Ceftiofur is a new broad spectrum cephalosporin marketed for the treatment of acute bovine respiratory disease. In this investigation ceftiofur was administered by intramuscular injection, at 24 h intervals, to healthy beef-bred calves for four days at dosages of 2.2 and 4.4 mg/kg of body weight, with 4 wk intervals between dosing regimens. Serum, tissue chamber fluid (TCF), and bronchial secretion (BS) concentrations of ceftiofur were measured by microbiological assay after the first and fourth dose of each dosing regimen. Peak serum concentrations (Cmax) of 8.8 micrograms/mL and 17.3 micrograms/mL were obtained approximately 2 h (Tmax), the time of mean peak concentration) after single injections of 2.2 mg/kg and 4.4 mg/kg, respectively. The Cmax was increased approximately twofold following multiple doses of 2.2 mg/kg (Cmax = 13.1 micrograms/mL) and 4.4 mg/kg (Cmax = 24.1 micrograms/mL). Ceftiofur accumulated slowly into TCF and peak concentrations were found to be approximately 14% of those observed in serum after the first dose and approximately 24% after multiple dosing. Concentrations of ceftiofur in BS were obtained rapidly with peak concentrations reaching 45% of the serum Cmax after the first dose. After multiple dosing the Cmax for BS was approximately 25% of the serum Cmax. This study found that both the 2.2 mg/kg and 4.4 mg/kg dosing regimens resulted in continuous serum, TCF and BS concentrations of ceftiofur that exceeded the minimal concentration required to inhibit the bacteria most frequently isolated from calves with acute bovine respiratory disease.  相似文献

Barley (Hordeum vulgare) and wheat (Triticum aestivum) are similar crops that can be grown in the arid regions of southwestern Asia. Generally, barley is favored over wheat in drier areas. However, the quantitative basis for the relative advantage of each crop under differing growing conditions has not been resolved. This study was undertaken to compare the two crops using the same model structure. A wheat model developed for southwestern Asia was readily adapted to simulate barley. Simulations of barley crops proved to be robust over a range of environments. Comparison of simulated yields in a number of environments showed that the critical difference between species was the cumulative effect of plant development traits that allowed barley to mature much earlier than wheat. Consequently, early maturity of barley in contrast to wheat often allowed barley to complete its growing cycle before water-deficit conditions developed in the spring. There was, however, little or no yield advantage for barley in seasons of high rainfall. Also, rare seasons with significant rainfall after barley matured allowed wheat to actually produce the greater yield. These simulation results indicated that development traits are crucial in the interaction between crop yield and exposure to water deficits at the end of the growing season.  相似文献

The impact of extreme events (such as prolonged droughts, heat waves, cold shocks and frost) is poorly represented by most of the existing yield forecasting systems. Two new model-based approaches that account for the impact of extreme weather events on crop production are presented as a way to improve yield forecasts, both based on the Crop Growth Monitoring System (CGMS) of the European Commission. A first approach includes simple relations – consistent with the degree of complexity of the most generic crop simulators – to explicitly model the impact of these events on leaf development and yield formation. A second approach is a hybrid system which adds selected agro-climatic indicators (accounting for drought and cold/heat stress) to the previous one. The new proposed methods, together with the CGMS-standard approach and a system exclusively based on selected agro-climatic indicators, were evaluated in a comparative fashion for their forecasting reliability. The four systems were assessed for the main micro- and macro-thermal cereal crops grown in highly productive European countries. The workflow included the statistical post-processing of model outputs aggregated at national level with historical series (1995–2013) of official yields, followed by a cross-validation for forecasting events triggered at flowering, maturity and at an intermediate stage. With the system based on agro-climatic indicators, satisfactory performances were limited to microthermal crops grown in Mediterranean environments (i.e. crop production systems mainly driven by rainfall distribution). Compared to CGMS-standard system, the newly proposed approaches increased the forecasting reliability in 94% of the combinations crop × country × forecasting moment. In particular, the explicit simulation of the impact of extreme events explained a large part of the inter-annual variability (up to +44% for spring barley in Poland), while the addition of agro-climatic indicators to the workflow mostly added accuracy to an already satisfactory forecasting system.  相似文献

