The aim of this study was to determine the pharmacokinetics and prostaglandin E2 (PGE2) synthesis inhibiting effects of intravenous (IV) and transdermal (TD) flunixin meglumine in eight, adult, female, Huacaya alpacas. A dose of 2.2 mg/kg administered IV and 3.3 mg/kg administered TD using a cross‐over design. Plasma flunixin concentrations were measured by LC‐MS/MS. Prostaglandin E2 concentrations were determined using a commercially available ELISA. Pharmacokinetic (PK) analysis was performed using noncompartmental methods. Plasma PGE2 concentrations decreased after IV flunixin meglumine administration but there was minimal change after TD application. Mean t1/2λz after IV administration was 4.531 hr (range 3.355 to 5.571 hr) resulting from a mean Vz of 570.6 ml/kg (range, 387.3 to 1,142 ml/kg) and plasma clearance of 87.26 ml kg?1 hr?1 (range, 55.45–179.3 ml kg?1 hr?1). The mean Cmax, Tmax and t1/2λz for flunixin following TD administration were 106.4 ng/ml (range, 56.98 to 168.6 ng/ml), 13.57 hr (range, 6.000–34.00 hr) and 24.06 hr (18.63 to 39.5 hr), respectively. The mean bioavailability for TD flunixin was calculated as 25.05%. The mean 80% inhibitory concentration (IC80) of PGE2 by flunixin meglumine was 0.23 µg/ml (range, 0.01 to 1.38 µg/ml). Poor bioavailability and poor suppression of PGE2 identified in this study indicate that TD flunixin meglumine administered at 3.3 mg/kg is not recommended for use in alpacas.  相似文献

The pharmacokinetic–pharmacodynamic (PK/PD) modeling of enrofloxacin data using mutant prevention concentration (MPC) of enrofloxacin was conducted in febrile buffalo calves to optimize dosage regimen and to prevent the emergence of antimicrobial resistance. The serum peak concentration (Cmax), terminal half‐life (t1/2K10), apparent volume of distribution (Vd(area)/F), and mean residence time (MRT) of enrofloxacin were 1.40 ± 0.27 μg/mL, 7.96 ± 0.86 h, 7.74 ± 1.26 L/kg, and 11.57 ± 1.01 h, respectively, following drug administration at dosage 12 mg/kg by intramuscular route. The minimum inhibitory concentration (MIC), minimum bactericidal concentration, and MPC of enrofloxacin against Pasteurella multocida were 0.055, 0.060, and 1.45 μg/mL, respectively. Modeling of ex vivo growth inhibition data to the sigmoid Emax equation provided AUC24 h/MIC values to produce effects of bacteriostatic (33 h), bactericidal (39 h), and bacterial eradication (41 h). The estimated daily dosage of enrofloxacin in febrile buffalo calves was 3.5 and 8.4 mg/kg against P. multocida/pathogens having MIC90 ≤0.125 and 0.30 μg/mL, respectively, based on the determined AUC24 h / MIC values by modeling PK/PD data. The lipopolysaccharide‐induced fever had no direct effect on the antibacterial activity of the enrofloxacin and alterations in PK of the drug, and its metabolite will be beneficial for its use to treat infectious diseases caused by sensitive pathogens in buffalo species. In addition, in vitro MPC data in conjunction with in vivo PK data indicated that clinically it would be easier to eradicate less susceptible strains of P. multocida in diseased calves.  相似文献

The growth performance of twenty poplar(Populus deltoides Bartr.) clones,planted in the central-plain region of Punjab in north-western India,was evaluated using a randomized block design with three replications and plot size of four plants.Significant differences among clones were observed for diameter at breast height(DBH),height and volume per tree at age five,six and eight years.Clone L-48/89 recorded significantly superior volume than the control(G-48) at all ages,with superiority of 51.4,43.9 and 48.5 per cent at age five,six and eight years,respectively.The phenotypic and genotypic coefficients of variation were the highest for volume(26.55-34.66% and 15.84-26.00%) and the minimum for tree height(8.43-12.13% and 4.71-7.59%).The broad sense heritability was relatively higher for DBH(0.42-0.55) and genetic advance as per cent of mean was highest for volume(19.48-40.18).All genetic parameters increased with age.All traits showed significant positive correlation with each other.  相似文献

