Sporamin is a soluble protein in sweet potato, and falls into two distinct homology groups, subfamilies A and B. In this research, a sporamin B was purified and its amino acid sequences, trypsin inhibitor activity (Ti activity) were analyzed. This sporamin B was isolated from sweet potato tubers [Ipomoea batatas (L.) Lam cv. 55-2] through extraction of the water-soluble fraction, dialysis, ultrafiltration and ion-exchange chromatography. Homology determined by polyacrylamide gel electrophoresis showed that mainly one bond appeared in gel after being reduced by SDS (sodium dodecyl sulfate), or by SDS and 2-mercaptoethanol, or in native situation. By comparing the data of the polypeptide mass Matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry with those of the mass of the theoretical amino acid sequences from NCBI protein database, it was revealed that it was Q40091|Q40091_IPOBA, sweet potato sporamin B - Ipomoea Batatas (sweet potato) (Batate). The sequence coverage was 70.6%. N-terminal sequence was SETPV (Ser-Glu-Thr-Pro-Val). There is a linear relationship between trypsin inhibitor activity (Ti activity) and amounts of this sporamin B (3-18 μg mL-1). The equation of linear regression was y = 2.5809x + 17.049 (r2 = 0.9966). There was a curvilinear relationship between Ti activity and amounts of this sporamin B (21-150 μg mL-1). The equation of curvilinear regression is y = 14.417ln(x) + 23.26 (r2 = 0.9924). The concentration of sporamin B with Ti activity after heating at 40°C may induce part denature of this sporamin B, and there was no statistic difference after heating at 40, 50, 60°C for 20 min. Heat treatment at more than 90°C leads to a dramatic decrease of trypsin inhibitor efficiency. The results suggested that Q40091|Q40091_IPOBA was the major sporamin B in sweet potato tubers [Ipomoea Batatas (L.) Lam cv. 55-2], which had strong Ti activity, and was stable to both thermal and DTT (DL-dithiothreitol) relatively.  相似文献

Studies of biological responses in the terrestrial environment to rapid changes in climate have mostly been concerned with above-ground biota, whereas less is known of belowground organisms. The present study focuses on enchytraeids (Oligochaeta) of heathland ecosystems and how the enchytraeid community has responded to simulated climate change in a long-term field experiment. Either increased temperature or repeated drought was applied for 13 years to field plots located in Wales, The Netherlands and Denmark representing a gradient in precipitation and annual temperature fluctuations thereby providing an opportunity to study biological responses on a local (within sites) and regional scale. Warming treatments increasing night-time temperature (0.5–1 °C higher than ambient at 5 cm soil depth) had no detectable effects on the enchytraeid communities. Increased intensity and frequency of drought had rather weak persistent effects on total enchytraeid abundance suggesting that ecosystem functions of enchytraeids may only be transiently impacted by repeated spring or summer drought. However, drought treatment had persistent negative effects on species richness and community structure across sites. Drought treated plots harboured only 35–65% of the species present in control plots, and the reduction of species richness was most pronounced at the driest sites. It is discussed that soil invertebrates, due to their weak migratory potential, may be more liable to extinction under changing climatic conditions than above-ground species, and therefore consequences of climate change to soil organisms need particular attention in future research.  相似文献

To estimate the possible effects of site factors and climatechange on forest growth in the Taihang Mountains, northern China,we assessed the factors influencing forest growth by using forestinventory data from 712 forest sample plots. Meteorologicaldata from 77 meteorological stations in the region were usedto estimate temperature and precipitation at each site fromelevation and longitude. Analyses showed that temperature, aspect,precipitation and soil thickness all significantly influencedforest growing stock (FGS), i.e. stem volume. When temperaturerose, FGS was reduced, possibly because increasing temperatureincreased evapotranspiration. Precipitation had a positive effecton FGS. The effect of aspect on FGS was perfectly expressedas a cosine function, with south-west- and south-facing slopeshaving the lowest FGS and north-facing slopes having the highest.We developed multifactorial regression models to predict changesin FGS in the Taihang Mountains. Temperature, forest age, forestcover, soil thickness, precipitation and aspect were well relatedto FGS. The effects of a temperature decrease and a precipitationincrease on FGS would be 2.5–8 per cent per degree centigradeand 10 per cent per 100 mm, respectively. The combination oftemperature increase and precipitation changes under futureclimate change is likely to result in a decrease of FGS, thoughthis does not take account the effect of increasing CO2. Wealso used multifactorial regression models to analyse the effectsof site factors on FGS of Pinus tabulaeformis Carr. and Robiniapseudoacacia L., two major species used in afforestation inthe Taihang Mountains. Although site factors had similar effectson FGS, diameter at breast height and tree height of both species,prediction accuracy (regression coefficient) was improved greatlywhen we treated the species separately.  相似文献

