Abrupt dietary transitions and feeding of rapidly fermentable diets are common practices in the horse industry and have been associated with digestive and metabolic disorders that can impair the performance of horses. The present study investigated the effect of dietary transition from pasture grazing to confinement with concentrate feeding, and back, on fecal pH and bacterial populations of Streptococcus spp and Lactobacillus spp. Six Thoroughbred fillies, previously grazing perennial ryegrass and white clover-based pasture, were housed in individual stalls and fed an increasing ratio of concentrate to conserved forages for 13 days (days 1-13), followed by an abrupt transition back to only pasture-grazing for 3 days (days 14-16). The concentrate was initially offered at 0.83 kg dry matter (DM)/d and increased to 5 kg DM/d, whereas ensiled alfalfa was initially offered at 0.61 kg DM/d, increasing to 1.22 kg DM/d. Meadow hay was initially offered at 6.73 kg DM/d, decreasing to 1.6 kg DM/d. Fecal specimens were collected daily for determination of pH, and every 2 days for quantitative analysis of Streptococcus spp and Lactobacillus spp. Mean fecal pH increased significantly from pasture baseline values (pH 6.18) during the initial confinement and supplementation on day 1 (6.37), day 2 (6.52), day 3 (6.58), and day 4 (6.43) (standard error of mean [SEM]: 0.056; P < .001). By day 5, mean fecal pH values had decreased to, and remained at, baseline values until the horses returned to pasture, when another increase occurred at day 15 (6.45). Fecal colony forming units (cfu) of Streptococcus spp and Lactobacillus spp increased linearly (r = 0.94; P < .001) from 6.0 and 6.1 log10 cfu/g on day -4, to 7.8 log10 cfu/g on day 14 (SEM: 0.2 P < .001), respectively. Fecal cfu decreased on return to a pasture-only diet (P < .001). In this study, the increment of bacterial populations was associated with a relatively stable fecal pH and highlights the difficulty in identifying the effects of dietary transition on the equine hindgut health, without microbial culture.  相似文献

本研究无菌采集唐山市某奶牛场临床型乳房炎病牛的奶样30份,对奶样进行了病原菌的分离培养、生化鉴定及药敏试验。从30份奶样中共分离到8种156株细菌,其中金黄色葡萄球菌58株(37.2%)、大肠杆菌17株(10.9%)、表皮葡萄球菌10株(6.4%)、无乳链球菌38株(24.4%)、乳房链球菌15株(9.6%)、停乳链球菌16株(10.3%)、克雷伯氏菌1株(0.6%)、变形杆菌1株(0.6%)。结果表明,葡萄球菌、链球菌为主要病原菌,其次是大肠杆菌,占总菌数的98.7%(154/156);药敏试验结果表明,主要致病菌对中药组方1(M、G)(蒲公英、连翘、瓜蒌、通草、川芎、土贝母、芙蓉叶、金银花、王不留行、当归)、中药组方2(G)(黄芪、党参、当归、通草、川芎、白术、木通、甘草、王不留行、路路通)、中药复方黄连乳腺炎注射液(M)、新霉素呈高度敏感。  相似文献

试验旨在探讨含水量和添加植物乳杆菌对辣木叶青贮品质、单宁含量的影响。采用双因素(水分×植物乳杆菌)完全随机设计,含水量分别为60%、70%和75%,植物乳杆菌设置添加组(1×10^6 cfu·g^-1鲜样)和对照组。分别于青贮的60和120 d,开袋取样测定发酵品质、蛋白组分和单宁含量。辣木叶干物质中粗蛋白(CP)含量为16.72%,可溶性碳水化合物(WSC)含量为10.72%。含水量对辣木叶青贮品质有显著影响,70%含水量组与60%水量组相比,氨态氮(NH3-N)、氨基酸类游离氮(FAA)、乙酸(AA)、丙酸(PA)和乳酸(LP)含量均显著降低(P<0.05);在70%含水量条件下,添加植物乳杆菌,有机酸(LA、AA、PA)含量显著升高(P<0.05),乳酸菌(LAB)和酵母菌(Yeast)数目显著降低(P<0.05);发酵60 d时,在60%和75%含水量条件下,添加植物乳杆菌,pH值显著降低(P<0.05);所有处理的大肠杆菌数量均小于2.0 log cfu·g^-1FM。不同含水量和植物乳杆菌对辣木叶青贮后单宁含量均有降低作用,但无显著差异(P>0.05)。辣木叶蛋白丰富,易于调制青贮饲料,70%含水量和添加植物乳杆菌对辣木叶青贮品质有明显的改善作用,对降解单宁含量并未产生显著影响。  相似文献

