河南产猫爪草的质量分析研究

为研究河南产猫爪草的质量,按照《中国药典》(2015年版)对河南产猫爪草进行来源鉴别、性状鉴别、横切面显微鉴别、薄层色谱鉴别,并增加粉末显微鉴别、粗多糖和多糖含量测定项目。结果表明:各新增项目简单、准确、可行、重现性好,可为河南产猫爪草质量评价及质量标准制定提供依据。

Study on Antiviral Effect of Polysaccharides from Lagenaria siceraria (Molina) Standl.on NDV in vitro

To research on polysaccharides from Lagenaria siceraria (Molina) Standl.against Newcastle disease virus (NDV).Total polysaccharide and four different solvent extractions of Lagenaria siceraria (Molina) Standl.were extracted by the methods of hot water extraction and ethanol precipition in this study.The content of polysaccharide was measured by phenol-sulfuric acid method and infrared spectroscopy method.The safe concentration and growth of chick embryo fibroblast (CEF) were assayed by MTT method,in order to facilitated the comparison under the same level,the safe concentration was united as 78.125 μg/mL.Under the safety range of concentration,detected the block-virus activity,anti-virus activity and virus-killing activity of polysaccharides through the ways of pre-adding polysaccharides,post-adding polysaccharides and adding polysaccharides with NDV.The results showed that the direct inactivation and propagation inhibition activity of total polysaccharide and four different solvent extractions were stronger than anti-absorption function.Anti-virus inhibition rate of 70%,80% gradient alcohol precipitation of polysaccharides from Lagenaria siceraria (Molina) Standl.(LSP₇₀,LSP₈₀) and total polysaccharide (LSP_t)were 40.41%,44.54% and 61.85%,virus-killing inhibition rate were 44.74%,58.76% and 59.38%.LSP₈₀ had the highest virus inhibition rate as 37.14% in the block-virus activity of those five polysaccharides.In summary,70%,80% gradient alcohol precipitation and total polysaccharide in polysaccharides possessed better activity and would be as the materials for further research.     

虫草参多糖提取工艺研究

研究探讨虫草参多糖的提取工艺,通过在单因素实验的基础上进行正交实验研究,实验结果表明:各因素对虫草参多糖提取率的影响从大到小依次为B>A>C,提取温度>料液比>提取时间.虫草参多糖提取工艺的最优组合条件为A2B2C3,即料液比1:30,提取温度60℃,提取时间4 h,在此条件下虫草参多糖提取率54.45%.     

蝉拟青霉多糖的纯化工艺

以多糖含量为指标,对除蛋白、脱色、透析纯化工艺参数进行优化,以期提高蝉拟青霉多糖的纯化工艺水平。结果显示:以木瓜蛋白酶提取液,Sevag法脱蛋白2次基本除蛋白完全;AB-8型大孔吸附树脂纯化效果最好,在p H为7、温度25℃、4mg/m L上样液浓度、2m L/min上样速度;透析时间为72h的纯化条件下,纯化效果最好。     

Establishment of Detection Methods for Dexamethasone Sodium Phosphate in Injections by UPLC-PDA

A detection method of dexamethasone sodium phosphate in houttuynia injection,bupleurum injection,vitamin C injection,lincomycin hydrochloride injection,and asragulus polysacharin injection was established by UPLC-PDA.The Waters ACQUITY UPLC^® BEH C18 column(2.1 mm×50 mm,1.7 μm) was used,the mobile phase consisted of 0.75% triethylamine phosphatic liquor/solution (pH 3.0)-methyl alcohol-acetonitrile(50:45:5),the flow rate was 0.3 mL/min,the column temperature had remained 25℃,photodiode array detector wavelength range was 190 to 400 nm,the recorded wavelength was 242 nm,the injection volume was 10 μL.The linear range of dexamethasone sodium phosphatel was 10.0 to 200.0 mg/L (R²=0.9999),the mean recovery and RSD for dexamethasone sodium phosphate in houttuynia injection,bupleurum injection,vitamin C injection,lincomycin hydrochloride injection and asragulus polysacharin injection were 97.54% and 1.14%,101.59% and 0.19%,100.92% and 0.92%,99.82% and 0.73%,100.08% and 0.62%,respectively.This method was accurate,easy and fast,it was suitable for detection of dexamethasone sodium phosphatein houttuynia injection and other four kinds of injections.     

仙草多糖提取工艺条件优化研究

采用水提法浸提仙草，以仙草多糖的提取率为指标，研究料液比、浸提温度、浸提时间及浸提次数4个因素对其提取率的影响，并从中挑取合适的因素水平进行正交试验及SPSS分析。结果表明：在这4个因素中，对提取率影响最大的因素是浸提温度，然后是浸提时间，再次是料液比，最后是浸提次数；在料液比1:60、温度100 ℃、浸提时间3 h的最优条件下，仙草多糖的提取率高达17.89 %，此时仙草多糖的含量为44.13 %。