Since 1985, the U.S. Department of Agriculture, Animal and Plant Health Inspection Service has maintained the 'Port Information Network' (PIN) database for plant pests intercepted at the U.S. ports of entry. As of August 2001, PIN contained 6825 records of beetles (Coleoptera) in the family Scolytidae that had been intercepted during the years 1985–2000 from countries outside of North America. Of the 6825 scolytid interceptions, 2740 (40%) were identified to the species level, 2336 (34%) to only the genus level, and 1749 (26%) were identified to only the family level. Of the 49 identified scolytid genera, the 10 most common were Hypothenemus (821 interceptions), Pityogenes (662), Ips (544), Coccotrypes (520), Orthotomicus (461), Hylurgops (327), Hylurgus (266), Tomicus (194), Dryocoetes (166), and Hylastes (142). The 10 most common identified species were Pityogenes chalcographus (565 interceptions), Orthotomicus erosus (385), Hylurgops palliatus (295), Ips typographus (286), Hylurgus ligniperda (217), Ips sexdentatus (157), Tomicus piniperda (155), Hylastes ater (75), Hypothenemus hampei (62), and Polygraphus poligraphus (48). Of these 10 species, H. palliatus, H. ligniperda, and T. piniperda are known to be established in the continental U.S. The scolytids were intercepted from 117 different countries; the top 12 countries were Italy (1090 interceptions), Germany (756), Spain (457), Mexico (425), Jamaica (398), Belgium (352), France (261), China (255), Russia (247), India (224), U.K. (151), and Portugal (150). The scolytids were intercepted in 35 U.S. states and 97 port cities. In general, there was a positive relationship between the number of scolytid interceptions from individual countries and the value of the imports from those countries. Overall, 73% of the scolytids were found in solid wood packing materials, 22% in food or plants, and 5% in other or unspecified materials. The products most commonly associated with scolytid-infested wood packing materials were tiles, marble, machinery, steel, parts, ironware, granite, aluminum, slate, and iron. The food products and plants that were commonly infested with scolytids included nutmeg, palms, coffee beans, kola nuts, and macadamia nuts.  相似文献

Specific-pathogen-free chickens inoculated with isolate VA (variant A) or isolate IM of infectious bursal disease virus (IBDV) were examined for mitogenic response to T-cell mitogens, primary and secondary antibody response to sheep erythrocytes and Brucella abortus, and gross and histologic lesions in thymus and bursa. Both isolates induced comparable depression in the mitogenic and antibody response, and both caused extensive gross and histologic lesions in the bursa of Fabricius. However, bursal necrosis induced by the IM isolate was accompanied by an inflammatory response, whereas the inflammatory component was lacking in the lesion induced by the VA isolate. Furthermore, the IM isolate induced extensive lesions in the thymus, but the VA isolate did not.  相似文献

This study used a cross-over design to compare the efficacy of 3 corticosteroids for the relief of airway obstruction and inflammation in 9 heaves-affected horses. The severity of airway obstruction and inflammation was quantified by measurement of lung function and by bronchoalveolar lavage fluid (BALF) cytology, respectively. Airway obstruction was induced by stabling the horses and they remained stabled during the 10 day treatment period. Lung function was measured before treatment (baseline), at Days 3, 7, and 10 of treatment, and after 30 days at pasture. BALF cytology was investigated at baseline, Day 10, and at pasture. All 9 horses received the following 4 treatments in random order: no treatment, daily oral prednisone tablets (1 mg/kg), daily i.v. dexamethasone solution (0.1 mg/kg), and i.m. dexamethasone-21-isonicotinate (0.04 mg/kg) every 3 days. When horses received no treatment, lung function did not change significantly during stabling but improved at pasture. In all horses, daily i.v. administration of dexamethasone solution improved lung function within 3 days to levels as good as or better than those measured at pasture. Dexamethasone-21-isonicotinate was rapidly effective in 8 of 9 horses. The other horse did not respond to this drug. Prednisone tablets were without effect on Days 3 and 7 of treatment, but by Day 10, 5 of 9 horses showed some improvement in lung function. Dexamethasone i.v. solution decreased the percent neutrophils in BALF at Day 10. Other treatments had no effect on BALF cytology. These results demonstrate that dexamethasone rapidly relieved airway obstruction in heaves-affected horses. Oral prednisone had inconsistent effects but may be beneficial in some horses after more than a week of treatment.  相似文献

