Marteilia sydneyi is the causative agent of QX disease in Sydney rock oyster, Saccostrea glomerata . It is responsible for disease outbreaks among oysters that occur during summer and can result in up to 95% mortality. QX disease has significantly decreased S. glomerata production in some areas of Australia's eastern seaboard over the past 30 years. Marteilia sydneyi sporulates in the digestive gland of oysters leading to complete disorganization of the infected tissues. The current study used proteomics to identify potential molecular markers of sporulating M. sydneyi infection during a field trial undertaken in the Georges River, Sydney, between December 2006 and May 2007. Early stages of M. sydneyi infection were detected by polymerase chain reaction, whilst cytological examination was used to identify sporulating M. sydneyi in the gut. Protein expression in oyster haemolymph was assessed during the M. sydneyi infection period by two dimensional electrophoresis. Proteome maps identified significant differences in the expression of four proteins in oysters with sporulating M. sydneyi infections.  相似文献

A 2‐year‐old neutered male domestic shorthair cat was presented to the emergency service of the National Veterinary School of Toulouse (France) for acute vomiting and diarrhea with lethargy, inappetence, and adypsia for the past 48 hours. Complete blood counts were performed with the ProCyte DX at the emergency department and with the Sysmex XT‐2000iV at the laboratory 2 weeks later. The scattergrams from the two analyzers revealed similar unusual and abnormal dot plots. The Sysmex XT‐2000iV DIFF scattergram also showed no clear separation between different leukocyte populations. The eosinophil cluster was in an abnormal location compared with that of the “typical” location in a normal cat. A blood smear evaluation revealed the presence of numerous mast cells. Thus, we hypothesized that the Sysmex XT‐2000iV had detected the mast cell population, and this led to errors in the differential counts. To explore this hypothesis, we manually gated on the DIFF scattergram and performed a manual differential on the blood smear. With this new gating strategy, the Sysmex XT‐2000iV and manual differentials were similar. Thus, in the case of systemic mastocytosis, mast cells can be located between the lymphocyte, monocyte, and eosinophil clusters on scattergrams.  相似文献

cDNA-SCoT差异显示分析是应用于植物基因差异表达研究的一项新技术.本研究以甘蔗品种桂糖11号根尖为材料,提取总RNA并反转录第1链cDNA,对影响甘蔗cDNA-SCoT差异显示PCR反应的cDNA模板、SCoT引物、dNTPs、Taq DNA聚合酶和Mg2+等因子进行优化,建立甘蔗cDNA-SCoT反应体系,同时应用优化后反应体系分离25%PEG6000胁迫诱导甘蔗根尖差异表达基因片段.结果表明:各项因子均对PCR扩增效果存在影响,20μL反应体系中各因子的最佳含量如下:cDNA 80 ng、引物浓度1.0 μmol/L、Taq DNA聚合酶2.0 U、dNTPs浓度200 μmo1/L、Mg2+浓度1.8 mmol/L.研究结果还显示,SCoT引物扩增多态性丰富,平均每条引物扩增条带30条,多态性条带比率为46.9%,6条引物分离得到PEG诱导甘蔗根尖特异表达片段45条,表明本研究所建立的甘蔗cDNA-SCoT反应体系具有操作简便、结果稳定、重复性好、成本低、多态性丰富等优点,可为甘蔗基因差异表达研究提供新的技术支持.  相似文献

This study presents four strategies of a novel evolutionary algorithm, multi-objective differential evolution algorithm (MDEA). The four strategies namely, MDEA1, MDEA2, MDEA3 and MDEA4 are adapted to solve the multi-objective crop planning model with multiple constraints in a farmland in the Vaalharts irrigation scheme (VIS) in South Africa. The three objectives of the model are to minimize the total irrigation water (m2) and to maximize both the total net income in South African Rand (ZAR) from farming and the total agricultural output in tons. The total area of the farm is 771,000 m2 and supplied with 704,694 m2 of irrigation water annually. Numerical results produce non-dominated solutions which converge to Pareto optimal fronts. MDEA1 and MDEA2 strategies with binomial crossover method are better for solving the crop planning problem presented than MDEA3 and MDEA4 strategies with exponential crossover method. MDEA1 found a solution with the highest total net income of ZAR 1,304,600 with the corresponding total agricultural output, total irrigation water and total planting areas of 316.26 tons, 702,000 m3 and 725,000 m2, respectively. The planting areas for the crops in the solution are 73,463 m2 for maize, 551,660 m2 for groundnut, 50,000 m2 for Lucerne and 50,000 m2 for Peacan nut. It can be concluded that MDEA is a good algorithm for solving crop planning problem especially in water deficient areas like South Africa.  相似文献

