Pastoral systems are regarded as complex social-ecological systems with components that interact and change over a range of spatial and temporal scales. As such, herd mobility has traditionally been used to respond to the dynamic nature of these systems. However, mobile pastoral systems around the world are becoming more constrained and increasingly fragmented with important implications for herd mobility. This study assessed the spatial distribution of 256 herds and their mobility patterns over a decade in the 10 villages that comprise the spatially constrained Leliefontein pastoral area in South Africa. We developed a hierarchical model of rangeland use, which showed that several stratified and connected socioeconomic, climatic, and environmental factors determined the spatial and temporal use of grazing areas in this 192 000-ha semiarid environment. At the highest level of use, access to the Leliefontein pastoral area is formally regulated. At the next level, the place of residence of herd owners largely defined which village commons was used by their livestock. At the third level of rangeland use, the wealth status of owners determined where in relation to human settlements their herds were located. At the lowest level in the hierarchy, the locations of water and croplands delineated seasonal grazing areas and the movement of stockposts. These stratified factors, together with the overall variability in grazing resource availability and the different decision making processes involved, resulted in high flexibility and diversity of herd mobility patterns at the lowest level of rangeland use. This, in turn, ensured heterogeneity in resource use over a range of spatial and temporal scales. It was concluded that policies should embrace the complexity of the pastoral system and enable the adaptive management of herds. This could reduce the level of vulnerability experienced by pastoralists to climate variability and wider societal change.  相似文献

AIM: To investigate the effects of curcumin (Cur) on the expression of High mobility group box 1 protein (HMGB1), interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α) in amyloid-β (Aβ)-induced primary rat microglial cells. METHODS: Microglia were derived from the cerebral cortices of postnatal rat brains. The cells were identified by immunocytochemistry using mouse anti rat Iba-1 monoclonal antibody. A cell model using primary rat microglial cells incubated with Aβ25-35 as an inflammation model of Alzheimer's disease (AD) was set up. The morphological characters of primary rat microglial cells were observed. The concentration of Aβ25-35 and the treatment concentration of curcumin were selected by CCK-8 assay. Cultured primary rat microglial cells were divided into 5 groups:normal cell group, Aβ25-35 group, Cur group, Aβ25-35+Cur group and Aβ25-35+DMSO group. The expression of HMGB1, NF-κB, and receptor for advanced glycation end products (RAGE) was detected by Western blot. The levels of HMGB1, IL-1β, and TNF-α in the culture supernatant were measured by ELISA. RESULTS: The purity of primary microglias determined by Iba-1 immunofluorescence was more than 95%. The protein levels of HMGB1, RAGE and NF-κB were significantly increased after Aβ25-35 stimulation. After treatment with Cur, the protein levels of HMGB1, RAGE and NF-κB were significantly decreased (P<0.05). The levels of HMGB1, IL-1β and TNF-α in the supernatant were significantly increased after Aβ25-35 stimulation. Cur significantly decreased the level of HMGB1, IL-1β and TNF-α in the supernatant. CONCLUSION: Curcumin significantly inhibits neuroinflammation stimulated by Aβ25-35 in primary rat microglial cells.  相似文献

Summary In screening to isolate a full-length copy of a previously isolated cDNA clone, a further three cDNAs were also isolated from a library prepared from sub-apical swelling-stolon tissue of potato (Solanum tuberosum L.). Sequence analysis showed these clones to be similar to extensin-like protein genes, acyl carrier protein thioesterase genes and high mobility group protein genes, respectively. A further cDNA, isolated by subtractive hybridisation, was similar to a tomato cDNA previously isolated on the basis of its down-regulation following nematode infection. While all the newly isolated genes were expressed in swelling stolons, for most, maximal expression was seen to be in stem tissue. Possible roles for these genes in the development of potato plants are discussed, as is the significance of gene expression in stems and stolons to the process of tuberisation.  相似文献

Intrinsic activities of monodisperse ethoxylated dodecanols (MEDs), diethyl suberate (DESU) and tributyl phosphate (TBP) were investigated using Stephanotis floribunda leaf cuticular membranes (CMs) and [14C]2,4‐dichlorophenoxy butyric acid (2,4‐DB) as a model solute. When sorbed in cuticular membranes, MEDs, DESU and TBP increase solute mobility and are called accelerators for this reason. With MEDs, dose‐effect curves (log mobility vs accelerator concentration) were linear but, with DESU and TBP, curves convex to the x axes were obtained that approached a maximum at 90 and 150 g kg−1, respectively. Accelerators increased the mobility of 2,4‐DB in the CMs by 9‐ to 48‐fold, and effects were larger at lower temperatures (range 15–30 °C). Activation energy for diffusion of 2,4‐DB was 105 kJ mol−1, decreasing with increasing accelerator concentrations to 26 kJ mol−1 with DESU at 90 g kg−1 and 64 kJ mol−1 with TBP at 150 g kg−1. Thus, the intrinsic activity of DESU was much higher than that of TBP, which implies that, for a given effect, less DESU than TBP would be needed. MEDs were also very effective accelerators, lowering activation energies to 36 kJ mol−1. Data are discussed in relation to increasing rates of foliar penetration of active ingredients at low temperatures. © 2001 Society of Chemical Industry  相似文献