Frozen samples of Finnish ready-mixed mink feed were analyzed for total bacterial count, the number of faecal streptococci, the coliform count, the number of haemolytic bacteria and the number of sulphite-reducing bacteria. The investigation comprised 242 feed samples from 38 central kitchens and larger private farm kitchens, the combined feed production of which is about 85 % of the yearly feed production of Finland.Of all samples 48.3 % had a total bacterial count of 106 … 6} × 106 bacteria per g of feed. The total bacterial count was relatively constant during the first four production periods of the year (December-August) and was elevated during the last period (September-November). The percentage of samples containing less than 2.5 × 104 faecal streptococci per g of feed was 49.8 %; 62 % of the samples contained less than 2.5 × 104 coliform bacteria per g. The content of coliform bacteria was lowest during the third production period (May); 48.5 % of the samples contained 5 × 103 … 105 haemolytic bacteria per g, and 4.6 % were negative. The content of haemolytic bacteria was relatively constant during the whole production year; 52.6 % of the feed samples contained 5 × 103 … 105 sulphite-reducing bacteria per g, and 17.2 % were negative. The mean content of sulphite-reducing bacteria was lowest during the second production period (March-April).The results are discussed and compared with corresponding results from Norway and Denmark.  相似文献

Objective To evaluate seasonal effects on the presence or absence of fungal and aerobic bacterial flora of the conjunctival fornix of normal Florida Thoroughbred horses. Sample population Both eyes of 100 horses. Procedure Horses with normal anterior segment ophthalmic examinations from three farms in north central Florida were included. Each animal had the ventral conjunctival fornix of each eye swabbed with sterile culturettes. Samples were taken in October, January, April, and July (1999–2000). Aerobic and fungal cultures were plated. Bacterial cultures were reviewed at 24 and 48 h. Fungal cultures were reviewed weekly for 4 weeks. Logistic regression analysis with season as a factor and age of the horse as a covariate was performed. Statistical significance was set at P < 0.01. Results Horses ranged from 3 to 24 years of age, with a median age of 9 years. Twenty‐four genera of bacteria and 35 genera of fungi were recovered. Corynebacterium sp., Staphylococcus sp., Bacillus sp. and Moraxella sp. were the bacteria most frequently isolated. Mold species, dematiaceous mold species, Chrysosporium sp., Cladosporium sp., and Aspergillus sp. were the most frequently recovered fungi. Season did not have a significant effect on the presence of microorganisms isolated for individual horses adjusted for age. Younger horses had an increased incidence of gram‐negative rods and fungal isolates. The number of bacteria and fungi isolated are not uniform across seasons. Conclusion There were no significant differences between the number or type of organisms cultured during the sampling seasons in normal Florida horses. A large range of normal bacterial and fungal flora were isolated from these horses. The number of bacteria and fungi isolated are not uniform across seasons. 耐冷菌及其分泌的耐热酶是原料乳中常见污染物,会导致乳制品腐败。通过16S rRNA基因测序分析了分离自上海地区原料乳的耐冷菌群,以及通过平板法研究不同耐冷菌的蛋白酶和脂肪酶活力。结果表明:所有的分离株属于9个菌纲,含33个菌属,其中数量占主要的菌属是假单胞菌(Pseudomonas)(55%)、不动杆菌(Acinetobacter)(10%)和黄杆菌(Flavobacterium)(5%),其余30个菌属占所有菌株数的30%。耐冷菌产酶能力平板法实验显示,大部分的耐冷菌都有产蛋白酶或脂肪酶的能力,不同菌属之间产酶特点存在差异。假单胞菌和黄杆菌主要表现为产蛋白酶活性,克雷伯氏菌(Klebsiella)主要表现为产脂肪酶活性,而大多数的不动杆菌没有产蛋白酶或脂肪酶活性。  相似文献