Pharmacokinetic and pharmacodynamic properties in goats of the non-steroidal anti-inflammatory drug tolfenamic acid (TA), administered both alone and in combination with the fluoroquinolone marbofloxacin (MB), were established in a tissue cage model of acute inflammation. Both drugs were injected intramuscularly at a dose rate of 2 mg kg−1. After administration of TA alone and TA + MB pharmacokinetic parameters of TA (mean values) were Cmax = 1.635 and 1.125 μg ml−1, AUC = 6.451 and 3.967 μg h ml−1, t1/2K10 = 2.618 and 2.291 h, Vdarea/F = 1.390 and 1.725 L kg−1, and ClB/F = 0.386 and 0.552 L kg−1 h−1, respectively. These differences were not statistically significant. Tolfenamic acid inhibited prostaglandin (PG)E2 synthesis in vivo in inflammatory exudate by 53-86% for up to 48 h after both TA treatments. Inhibition of synthesis of serum thromboxane (Tx)B2 ex vivo ranged from 16% to 66% up to 12 h after both TA and TA + MB, with no significant differences between the two treatments.From the pharmacokinetic and eicosanoid inhibition data for TA, pharmacodynamic parameters after dosing with TA alone for serum TxB2 and exudate PGE2 expressing efficacy (Emax = 69.4 and 89.7%), potency (IC50 = 0.717 and 0.073 μg ml−1), sensitivity (N = 3.413 and 1.180) and equilibration time (t1/2Ke0 = 0.702 and 16.52 h), respectively, were determined by PK-PD modeling using an effect compartment model. In this model TA was a preferential inhibitor of COX-2 (COX-1:COX-2 IC50 ratio = 12:1). Tolfenamic acid, both alone and co-administered with MB, did not affect leucocyte numbers in exudate, transudate or blood. Compared to placebo significant attenuation of skin temperature rise over inflamed tissue cages was obtained after administration of TA and TA + MB with no significant differences between the two treatments. Marbofloxacin alone did not significantly affect serum TxB2 and exudate PGE2 concentrations or rise in skin temperature over exudate tissue cages. These data provide a basis for the rational use of TA in combination with MB in goat medicine.  相似文献

A field experiment was conducted to study the N2 fixation efficiency of Sesbania rostrata and S. cannabina as affected by agronomic practices in semi-arid subtropical climate, Sowing seeds resulted in smaller numbers of nodules, lower dry weight, lower total biomass, less N uptake, and less N2 fixation for S. rostrata than S. cannabina, while cut-stem planting improved the symbiotic efficiency. Flooding the soil increased the relative humidity of the crop micro-environment by 4–11% and induced early appearance of stem nodules in S. rostrata. Only 67 kg N ha-1 was fixed by S. rostrata compared to 160 kg N ha-1 by S. cannabina when normal agronomic practices (sowing and non-flooding) were followed. In contrast, planting stem cuttings and flooding resulted in greater biological N2 fixation, 307 and 209 kg N ha-1 by S. rostrata and S. cannabina, respectively. Therefore, S. rostrata can be successfully exploited as a green manure when stem cuttings are planted under flooded conditions.  相似文献

Sustainability index was calculated to assess soil quality under the influence of different fertilizer management practices. It is based on the area of the triangle in which nutrient index, microbial index and crop index of soil represented the three vertices of a triangle. Nutrient index reflected the nutrient status of soil and was calculated from the measurements of various soil chemical parameters. Microbial index was calculated by determining various soil microbial and biochemical activities and crop index by measuring of crop yield parameters. Eighteen soil indicators were determined to assess nutrient index, microbial index and crop index in order to compare the effect of different sources of nutrients such as green manure, farmyard manure and chemical fertilizer in a rice/corn–wheat rotation. The indices were applied to assess the sustainability of five field experiments with respect to the different fertilizer treatments. The long-term application of organic manures in rice/corn–wheat cropping system increased the index value because it increased the nutrient index, microbial index and crop index of soils. The use of only chemical fertilizers in the rice–wheat cropping system resulted in poor soil microbial index and crop index. In corn–wheat system, additional application of FYM at 10 t ha–1 before sowing corn made the system more sustainable than application of 100%NPK; the sustainability index values were 2.43 (the highest for this system) and 0.93, respectively.  相似文献

In a four-period, cross-over study, the fluoroquinolone antibacterial drug marbofloxacin (MB) was administered to calves, alone and in combination with the nonsteroidal anti-inflammatory drug tolfenamic acid (TA). Both drugs were administered intramuscularly (IM) at doses of 2 mg/kg. A tissue cage model of inflammation, based on the actions of the mild irritant carrageenan, was used to evaluate the pharmacokinetics (PK) of MB and MB in combination with TA. MB mean values of area under concentration-time curve (AUC) were 15.1 μg·h/mL for serum, 12.1 μg·h/mL for inflamed tissue cage fluid (exudate) and 9.6 μg·h/mL for noninflamed tissue cage fluid (transudate). Values of C(max) were 1.84, 0.35 and 0.31 μg/mL, respectively, for serum, exudate and transudate. Mean residence time (MRT) of 23.6 h (exudate) and 22.6 h (transudate) also differed significantly from serum MRT (8.6 h). Co-administration of TA did not affect the PK profile of MB. The pharmacodynamics of MB was investigated using a bovine strain of Mannheimia haemolytica. Time-kill curves were established ex vivo on serum, exudate and transudate samples. Modelling the ex vivo serum time-kill data to the sigmoid E(max) equation provided AUC(24 h) /MIC values required for bacteriostatic (18.3 h) and bactericidal actions (92 h) of MB and for virtual eradication of the organism was 139 h. Corresponding values for MB + TA were 20.1, 69 and 106 h. These data were used to predict once daily dosage schedules for a bactericidal action, assuming a MIC(90) value of 0.24 μg/mL, a dose of 2.6 mg/kg for MB and 2.19 mg/kg for MB + TA were determined, which are similar to the currently recommended dose of 2.0 mg/kg.  相似文献