The use of persistent herbicides has increased the potential for contamination of soil, soil water and groundwater. The mobility, dissipation and fate of four herbicides, norflurazon, oxadiazon, oxyfluorfen and trifluralin, used in South Australian viticulture, have been studied in a typical sand‐over‐clay vineyard soil. Following herbicide application at field rates to plots up‐slope of miniature lysimeters, surface soil and soil water were sampled regularly over the period of annual rainfall. The concentration of each herbicide in the soil cores, surface soil and soil water was determined by GLC‐NPD following solid‐phase concentration procedures where necessary. Oxadiazon dissipated more quickly than the other three herbicides in the soil. Norflurazon was the most mobile of these herbicides in this soil. However all four herbicides were found in the soil water within the first year, though only norflurazon was found in the soil water in the subsequent year. Migration of different mineral particles within columns of soil‐sand mixtures containing 10 or 20 mass % of soil was investigated by establishing differences in the mineral suite between the ”︁bulk clay” and the ”︁mobile fine material” fractions. The ”︁bulk clay” fractions of all soils contained smectite, palygorskite, kaolinite, quartz, feldspar, and calcite. The soils were saturated with sodium by leaching with NaCl solution, and then leached with distilled water. Clay dispersion and particle migration occurred in the columns. Values of SAR (sodium adsorption ratio) of the effluent decreased with time due to carbonate dissolution. At a certain SAR value, the clays apparently formed aggregates, and as a consequence particle migration stopped in the column. In addition to clay‐sized particles (< 2 μm), very‐fine‐silt‐sized particles (2— 5 μm) were able to migrate in the soil‐sand mixtures, too, and to some extent fine‐silt‐sized particles (5—10 μm) as well. Average size of mobile particles decreases with increase of soil content in the soil‐sand mixtures. The mineralogical composition of the ”︁mobile fine material” changed during the experiment. At the beginning of the experiment, the ”︁mobile fine material” was enriched in the non‐phyllosilicates (especially in calcite, and in some cases in quartz, feldspar and dolomite) and contained low concentrations of phyllosilicates (smectite, palygorskite and kaolinite). At the end of the experiment, the proportion of non‐phyllosilicates decreased, and as a consequence, the proportion of phyllosilicates increased. Among the non‐phyllosilicates, calcite was the most mobile mineral. Among the phyllosilicates, palygorskite was preferentially mobilized in topsoil horizons. In subsoil horizons, on the other hand, kaolinite was preferentially mobilized. This difference was explained by the different nature of carbonates in the topsoil and subsoil horizons. Palygorskite is preferentially occluded within the soil carbonates of lacustrine origin over smectite and kaolinite. These carbonates are present mainly in the subsoil horizons. As a consequence, the presence of these carbonates in the subsoil horizons decreases the migration of mainly palygorskite.  相似文献

