微藻水热液化有机相最佳回收工艺研究

以微拟球藻为原料,研究微藻水热液化过程中多种参数对水热液化后有机物回收率的影响,包括温度、时间、溶剂比和pH等,从而获得微藻水热液化后有机物的最佳回收工艺条件。结果显示,在室温下进行有机相回收,保证回收溶剂比为20∶1(溶剂/水相不溶物;v/wt)、停留时间为10min、回收pH7,即可达到最佳回收效果。最佳条件下水热液化有机相的回收率为34%左右,且不同的回收温度和pH值对回收的有机相组分均会产生影响。     

Expression and mechanisms of hsa_circ_087631 in patients with primary biliary cholangitis

AIM To explore the expression and mechanisms of circular RNA hsa_circ_087631 in the patients with primary biliary cholangitis （PBC）. METHODS RT-qPCR was used to detect the expression of hsa_circ_087631 in PBC patients and healthy controls. Hsa-miR-346-overexpressing lentiviral vector pLenti-EF1a-EGFP-F2A-Puro-CMV-MCS was constructed and transfected into human acute T cell leukemia Jurkat cells， and then the expression levels of hsa_circ_087631， Bcl-6 mRNA and interleukin-21 （IL-21） mRNA were detected by RT-qPCR. Dual-luciferase reporter assay was performed to identify the interactions between hsa_circ_087631 and hsa-miR-346. RESULTS The expression of hsa_circ_087631 in the PBC patients was significantly increased compared with the healthy subjects. Hsa-miR-346-overexpressing lentiviral vector pLenti-EF1a-EGFP-F2A-Puro-CMV-MCS was successfully constructed. The expression of hsa-miR-346 was significantly increased after the hsa-miR-346-overexpressing lentiviral vector was transfected into the Jurkat cells， while the expression levels of hsa_circ_0087631， Bcl-6 mRNA and IL-21 mRNA were significantly decreased. After wild-type or mutant hsa_circ_087631 vector and hsa-miR-346 mimics were transfected into 293T cells， the results of dual-luciferase reporter assay showed that hsa-miR-346 significantly decreased the luciferase activity of wild-type hsa_circ_087631 （P<0.01）， but the regulation did not change significantly after mutation of the predicted binding site. CONCLUSION Peripheral blood hsa_circ_087631 level is elevated in the PBC patients. The hsa_circ_087631/hsa-miR-346/Bcl-6 signaling may take effect in human T cells. Hsa-miR-346 significantly reduces the expression of hsa_circ_087631， but it may not be regulated by predicted binding sites.     

基于超平面构建的高光谱异常目标检测方法

高光谱遥感影像具有光谱分辨率高、图谱合一的特点,影像中的地物组成也多种多样,针对图像数据中目标出现概率较小、检测概率较低等问题,本文提出了基于超平面构建的高光谱数据目标异常检测方法。它通过对高光谱数据的结构分析,构建出高维空间中的背景超平面算子,以待测像元在其正交子空间的投影值来表征异常程度的大小。通过OMIS和AVIRIS实测数据验证,并与RX算法相比较,基于超平面构建的高光谱异常检测算法能够在多种背景环境下检测出纯度高、光谱差异大的异常像元,并具有较高的检测率和较低的虚警率,因此具备了有效性和较好的实用性。     

Integration of pre- and postharvest treatments for the control of black spot caused by Alternaria alternata in stored persimmon fruit

In Israel, black spot caused by Alternaria alternata is the main postharvest factor that impairs the quality and reduces the storability of persimmon fruit (Diospyros kaki cv. Triumph). The fungus infects the fruit in the orchard and remains quiescent until harvest. After harvest, the pathogen slowly colonizes the fruit during storage at 0 °C, which elicits black spot symptom development 2–3 months after storage entry. A commercial postharvest dip treatment in chlorine at 500 mg L^?1, released from sodium troclosene tablets, effectively controlled black spot in fruit stored for up to 2 months. However, decay incidence increased as the length of storage was extended beyond 2.5 months. The long incubation period that precedes black spot symptom development after harvest enabled the development of a series of integrative approaches for application at the pre- and postharvest stages, in combination with the commercial chlorine dip treatment, to improve the control of black spot disease. Preharvest treatments included treatment with the cytokinin-like N₁-(2-chloro-4-pyridyl)-N₃-phenylurea (CPPU) 30 d after fruit set, or a single spray with the curative fungicide polyoxin B 14 d before harvest, and when one of these was applied in combination with the postharvest chlorine dip treatment, the black spot infected area was reduced by 3 and 60%, respectively, compared with the chlorine dip alone. At the postharvest stage, fogging during storage, or post-storage on-line spraying with sodium troclosene, when applied in combination with the postharvest chlorine dip, improved the percentage of marketable fruit by 2 or 10%, respectively, compared with the chlorine dip alone. The results indicate that postharvest pathogens that show a slow colonization pattern might enable the integration of pre- and postharvest disease control methods to improve quality and reduce postharvest disease development.     