利用实验室保藏的多株菌种,对苹果渣进行三元混菌固态发酵产复合酶。以纤维素酶、木聚糖酶、果胶酶活性大小为测定指标,对筛选出单菌活性较高的菌株,按照1:1:1进行3种菌混合发酵,确定混菌后效果较好的3种菌。考虑到试验因素之间的交互作用,以3种菌的接种比例及接种时间间隔作为试验因素,通过正交表交互作用设计试验及分析,确定3种菌混合活性最佳时间间隔为菌种#1、#2、#3同时接种;接种#1体积1ml、接种#2体积2ml、接种#3体积1ml。混菌后酶活性比3种单菌的酶活分别提高了10.9%、2%、20%,达到了优化的目的。  相似文献

为了分析前期筛选出较耐低温不同乳酸菌(Lactic acid bacteria,LAB)对燕麦青贮发酵的影响,本试验以'陇燕5号’燕麦为原料,设置对照(CK)、商用菌株SLI和较耐低温菌株OL77,OL54及OL122共5个处理,在15℃,10℃和5℃低温下自然发酵45 d后进行营养成分、发酵品质和微生物数量的测定。结果表明:乳酸菌添加及温度对燕麦青贮的营养成分、发酵品质、微生物数量均有极显著影响。随着温度从15℃降至5℃,CK的粗蛋白(Crude protein,CP)降低了18.92%,氨态氮(Ammonia nitrogen,NH3-N)增加了58.47%;与SLI相比,耐低温菌株OL77在提高干物质(Dry matter,DM)和CP、降低NH3-N水平方面的效果优于OL54和OL122;添加剂在各温度处理下均提高了燕麦青贮发酵品质,15℃下添加OL77后燕麦青贮的pH值较CK和SLI分别降低了8.94%和1.57%,乳酸分别增加了79.23%和29.41%,乳酸菌数量分别增加了50.24%和15.61%。3个耐低温菌株中,OL77总体表现最优,促进燕麦低温发酵的效果最好。  相似文献

本试验旨在研究基于不同地区取样点土壤成分条件下,木质素降解菌的生长特性,并为筛选出降解农作物秸秆中木质素的优势菌奠定基础.通过对扬州、南京、临沂、潍坊和连云港地区5个取样点的土壤成分测定,并利用选择培养基和马铃薯葡萄糖琼脂(PDA)-RB亮蓝与PDA-Bavendamm平板显色反应鉴定不同土壤中木质素降解菌生长量及其木质素降解酶特性,依据各取样点菌株的生长特性最终选择优势菌株.结果表明,扬州取样点土壤中木质素降解菌密度[(5.67±0.58) 105g-1]极显著高于另外4个取样点(P<0.01),所获得的木质素降解菌的产酶特性亦优于其他取样点.可知,5个取样点土壤中木质素降解菌密度存在明显差异,并与土壤中总有机碳含量呈强正相关(r=0.84,P<0.01).  相似文献

为了建立一种快速区分样品中沙门氏菌的死细菌与活细菌的检测方法,将叠氮溴化乙锭(EMA)与PCR技术相结合。以沙门氏茵的invA为靶基因,以沙门氏菌的纯培养细胞做模板进行扩增,并进行灵敏度、特异性、曝光时间及EMA浓度试验。结果显示,灵敏度为14CFU/mL,最佳曝光时间为2min,当EMA的浓度小于18μg/mL时.EMA对活菌靶基因的扩增没有明显的抑制,而终浓度为1μg/mL的EMA,能有效抑制lxlosCFU/mL沙门氏菌死菌的扩增。EMA—PCR能有效降低沙门氏菌检测过程中的假阳性。  相似文献