The persistence and dissipation behaviour of tebufenozide, an ecdysone agonist, were investigated: (1) under laboratory conditions in aquatic models set up in glass aquaria, and (2) under field conditions in in-situ aquatic enclosures deployed in a mixed-wood boreal forest lake. Two models were set up in the laboratory study (Study I), which was conducted at constant conditions of temperature, water pH and photoperiod. In Model I, partitioning of tebufenozide from sediment, treated at a concentration of 1400 μg kg-1, into untreated water was examined. The results showed that the chemical moved very little from the treated sediment into water. The concentration in sediment and water decreased gradually during the 90-day incubation period. Tebufenozide disappeared faster from the top layer of sediment than from the middle and bottom layers. The half-lives of disappearance were 64 days for the top layer but >90 days for the middle and bottom layers respectively. In Model II, partitioning from water, treated at a concentration of 350 μg litre-1, into untreated sediment was investigated. The results showed that the chemical moved from treated water into sediment due to adsorption. Little vertical downward movement of the adsorbed residues from the top layer of sediment occurred into layers beneath. The adsorbed residues were also not released readily back into water. The concentration in water and sediment decreased gradually during the 90-day incubation period. The half-life of dissipation from water was 67 days. The field microcosm study (Study II), conducted under fluctuating conditions of temperature, water pH and photoperiod, involved application of tebufenozide onto aquatic enclosures at four concentrations of 0·05, 0·10, 0·26 and 0·5 mg litre-1. This study also showed that the chemical moved downwards from the applied location and was adsorbed onto sediment. The chemical persisted longer in Study II than in Study I. Tebufenozide, being photo-labile, probably degraded faster after constant exposure to light in Study I than after exposure to fluctuating light in Study II. At 90 days after treatment in Study I, only about 55% of the applied material persisted in the sediment, and there was little accumulation. In Study II, the material not only persisted but also was accumulated in the sediment, since at 92 days post-treatment the residues were about 25 times higher than the applied concentration level. Residues in water also decreased more rapidly in Study I than in Study II, because the concentration at 90 days post-treatment was about 41% of the applied value. In Study II, however, about 65% of the applied chemical persisted in water at 92 days post-treatment. While the long persistence of tebufenozide in both the laboratory and field studies was attributable to its low vapour pressure, low water solubility, high octanol/water partition coefficient etc., the differences in the persistence characteristics observed in the two studies were due to the fluctuating environmental conditions and water pH encountered in the field study, compared with the constant environmental conditions and water pH utilized in the laboratory study. © 1997 SCI.  相似文献

用肌肉注射HCG的处理方式(剂量为500U/㎏•体重,每周注射1次,注射时间为6周)诱导雄性花鳗鲡性腺发育成熟,成熟率达80.0%。对人工催熟花鳗鲡精子的生物学特性研究结果表明:花鳗鲡精子头部长径为3.81±0.69µm,短径为1.24±0.15µm;尾部长度为24.83±3.05µm;精液pH为7.3~7.5,精子密度为1.02×1010尾/ mL。精子的适宜盐度为15~20,其中盐度为15时,精子激活比率最高,快速运动时间以及精子的寿命最长。精子的pH适宜范围为6.0~8.0,pH值过高或过低都会影响精子的活力与寿命。另外,4 种金属离子(Mg2+、Ca2+、Na+和K+)对花鳗鲡精子活力与寿命的影响趋势基本一致,金属离子浓度过高或过低都会抑制精子的活力、缩短快速运动时间和寿命。而MgCl2、CaCl2、KCl、NaCl溶液浓度为0.4~0.6g/mL时,精子活力最好,最高激活比率为3级(41.0%~60.0%)。  相似文献

The mobility of alachlor from alginate-encapsulated controlled-release (CR) formulations was investigated in two contrasting soil profiles. Two CR formulations of alachlor were prepared with the following components (1) base—sodium alginate+kaolin+‘Tween’ 20 (1+10+0·5 by mass) and (2) base+40 g kg−1 linseed oil. These were compared to technical grade alachlor and to a commercial alachlor EC formulation (‘Lasso’ 4EC). All herbicide treatments were labeled with [14C]alachlor and were applied to duplicate soil columns that were composed of a surface and a subsoil horizon. Each horizon was packed to a depth of 12·5 cm, giving a total column length of 25 cm. The columns were leached with 21 cm (420 ml) to 30 cm (600 ml) of 0·01M calcium chloride for a period of 7 to 10 days. Alachlor leaching from the EC formulations was the same as that from the technical material in both soils: 33% in the Evesboro and 10% in the Conover soil. The CR-Oil formulation leached 4 and 2% of the applied [14C]alachlor, compared to 12 and 3% for the CR-N formulation for the Evesboro and Conover soils, respectively. The CR-Oil formulation also increased the amount of [14C]alachlor retained in the soil surface horizon (105–114%), compared to CR-N (39–45%), technical material (14–23%) and EC (12–17%).  相似文献

The distribution of FCoV Types I and II in a Portuguese cat population was studied by a RT-PCR assay targeting the 3′-end of the viral RNA. For a period of 3 years, 120 samples were collected and 57 were found positive for FCoV RNA. Within the positive samples the presence of FCoV Type I was found in 79%. Type II was only detected in 3.5% in animals with Feline Infectious Peritonitis. The remaining 17.5% could not be differentiated. These viral sequences, comprising a region within gene S were further subjected to a heteroduplex mobility assay (HMA) detecting the presence of viral quasispecies in 17% of the samples. Phylogenetic analysis for FCoV Type I revealed high genetic diversity between the Portuguese sequences and other previously characterized strains, while Type II tree showed a higher genetic homogeneity. This study confirmed the presence of FCoV Types I and II circulating in Portugal and detected high genetic diversity between circulating strains suggesting that the virus persists within the host as mixed viral populations.  相似文献
