of lines of the transgenic N. benthamiana were as resistant to two Swedish and two Danish PMTV isolates as to a Scottish isolate, and of five lines tested, greater than 93.5% of transgenic plants were immune. The coat protein gene sequences of these four Scandinavian isolates were very similar to those of the two Scottish isolates. The greatest divergence between the isolates was three amino acid changes and there was less than 2% change in CP gene nucleotide sequence. It is concluded that the PMTV CP transgene used in these experiments could confer resistance against isolates from different geographical areas because it is becoming apparent that the CP genes of PMTV isolates are highly conserved.     

替米考星治疗人工感染巴氏杆菌病仔猪试验

研究了国产替米考星注射用药对人工感染多杀性巴氏杆菌病猪的治疗效果,用断奶仔猪进行试验,为临床使用提供了依据.结果表明,替米考星治疗急性猪肺疫效果确实, 推荐剂量为10 mg/kg体重.     

花粉多糖注射剂质量标准中几个项目测定

根据中国药典对注射剂的要求,对花粉多糖的蛋白质、鞣质、树脂、草酸盐、炽灼残渣、钾离子、砷盐、重金属含量进行测定,均符合规定,达到质量标准。     

重组鸡IL-2增强鸡四联灭活苗免疫效果的研究

使用在原核表达系统中表达的重组鸡白细胞介素 2 (IL 2 )蛋白 ,经过初步的分离纯化 ,与鸡新城疫 禽流感 法氏囊 传染性支气管炎四联油乳剂灭活苗同时使用 ,检测其对疫苗的免疫增强效果。试验共分 5组 ,每组 1 0只鸡。其中对照组只注射四联油乳剂灭活苗 ;4个试验组 ,在注射疫苗的同时分别注射重组鸡IL 2蛋白 0 0 5mg、 0 0 1mg、 0 1 5mg、 0 2 0mg。试验组与对照组同时各注射新城疫等多联油苗 0 5mL。采用HA、HI试验测定鸡新城疫、禽流感抗体滴度。结果表明 :重组鸡IL 2对该四联油乳剂灭活苗有较强的免疫增强作用。以第 3、 4组的增强效果较为明显 ,其中对鸡新城疫病毒和禽流感抗原的抗体效价 ,与对照组相比分别提高 2 42 8和 2 2 3 0个滴度。重组鸡IL 2在使用的过程中 ,没有发现任何毒副作用 ,证明是一种安全高效的免疫增强剂。
