A procedure was developed for marker-assisted selection of complex traits for common bean (Phaseolus vulgarisL.) using an index based on QTL-linked markers and ultrametric genetic distances between lines and a target parent. A comparison of the mean seed yields of the top five lines selected by different schemes demonstrated that the highest yielding group was selected on the basis of a combination of phenotypic performance and a high QTL-based index,followed by groups identified by a high QTL-based-index, conventional selection,and a low QTL-based-index. This study demonstrated a simple way to use information obtained from QTL studies to make selection decisions. The study also showed that the use of the QTL-based-index in conjunction with the ultrametric genetic distance to the target parent would enablea plant breeder to select lines that retain important QTL in a desirable genetic background. Therefore, this type of MAS would be expected to be superior to the phenotypic selection. This revised version was published online in August 2006 with corrections to the Cover Date.  相似文献

Genetic progress through selection is directly related to the amount of variability present in the population and the quality of genes contributed by the parents. Genetic relationships between lines were studied using DNA marker-based estimates of genetic similarity. A statistical methodology using the width of a confidence interval was developed to determine the number of probes to be surveyed and the precision in the estimation of genetic distance between pairs of cultivars. Precision was affected by type of genetic distance used, the number of cultivars, and amount of genetic diversity present in the studied group. The width of a (1-α)% confidence interval decreased as the number of RFLP fragments increased. Oat and wheat diversity studies were used to illustrate the methodology. The sexual propagation of genebank accessions by hand pollination in the greenhouse is a very expensive and time-consuming routine task. The self-incompatibility of most diploid potato species allows their multiplication in isolated field plots, which is coupled with the advantage of the presence of natural pollinators and maintaining a high level of heterozygosity. The aim of this study was to determine the isolation distance required for minimizing pollen flow between neighbouring multiplication plots. The Solanum phureja clone IVP48 was used as the pollen donor since it possesses a dominant seed marker (‘embryo spot’). The centrally located pollen donor was surrounded by S. phureja recipient plots at distances of 10,20,40, and 80 m, respectively. The average pollen contamination ranged from 0.2% in the plots 80 m from the pollen donor to 5.1% in those 10 m away. In this study, procedures are suggested which will help to reduce cross-pollination between accessions.  相似文献

Significant relative midparent heterosis (MPH%) for grain yield in triticale (×Triticosecale Wittm.) has generated interest in the development of hybrid cultivars. The objectives of this study were to (i) examine the association between parental genetic distance (GD) and specific combining ability (SCA), (ii) investigate the existence of genetically distant heterotic groups in elite germplasm, and (iii) draw conclusions for future hybrid breeding in winter triticale. Genetic distance between 61 lines was estimated, based on 93 polymorphic simple sequence repeat (SSR) marker loci and 10 AFLP (amplified fragment length polymorphism) primer‐enzyme combinations (PEC). Agronomic data of 206 F1 crosses and their 61 parental lines grown in six German environments were published recently in a companion study. Correlations were made between SCA for grain yield, number of spikes/m2, 1000‐kernel weight and number of kernels per spike with GD estimates of the 56 female and five male parents (testers). Principal co‐ordinate analyses (PCoA) based on SSR data revealed no distinct subgroups in the germplasm. Correlations between GD and SCA were low for all traits (|r| ≤ 0.31), which hampers the prediction of SCA from molecular data. A multi‐stage procedure is recommended for future hybrid breeding in triticale by applying a pragmatic approach for the grouping of germplasm following the early history of hybrid breeding of maize.  相似文献

用一年生墨西哥大刍草(Zea Mexican Schard)与玉米G综合种远缘杂交选育的10个自交系作母本,以7个骨干自交系为父本,按10×7不完全双列杂交设计组配70个杂交组合为供试材料,研究了远缘杂交选系主要农艺性状和经济性状的遗传组成、遗传力和配合力效应。结果表明,抽丝期、株高、茎粗、穗长和百粒重主要受加性基因效应影响,狭义遗传力高,适合早代选择;茎强度、穗粗、穗行数和行粒数受加性基因和非加性基因效应同等影响;而穗位高、穗重、穗粒重和出籽率主要受非加性基因效应影响,狭义遗传力低,宜晚代选择。一般配合力效应分析结果,GT107可以作为很好的早熟、矮杆、抗倒和高产育种潜力亲本。  相似文献

Summary Three groups of characters selected using three methods were used to calculate genetic distance among two groups of winter wheat lines using Euclidean distance based on principal components. These wheat lines were then clustered by the unweighted pair-group arithmetic average method. The relationship between genetic distance and standardized potence was seemingly influenced by genetic materials. In order to obtain promising crosses, it is advisable to select parents from intercluster rather than intracluster materials. The results suggested that the repeatability method was better than the conventional and weighting methods for selecting characters for distance analysis.  相似文献

Crosses were made between four spring barley (Hordeum vulgare L.) cultivars and five F1, hybrids with one genotype of Hordeum bulbosum L. in two locations to investigate further previous low crossabilities which had been found in the barley cultivar ‘Apex’ with H. bulbosum. Data at all the main steps of the H. bulbosum technique were recorded and statistically analyzed. Significant differences between barley genotypes were demonstrated for all characters. It was confirmed that ‘Apex’ has poor crossability with H. bulbosum. Out of the three F1 hybrids having ‘Apex’ as one parent, two exhibited low crossability similarly to ‘Apex’ but one showed significantly higher seed setting than ‘Apex’. The effect of the location was only significant on seed setting, while genotype X location interactions were significant on seed setting, seed quality and rate of haploid plants in relation 10 florets pollinated. Another problem which has influenced the success rate of the H. bulbosum method was found in the cultivar ‘Havilla’. Although seed setting and seed quality were high for this cultivar, embryo differentiation was low. However, this latter problem was found to influence less the overall success rate than poor crossability. Mahalanobis's distances were calculated and the dendrite of the shortest distances between barley genotypes was plotted.  相似文献

Root-knot nematode is a significant root-parasite of common wheat. A dominant gene Rkn-mnl for resistance was transferred into wheat from Acgilops variabilis. This gene was shown to be on chromosome 3B. Further analysis indicated that it is on the long arm of this chromosome and independent of the centromere.  相似文献
