The freshwater snails belonging to the genus Melanoides Olivier, 1804 are widespread across tropical regions of the world and endemic species have evolved in the African Lakes Malawi, Mweru and Tanganyika. The endemic Melanoides species of Lake Malawi have been investigated several times during the last century, due to their large conchological variation, but no unambiguous answer regarding the number of species has been given. The phylogenetic relationship between morphs or genetic clones of Melanoides in Lake Malawi was inferred by phylogenetic analyses of DNA sequence data from the mitochondrial genes 16S and COI. Additional sequences from GenBank were included to investigate the relationship to other morphs from different parts of the world. For the first time, a putative secondary structure was developed for a partial region of 16S in this genus to identify the variability of the secondary structure in stems and loops. The molecular analyses indicated that several genetic clones exist in Lake Malawi and that M. tuberculata is a paraphyletic taxon. It is not clear from the results whether invasions or dispersals account for the complex situation in Lake Malawi. The basal position of M. admirabilis, endemic to Lake Tanganyika, in the inferred phylogeny indicates that Africa might be the origin of the genus. The results further indicate that three major clades of Melanoides, consisting of several genetic clones, are present in Lake Malawi; one clade consisting of invasive M. tuberculata, another of native M. tuberculata and a third consisting of the M. polymorpha- complex. It appears as if the unique development of morphs within the Melanoides genus in Lake Malawi has evolved primarily by divergence of genetic clones instead of species differentiation. 相似文献

The phylogenetic relationship between different Cichorium intybus cultivars and cultivar groups was studied. A total of 127 random amplified polymorphic DNA markers have been used to distinguish between 51 Cichorium intybus cultivars belonging to three different cultivar groups. A principal component analysis was performed on the complete data set and a clear grouping of chicory cultivar groups was observed. The three chicory groups tested (root chicory, witloof and pain de sucre) were separated from one another in the principal component plots. The first two axes accounted for 43.9% of the total variation. Although the chicory used in traditional forcing in soil and the red chicory cultivars were related more to each other and to the chicory cultivars from the witloof group, they still formed separate groups in the principal component plot. 相似文献

Genetic diversity in 49 plants of 16 accessions representing one wild and two cultivated taxa of Cynara cardunculus L. (wild cardoon, cultivated cardoon and globe artichoke) was investigated using amplified fragment length polymorphic (AFLP) analysis. Four pairs of primers identified a total of 264 scorable loci, 196 of which were polymorphic. The number of markers scored per primer combination ranged from 41 to 107, with an average of 66 markers per primer combination. All the primer combinations detected polymorphism. The data were used for cluster analysis and AMOVA. The clusters obtained are in agreement with the current botanical classification and the differences among the taxa were substantial. The eco‐geographical groups within wild cardoon are clearly separated in relative subclusters, and reflect the geography of the collection areas. The six clones of the four cultivated artichoke varieties were clearly separated into four subclusters, corresponding to the current classification which is based on morphological traits. Genetic relationships among 18 accessions, including 16 of Ananas and two of Pseudananas, were investigated using RAPD molecular markers. The procedure for DNA extraction was adapted from the method of Dellaporta
et al. (1983) where an incubation in proteinase K and a purification step were included. From the total of 148 markers scored,132
(89.2%) were polymorphic. The similarity matrix was used for cluster analysis. The phenogram developed from the RAPD bands
showed that for most of the cases, the accessions within a species grouped together. Nevertheless, a moderate infraspecific
genetic variation was observed. For example, DNA data grouped all A. comosus accessions with a mean similarity coefficient of 0.85. Comparable results were obtained with all other species investigated.
The highest genetic divergence was found withinA. lucidus where the mean similarity coefficient among accessions was0.75. A similar level of genetic polymorphism was observed among
species,therefore, a definition about which species were involved in the constitution of A. comosus genotypes was not possible. These results agree with the breeders standpoint suggesting that all Ananas species belong to the primary gene pool of pineapple.

Summary Fifty four hybrid plants between Elymus canadensis and Psathyrostachys juncea were obtained by handpollination and embryo culture. The average cross compatibility between both species was 31.2 percent. One amphiploid plant was induced by colchicine treatment. The hybrid and amphiploid plants resembled P. juncea in appearance but showed a higher plant height and dry matter yield than the parents. The hybrids showed extremely low pollen stainability and were completely sterile. With the exception of one plant (2n=3x+1=22), all hybrid plants were allotriploids (SHN, 2n=3x=21). The amphiploid plant (SSHHNN, 2n=6x=42) showed 58.9% pollen stainability and 11.6% seed fertility.Mean chromosome associations of the hybrids and amphiploid at metaphase I were 0.02IV+0.06III+2.03II+16.91I and 0.07III+18.00II+5.85I, respectively. Lagging chromosomes, chromosome bridges, abnormal cytokinesis, and micronuclei were occasionally observed at the anaphase, telophase, or tetrad stage. 相似文献

Summary Male meiosis of the interspecific hybrid between Allium roylei and A. cepa is undisturbed relative to its parents. Based on meiotic data, A. roylei is concluded to be a closer relative of A. cepa than A. fistulosum. Segregation ratios for downy mildew resistance among BC1 and F2 progenies from the F1 between A. roylei and A. cepa indicate the presence of two dominantly inherited, weakly linked nuclear resistance genes, Pd
1 and Pd
2, in A. roylei (recombination frequency 0.32±0.03). Presumably Pd
1 and Pd

Summary The genetic variation among 23 accessions of 5 species in the subgenus Ceratotropis, genus Vigna, were investigated by random amplified polymorphic DNA (RAPD) analysis. A total of 404 fragments amplified with 24 primers were scored and analyzed by cluster analysis. The accessions used were separated into two main groups with an average of 70% differences. Within the main groups, five subgroups were recognized, which are in complete agreement with taxonomic species. Wild forms were always grouped with their most closely related cultivated forms and they showed variation in each species. The largest intraspecific variation was found in V. radiata (mungbean), in which wild forms (V. radiata var. sublobata) were highly different from each other and from cultivated forms. V. angularis (adzuki bean) showed the least variation and thus, was probably differentiated in relatively recent times. 相似文献

In this study, we report the use of ISSR to assess genetic diversity and to determine the relationships among ten cultivars
of common bean developed in Argentina and three materials from France. ISSR markers resolved two major groups corresponding
to the Andean and Mesoamerican gene pools of common bean. We compared the results of previous analysis, performed with RAPD
markers (Galván et al., 2001), with the results generated by means of ISSR. It appears that ISSR are better tools than RAPDs
to identify beans by gene pool of origin though they did not revealed as many differences between individuals as RAPDs.