The pharmacokinetics (PK) and pharmacodynamics (PD) of marbofloxacin (MBF) were determined in six healthy female goats of age 1.00–1.25 years after repeated administration of MBF. The MBF was administered intramuscularly (IM) at 2 mg kg?1 day?1 for 5 days. Plasma concentrations of MBF were determined by high‐performance liquid chromatography, and PK parameters were obtained using noncompartmental analysis. The MBF concentrations peaked at 1 hr, and peak concentration (Cmax) was 1.760 µg/ml on day 1 and 1.817 µg/ml on day 5. Repeated dosing of MBF caused no significant change in PK parameters except area under curve (AUC) between day 1 (AUC0–∞D1 = 7.67 ± 0.719 µg × hr/ml) and day 5 (AUC0‐∞D5 = 8.70 ± 0.857 µg × hr/ml). A slight difference in mean residence time between 1st and 5th day of administration and accumulation index (AI = 1.13 ± 0.017) suggested lack of drug accumulation following repeated IM administration up to 5 days. Minimum inhibitory concentration (MIC) demonstrated that Escherichia coli (MIC = 0.04 µg/ml) and Pasturella multocida (MIC = 0.05 µg/ml) were highly sensitive to MBF. Time‐kill kinetics demonstrated rapid and concentration‐dependent activity of MBF against these pathogens. PK/PD integration of data for E. coli and P. multocida, using efficacy indices: Cmax/MIC and AUC0–24hr/MIC, suggested that IM administration of MBF at a dose of 2 mg kg?1 day?1 is appropriate to treat infections caused by E. coli. However, a dose of 5 mg kg?1 day?1 is recommended to treat pneumonia caused by P. multocida in goats. The study indicated that MBF can be used repeatedly at dosage of 2 mg/kg in goats without risk of drug accumulation up to 5 days.  相似文献

Mean λz‐HL after IV administration was 6.032 hr (range 4.735–9.244 hr) resulting from a mean Vz of 584.1 ml/kg (range, 357.1–1,092 ml/kg) and plasma clearance of 67.11 ml kg?1 hr?1 (range, 45.57–82.35 ml kg?1 hr?1). The mean Cmax, Tmax, and λz‐HL for flunixin following TD administration was 0.134 μg/ml (range, 0.050–0.188 μg/ml), 11.41 hr (range, 6.00–36.00 hr), and 43.12 hr (15.98–62.49 hr), respectively. The mean bioavailability for TD flunixin was calculated as 24.76%. The mean 80% inhibitory concentration (IC80) of PGE2 by flunixin meglumine was 0.28 μg/ml (range, 0.08–0.69 μg/ml) and was only achieved with IV formulation of flunixin in this study. The PK results support clinical studies to examine the efficacy of TD flunixin in goats. Determining the systemic effects of flunixin‐mediated PGE2 suppression in goats is also warranted.  相似文献

A greenhouse experiment with soybean grown on sulfur (S) and boron (B) deficient calcareous soil was conducted for two years in northwest India to study the influence of increasing sulfur and boron levels on yield and its attributing characters at different growth stages (55 days, maturity). The treatments included four levels each of soil applied sulfur viz. 0, 6.5, 13.4, 20.1 mg S kg?1 and boron viz. 0, 0.22, 0.44, 0.88 mg B kg?1 at the time of sowing. The highest dry matter yield at 55 days after sowing, DAS (19.3 g pot?1) and maturity (straw yield ?25.2 g pot?1 and grain yield ?7.3 g pot?1) was recorded with B0.44 S13.4 treatment combination. The combined applications of sulfur and boron yielded highest oil content with B0.44S13.4 (21.7%) treatment level. Chlorophyll ‘a’ and ‘b’ increased significantly with successive levels of sulfur and boron addition at 55 DAS. The mean sulfur and boron uptake in straw and grains increased significantly with increasing levels of sulfur and boron up to 13.4 mg kg?1 and 0.44 mg kg?1 and decreased non-significantly thereafter. At both the growth stages, a synergistic interactive effect of combined application of sulfur and boron was observed with B0.44 S13.4 treatment level for sulfur and boron uptake in straw and grains.  相似文献