提高玉米产量的“四防六技巧”

玉米在生长过程中如果管理不善，容易出现早衰、空秆、缺粒、倒伏等现象，这样就会严重影响到玉米品质和产量。如何防止这种现象发生，应着重做好“四防”。玉米生长的前期如何做到“促下控上”，中后期为授粉结实创造良好的生长环境，提高光合效率，延长根和叶的生理活性，达到粒重饱满高产的目的，应做好“六技巧”。     

冷却剂参数对铣槽喷管低周疲劳寿命的影响

采用有限体积流固耦合计算方法、非线性有限元热结构耦合分析方法和局部应变法研究大面积比铣槽喷管三维再生冷却槽道在循环工作条件下的热结构变形与低周疲劳寿命,并对比分析了冷却剂质量流量与入口温度对铣槽喷管疲劳使用寿命的影响。计算结果表明,铣槽喷管热结构响应呈现复杂的三维效应,应变较大位置主要分布在与肋连接的内衬区域,喷管中部的残余应变量最大；冷却槽道低周疲劳寿命分布和热结构响应基本一致,最小寿命位于喷管中部与肋相连的内衬区域燃气侧；随冷却剂质量流量增加,铣槽喷管低周疲劳寿命不断提高；随冷却剂入口温度增加喷管尾部低周疲劳寿命值不断降低,而喷管中前部的低周疲劳寿命值却不断提高,当冷却剂入口温度为280K左右时,本文的铣槽喷管总体使用寿命达到最大。      

Xanthone derivatives from the fermentation products of an endophytic fungus Phomopsis sp.

Three new xanthones, 1,5-dihydroxy-3-hydroxyethyl-6-methoxycarbonylxanthone (1), 1-hydroxy-5- methoxy-3-hydroxyethyl-6-methoxycarbonylxanthone (2), and 1-hydroxy-3-hydroxyethyl- 8-ethoxycarbonylxanthone (3), along with seven known xanthones (4–10) were isolated from the fermentation products of an endophytic fungus Phomopsis sp.. Their structures were elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1–10 were also tested for their cytotoxicity against five human tumor cell lines (NB4, A549, SHSY5Y, PC3, and MCF7) by MTT method using paclitaxel as positive control. Compounds 1 and 3 showed cytotoxicity against A549 cell lines with IC50 values of 3.6 and 2.5 μM, respectively. In addition, 1 was cytotoxic to MCF7 cells with IC50 value of 2.7 μM.     

Pestaloficiols Q–S from the plant endophytic fungus Pestalotiopsis fici

Two new isoprenylated chromone derivatives, pestaloficiols Q (1) and R (2), and one new benzofuran derivative, pestaloficiol S (3), along with three known metabolites, anofinic acid (4), siccayne (5), and pyrenophorol (6) were isolated from solid cultures of the plant endophytic fungus Pestalotiopsis fici. Their structures were elucidated primarily by NMR spectroscopy, and the absolute of the C-6 secondary alcohol in 1 was deduced on the basis of circular dichroism (CD) data. Compound 5 showed cytotoxic activity against the human cancer cell lines, HeLa and HT29, with IC₅₀ values of 48.2 and 33.9 μM, respectively.     

Alkyl and phenolic glycosides from Saussurea stella

One alkyl glycoside, saussurostelloside A (1), two phenolic glycosides, saussurostellosides B1 (2) and B2 (3), and 27 known compounds, including eleven flavonoids, seven phenolics, six lignans, one neolignan, one phenethyl glucoside and one fatty acid, were isolated from an ethanol extract of Saussurea stella (Asteraceae). Their structures were elucidated by NMR, MS, UV, and IR spectroscopic analysis. Of the known compounds, (+)-medioresinol-di-O-β-d-glucoside (7), picraquassioside C (10), and diosmetin-3′-O-β-d-glucoside (27) were isolated from the Asteraceae family for the first time, while (+)-pinoresinol-di-O-β-d-glucoside (6), di-O-methylcrenatin (11), protocatechuic acid (14), 1,5-di-O-caffeoylquinic acid (17), formononetin (28), and phenethyl glucoside (29) were isolated from the Saussurea genus for the first time. The anti-inflammatory activities of three new compounds (1–3), five lignans ((−)-arctiin (4), (+)-pinoresinol-4-O-β-d-glucoside (5), (+)-pinoresinol-di-O-β-d-glucoside (6), (+)-medioresinol-di-O-β-d-glucoside (7) and (+)-syringaresinol-4-O-β-d-glucoside (8)), one neolignan (picraquassioside C (10)), and one phenolic glycoside (di-O-methylcrenatin (11)) were evaluated by testing their inhibition of the release of β-glucuronidase from PAF-stimulated neutrophils. Only compound 5 showed moderate inhibition of the release of β-glucuronidase, with an inhibition ratio of 39.1%.
