Xylooligosaccharide (XO) production was performed from xylan, which was obtained by alkali extraction from cotton stalk, a major agricultural waste in Turkey. Enzymatic hydrolysis was selected to prevent byproduct formation such as xylose and furfural. Xylan was hydrolyzed using a commercial xylanase preparation, and the effects of pH, temperature, hydrolysis period, and substrate and enzyme concentrations on the XO yield and degree of polymerization (DP) were investigated. Cotton stalk contains about 21% xylan, the composition of which was determined as 84% xylose, 7% glucose, and 9% uronic acid after complete acid hydrolysis. XOs in the DP range of 2-7 (X6 approximately X5>X2>X3) were obtained with minor quantities of xylose in all of the hydrolysis conditions used. Although after 24 h of hydrolysis at 40 degrees C, the yield was about 53%, the XO production rate leveled off after 8-24 h of hydrolysis. XO yield was affected by all of the parameters investigated; however, none of them affected the DP of the end product significantly, except the hydrolysis period. Enzyme hydrolysis was maintained by the addition of fresh substrate after 72 h of hydrolysis, indicating the persistence of enzyme activity. The optimal hydrolysis conditions were determined as 40 degrees C, pH 5.4, and 2% xylan. The obtained product was fractionated via ultrafiltration by using 10, 3, and 1 kDa membranes. Complete removal of xylanase and unhydrolyzed xylan was achieved without losing any oligosaccharides having DP 5 or smaller by 10 kDa membrane. This study was aimed at elucidating the importance of original Pb-speciation versus soil-characteristics to mobility and distribution of Pb in industrially polluted soils. Ten industrially polluted Danish surface soils were characterized and Pb speciation was evaluated through SEM-EDX studies, examination of pH-dependent desorption, distribution in grain-size fractions and sequential extraction. Our results show that the first factors determining the speciation of Pb in soil are: (1) the stability of the original speciation and (2) the contamination level, while soil characteristics are of secondary importance. In nine of ten soils Pb was concentrated strongly in the soil fines (< 0.063 mm). In all soils, particles with a highly concentrated Pb-content were observed during SEM-EDX. In eight of the soils, the particles contained various Pb-species with aluminum/iron, phosphate, sulfate and various metals (in solder and other alloys) as important associates. In the one soil, where Pb was not concentrated in the soil fines, Pb was precipitated solely as PbCrO4, while pure (metallic) Pb was repeatedly observed in the last soil. Pb was bound strongly to the soils with > 50% extracted in step III (oxidizing) and IV (residual) of sequential extraction for all soils but one. A significant amount of exchangeable Pb existed only in severely contaminated soils, where the bonding capacity of organic matter and oxides was exceeded. Among soil constituents, Pb was observed to adsorb preferentially to feldspars and organic matter while presence of phosphate increased the strength of the Pb-bonding in phosphate-rich soils. 相似文献

The essential oil from the gum of Pistachio (Pistacia vera L. (Anacardiaceae)) grown in Turkey was obtained by the hydro-distillation method, and its chemical composition was analyzed by GC and GC-MS. Moreover, the antimicrobial activities of the oil against the growth of 13 bacteria and 3 pathogenic yeasts were evaluated using the agar-disk diffusion and minimum inhibitory concentration (MIC) methods. The results showed that the essential oil contained about 89.67% monoterpenes, 8.1% oxygenated monoterpenes and 1.2% diterpenes. alpha-Pinene (75.6%), beta-pinene (9.5%), trans-verbenol (3.0%), camphene (1.4%), trans-pinocarveol (about 1.20%), and limonene (1.0%) were the major components. The antimicrobial results showed that the oil inhibited nine bacteria and all the yeasts studied, and the activities were considerably dependent upon concentration and its bioactive compounds such as carvacrol, camphene, and limonene. Moreover, the essential oil of the gum was found to be more effective yeastcide than Nystatin, synthetic yeastcide. Furthermore, the antibacterial activities of the oil were lower than those of standard antibiotics, ampicillin sodium, and streptomycine sulfate under the conditions studied. 相似文献

The gazelles (Gazella subgutturosa), which are at the risk of extinction and are legally protected, have a cultural and aesthetic value in Turkey. It becomes incredibly important to increase their numbers in order to ensure their survival. In this context, it is required to examine thoroughly the pelvic cavity which contains reproductive organs and is used as the birth canal, as well. This study was conducted to determine the pelvimetric data of the gazelles by scanning their pelvic cavity via a multidetector computed tomography (MDCT) and preparing three-dimensional models of the pelvic cavity. Twelve adult (6 females, 6 males) gazelle carcasses were used in the study. After the pelvic cavity was scanned using the MDCT device with 64 detectors at 80 kV, 200 mA, 639 mGY and 0.625 mm cross-section thickness, the MDCT images were obtained. The three-dimensional models of the images obtained using MIMICS 20.1 (The Materialise Group, Leuven, Belgium) program were prepared. Pelvimetric measurements of the pelvic cavity were taken using these models. It was determined based on the pelvimetric examinations that there was a significant difference between the female and male gazelles in terms of conjugate diameter, vertical diameter, intermediate traversal diameter, and cranial and medial transverse diameter data at p < .05. Also, there was a significant difference in their data related to caudal transverse diameter and ischial arch at p < .01. Consequently, it is thought that the use of pelvimetric values belonging to the pelvic cavity of gazelles would reveal the sexual dimorphism of the species and its differences with other species and would contribute to clinical sciences. 相似文献

The objective of this study was to evaluate the influence of an experimentally induced acute inflammation on serum adiponectin and insulin-like growth factor 1 (IGF-1) levels in the dog, and to compare their evolution with other well-established acute phase proteins (APPs) such as C-reactive protein (CRP), and haptoglobin (Hp). Therefore levels of adiponectin, IGF-1 and a profile of APPs were measured in healthy dogs after intravenous administration of E. coli LPS (0.02 mg/kg) and compared with dogs injected with saline solution (0.2 mL/kg). Adiponectin and IGF-1 were both decreased in response to endotoxins in the dog. Significant positive correlations were found between adiponectin and IGF-1 (r=0.31; p<0.05). Adiponectin had also a significant negative correlation with CRP (r=-0.39; p<0.05) and Hp (r=-0.27; p<0.05), whereas IGF-1 had significant negative correlation with CRP (r=-0.52; p<0.001). The results obtained in the present study indicate that adiponectin and IGF-1 behave as negative acute phase proteins after acute inflammatory stimulus in dogs. 相似文献

Erythropoietin (EPO) is both hematopoietic and tissue protective, putatively through interaction with different receptors. We generated receptor subtype-selective ligands allowing the separation of EPO's bioactivities at the cellular level and in animals. Carbamylated EPO (CEPO) or certain EPO mutants did not bind to the classical EPO receptor (EPOR) and did not show any hematopoietic activity in human cell signaling assays or upon chronic dosing in different animal species. Nevertheless, CEPO and various nonhematopoietic mutants were cytoprotective in vitro and conferred neuroprotection against stroke, spinal cord compression, diabetic neuropathy, and experimental autoimmune encephalomyelitis at a potency and efficacy comparable to EPO. 相似文献