Salinity has a two‐phase effect on plant growth, an osmotic effect due to salts in the outside solution and ion toxicity in a second phase due to salt build‐up in transpiring leaves. To elucidate salt‐resistance mechanisms in the first phase of salt stress, we studied the biochemical reaction of salt‐resistant and salt‐sensitive wheat (Triticum aestivum L.) genotypes at protein level after 10 d exposure to 125 mM–NaCl salinity (first phase of salt stress) and the variation of salt resistance among the genotypes after 30 d exposure to 125 mM–NaCl salinity (second phase of salt stress) in solution culture experiments in a growth chamber. The three genotypes differed significantly in absolute and relative shoot and root dry weights after 30 d exposure to NaCl salinity. SARC‐1 produced the maximum and 7‐Cerros the minimum shoot dry weights under salinity relative to control. A highly significant negative correlation (r2 = –0.99) was observed between salt resistance (% shoot dry weight under salinity relative to control) and shoot Na+ concentration of the wheat genotypes studied. However, the salt‐resistant and salt‐sensitive genotypes showed a similar biochemical reaction at the level of proteins after 10 d exposure to 125 mM NaCl. In both genotypes, the expression of more than 50% proteins was changed, but the difference between the genotypes in various categories of protein change (up‐regulated, down‐regulated, disappeared, and new‐appeared) was only 1%–8%. It is concluded that the initial biochemical reaction to salinity at protein level in wheat is an unspecific response and not a specific adaptation to salinity.  相似文献

Summary Changes in the physicochemical properties of three kinds of litter (Prunus serotina leaves, Carpinus betulus leaves, and Pinus sylvestris needles) were analyzed by differential scanning calorimetry and differential thermogravimetry after decomposition for 12 to 27 months under field conditions. As expected, holocellulose was always decomposed to a larger extent than the corresponding lignin components, leading to an enrichment of lignin in the residue. These lignins were more or less modified depending on the plant species. Moreover, the results suggest that energy-rich crystalline cellulose accumulates during decomposition at the expense of easier degradable amorphous cellulose and hemicelluloses. The quotient Q, from the corresponding calorimetry and thermogravimetry values, was introduced to estimate the specific energy content as a measure for the decomposition of litter components.Dedicated to the late Prof. Dr. W. Wheat flour replacement from 0 to 40% by single tef flours from three Ethiopian varieties DZ-01-99 (brown grain tef), DZ-Cr-37 (white grain tef) and DZ-Cr-387 (Quncho, white grain tef) yielded a technologically viable ciabatta type composite bread with acceptable sensory properties and enhanced nutritional value, as compared to 100% refined wheat flour. Incorporation of tef flour from 30% to 40% imparted discreet negative effects in terms of decreased loaf volume and crumb resilience, and increase of crumb hardness in brown tef blended breads. Increment of crumb hardness on aging was in general much lower in tef blended breads compared to wheat bread counterparts, revealing slower firming kinetics, especially for brown tef blended breads. Blended breads with 40% white tef exhibited similar extent and variable rate of retrogradation kinetics along storage, while brown tef-blended breads retrograded slower but in higher extent than control wheat flour breads. Breads that contains 40% tef grain flour were found to contain five folds (DZ-01-99, DZ-Cr-387) to 10 folds (DZ-Cr-37) Fe, three folds Mn, twice Cu, Zn and Mg, and 1.5 times Ca, K, and P contents as compared to the contents found in 100% refined wheat grain flour breads. In addition, suitable dietary trends for lower rapidly digestible starch and starch digestion rate index were met from tef grain flour fortified breads.  相似文献