A prospective cohort study design was used to evaluate the impact of subclinical Mycobacterium paratuberculosis infection on days open in a sample of Michigan dairy herds with a history of cows positive for M. paratuberculosis diagnosed by fecal culture. Participating herds were tested and productivity and reproduction records were monitored for 18 months. All cows > or = 24 months old were tested for M. paratuberculosis infection using the ELISA and radiometric fecal-culture (RFC) techniques. Test-negative cows were re-tested at the conclusion of the monitoring period. Multivariable regression models were used. Using both tests in parallel, the overall sample apparent prevalence for M. paratuberculosis infection was 41.8% (223/533 animals tested). Adjusting for diagnostic sensitivity and specificity, this resulted in a calculated sample true prevalence of 59.9%. ELISA-positive cows (on average) had a 28-day increase in days open when compared to ELISA-negative cows (p=0.02). The diagnostic method used to define a case altered the apparent association between paratuberculosis test status and days open. Fecal culture was a less-effective diagnostic tool for use in herds with a high prevalence of infected animals. The increase in days open in the ELISA-positive cows was an indication that perhaps reduced estrus expression or an increased post-partum anestrous period occurred in the subclinically infected ELISA-positive animals. This might have been due to a negative energy balance associated with M. paratuberculosis infection.  相似文献

Elbow dysplasia is a heritable disease that is a common cause of lameness and progressive elbow osteoarthritis in young large breed dogs. The Orthopedic Foundation for Animals (OFA) screens elbow radiographs, and assigns grades 0–3 based on presence and severity of bony proliferation on the anconeal process. Grade 1 is assigned when less than 3 mm is present and considered positive for dysplasia. We investigated the incidence of elbow dysplasia and progression of osteoarthritis in elbows with grades 0 and 1 in 46 elbows screened at least 1 year previously, using CT as a gold standard and with the addition of CT absorptiometry. The incidence of dysplasia based on CT was 62% in grade 0, and 75% in grade 1 elbows, all of which had medial coronoid disease. Progressive osteoarthritis at recheck was consistent with elbow dysplasia. The sensitivity and specificity of the OFA grade for elbow dysplasia compared to CT findings was 75% and 38%, respectively. Increased bone mineral density of the medial coronoid process as characterized by osteoabsorptiometry warrants further investigation with respect to elbow dysplasia. Proliferation on the anconeal process without CT evidence of dysplasia or osteoarthritis was present in 20% of the elbows, and is theorized to be an anatomic variant or enthesopathy of the olecranon ligament/synovium. Results of our study suggest that the “anconeal bump” used for elbow screening by the OFA is a relatively insensitive characteristic, and support the use of CT for identifying additional characteristics of elbow dysplasia.  相似文献

Slow-feathering (SF) white leghorn dams harboring the endogenous viral gene ev21, which encodes for complete endogenous virus-21 (EV21), and rapid-feathering (RF) dams lacking EV21 were immunized with a live field strain of avian leukosis virus (ALV) subgroup A. One group of SF dams and one group of RF dams were not immunized and were maintained to produce chicks lacking maternal ALV antibody. When the SF dams were crossed with line 15B1 males, the resulting male progeny were SF, EV21-positive, and the females were RF, lacking EV21 or congenitally infected with EV21. EV21-positive and -negative progeny of immunized and unimmunized SF and RF dams were exposed to ALV at hatching. Viremia, antibody development, cloacal shedding, and tumors in chickens lacking EV21 were compared with those in chickens with EV21. Congenital transmission of EV21 from SF dams to RF female chicks was significantly higher in immunized dams than in unimmunized dams. Maternal ALV antibody delayed infection with ALV and reduced viremia and cloacal shedding of virus in progeny. The effect of maternal antibody on ALV infection was much more pronounced in progeny lacking EV21 than in progeny harboring EV21. The data suggest that the development of ALV infection and tumors may be influenced by status of infection with EV21 and by the immune status of dams.  相似文献