The establishment of woody fallow systems is an option for the improvement of degraded agricultural landscape within the ecologically deteriorated Aral Sea area in northwest Uzbekistan. Growth and development of ten tree species, differing in tolerance to drought and salinity, were studied over 24 months. To determine species suitability for afforestation, conventional diameter and height measurements were compared to Relative Growth Rate (RGR) and its underlying components, Net Assimilation Rate (NAR), Specific Leaf Area (SLA) and Leaf Weight Ratio (LWR), as well as the Crop Growth Rate (CGR) as a function of NAR and Leaf Area Index (LAI). RGRs varied between 0.46 and 2.16 mg g−1 day−1 and showed highly significant differences among species, but also between years, which reduces the parameter's suitability for species selection. The same is true for NAR and SLA. CGR values ranged from 0.01 to 0.71 g m−2 day−1, increased with age of the trees, and showed significant species differences. CGR correlated better with NAR (r = 0.89) and SLA (r = 0.86) than RGR. Overall CGR correlated highly with the RGRs in height and diameter (in 80% and 71% of all cases, respectively). Thus, CGR appears to be a suitable indicator complementing RGR, NAR and SLA estimates to decide on species suitability for afforestation. The intensive measurements for RGR should be done later, rather than earlier in the tree life cycle. At an early age, CGR and diameter measurements are more meaningful. A combination of estimates, not a single factor, best assesses the long-term sustainable growth under natural conditions and allows early selection of species suitable for afforestation of degraded patches in the agricultural landscape. Ranking of all parameters concurrently showed the high potential of Elaeagnus angustifolia and Populus euphratica, which matched previous rankings based on total biomass production and financial added value.  相似文献

Multi-species tree-shrub-grass riparian buffer systems have been recognized as one of the most cost-effective bioremediation approaches to alleviate nonpoint source agricultural pollution in heavily fertilized systems. However, highly concentrated herbicides in surface and subsurface water and shade cast by trees along the stream bank usually compromise the effectiveness of these systems. Greenhouse trials and field lysimeter studies were conducted to evaluate the tolerance of orchard grass (Dactylis glomerata), smooth bromegrass (Bromus inermis), tall fescue (Festuca arundinacea), timothy (Phleum pratense), and switchgrass (Panicum virgatum) ground covers to atrazine and Balance™ (isoxaflutole) plus their capacity to sequester and degrade these herbicides and their metabolites. Their ability to remove soil nitrate was also quantified. Concentrations of atrazine, Balance™ and their metabolites in the leachate, soil and plant samples were determined by solid phase extraction followed by high performance liquid or gas chromatographic analyses. Distribution of the herbicides and metabolites in the system was calculated using a mass balance approach. Herbicide bioremediation capacity of each lysimeter treatment was determined by the ratio of metabolites to parent herbicide plus metabolites. Bioremediation of nitrate was quantified by comparing nitrate reduction rates in grass treatments to the bare ground control. Based on this herbicide tolerance, bioremediation data and shade tolerance determined in a previous study, it was established that switch grass, tall fescue and smooth bromegrass are good candidates for incorporation into tree-shrub-grass riparian buffer systems designed for the bioremediation of atrazine, Balance™ and nitrate.  相似文献

Pre-marked skid trails, directional felling and climber cutting when logging in tropical rainforests may be important ways of reducing damage to the forest, thus creating a healthier stand and improving future yields.This study, carried out in a virgin dipterocarp rainforest in the south of Sabah, Malaysia, compared two types of logging (both with and without pre-cutting climbers): conventional selective logging (CL) and supervised logging (SL). The latter is a selective logging system in which both pre-marked skid trails and directional felling were implemented. The pre-marked skid trails were aligned parallel to each other, spaced 62 m apart. A randomised complete block 2 × 2 factorial design was used in the experiment, consisting of 16 gross treatment plots, each of 5.76 ha with a 1 ha net plot in the centre.Fewer trees tended (0.050 < P  0.100) to be logged in SL plots than in CL plots (on average 9.4 and 13.0 trees ≥60 cm diameter breast height ha−1). Pre-felling of climbers resulted in four more dipterocarp trees being logged ha−1, compared with no climber cutting: a statistically significant difference (P  0.050). The basal areas lost of both large trees (≥ 60 cm dbh) and small dipterocarp trees (10–29 cm dbh) tended to differ between the logging systems, with CL leading to greater losses.There were significant differences in the residual stands left by the logging systems, with respect to the number of dipterocarps and their basal area in the diameter class 10–29 cm; ca 30% more stems being found after SL. No significant differences (or tendencies) in these variables were found in the residual stands in other diameter classes, or when trees of all species were considered.  相似文献

The goal of this study was to assess the anaesthetic induction and recovery time in kutum (Rutilus frisii kutum) after exposure to various concentrations (0.1, 0.3, 0.5, 0.7 and 0.9 ml/l) of 2‐PE as an anaesthetic, as well as the effects of optimal concentration (0.7 ml/l) of 2‐PE in relation to different exposure time (3, 10, 15 min) on some haematological and serum biochemical indices in this species. Moreover, the effects of 0.7 ml/l on blood parameters were assessed 24 h after the longest exposure. Significant increase was determined in Hb, MCH and MCHC after 10‐min exposure to 2‐PE (p < 0.05). Moreover, Hct, Hb and RBC levels increased significantly after 15 min‐exposure to 2‐PE (p < 0.05). There were no prominent changes in WBC and MCV. The plasma concentrations of glucose, cholesterol and cortisol increased significantly after 10‐ and 15‐min exposure to 2‐PE (p < 0.05) compared with the control group and all other exposure times. The activity of ALT and AST were significantly increased after 10‐ and 15‐min exposure respectively (p < 0.05). In this study, it appears that anaesthetizing kutum with 2‐PE at 0.7 ml/l for 3 min had no effect on the stress markers.  相似文献
